선택적 리그닌 분해능을 가지고 생물펄프공정에 사용가능한 백색부후균을 얻기 위하여 94종류의 목재부후균을 검정하였고 선발된 7개종에 대하여 활엽수와 침엽수의 부후능력을 측정하였다. 우선 백색부후균은 셀룰로오즈 분해효소, 페놀산화효소, laccase, peroxidase 등의 효소활성을 간단한 방법으로 검정하여 선발하였는데, Bavendamm test에서 양성을 나타내는 대부분의 균들은 syringaldazine을 사용한 laccase test에서도 강한 반응을 나타낸 반면, 음성반응을 나타낸 대부분의 균들은 laccase와 peroxidase test에서도 음성반응을 나타내었다. 선택적 리그닌 분해능력을 지닌 부후균을 선발하기 위하여 부후균을 은사시나무와 일본잎갈나무(낙엽송) 목재블럭에 접종하여 12주간 배양한 후에 부후된 목재의 중량감소율, 리그닌 량의 감소, 형태적 변화들을 화학분석과 주사전자현미경을 통하여 분석하는 목재부후 실험을 실시하였다. 이 실험에서 사용한 거의 모든 균주는 목재블럭의 중량감소율이 일본잎갈나무 보다 은사시나무에서 2배이상 높게 나타났으며 균을 접종하지 않은 목재블럭에서는 중량감소가 전혀 나타나지 않았다. Ceriporiopsis subvermispora와 Phanerochaete chrysosporium이 다른 균주에 비해서 침엽수와 활엽수의 리그닌을 모두 잘 분해시키는 것으로 나타났으나 분해 능력은 Ceriporiopsis subvermispora가 더욱 우수하였다. Bjerkandera adusta와 미동정된 2균주는 은사시나무에서만 상대적으로 높은 리그닌 분해능력을 나타내었다. B. adusta는 모든 세포벽 성분을 동시에 분해시켜서 2차세포벽을 얇게 만들었으나 다른 균주들은 선택적 리그닌 분해력을 나타내어 두 세포의 세포벽 사이에 위치하는 중벽에 존재하는 리그닌을 분해시켜서 세포를 분리시키는 것이 관찰되었다.
백색부후균(Trametes versicolor)과 갈색부후균(Tyromyces palustris)은 목조문화재 및 건축물 등의 목재 조직의 건조, 부패과 균열, 스펀지화, 변색 등 다양한 손상을 발생시켜 구조적, 미학적인 관점에서 심각한 문제를 나타내고 있다. 그리고 가장 피해를 많이 주는 해충인 흰개미(Reticulitermes speratus Kyushuensis Morimoto)는 목조건축물의 옥외 기둥이나 대들보 등의 표층을 얇게 남겨두고 내부를 침해함으로써, 결국은 속이 빈 기둥이 되어 건축물이 무너져버리는 피해를 줄 수 있다. 대표적 피해 미생물인 목재부후 균이 흰개미의 서식에 미치는 영향을 파악하기 위하여 부후균이 만연된 곳에 흰개미를 넣어 공존 가능성을 실험해 보았다. 목재의 중량으로 피해율을 산정한 결과, 전반적인 총질량은 두 종류의 부후균에서 모두 감소하였다. 목재의 질량은 백색부후균에서는 모두 감소하였으나, 갈색부후균에서는 오히려 증가한 경향을 볼 수 있었다. 그리고 섭식량을 측정하기 위한 목재의 건조 후 질량에서는 갈색부후균에 넣었던 목재의 질량 감소가 더 크게 나타났다.
