Isoeugenol prevents N-methyl-D-aspartate(NMDA)-induced neurotoxicity and convulsion

  • Isoeugenol, one of the phenylpropanoid derivatives has been known to inhibit the lipid peroxidation via scavenging effect on hydroxyl or superoxide radical production. We examined whether isoeugenol has a inhibitory effect against N-methyl-D-aspartate(NMDA)-, oxygen/glucose deprivation- and xanthine/xanthine oxidase(X/XO)-induced neurotoxicity or NMDA-induced $^{45}Ca^{+2}$ uptake elevation in primary mouse vertical cultures. We also evaluated whether isoeugenol exhibits inhibitory action on NMDA-induced convulsion in mice. Isoeugenol ($30{\sim}300{\mu}M$) attenuated NMDA- and X/XO-induced neurotoxicity by 11~85% and 83~92%, respectively. In the oxyge/glucose deprivation(60 min)-induced neurotoxicity, isoeugenol significantly(p<0.05) reduced by 32% at the maximal concentration. However, it failed to ameliorate NMDA-induced $^{45}Ca^{+2}$ uptake elevation. Isoeugenol(0.5g/kg, i.p.) delayed 6.5 times on the onset time of convulsion evoked by NMDA($0.1{\mu}g$) compared to that of control. These results suggest that the neuroprotective action of isoeugenol may be ascribed to the modulation of massive generation of reactive oxygen species(ROS) occurred during the ischemic or excitotoxic damage, not by directly affecting the NMDA receptor.

Effect of Ethanol-pretreatment on the Liver Xanthine Oxidase Activity in Xylene-treated Rats (에탄올 전처치한 흰쥐에 Xylene 투여가 간조직 중 Xanthine Oxidase 활성 변동에 미치는 영향)

  • To evaluate an effect of ethanol pretreatment on the liver xanthine oxidase(XO) activity, 0.25ml of xylene(50% in olive oil) per 100g body weight was daily given four days to the rats at 2hrs after aministration of ethanol each day, while each control group(ethanol, xylene, olive oli) was treated as the same dose described as above. The animals were sacrificed at 24hrs after last injection. Xylene-treated rats showed the more decreased activity of liver XO compared to the control. But the pretreatment of ethanol to the xylene-treated rats enhanced the liver XO activity. Furthermore, the xylene-treated rats led to more increased Vmax value in liver XO compared to the only xylene-treated rats. On the other hadn, hepatic aldehyde dehydrogenase activity was more decreased in xylene-treated rats pretreated with ethanol than in xylene-treated rats. And the intermediated xylene metabolites, methyl benzylalcohol or aldehyde inhibited the XO activity "in vitro". In conclusion, the phenomenon that pretreatment of ethanol to the xylene-treated rats led to the enhancement of liver XO activity, may be due to an influence of acetaldehyde.taldehyde.

  • Introduction : Methanethiol is a toxicant that is a byproduct in the industrial process (oil refinery), and it is produced in vivo from methionine via transamination in case of its overintake. And it also can be generated by the action of mucosal thiol Smethyltransferase on hydrogen sulfite which is formed by anaerobic bacteria in the intestinal tract. The toxicity of methanethiol has often been suggested as one of endogenous factors involved in the pathogenesis of hepatic encephalopathy. Furthermore, methanethiol could cause the membrane damage and inhibition of some membrane protective enzymes.

Lipoxygenases, Hyaluronidase, and Xanthine Oxidase Inhibitory Effects Extracted from Five Hydrocotyle Species

  • Hydrocotyle is a genus of prostrate, perennial aquatic or semi-aquatic plants formerly classified in the family Apiaceae, now in the family Araliaceae. Lipoxygenases (LOX) are present in the human body and play an important role in the stimulation of inflammatory reactions. Ethanolic extracts of five Hydrocotyle species (H. ramiflora, H. maritima, H. nepalensis, H. sibthorpioides, and H. yabei) showed inhibition of 23.5~50.6% at 2.0 mg/mL. Their extracts showed LOX inhibition in half maximal effective concentration (EC50) range 15.1~15.7 ㎍/mL. Hyaluronic acid is a glycosaminoglycan, a major component of the extracellular matrix Five extracts of these species inhibited less than 23.0% of Hyaluronidase (HAase) activity at a concentration of 2.0 mg/mL Xanthine oxidase (XO) is a form of xanthine oxidoreductase, a type of enzyme that generates reactive oxygen species. Five Hydrocotyle species were found to have inhibitory activity of XO at 2.0 mg/ml, with 65% having greater than 50% inhibition. H. ramiflora exhibited the highest activity with an inhibition of 80.0%. The results suggested that Lipoxygenases, Hyaluronidase, and Busan 47340, Republic of Korea from five Hydrocotyle species might be multifunctional and prevent the degradation of allergic reactions and inflammation.

