제주지역 양돈장에서 발생한 자돈 설사형 바이러스의 면역조직화학적 확인 (Immunohistochemical study of causative agents for the porcine viral diarrhea disease on newborn piglets in Cheju)

  • In an outbreak of acute porcine diarrhea in newborn piglets, an etiological study was carried out using piglets submitted in Cheju Province Institute for Livestock Promotion(Cheju Veterinary Service for the disease diagnosis). Sixteen piglets(2-7 days old) were collected from 4 farms during outbreaks of diarrhea disease(from January to April 2000). Specimens were taken after necropsy and examined by immunohistochemistry using of monoclonal antibodies for porcine epidemic diarrhea(PED) virus, transmissible gastroenteritis(TGE) virus, and porcine rotavirus. Immunohistochemistry showed that PED virus antigens, but both TGE virus and rota virus antigens not, were localized in the some epithelial cells of the intestines of 14 animals among 16 piglets examined. PEB virus antigens were mainly detected in the cytoplasm of enterocytes. Infected cells, which were most abundant in the villous epithelial cells of the jejunum and ileum, were uncommon in the crypt, epithelial cells, the lamina propria and Peyer's patches of piglets examined. The results suggest that PED virus is one of the most prevailing agents in an outbreak of fatal diarrhea in newborn piglets on Cheju island and PED virus was need to further study to prevent this disease.

Sustained Viral Activity of Epstein-Barr Virus Contributes to Cellular Immortalization of Lymphoblastoid Cell Lines

  • EBV-transformed lymphoblastoid cell lines (LCLs) are used as a resource for human genetic, immunological, and pharmacogenomic studies. We investigated the biological activity of 20 LCL strains during continuous long-term subculture up to a passage number of 160. Out of 20 LCL strains, 17 proliferated up to a passage number of 160, at which point LCLs are generally considered as "immortalized". The other three LCL strains lost the ability to proliferate at an average passage number of 41, during which these LCLs may have undergone cellular crisis. These non-immortal LCL strains exhibited no telomerase activity, decreased EBV gene expression, and a lower copy number of the EBV genome and mitochondrial DNA when compared with immortal LCLs. Thus, this study suggests that sustained EBV viral activity as well as telomerase activity may be required for complete LCL immortalization.

Identification of Korean native cattle persistently infected with BVDV using Ear-notch method

  • Bovine viral diarrhea Virus (BVDV) infections cause respiratory, gastrointestinal, and reproductive problems, such as infertility, abortion, stillbirth, and sickly offspring. Many countries have reduced the economic damage through the application of different control programmes, and some have successfully eradicated BVD. Detection and elimination of cattle persistently infected (PI) with BVDV is important for BVD eradication because PI cattle are a main source of BVD transmission. In this study, the prevalence of Korean native cattle persistently infected (PI) with BVDV was investigated and determined in 49 farms with 3,050 cattle. The all samples were collected by ear notch sampling. Korean native cattle with initial positives on antigen-ELISA (Ag-ELISA) were sampled again after 3~4 weeks and cattle with second positives in both Ag-ELISA and immunohistochemistry (IHC) were identified as PI cattle. Among the 49 farms, 14 (28.6%) farms had at least more than one PI cow and 21 (0.69%) of 3,050 cattle were determined as PI cattle. As a result of this work, it is suggested that national BVD eradication program is required to reduce economic losses by BVDV infection in Korean cattle industries.

  • Coronavirus, known as one of the causes of colds including mild upper respiratory tract disease in humans, has mutated into the infectious severe disease, COVID-19 through SARS and MERS. The mortality and symptoms of COVID-19 are related to the ability to regulate innate immunity, which acts as the first barrier against microorganisms and viruses. During the COVID-19 pandemic, the demand for food that helps to strengthen immunity is rapidly increasing. Functional foods promote general health and alleviate the risk of disease symptoms by activating multiple biological functions. A recent, there is an interest in discovering functional substances that can induce enhancement of immunity and prevent viral infection as well as relieve disease symptoms. Therefore, this article focus to understand the concept of immune response and highlights the recent status of functional foods and research trends that can help prevent and treat viral infections by inducing the enhancement of immune function.