난분해성물질 함유폐수에 대해 유지관리가 용이하며 저비용-저에너지 고효율적인 새로운 수 처리 방식인 대기노출형 백색부후균 생물막 공법을 적용하기 위한 기초실험을 실시하였다. 먼저 난분해성물질을 함유한 오 폐수 처리에 가장 효율적인 백색부후균 종을 선정하기 위해 백색부후균 3종 및 활성슬러지에 대한 처리특성을 조사하였으며, 선정된 백색부후균 P. chrysosporium PSBL-1에 대해서 HBC링 여재에 대한 부착 및 탈착특성을 조사하였다. 백색부후균 3종(P. chrysosporium PSBL-1, P. chrysosporium KCTC 6147, Trametes sp. KFCC 10941)의 pH에 따른 처리특성을 조사한 결과 NBDCOD 제거율은 P. chrysosporium PSBL-1과 P. chrysosporium 6147가 pH 3.5$\sim$5.5에서 각각 51$\sim$59.8%, 57.5$\sim$60.3%로 비슷한 결과를 나타냈으나, TN 제거율은 pH 4.5$\sim$11.5에서 각각 39.3$\sim$85.3%, 3.4$\sim$7.6%로 현저한 차이를 나타내고 있다. 유기물 및 질소 동시제거 특성을 고려하여 백색부후균 P. chrysosporium PSBL-1을 선정하였다. 백색부후균 P. chrysosporium PSBL-1은 부유상태의 초기농도 4,600 mg/L에서 HBC링 여재 부착시작 5분후 pH 4, 7, 10에서 각각 4,538 mg/L, 4,546 mg/L, 4,531 mg/L, 10분 후 4,575 mg/L, 4,573 mg/L, 4,568 mg/L로 5분 이후에는 미생물 부착이 거의 되지 않았다. HBC링 여재에 부착된 P. chrysosporium PSBL-1을 10일 동안 1일 간격으로 탈착량을 조사한 결과 탈착량은 거의 미미하였다.

White-rot fungi of the genus Bjerkandera are cosmopolitan and have shown potential for industrial application and bioremediation. When distinguishing morphological characters are no longer present (e.g., cultures or dried specimen fragments), characterizing true sequences of Bjerkandera is crucial for accurate identification and application of the species. To build a framework for molecular identification of Bjerkandera, we carefully identified specimens of B. adusta and B. fumosa from Korea based on morphological characters, followed by sequencing the internal transcribed spacer region and 28S nuclear ribosomal large subunit. The phylogenetic analysis of Korean Bjerkandera specimens showed clear genetic differentiation between the two species. Using this phylogeny as a framework, we examined the identification accuracy of sequences available in GenBank. Analyses revealed that many Bjerkandera sequences in the database are either misidentified or unidentified. This study provides robust reference sequences for sequence-based identification of Bjerkandera, and further demonstrates the presence and dangers of incorrect sequences in GenBank.
본 연구의 목적은 회전생물반응기에 고정화한 P. chrysosporium IFO 31249를 이용하여 LiP 생산의 최적 조건을 조사하는 것이다. 회전생물반응기에서 batch culture시 최대 LiP 활성도는 300 U/L이었다. LiP 생산의 최적 조건은 드림의 회전 속도는 1 rpm, 담체 충진율은 20%이었다. MnP 생산을 위한 $MnSO_4$$\cdot$$H_2O$ 의 최적 농도는 50 ppm이었다. 그리고 산소의 충분한 공급은 LiP 생산의 가장 중요한 인자이었다. 또한 LiP 및 MnP의 생산에 있어 repeated-batch culture가 3번이나 가능하였다.
대표적인 azo계 반응성 염료인 Remazol Black B에 대한 탈색능이 우수한 신규 백색부후균인 Phanerochaete sp. EJ-31L의 탈색 특성을 평가해 보았다. 보조 탄소원 농도, 질소원, pH 등의 배양조건이 염료 탈색율에 미치는 영향을 분석한 결과, 3%(w/v)의 sucrore, 0.05%(w/v)의 $(NH_4)_2SO_4$를 첨가한 배지에서 pH 6.5의 조건에서 탈색능이 우수하였다. 혐기적 조건에서는 호기적 조건 대비 27% 수준의 탈색 효율을 보였으며, 진탕배양에서의 탈색율이 정치배양 대비 2배이상 높아 Phanerochaete sp. EJ-31L은 호기적 조건에서 탈색능이 우수함을 알 수 있었다. 초기 Remazol Black B 농도 50 ppm에 대한 회분식 처리시 약 95% 이상의 탈색율을 보여 향후 azo계 염료 처리용 생물공정에 백색부후균인 Phanerochaete sp. EJ-31L을 활용할 수 있을 것으로 기대된다.
