옥수수 calreticulin 과발현 토마토에서 tobamovirus의 상엽 이동 억제 (Suppression of tobamovirus movement toward upper leaves in the tomato plant over-expressing a maize calreticulin)

  • To ascertain the effect of over-expressed maize calreticulin in tomato plant on tobamovirus movement in addition to validating potentiality of the gene (ZmCRT) as a means for the virus-resistance resource, four ZmCRT-expressing homozygous lines were generated from the T0 plants as using an Agrobacterium-mediated transformation, nucleic acid analyses, and a conventional breeding method. Of them, a line was subjected to the bioassay for tolerances to tobacco mosaic virus-U1 (TMV-U1) and tomato mosaic virus (ToMV) followed by RT-PCR and a chlorophyll fluorescence quenching analyses. Both transgenic plants transcribing ZmCRT and wild-type plants showed no symptom by 20 days after viruses inoculation, however the photosystem II quantum yield parameter measured from the upper leaves of ToMV-inoculated plants revealed that ZmCRT transgenic plants have higher photosynthetic ability than wild-type ones at that time, which indirectly implies that over-expressed ZmCRT product acts as a barrier to the cell-to-cell and/or systemic movement of ToMV. Moreover, ZmCRT transgenic plants showed remarkably longer shoot length than wild-type ones in 40 days after TMV-U1 or ToMV inoculation each, which might be resulted from higher photosynthetic ability during the phase not yet showing any external symptoms. Collectively, over-expressed ZmCRT protein in tomato plants is able to interrupt the systemic movement of infected TMV-U1 and ToMV even though not perfect.

Capsicum annuum NAC4 (CaNAC4) Is a Transcription Factor with Roles in Biotic and Abiotic Stresses

  • Transcription factors (TFs) regulate gene expression by binding to DNA. The NAC gene family in plants consists of crucial TFs that influence plant development and stress responses. The whole genome of Capsicum annuum shows over 100 NAC genes (CaNAC). Functional characteristics of the most CaNAC TFs are unknown. In this study, we identified CaNAC4, a novel NAC TF in C. annuum. CaNAC4 expression increased after inoculation with the pathogens, Xanthomonas axonopodis pv. vesicatoria race 3 and X. axonopodis pv. glycines 8ra, and following treatment with the plant hormones, salicylic acid and abscisic acid. We investigated the functional characteristics of the CaNAC4 gene and its roles in salt tolerance and anti-pathogen defense in transgenic Nicotiana benthamiana. For salt stress analysis, the leaf discs of wild-type and CaNAC4-transgenic N. benthamiana plants were exposed to different concentrations of sodium chloride. Chlorophyll loss was more severe in salt stress-treated wild-type plants than in CaNAC4-transgenic plants. To analyze the role of CaNAC4 in anti-pathogen defense, a spore suspension of Botrytis cinerea was used to infect the leaves. The disease caused by B. cinerea gradually increased in severity, and the symptoms were clearer in the CaNAC4-transgenic lines. We also investigated hypersensitive response (HR) in CaNAC4-transgenic plants. The results showed a stronger HR in wild-type plants after infiltration with the apoptosis regulator, BAX. In conclusion, our results suggest that CaNAC4 may enhance salt tolerance and act as a negative regulator of biotic stress in plants.

Occurrence of Stem Rot of Wild Aster(Aster koraiensis) Caused by Sclerotium rolfsii in Korea

  • A destructive stem rot of wild aster(Aster koraiensis) occurred sporadically some farmers' fields in Guman-myon, Kosonggun, Kyongsangnam-do in 2000. One of the most severely infected field in Kosong showed 28.6 percent of infection rate. The fungus also caused stem or crown rot and systemic wilt or blight of the plants. White mycelium spread over stems and petioles of infected plants and sclerotia formed on the old lesions and near the soil surface. The fungus showed maximum mycelial growth around $30^{\circ}C$ and did not grow under $5^{\circ}C$ and over $45^{\circ}C$ and mycelial width were $4.3{\sim}10.2{\mu}m$. Colony was white, usually many narrow mycelial stand in the aerial mycelium and formed clamp connection. Numerous sclerotia were formed on PDA at $30^{\circ}C$. The shape sclerotia were globoid and $0.8{\sim}3.0{\times}0.9{\sim}3.4$ mm in size. The fungus was isolated repeatedly from the infected tissues and confirmed its pathogenecity to wild aster and identified as Sclerotium rolfsii. This is the first report on the stem rot of wild aster caused by S. rolfsii in Korea.

Newly developed MSAP analysis reveals the different polymorphism patterns in transgenic tobacco plants with the dsRNA MET1 gene

  • DNA methylation is known to play an important role in the regulation of gene expression in eukaryotes. In this study, we isolated NtMET1 from Nicotiana tabacum cv. Havana (SR1) and obtain transgenic plants that reduced MET1 expression level with the double-strand RNA (dsRNA) MET1 gene. Transgenic tobacco plants showed dwarf and abnormal flower development when compared with the wild type. Using methylation-sensitive amplified polymorphism (MSAP) analysis, the patterns of cytosine methylation in transformed plants and the wild type were compared. MseI/HpaII selection primers showed an interesting polymorphism, and 153 DNA bands of interest were detected. Among these, 30 selective fragments were sequenced and analyzed with a BLAST search by successful MSAP modifications. The homology search showed that the transposons and tandem repeated sequences were related to the phenotypes. These results suggested that the decreased degree of methylation by dsRNA strategy caused abnormal growth and development in N. tabacum.

