Water storage cells (WSCs) in the leaf succulent Orostachys malacophyllus have been studied to understand their adaptive nature to its coastal habitat employing the electron microscopy. Attention has been paid to the features of vacuoles and plasmodesmata in this study, since leaf tissues in O. malacaphyllus are under continous physiological drought due to its occurrence in the shore-line environment. The WSCs occupied almost all of the leaf volume and appeared empty at low magnifications. Among the WSCs, small rudimentary vascular bundles were scattered throughout the internal volume. However, in high magnification the WSCs were vacuolate in most cases and vacuolization into a well-developed huge central vacuole was very common phenomenon. Such vacuolization has been detected within the vacuoles as well as within the cytoplasms. Well-developed plasmodesmata were often found in cells appeared to be mucilagenous. Moreover, plasmodesmata being involved in the secretion of materials or structures were even encountered. Thus, vacuolization from various sizes of vacuoles in the WSCs to have a huge central vacuole seems playing an important role in adapting the plant itself to its coastal habitat.
석유 제품 지하 저장 공동의 지하수 수질은 공동 인근 매립지 수질의 영향을 받는다. 매립지 주변의 지하수 수질 분석 결과, 시료의 반 정도에서 지하공동 수질에 영향이 미미한 농도의 As, Cu 및 Pb가 검출되었으며, Cd, Hg, $Cr^{6+}$, $CN^-$, TCE, PCE 및 Pheno은 검출되지 않았다. $COD^{Mn}$의 농도는 모든 시료에서 $8.0mg/{\ell}$이하로 유기물질에 의한 지하수 오염은 거의 없는 것으로 분석되었다. 1차, 2차 미생물 분석결과 총박테리아는 각각 $94.84{\times}10^4cells/{\mu}g/m{\ell}$과 $146.26{\times}10^{-4}cells/{\mu}g/m{\ell}$이고 황산환원 박테리아는 $2cells/m{\ell}$이하로 분석되어 지하공동의 수질에 영향이 미미할 것으로 분석되었다. 저장 공동 인근 지역의 지하수 수질 분석은 공동의 수리 지질의 안정성을 향상시키기 위하여 지속적인 조사와 관리가 필요할 것으로 판단된다.

W/O/W (water-in-oil-in-water) type multiple emulsion was applied to improve the storage stability of an antagonistic microorganism, Burkholderia gladioli. Encapsulation of microorganism into a W/O/W emulsion was conducted by using a two-step emulsification method. W/O/W emulsion was prepared by the incorporation of B. gladioli into rapeseed oil and the addition of polyglycerin polyriconolate (PGPR) and castor oil polyoxyethylene (COG 25) as the primary and secondary emulsifier, respectively. Microcrystalline cellulose was used as an emulsion stabilizer. To evaluate the usefulness of W/O/W emulsion formulation as a microbial pesticide for controlling the bacterial wilt pathogen (Ralstonia solanacearum), the storage stability and antagonistic activity of emulsion formulation were tested in vitro. The storage stability test revealed that the viability of formulated cells in emulsion was higher than that of unformulated cells in culture broth. At $4^{\circ}C$, the viabilities of formulated cells and unformulated cells at the end of 20 weeks decreased to about 2 and 5 log cycles, respectively. At $37^{\circ}C$, the viability of formulated cells decreased to only 2 log cycles at the end of storage. On the other hand, the viable cells in culture broth were not detected after 13 weeks. In activity test, formulated cells in emulsion were more effective in inhibiting the growth of pathogen than unformulated cells in culture broth. Unformulated cells completely lost their antagonistic activity during storage under similar conditions. The W/O/W multiple emulsion formulation was shown to be useful as the novel liquid formulation for biological control.
This study analyzed the relationship between harmful cyanobacterial abundance and environmental factors in order to figure out the causes of the recent increase of cyanobacteria in Lake Dukdong from 2019 to 2021. Lake Dukdong, which is used as a drinking water source for Gyeongju City, has an algae alert system in place. Lake Dukdong has maintained good water quality, but algae alert level 1 (over 1,000 cells/mL) has been issued in recent years. As a result of Pearson correlation analysis (from May to Oct.), the cell density of Microcystis and Aphanizomenon, which form part of the most harmful cyanobacteria genus, were significantly positively correlated with the water temperature and water storage volume. T-test was performed to compare the data from 2016-2018 and 2019-2021 (from May to Oct.). The average density of harmful cyanobacteria cells increased about six-fold from 54 to 344 cells/mL. There were significant differences in water temperature, pH, total nitrogen (TN), total phosphorus (TP), TN/TP ratio, water storage volume, and cyanobacterial cell density. Water temperature increased from 19.2 to 22.8 ℃. TP concentration increased from 0.017 to 0.028 mg/L. The main cause of the recent increase of harmful cyanobacteria in Lake Dukdong is thought to be the increase in water temperature, TP concentration, and water storage volume from 2019 and 2021, resulting in more favorable conditions for cyanobacterial growth.
