Morphological Characteristics of the Fruiting Bodies of Wild Lentinula edodes Strains Cultivated on Sawdust Blocks in Korea (국내에서 수집한 야생 표고의 사각 톱밥배지 재배에서 자실체의 형태적 특성)

  • Wild Lentinula edodes (shiitake) strains are an important breeding material for developing new varieties suitable for various consumer preferences and environmental conditions. The morphological characteristics of 100 wild shiitake strains collected from 14 regions were investigated in a sawdust block cultivation. The characteristics of their fruiting bodies were diverse. Among them, 38% of the caps was convex, 33% was broadly umbonate, 27% was applanate, and 2% was shallowly depressed. In terms of their stipes, 55, 23, and 22% were the column, clavate, and funnel type, respectively. The shape of the cap was the most diverse on Mt. Hwaak, whereas the shape of the stipe showed various phenotypes from various regions. Even among strains collected from the same forest, different types of mushrooms were observed. In conclusion, wild shiitake strains that have adapted to various environmental stress can be used to develop new varieties. Thus, securing various genetic resources and studying their traits are important for researching breeding technology.

Isolation of a Variant Strain of Pleurotus eryngii and the Development of Specific DNA Markers to Identify the Variant Strain

  • A degenerated strain of Pleurotus eryngii KNR2312 was isolated from a commercial farm. Random amplified polymorphic DNA analysis performed on the genomic DNA of the normal and degenerated strains of this species revealed differences in the DNA banding pattern. A unique DNA fragment (1.7 kbp), which appeared only in the degenerated strain, was isolated and sequenced. Comparing this sequence with the KNR2312 genomic sequence showed that the sequence of the degenerated strain comprised three DNA regions that originated from nine distinct scaffolds of the genomic sequence, suggesting that chromosome-level changes had occurred in the degenerated strain. Using the unique sequence, three sets of PCR primers were designed that targeted the full length, the 5' half, and the 3' half of the DNA. The primer sets P2-1 and P2-2 yielded 1.76 and 0.97 kbp PCR products, respectively, only in the case of the degenerated strain, whereas P2-3 generated a 0.8 kbp product in both the normal and the degenerated strains because its target region was intact in the normal strain as well. In the case of the P2-1 and P2-2 sets, the priming regions of the forward and reverse primers were located at distinct genomic scaffolds in the normal strain. These two primer sets specifically detected the degenerate strain of KNR2312 isolated from various mushrooms including 10 different strains of P. eryngii, four strains of P. ostreatus, and 11 other wild mushrooms.

Medium Development of Pleurotus Ostreatus by Using Korean Herb Medicine Materials (한방재료를 이용한 느타리버섯의 배지개발)

  • To reduce the production cost of $Pleurotus$ $ostreatus$, discarded medicinal sludge was collected from oriental medical clinics to develop the $Pleurotus$ $ostreatus$ culture medium. According to the analysis of the proximate composition of the materials used in Korean herb medicine, the crude ash contents of $Carthamus$ $tinctrius$ L stem and $Acanthopanax$ $chiisanensis$ were 11.6% and 10.1% respectively, which were relatively higher than the 9.6% of the control medium, waste cotton. Crude protein was detected in 9.8% of the waste cotton medium, whereas it was detected in 14.9%, 13.9%, 13.4%, and 11.5%, of wild mugwort, $Acanthopanax$ $\underline{chiisanensis}$, medicinal sludge, and $Carthamus$ $tinctrius$ L stem, respectively, which are all higher than the control. The pH of medicinal sludges, wild mugwort, and $Aacanthopanax$ $chiisanenses$ ranged from 5.27 to 5.72, which was similar to the 5.70 pH value of waste cotton. In the case of addition concentration of each Korean herb medicine material influencing mycelial growth of the $Pleurotus$ $ostreatus$, the 9% concentration was more favorable compared to that of 3% and 6%. However, the addition of Korean herb medicine materials did not significantly affect the growth of $P.$ $tolaassi$ and $Trichoderma$ $sp.$ According to a field experiment that added 9% of medicinal sludge into the waste cotton medium, the mycelial growth of mushrooms was facilitated by approximately 2 days, and the mushroom yield was increased by 10~15%. Furthermore, pileus and stipe of the mushrooms were even and superior in quality.

  • Present experiments were conducted to determine the possibility of cultivation of 9 edible wild species selected among the higher fungi growing in Korea. In the investigation on the mycelial growth according t6o the different media, the mycelial growth of Coprinus comatus was fast on the CSA medium, when malt extract was added to the basal medium PSA, the mycelial of Lapista nuda and Auricularia auricula-judae was fast in growth and density. In the spawning, the mycelial growth of Pholiota squarrosa on the oak tree's sawdust, Pleurotus cornucopiae on the broad-leaves' sawdust, and Coprinus comatus on the compost was respectably fast and also it shown to be possibility of artificial cultivation owing to their carporphore budding when Coprinus comatus and Lepiota alborubescens cultivated on the rice straw, Auricularia auriculajudae and Pleurotus cornucopiae on the sawdust of the popla and Pholiota squarrosaon the sawdust of the oak tree.

Genetic variation of local varieties and mutants groups induced by gamma ray in Hypsizigus marmoreus (느티만가닥버섯의 재래종과 감마선 돌연변이체들의 유전적 변이)

  • This research was carried out to analyze the genetic variation of 18 wild strain, 2 breed varieties and 20 mutants of Hypsizygus marmoreus by random amplification of polymorphic DNA(RAPD). Also, 3 strains of Lyophyllum decartes and 1 strain of Lyophyllum shimeji were used. These mushrooms were collected from korea, china, Taiwan and Japan. Spores of H. marmoreus JV-2 strain were irradiated by gamma ray for mutagenesis. 40 kind of primers were used for this reaserch. Number of reaction primer were 31. Electrophorectic patterns of RAPD showed genetic variation. In phylogenetic tree, they were divided into seven group. Discriminative differences were observed between wild strain and mutants in H. marmoreus. These results might suggest that these primers and gamma ray irradiation of spores were useful tools for developing new strain for mushroom.