Effect of Leegapunsusan on galactosamine induced hepatoxity in rats (이간분수산(利肝分水散)이 Galactosamine에 의해 유발(誘發)된 백서(白鼠) 간기능손상(肝機能損傷)에 미치는 영향(影響))

  • The purpose of this study is to observe tile protective effect of Leeganpunsusan on serum reaction and hepatic tissue in galactosamine treated rats. In this study, the experimental rats divided three group(Normal, Control and sample group). Under the same condition, normal and control groups were administered water, sample group was administered Leeganpunsusan for 15 days. The last day, both normal and control goups were injected to abdomen with galactosamine. The rates of lipid peroxide, SOD(activity) and xanthine oxidase(activity) were measured. The results were obtained as follows : Effects of the extract of Leeganpnnsusan on the hepatic lipid peroxidation, xanthine oxidase activity in VITRO, as compared with control group were significantly decreased with the level of concentration of extract prepared from Leeganpunsusan. In VIVO, the hepatic content of lipid peroxide, the rate of type changing(type D to O) and xanthine oxidase activity were significantly decreased in galactosamine-treated rats. Effect of on the hepatic cytosolic superoxide dismutase activity was significantly increased.

Effects of Vitamin E on Liver Cytochrome $P_{450}$ Content and Xanthine Oxidase Activity in Acute Cadmium-Poisoned Rats (식이 Vitamin E가 급성 카드뮴중독 흰쥐 간조직의 Cytochrome $P_{450}$ 함량과 Xanthine Oxidase 활성에 미치는 영향)

  • The purpose of this study was to investigate the effects of vitamin E on liver microsomal cytochrome P450 contents and xanthine oxidase activity in acute cadmium poisoned rats. Sprague Dawley male rats weighing 100$\pm$10g were randomly assigned to one normal and three cadmium injected groups. Cadmium injected groups were fed vitamin E free diet(0E Cd group), 40mg vitamin E per kg diet(40E Cd group) or 400mg vitamin E per kg diet(400E Cd group). Vitamin E level of normal group was 40mg per kg diet. Animals were injected intraperitoneally with 2.0mg Cd2+/kg bw for 4 days after the rats were fed diets with three different levels of vitamin E for 2 and 4 weeks. Body weight, food intake and feed efficiency ratio of cadmium injected animals, were decreased compared with those of normal group. The weights of liver and kidney in cadmium injected groups were not different from those of normal group. Cadmium contents of liver in cadmium groups were 160 fold higher those that of normal group. Accumulation of cadmium poisoned rat liver was reduced by vitamin E supplementation. Contents of blood hemoglobin and hematocrit in 0E Cd groups were decreased to 2~ 14% of those of the other groups. Contents of serum triglyceride in all experimental groups were not significantly different each other. Levels of serum total cholesterol, LDL cholesterol and antherogenic index in 0E Cd and 40E Cd groups were higher than those of normal group, while the contents of HDL cholesterol in 0E Cd and 40E Cd groups were lower than those of normal group. Xanthine oxidase(XOD) activity and cytochrome P450 contents in the liver were significantly increased in cadmium injected groups but these were reduced by vitamin E supplementations. The present results indicate that acute cadmium poisoning in rats causes increasing free radical generation systems in the liver and that leads to liver tissue damage. But these abnormalities can be reduced by dietary vitamin E supplementations.

Effect of Occlusion on the Activities of Dermal Xanthine Oxidase in Rats (흰쥐에 있어서 피부노출 차단이 피부조직의 Xanthine Oxidase 활성에 미치는 영향)

  • To investigate an impact of skin occlusion on the dermal xanthine oxidase (XO) activity, the dorsal part in rats was covered with closed petri dish-shaped chamber, 46 mm in diameter and 10 mm in height, which was made of glass. The crack between top of chamber and skin was sealed by an adhesive agent. After 5 days, the quantity of sweat accumulation was about 400 mg, whereas after 10 days that was decreased about to 21 mg. The 5 days skin occlusion showed the more increased activity of dermal XO compared with the control, and the increased ratio of enzyme activity to the control was higher than that of 10 days skin occlusion, with the increase being associated with sweat accumulation in chamber. Furthermore, the V$_{max}$ of dermal XO in 5 days skin occlusion was higher than that in the control. In conclusion, it may be hypothesized that the XO system may play an role for defence mechanism in dermal tissue.

