Effects of Prostaglandin E2 Analogue, Enprostil, on Lipid Metabolism in Mice

  • This study was conducted to investigate the effects of effects of enprostil, a prostaglandin $E_2$, analogue, on liver triacylglycerol content and factors that regulate liver lipid metabolism in mice. Mice received vehicle or $10{\mu}g$ enprostil/kg body weight intraperitoneally every 6 h, and were killed at 0, 6, 12, 18 and 24 h after the first injection. Enprostil significantly lowered liver triacylglycerol content after 12 h of the first injection. However, the peroxisomal ${\beta}$-oxidation activity was inconsistent with the result of liver triacylglycerol content, because its activity was lovered by enprosil. In another experiment, the effect of enprostil on lipid metabolism in mice was investigated in a short period. Mice received $10{\mu}g$ enprostil/kg body weight intraperitoneally, and were killed after 0, 5, 10, 30 and 60 min. After 30 min, malic enzyme activity was significantly increased by the administration of enprostil compared with the activity at 5 min after. No significant changes in liver carnitine palmitoyltransferase and peroxisomal ${\beta}$-oxidation activities were observed. Plasma free fatty acid concentrations were markedly reduced from 5 through 60 min after the administration of enprostil. Consequently, enprostil suppressive effect on liver triacylglycerol concentration might result from the decreased entry of free fatty acid into liver.

Idescarpin Isolated from the Fruits of Idesia polycarpa Inhibits Melanin Biosynthesis

  • Tyrosinase is an enzyme that catalyzes the biosynthetic pathway of melanin pigments participating in the coloring of skin, hair, and eyes, and is widely distributed in nature. The inhibitory compounds of tyrosinase have been extensively used as a cosmetic agent with a skin-whitening effect. In this paper, several plant extracts were screened using Melan-a cells for the melanin biosynthesis inhibition activity, and Idesia polycarpa was selected. A melanin biosynthesis inhibitor was isolated from I. polycarpa fruits by activity-guided fractionation, and the inhibitor was identified as 6-hydroxy-2-[[[(1-hydroxy-6-oxo-2-cyclohexenl-yl)carbonyl]oxy]methyl]phenyl$\beta$-D-glucopyranoside (idescrapin) by comparing it with reported spectral data. Idescarpin $(IC_{50}=8{\mu}g/ml)$ reduced melanin content compared with the vehicle. In addition, the inhibitory activity of idescarpin for melanin synthesis is mediated by decreasing tyrosinase protein rather than directly inhibiting the tyrosinase activity. These results suggest that idescarpin isolated from I. polycarpa fruits may be used as a skin-whitening agent.

Bitter Melon Seed Extract does not Alter Photoperiodic Effects on Reproduction of Male Golden Hamsters

  • Bitter melon (Momordica charantia, MC) has been used in traditional Korean medicine in treating diabetes. In addition, some reports were emerged, showing the antifertility activities of MC in mammals. We investigated the effects of ethanolic MC extract on the reproductive activity of golden hamsters whose spermatogenetic capacity is controlled by their photoperiods. The animals were divided into 4 groups: long photoperiod (LP) control, short photoperiod (SP) control, and LP animals treated with MC. The animals were orally ingested with low (0.03 g/kg) or high (0.15 g/kg) concentrations of the ethanolic extracts for 8 weeks on the daily basis. The control animals received the vehicle. The animals were then mated with age-matched females, experienced pregnancy. As results, the LP control animals showed active large testes but SP control animals displayed remarkably reduced testes. The animals treated with both concentrations of MC extracts demonstrated large testes, indicating fertile activity as animals in LP. LP control animals had litters as expected, but SP controls had no litters at all. MC extract showed the same results as LP animals in generating offsprings. These results suggest that the MC extract does not change the photoperiodic influence on reproductive activity of male golden hamsters.