  • It was attempted in the present study to determind the effect of Korean ginseng on acute viral(B type) hepatitis by double blind test clinically and the results were as follows. 1. The double blind test of 4 week period showed no statistical significance of the effect of ginseng on the disease. 2. The double blind test of 2 week period, however, showed a significant effect in such clinical symptoms as appetite, stomach ache, headache, chillness dizziness, nausea and vomitting, stool habit change and jaundice. Improvement of the blood serum level of transaminases, bilirubin. alkaline phosphatase and cholesterol Chillness feeding period was observed.

Acupuncture and blood borne viral infections: a brief summary

  • Blood borne infectious diseases are usually a public concern. The transmissions of many diseases are via blood borne mode. Several activities are related to this transmission such as injection, blood transfusion and transplantation. Also, the acupuncture practice can be a possible route for blood borne infectious disease transmission. In this specific review, the author briefly reviews acupuncture and blood borne viral infections.

  • 목 적 : Kikuchi-Fujimoto disease는 histiocytic necrotizing lymphadenitis라고도 불리는 질환으로 발열과 경부 림프절염을 특징으로 하며 자연 호전되는 경과를 보이는 질환이다. 원인에 대해서는 아직 잘 밝혀지지 않았다. 자연 호전되는 경과 등은 이 질환의 원인으로 바이러스 감염을 추측하게 되고, 고려되는 바이러스로는 대표적으로 EBV, HHV6, HHV8, CMV 등이 있다. 본 연구는 그 중에서 EBV, HHV6를 선택하여 HNL와의 연관성을 밝히고자 하였다. 방 법 : 1999년에서 2005년 사이 부산대학교 병원에서 조직검사에서 KFD으로 진단된 환자 51명을 대상으로 나이, 성별, 발열기간, 침범된 림프절 등에 대하여 의무기록지를 바탕으로 후향적으로 분석하였고 이들의 조직을 이용하여 EBV에 대한 ISH을 시행하고, HHV6에 대한 면역조직화학염색을 각각 시행하였다. 결 과 : 대상환자는 남자가 24명, 여자가 27명이었고 평균 연령은 25.9세였다. EBV에 대한 ISH에서 양성을 나타낸 환자는 51명 중에서 8명으로 15.7%였으며 HHV6에 대한 면역조직화학염색에서 양성을 나타낸 환자는 15명으로 29.4%였다. EBV의 경우 혈청검사(VCA IgG와 VCA IgM)가 같이 시행된 경우가 23명이었고 이중 한 명에서 EBV VCA IgM이 양성이면서 EBV ISH에서도 양성이었다. 결 론 : 본 연구는 HNL의 원인으로서 EBV와 HHV6의 역할을 증명하지 못하였으나, 드물게 HNL의 원인으로 바이러스 감염이 관여할 것으로 추측된다.

새로운 국내 분리 토끼출혈병바이러스(RHDVa)를 감염시킨 토끼에서의 경시적인 병리학적 변화와 조직 내 바이러스 항원 분포 (Sequential pathologic changes and viral distribution in rabbits experimentally infected with new Korean strain of rabbit hemorrhagic disease virus (RHDVa))