외상성 손상 후 치아 탈구 시 치아를 저장하는 저장용액의 종류와 온도가 치주인대세포의 생존율에 어떠한 영향을 미치는지 연구하기 위하여 치주인대세포를 10% fetus bovine serum(FBS) 함유 ${\alpha}-minimal$ essential medium$({\alpha}-MEM)$에서 $37^{\circ}C$ 5% $CO_2$ 공기 혼합 배양기에서 배양하고 4 25, $37^{\circ}C$의 Hank's balanced salt solution(HBSS) 과 ${\alpha}-MEM$, 우유(S회사, P회사), tap water에 저장하고 60분이 지난 후 각 군에 대해 치주인대세포의 생존율을 측정하기 위하여 MTT assay를 시행하여 다음과 같은 결과를 얻었다. 1. $4^{\circ}C$ 저장용액의 치주인대세포의 생존율은 ${\alpha}-MEM$과 P회사우유에서 가장 높았고 HBSS, S회사우유, tap water 순으로 낮았다. 2. $25^{\circ}C$ 저장용액의 치주인대세포의 생존율은 ${\alpha}-MEM$에서 가장 높았고 P회사우유, HBSS, S회사우유, tap water 순으로 낮았다. 3. $37^{\circ}C$ 저장용액의 치주인대세포의 생존율은 ${\alpha}-MEM$과 P회사우유, HBSS, S회사우유에서 높았고 tap water에서 가장 낮았다. 4. ${\alpha}-MEM$의 치주인대세포의 생존율은 $4^{\circ}C$에서 가장 높았고 $25^{\circ}C$와 $37^{\circ}C$순으로 낮았으며 HBSS에서는 $4^{\circ}C$에서 높았고 $25^{\circ}C$와 $37^{\circ}C$에서 낮았다. 5. S회사와 P회사우유에서 치주인대세포의 생존율은 $4^{\circ}C$와 $25^{\circ}C$에서 높았고 $37^{\circ}C$에서 낮았다. 이상의 결과를 종합해볼 때 외상으로 인해 치아가 탈구되었을 때 탈구된 치아의 저장용액으로 HBSS용액이 추천되고 있으나 이 연구에서 $4^{\circ}C$와 $25^{\circ}C$ 우유에서 치주인대세포의 생존율이 높았으므로 사고 현장에서 쉽게 구할 수 있고 치주인대세포의 생활력 보존에도 유리한 낮은 온도의 우유에 탈구된 치아를 보존하는 방법도 좋을 것으로 생각된다.
This study was carried out to investigate the microbiological and chemical properties of natural water during storage. The water samples were taken at the time of purchase and the opened bottles and unopened bottles stored at the temperature of 4$^{\circ}C$, 18$^{\circ}C$, and 3$0^{\circ}C$. The bacterial content normally rose to 2.06$\times$102 CFU/$m\ell$ for the unopened bottles and 2.91$\times$102 CFU/$m\ell$ for the opened bottles after 2 weeks of storage, and 1.21$\times$102 CFU/$m\ell$ and 2.64$\times$102, respectively, after 24 weeks of storage. The number of viable cells of bacteria peaked more rapidly at the storage temperature of 3$0^{\circ}C$ than 18$^{\circ}C$. But the total samples were found to be negative for coliforms test during the study period. The average range of pH value was from 7.39 to 7.76. The results showed that the nitrates and chlorides satisfied the Korea Drinking Water Quality Standards during the storage period of 24 weeks. However, the undesirable changes of the taste and odor were detected within 2 weeks and 3 weeks, respectively.