Synthesis of Palladium Nanocubes/Nanorods and Their Catalytic Activity for Heck Reaction of Iodobenzene

  • Palladium has been used as a catalyst not only in Suzuki and Heck cross coupling reaction in organic chemistry, but also in automobile industry for the reduction of vehicle exhausts. The catalytic activity of Pd nanoparticles depends strongly on their size and exposed crystalline facets. In this study, the single crystalline palladium nanocubes/nanorods were prepared in the presence of polyvinyl pyrrolidone (PVP) and potassium bromide (KBr) using the polyol method. Selected area diffraction pattern and high-resolution transmission electron microscopy (TEM) were performed by TEM. The result shows that the ratio of KBr/PVP is the key factor to determine whether the product is cubes or rods. The as-prepared Pd nanocubes were highly uniform in both size and shape. The ordered packing structures including monolayer and multilayer can be fabricated via the rate-controlled evaporation of solution solvent. The catalytic activity of these Pd nanocubes towards heck reaction of iodobenzene with acrylate or acrylic acid was found to be higher than that of Pd nanorods. We suspect it is caused by the difference of energy state while Pd nanocubes is {100} plane and nanorods is {111} plane.

  • Nowadays, with improvement of economical income and life qualities, life pattern changes have been brought such as increasing of avocational activities. Consequently, following those life trends, utilization of car is getting increased. Thus, the perceptions of car have been changed from the only means of transport in the past to a 2nd residental space. that is why the car's endo environmental factors are getting so important. Air conditioner regulating air ventilization in vehicle's indoor automatically sets the right temperature based on the differences of indoor and outdoor's temperature with development of advanced functions to provide better environmental qualities in vehicle. However, even those advanced techniques for functional development are got so diverse though, the essential technique for preventing the growth of bateria and mold inside of the air conditioner are not even severals. Especially, evaporator one of the vehicle air conditioning equipments generates cooled air by vaporizing refrigerant in liquid state with the water as the adduct for this reactions. It has structural difficulties for water vaporation then cause the growth of germs. That's why this reseach was focused on the way of eliminating germs in the vehicle air conditioner efficiently. Direct air sterilizer by using UVC(Ultraviolet C) is manufactured and that performances are evaluated.

Curcumin Increase the Expression of Neural Stem/Progenitor Cells and Improves Functional Recovery after Spinal Cord Injury

  • Objective : To investigates the effect of curcumin on proliferation of spinal cord neural stem/progenitor cells (SC-NSPCs) and functional outcome in a rat spinal cord injury (SCI) model. Methods : Sixty adult male Sprague-Dawley rats were randomly and blindly allocated into three groups (sham control group; curcumin treated group after SCI; vehicle treated group after SCI). Functional recovery was evaluated by the Basso, Beattie, and Bresnahan (BBB) scale during 6 weeks after SCI. The expression of SC-NSPC proliferation and astrogliosis were analyzed by nestin/Bromodeoxyuridine (BrdU) and Glial fibrillary acidic protein (GFAP) staining. The injured spinal cord was then examined histologically, including quantification of cavitation. Results : The BBB score of the SCI-curcumin group was better than that of SCI-vehicle group up to 14 days (p<0.05). The coimmunoreactivity of nestin/BrdU in the SCI-curcumin group was much higher than that of the SCI-vehicle group 1 week after surgery (p<0.05). The GFAP immunoreactivity of the SCI-curcumin group was remarkably lower than that of the SCI-vehicle group 4 weeks after surgery (p<0.05). The lesion cavity was significantly reduced in the curcumin group as compared to the control group (p<0.05). Conclusion : These results indicate that curcumin could increase the expression of SC-NSPCs, and reduce the activity of reactive astrogliosis and lesion cavity. Consequently curcumin could improve the functional recovery after SCI via SC-NSPC properties.

On Visualization of Trajectory Data for Traffic Flow Simulation of Urban-scale (도시 스케일의 교통 흐름 시뮬레이션을 위한 궤적 데이터 시각화)

  • As traffic volume increases and road networks become more complicated, identifying for accurate traffic flow and driving smooth traffic flow are a concern of many countries. There are various analytical techniques and studies which desire to study about effective traffic flow. However, the necessary activity is finding the traffic flow pattern through data visualization including location information. In this paper aim to study a real-world urban traffic trajectory and visualize a pattern of traffic flow with a simulation tool. Our experiment is installing the sensor module in 40 taxis and our dataset is generated along 24 hours and unscheduled routes. After pre-processing data, we improved an open source traffic visualize tools to suitable for our experiment. Then we simulate our vehicle trajectory data with a dots animation over a period of time, which allows clearly view a traffic flow simulation and a understand the direction of movement of the vehicle or route pattern. In addition we further propose some novel timelines to show spatial-temporal features to improve an urban environment due to the traffic flow.