와일드 가든(Wild Garden)의 개념적 모형 유추 - 윌리암 로빈슨(William Robinson)과 거투르드 제킬(Gertrude Jekyll)의 비교 연구 - (Inference of the Conceptual Model of Wild Gardens - A Comparative Study of William Robinson and Gertrude Jekyll -)

  • 자연적, 환경적 문제를 가진 현대에서 빛을 발하고 있는 자연식재는 와일드 가든(wild garden)에서 그 원류를 찾을 수 있다. 윌리암 로빈슨(William Robinson)이 시초가 되고, 거투르드 제킬(Gerturde Jekyll)이 구체적으로 형상화하였다. 특히, 와일드 가든은 현대정원의 가장 중요한 화두가 될 수 있는 생태적 설계의 예고와 내츄럴리즘의 분화과정에 중요한 초석이 되었다는 점에서 재조명해 볼 필요가 있다. 본 연구는 로빈슨의 주택 그래비티 매너(Gravety Manor)와 제킬의 주택 먼스태드우드(Munstead Wood)를 통해 와일드 가든의 개념적인 모형을 정확히 유추해 보고, 역사적인 의의를 찾고자 하는 것이다. 연구의 결과를 요약하면, 첫째, 공간 구성 면에서는 기본적인 코타지 가든의 영감을 이어받아 비정형의 형식을 도입하였다. 둘째, 소재의 사용 면에서는 여행을 통해 다양한 기후를 접하면서 식물의 내한성과 외래종에 대한 이해를 바탕으로 자생식물과 귀화식물을 같이 사용하였다. 또한 숲 하부 식재와 산림지역에 관심을 가졌다. 셋째, 디자인 기법 면에서는 색감과 질감에 대해 각 개별 식물 및 상호간의 관계에 관해 연구하여 자연의 축소판과 같은 다양한 경관을 연출하였다. 넷째, 유지관리 면에서는 식물이 가진 본래의 성질대로 살 수 있는 최소한의 유지관리를 위한 방식을 기본으로 하였다.

Effects on the Development of Plutella xylostella and Spodoptera litura after Feeding on Transgenic Cabbage Expressing Potato Proteinase Inhibitor II and Bar Genes

  • Cabbage plants were transformed with the potato proteinase inhibitor II (PINII) gene, bar gene, and hpt gene using Agrobacterium. The expression of the PINII gene was driven by its own promoter which was wound-inducible. Ten transgenic plants were obtained from medium containing hygromycin as a selection antibiotic. The integration and expression of PINII and bar genes were confirmed by Southern and Northern hybridization. Growth and development of diamondback moths (Plutella xylostella) and tobacco cutworm (Spodoptera litura) larvae were examined on $T_1$ plants. The weight of the larvae and pupae of these two insects grown on transgenic plants was not different compared to those grown on wild type plants. However, the pupation and emergence rate of diamondback moths and tobacco cutworms fed on some transgenic plants was lower than on wild type plants. These results suggest that the PINII transgene under the control of a wound-induced promoter may be used for control of insects in transgenic cabbage through reduction of insect progeny number.

Increase of resistance to oxidative stress induced by methyl viologen in progeny from a cross between two transgenic Petunia lines with NDPK and SOD genes

  • This study was conducted to investigate how to enhance resistance to oxidative stress in petunia progeny obtained by a crossing between transgenic plants, MnSOD (SOD2) ($T_4$) and NDPK2 ($T_2$), to develop transgenic petunia much more resistant to environmental stress. At the treatment of MV 200 ${\mu}M$, the progeny was significantly less damaged than its parental plants (SOD2- or NDPK2-transgenic lines) as well as wild type plants, implying its resistance to oxidative stress was enhanced compare to that of SOD2- or NDPK2- transgenic plants. In an expression of 11 quantitative traits, the progeny remained similar to control plants, although it infrequently displayed slightly longer or wider than either parental or wild type plants. In the expression of 6 qualitative traits, there was no significant difference between parental or non-transgenic control plants.

비탈면 침식방지를 위한 자생초본식물의 뗏장 형성에 관한 연구 (Study on the Sod Forming Effects of the Native Plants for the Erosion Control in Slopes)

  • This study was conducted to suggest vegetation mat forming methods with native wildflowers, introduced wild flower, grasses and native herb plants which can be used for erosion control of the disturbed slopes. The main results are summarized as follows. In forming the vegetation mat at early stage, foreign grasses, native wild plants shows more effective than the other seed mixture types. But, after 12 weeks, using native wild flowers shows more effective than foreign grasses. Also, the mixture of the native grasses and kentucky bluegrasses (seed mixture type F) are the best in ground coverage among the seed mixtures types. In order to restore slopes more naturally, more diverse plants should be used. The native grasses such as Marrubium incisum Bentham, Lutos comiculatus L. var. japonicus Regel, and Artemisia princeps Pampan seems to be useful to forming the vegetation mat, also it will be more easy if mixing with foreign grasses such as Kentucky bluegrass. The mixture of native grasses and introduced wild flowers are not proper in forming the vegetation mat. Also, pot planting of the native grasses are not proper in forming the vegetation mat.