Unusual bloom of toxic cyanobacteria in water bodies have drawn attention of environmentalists world over. Major bloom of Anabaena, Microcystis in water storage reservoir, rivers and lake leading to adverse health effects have been reported from Australia, England and many part of the world. These cyanobacterial cells can release intercellular matter like toxin in water and these intercellular matter can increase the concentration of organic matter. Cellysis can occur when algal cells meet the disinfectants like chlorine in water treatment plant and the resultant rising up of DOC(Dissolved Organic Carbon) or TOC(Total Organic Carbon) can increase the formation of disinfection by products. Disinfectants that kill microorganisms react with the organic or inorganic matter in raw water. In general disinfectants oxidize the matter in raw water and the resultant products can be harmful to human. There are always conflict about which is more important, disinfection or minimizing disinfection by products. The best treatment process for raw water is the process of the lowest disinfection by products and also the the lowest microorganism. In this study the cultured cells, Microcytis Aeruginosa(MA), Anabaena Flos-aquae(AF), Anabaena Cylindrica(AC), and the cells obtained in Daechung Dam(DC) whose dominant species was Anabaena Cylindrica were subjected to chlorination. Chlorination oxidizes inorganic and organic compounds and destruct live cells in raw water. Chloroform was analyzed for the cultured cells which were treated with $20mg/\ell$ dose of chlorine. In general chloroform is easily formed when dissolved organic matter react with chlorine. The cultured cells contributes the concentration of dissolved organic carbon and also that of total organic carbon which might be potent precusors of chloroform formed. The correlations of the concentration of chloroform, DOC and TOC were investigate in this study.

The effect of cryoprotectants (maltodextrin+glycerol) and cryoprotectants+antioxidant [ascorbic acid and/or butylated hydroxytoluene (BHT)] mixtures on the survival, electrolyte leakage, and lipid degradation of freeze-dried Weissella paramesenteroides LC11 during storage was investigated and compared with that of the control (cells without additives) over a 90-day storage period at 4 or $20^{\circ}C$ in glass tubes with water activity ($a_w$) of 0.23. The survival, electrolyte leakage, and lipid degradation were evaluated through colony counts, electrical conductivity, and thiobarbituric acid reactive substances (TBARS) content, respectively. The fatty acids composition was determined by gas chromatography, in both the total lipid extract and the polar lipid fraction, and compared with that of the control after the 90-day storage period. As the storage proceeded, increases in leakage value and TBARS content, as well as a decrease in viability, were observed. After 90 days of storage, the major fatty acids found in both the total lipid extract and the polar lipid fraction were palmitic (16:0), palmitoleic (16:1), stearic (18:0), oleic (18:1), linoleic (18:2), and linolenic (18:3) acids. The survival, leakage value, TBARS content and 18:2/16:0 or 18:3/16:0 ratio were the greatest for the protected strain held at $4^{\circ}C$. Cells with the cryoprotectants+BHT mixture showed the highest percentage of survival and 18:2/16:0 or 18:3/16:0 ratio in both lipid extracts, as well as the lowest leakage value and TBARS content after the 90-day storage period. Drying cells with the cryoprotectants+BHT mixture considerably slowed down polar lipid degradation and loss of membrane integrity, resulting in improved viability during storage.
태양에너지를 활용하여 전력을 생산하는 시스템인 Solar water battery는 광전기화학전지와 에너지저장시스템을 결합한 것으로 추가적인 외부 전압 없이 태양에너지의 전환과 저장을 동시에 할 수 있다. Solar water battery는 광전극, 저장전극 그리고 상대전극으로 구성되어 있고, 이들의 선택과 조합은 시스템의 성능과 효율에 있어 중요한 역할을 한다. 본 연구에서는 Solar water battery의 구성요소들을 변화시켜 시스템에 미치는 영향을 알고자 하였다. 상대전극이 방전 시 미치는 영향, 광전극과 저장전극의 전극 재료, 전해질의 종류에 따른 태양에너지 전환 효율과 저장 용량에 미치는 영향에 대해 연구하였다. 이들의 최적화된 구성(TiO2 : NaFe-PB : Pt foil)에서 15시간동안의 광조사 후의 방전 용량이 72.393 mAh g-1으로 시스템 구성 조건에 따라 광전환/저장 효율이 크게 영향을 받음을 확인 할 수 있었다. 또한, 유기 오염물질을 광전극 반응조내 전해질에 첨가하여 광전하를 효율적으로 분리시킴으로써 광전류 증가시켰으며, 이로 인해 저장용량이 향상되고, 동시에 오염물질도 분해시킬 수 있음을 확인하였다. 이처럼 Solar water battery는 추가적인 외부 전압이 필요없는 새로운 친환경 태양에너지 전환/저장 시스템이며, 나아가 수처리에도 활용할 수 있을 것으로 기대된다.