  • Rabbit hemorrhagic disease is a highly acute and fatal viral disease caused by rabbit hemorrhagic disease virus (RHDV). Since first outbreak in Korea 1987, RHDV has been continually affected in the country, but the pattern of outbreak seem to be changed. In this study, to understand the pathogenesis of the new RHDVa serotype, we therefore carried out to inoculate RHDVa to rabbits, and to examine the sequential histopathologic changes and viral distribution. Macroscopically, various sized dark red or white spots or appearance were observed in the liver, lung, kidney uterus and ureter. In euhanized rabbits, significant pathologic findings such as infiltration of heterophils and mononuclear cells were observed at 24 hours after inoculation (HAI), and these were sequentially extended periportal to centrilobular area. However, in dead rabbits, severe hepatic degeneration and/or necrosis with relatively weak inflammatory responses were observed. RHDV antigens began to detect in liver, spleen, and lung from 12 HAI by PCR. Immunohistochemically, RHDV positive cells were seen in only liver from 24 HAI, and the degree of immunogen reactivity was stronger in dead rabbits than in euthanized ones. In conclusion, RHDVa caused the subacute or chronic infection accompanying low mortality and moderate to severe inflammatory reaction in rabbits, suggesting the possibility that RHD could become endemic.

오제스키병의 생체 조기진단을 위한 면역세포화학, In situ hybridization 및 전자현미경적 연구 (Immunocytochemistry, In situ hybridization and electron microscopy for early diagnosis of Aujeszky's in living pigs)

  • The purpose of this study was to establish early diagnostic methods for the detection of Aujeszky's disease viral antigens and nucleic acid in nasal cells, and buffy coats from experimentally infected living pigs by a combination of immunocytochemistry, in situ hybridization with digoxigenin(DIG)-labled probe and electron microscopy. Forty days old piglets were inoculated intranasally with $10^{7.0}TCID_{50}$ of Aujeszky's disease virus (ADV, NYJ-1-87 strain). The viral antigens and nucleic acid of ADV were detected in nasal cells, and buffy coat for 20 days after inoculation by immunocytochemistry, in situ hybridization with DIG-labeled probe and electron microscopical method. The results were compared with conventional methods such as a porcine Aujeszky's disease serodiagnostic(PAD) kit, neutralization test(NT) and virus isolation. 1. The viral antigens, nucleic acids and capsids of ADV were detected in nasal cells, buffy coats from 3 days to 20 days after inoculation by immunocytochemistry, in situ hybridization with DIG-labeled probe and electron microscopy, respectively. 2. When viral antigens were detected by the immunocytochemical technique, a diffuse brown deposit was observed in the nucleus and cytoplasm of nasal cells, buffy coats and PK-15 cells under a microscope. 3. DIG-labeled DNA probe was prepared by amplification of conserved sequence of recombinant ADV-gp50 clone with polymerase chain reacction. When ADV-DNA was detected by ISH with DIG-labeled probe, purplish blue pigmentation were observed in the nuclei and cytoplasms of ADV-infected cells under a microscope. Positive signals were observed in nasal cells and in the buffy coat and PK-15 cells at the first day after inoculation. 4. Where ADV-capsids were detected by transmission electron microscopical method, aggregation of capsids was observed in the nuclei and cytoplasms of nasal cells, buffy coats and PK-15 cells. The results suggested that these methods were considered as the highly sensitive and reliable tools for rapid and confirmative diagnosis of Aujeszky's disease in living pigs.

암 유전자 치료제의 개발 현황 (Cancer Gene Therapy. History and Major Developments)

  • Medicine is undergoing a revolution in the understanding of the mechanisms through which disease processes develop. The advent of genetics and molecular biology to oncology not only is providing surrogate predictors of therapy response and survival which are forming the basis for selection among established treatment options, but is providing targets for new directions in therapy as well. Molecular modification of somatic cells for the purposes of protecting the normal cells from the toxicity of cancer chemotherapy, for the sensitization of the tumor cells to therapy and use of conditionally replicating viral vector have been new directions of cancer treatment which have reached the clinical arena. Advances in molecular pharmacology and vector design summarized in this paper may provide solutions to some of the existing problems in the technology of gene transfer therapy. Continued basic research into the biological basis of human disease, systemic studies of the application of these discoveries to therapy and the improvement of vector for gene delivery all combined may result in advances in this important field of therapy over the next few years.