The effect of cadmium ions on ligninolytic and decolourizing activities in cultures of two white-rot fungi, Cerrena unicolor and Trametes versicolor, were examined. Cadmium was added to the shallow stationary cultures growing on a liquid mineral medium. Both examined strains sorbed Cd ions in the first 24 hr of incubation. An appreciable stimulation of the activity of extracellular laccase (LAC) and inhibition of the extracellular manganese-dependent peroxidase (MnP) were simultaneously observed when 25 mgL-1 and 50 mgL-1 of cadmium ions were added to the cultures. On the other hand, the addition of cadmium ions also resulted in stimulating the decolorization activity of C. unicolor to decolorize Remazol Brilliant Blue R (RBBR) in the cultures, but decreasing it in the culture of T. versicolor, which is compared to the inhibition of MnP activity in this fungus. Our data indicate that the presence of Cd(II) ions can affect the ligninolytic activity of white-rot fungi. It was found that C. unicolor is a strain resistant to the presence of Cd ions in the liquid culture media, and has a potential to use this strain for bioremediation of sites contaminated with both heavy metals and aromatic pollutants.
Degradation and glucose production from wood chips of white pine (Pinus strobus) and tulip tree (Liriodendron tulipifera) by several white rot fungi were investigated. The highest weight losses from 4 g of wood chips of P. strobus and L. tulipifera by the fungal degradation on yeast extract-malt extract-glucose agar medium were 38% of Irpex lacteus and 93.7% of Trametes versicolor MrP 1 after 90 days, respectively. When 4 g of wood chips of P. strobus and L. tulipifera biodegraded for 30 days were treated with cellulase, glucose was recovered at the highest values of 106 mg/g degraded wood by I. lacteus and 450 mg/g degraded wood by T. versicolor. The weight loss of 10 g of wood chip of L. tulipifera by T. versicolor on the nutrient non-added agar under the nonsterile conditions was 35% during 7 weeks of incubation, and the cumulative amount of glucose produced during this period was 239 mg without cellulase treatment. The activities of ligninolytic enzymes (lignin peroxidase, manganese peroxidase, and laccase) of fungi tested did not show a high correlation with degradation of the wood chips and subsequent glucose formation. These results suggest that the selection of proper wood species and fungal strain and optimization of glucose recovery are all necessary for the fungal pretreatment of woody biomass as a carbon substrate.
One of the most economically viable processes for the bioconversion of many lignocellulosic waste is represented by white rot fungi. Phanerochaete chrysosporium is one of the important commercially cultivated fungi which exhibit varying abilities to utilize different lignocellulosic as growth substrate. Examination of the lignocellulolytic enzyme profiles of the two organisms Phanerochaete chrysosporium and Rhizopus stolonifer show this diversity to be reflected in qualitative variation in the major enzymatic determinants (ie cellulase, xylanase, ligninase and etc) required for substrate bioconversion. For example P. chrysosporium which is cultivated on highly lignified substrates such as wood (or) sawdust, produces two extracellular enzymes which have associated with lignin deploymerization. (Mn peroxidase and lignin peroxidase). Conversely Rhizopus stolonifer which prefers high cellulose and low lignin containg substrates produce a family of cellulolytic enzymes including at least cellobiohydrolases and ${\beta}-glucosidases$, but very low level of recognized lignin degrading enzymes.
This study was conducted to evaluate the degradation of aromatic dyes and the production of ligninolytic enzymes by 10 white rot fungi. The results of this study revealed that Pycnoporus cinnabarinus, Pleurotus pulmonarius, Ganoderma lucidum, Trametes suaveolens, Stereum ostrea and Fomes fomentarius have the ability to efficiently degrade congo red on solid media. However, malachite green inhibited the mycelial growth of these organisms. Therefore, they did not effectively decolorize malachite green on solid media. However, P. cinnabarinus and P. pulmonarius were able to effectively decolorize malachite green on solid media. T. suaveolens and F. rosea decolorized methylene blue more effectively than any of the other fungi evaluated in this study. In liquid culture, G. lucidum, P. cinnabarinus, Naematoloma fasciculare and Pycnoporus coccineus were found to have a greater ability to decolorize congo red. In addition, P. cinnabarinus, G lucidum and T. suaveolens decolorized methylene blue in liquid media more effectively than any of the other organisms evaluated in this study. Only F. fomentarius was able to decolorize malachite green in liquid media, and its ability to do so was limited. To investigate the production of ligninolytic enzymes in media containing aromatic compounds, fungi were cultured in naphthalene supple mented liquid media. P. coccineus, Coriolus versicolor and P. cinnabarinus were found to produce a large amount of laccase when grown in medium that contained napthalene.