Isolation of Antioxidative Compound from Scutellaria baicalensis G. (황금으로부터 항산화 활성 성분의 분리)

  • Root of Scutellaria baicalensis G. was extracted with methanol and water to give the yield of 30.0% in order to find the antioxidant substance. The extract was fractionated with diethyl ether, n-butanol and water to test the inhibitive activity against xanthine oxidase. Three fractions inhibited the activities of xanthine oxidase by 48.2%, 10.2% and 2.8%, respectively, at the amount of $0.1\;{\mu}g$. A component that exhibited strong inhibition of xanthine oxidase was isolated from diethyl ether fraction (SE Fr.) by silica gel column chromatography and HPLC, and then identified by $^1H-NMR$, $^{13}C-NMR$ and MS spectrophotometry. EDA (Electron Donating ability) of the compound was 28.5% at the concentration $100\;{\mu}g/3\;ml$. That was identified to be 3,5,7-trihydroxy-2'-methoxyflavanone by spectrophotometric analysis using $^1H-NMR,\;^{13}C-NMR$ and Mass spectrophotometry.

Effects of $\beta$-Mercaptoethanol on lipid Peroxidation and Fertilization Ability In Vitro by Xanthine-Xanthine Oxidase System in Pig (Xanthine-Xanthine Oxidase System,하에서 돼지 동결-융해정자의 Lipid Peroxidation과 체외수정능력에 대한 $\beta$-Mercaptoethanol의 영향)

  • This study was undertaken to evaluate the effects of $\beta$-mercaptoethanol ($\beta$-ME) on lipid peroxidation and fertilization ability in vitro by xanthine (X) - xanthine oxidase (XO) system in boar spermatozoa frozen-thawed. The boar spermatozoa were treated with X and/or XO, and the spermatozoa viability were measured by the eosin-nigrosin stain method. In control group, level of vitality in boar spermatozoa were higher than in medium with X, XO and X+XO groups. No significant differences, however, were observed under the all conditions. The percentage of spermatozoa that reached acrosome reaction were significantly (P<0.05) higher in sperm treated without that than with $\beta$-ME under the all conditions. On the other hand, when spermatozoa were inseminated in medium with X and/or XO, the penetration rates in all conditions were higher in medium with that than without $\beta$-ME. However, significant differences were not observed between medium with and without $\beta$-ME. The lipid peroxidation of sperm was evaluated on the basis of malondialdehyde (MDA) production. The MDA were higher in sperm treated without that than with $\beta$-ME under the above all conditions. However, significant differences were not observed between medium with and without $\beta$-ME. Sperm-SH group were higher detected in medium with that than without $\beta$-ME under the all conditions. The activity of sperm binding to Bona pellucida was also evaluated through binding to salt-stored porcine oocytes. In control group, sperm binding to zona pellucida were significantly (P<0.05) higher than in medium with X+XO groups. The sperm binding in all conditions were higher in medium with that than without $\beta$-ME. However, significant differences were not observed between medium with and without $\beta$-ME. These results suggest that addition of $\beta$-ME in X-XO system may play a positive role in improving of fertilization ability in vitro.

Use of a Xanthine-Xanthine Oxidase System on in vitro Maturation and Fertilization in the Pig (돼지난자의 체외성숙과 수정에 있어서 Xanthine-Xanthine Oxidase System의 이용)

  • This study was undertaken to evaluate the effects of catalase using xanthine (X)-xanthine oxidase (XO) system on in vitro maturation and fertilization in the pig. When follicular oocytes were cultured with X or XO, the maturation rates were not significantly different between in medium with and without catalase despite of different culture periods. However, significantly (P<0.05) higher maturation rates were obtained in culture with X-XO-catalase system. The rates of degenerated oocytes were increased with culture periods prolonged, and were significantly (P<0.05) higher in medium without that than with catalase at 120 h of culture. On the other hand, the parthenogenetic oocytes were observed with high proportions at 72 h of culture, but were not different between the medium with and without catalase at various times of culture. In another experiment, the frozen-thawed boar spermatozoa treated with X-XO system for in vitro fertilization. The penetration rates were higher in medium with that than without catalase during the in vitro fertilization with none (P<0.05), XO and X+XO. On the other hand, when sperm were treated with none, X, XO and X+XO, lipid peroxidation were produced with higher rates in medium without that than with catalase, and consequently the changes in sperm penetration and lipid peroxidation showed opposite patterns. Under the above all conditions, however, sperm-SH group were higher detected by catalase. When the activity of sperm binding to zona pellucida was evaluated through binding to salt-stored porcine oocytes, sperm binding to zona pellucida in control group were higher than in medium with X, XO and X+XO groups. No significant differences, however, were observed between medium with and without catalase. In conclusion, the exposure of follicular oocytes and spermatozoa to X-XO-catalase system may be caused stimulating in vitro maturation and fertilization in the pig.