Factors that Influence the Sharing Intention of Vehicle and Sharing Behavior in China (중국에서의 자동차 공유의도 요인과 공유 행동에 관한 연구)

  • Sharing economy which is sharing and borrowing goods, space, and services as a social and economic activity based on the Internet and smart phones is a new research topic in convergence area. The most representative areas of shared economy are space sharing and vehicle sharing. This paper aims to explore the factors affecting sharing intention of vehicle. We have identified the independent variables which include economic benefit, social benefit, social influence, psychological benefit and perceived risk. We also have done how these variables affect on sharing intention. Unlike Korea, in China vehicle sharing is legally allowed and has a large scale economy. To prove the hypothesis, we performed questionnaire investigation in China and analyzed with SPSS program. The result of hypothesis is the followings: The economic benefit, psychological benefit, social influence and perceived risk are affects sharing intention, and social benefit is not affect sharing intention and sharing intention is affect sharing behavior.

Effect of bitter melon (Momordica Charantia) on anti-diabetic activity in C57BLI/6J db/db mice (C57BL/6J db/db생쥐에서 여주 (Momordica Charantia)의 항당뇨 효과)

  • Many herbal extracts have been reported to have a preventive or therapeutic effect of on diabetes mellitus. Momordica Charantia commonly known as bitter melon or karela has been reported to be a medicinal plant for treating various diseases including cancers and diabetes. The objectives of this study were to investigate anti-diabetic effects of bitter melon (BM) as determined by blood glucose levels, glucose tolerance test (GTT), insulin tolerance test (ITT), insulin and HbA1C activities in serum, serum biochemical and lipid levels, histopathology, immunohistochemistry and AMPK-${\alpha}2$ expression of skeletal muscle in male C57BL/6J db/db mice. There were four experimental groups including vehicle control, BM 10 mg/kg, BM 50 mg/kg, and BM 250 mg/kg. BM at doses of 10, 50, and 250 mg/kg was orally administered to the diabetic mice everyday for 8 weeks. The treatments of BM 10, 50, and 250 mg/kg significantly decreased the blood glucose level in the diabetic mice compared with vehicle control (p < 0.05). The treatments of BM 10 and 50 mg/kg significantly decreased the GTT, ITT and HbA1c levels in the diabetic mice compared with vehicle control (p < 0.05). All BM groups significantly decreased GOT, GPT, BUN, LDL and glucose levels in the diabetic mice compared with the vehicle control mice (p < 0.05). The livers of mice treated with the BM 10, 50, and 250 mg/kg showed a remarkable decrease in the number of lipid droplets compared with the vehicle control. The pancreas of mice treated with the BM 10, 50, and 250 mg/kg showed a remarkable increase in insulin concentration of ${\beta}$-cells compared with the vehicle control. In addition, the treatments of BM 10, 50, and 250 mg/kg actually increased the expression of AMPK-${\alpha}2$ compared with vehicle control. These results suggest that BM has a respectable anti-diabetic effect resulting from inhibition of blood glucose level and lipid level in serum and that consumption of BM may give a benefit for controlling diabetes mellitus in humans.

  • The present study was done to determine the effect of trolox C. a hydrophilic analogue of vitamin E. on alteration in cytochrome P-450 (CYP)-dependent drug metabolism during ischemia and reperfusion. Rats were subjected to 60 min of hepatic ischemia and 5 h of reperfusion. Rats were treated intravenously with trolox C (2.5 mg/kg) or vehicle (PBS. pH 7.4), 5 min before reperfusion. Serum alanine aminotransferase and lipid peroxidation levels were markedly increased after ischemia and reperfusion. This increase was significantly suppressed by trolox C. (omitted)