Detection of Biological Activities of Wild Mushroom Methanol Extracts (야생버섯 메탄올추출물의 생리활성 검색)

  • In this study, the biological activities of 33 kinds of wild mushroom methanol extracts were investigated. Boletellus elatus showed the greatest fibrinolytic activity (1.08 plasmin units/mL) in a fibrin plate assay, and the activity of Heterobasidion insulasis was 0.89 plasmin units. The thrombin inhibitory activities of Boletellus elatus and Heterobasidion insulasis were 93.32% and 93.69%, respectively. In a ${\alpha}$-glucosidase inhibitory activity test, Laccaria amethystina showed the greatest inhibitory activity at 81.25%. The antioxidative activities of Gomphus sp. and Geastrum lageniforme were 91.37% and 90.42%, respectively. Since Boletellus elatus and Heterobasidion insulasis have strong fibrinolytic and thrombin inhibitory and antioxidative activities, the two mushrooms can be used as material for the development of biofunctional foods for cardiovascular diseases.

Ergostane-Type Steroids from Korean Wild Mushroom Xerula furfuracea that Control Adipocyte and Osteoblast Differentiation

  • As part of our current work to discover structurally and/or biologically novel compounds from Korean wild mushrooms, we isolated five ergostane-type steroids (1-5) from the fruiting bodies of Xerula furfuracea via repeated column chromatographic separations and HPLC purification. The chemical structures of the isolated steroids were shown to be (22E,24R)-24-methylcholesta-4,22-diene-3,6-dione (1), ergosta-7,22-diene-3β,5α,6β-triol (2), ergosta-7,22-diene-3β,5α,6β,9α-tetraol (3), (22E,24R)-5α,8α-epidioxyergosta-6,22-diene-3β-ol-3-O-β-D-glucopyranoside (4), and (22E,24R)-5α,8α-epidioxyergosta-6,9,22-triene-3β-ol-3-O-β-D-glucopyranoside (5)based on comparison of the data regarding their spectroscopic and physical properties with those of previous studies. Notably, this is the first report on the presence of the identified steroids (1-5) in this mushroom. We tested compounds 1-5 to determine their effects on adipogenesis and osteogenesis in the mouse mesenchymal stem cell line C3H10T1/2 and found that compounds 4 and 5 suppressed the differentiation of stem cells into adipocytes. Notably, in addition to its suppressive effect on adipogenesis, compound 5 was also shown to promote the osteogenic differentiation of stem cells. These findings demonstrate that the bioactive compounds isolated might be effective for the treatment of menopause-associated syndromes, such as osteoporosis and obesity, as the isolated compounds were shown to suppress adipogenesis and/or promote osteogenesis of stem cells.

Comparative mitogenomics of Pleurotus ostreatus Gonji7ho and its cap color mutant

  • Pleurotus ostreatus is a globally cultivated mushroom crop. Cap color is a quality factor in P. ostreatus. However, cap color can spontaneously mutate, degrading the quality of the mushroom on the market. Early detection and removal of mutant strains is the best way to maintain the commercial value of the crop. To detect the cap color mutant Gonji7ho, molecular markers were developed based on insertion/deletions (InDels) derived from the comparison of mitogenomes of Gonji7ho and Gonji7hoM mushrooms. Sequencing, assembly, and comparative analysis of the two mitogenomes revealed genome sizes of 73,212 bp and 72,576 bp with 61 and 57 genes or open reading frames (ORFs) in P. ostreatus Gonji7ho and Gonji7hoM, respectively. Fourteen core protein-encoding genes, two rRNA, and 24 tRNA with some OFRs were predicted. Of the 61 genes or OFRs in the wild type, dpo, rpo, and two orf139 were missing (or remnant) in the mutant strain. Molecular markers were developed based on the sequence variations (InDels) between the two mitogenomes. Six polymorphic molecular markers could detect the mutated mitochondria by PCR. These results provide basic knowledge of the mitogenomes of wild-type and mutant P. ostreatus, and can be applied to discriminate mutated mitochondria.

  • For the purpose of investigating the distribution and flora of wild mushrooms in Korea, this study was conducted primarily on the genus Amanita. Through survey of Suweon, Kwangneung, Mt. Chiri, Taegwanryung, Yeongdong district and Mt. Hanta, 25 species of Amanita including 3 unrecorded species were discovered and are described in this paper. The author proposes to group 25 Korean species of Amanita into 3 subgenera according to morphological characteristics by the modern taxonomic system, and also designated the 3 unrecorded species of Amanita including one variety in Korean common name.

A Preliminary Screening of 46 Korean Basidiomycetes Including Hebeloma crustulineforme for Their Hemolytic Activities (무우자갈버섯(Hebeloma crustulineforme)을 위시한 한국산 담자균류 46종의 용혈활성 검색)

  • To elucidate the hemolytic toxicities of Korean basidiomycetes, the cold-water extracts of 46 wild mushrooms were tested for hemolytic activity on mouse erythrocytes. Twelve species including Amanita esculenta, A. griseofarinosa, A. longistriata, A. melleiceps, A. phalloides, A. rubescence, A. spissacea, A. vaginata, Hebeloma crustuliniforme, Lepiota cygnea, Macrolepiota procera, and Oudemansiella platyphylla showed strong hemolytic activities. Of these, A. griseofarinosa and H. crustuliniforme showed no decrease in their hemolytic activity even after five minutes' boiling.
