Studies on the Nutritional Components of Dandelion(Taraxacum officinale) (민들레의 영양성분에 관한 연구)

  • The free sugars in leaf and root of dandelion were composed of sucrose, glucose and fructose. The contents of total free sugars was higher in root than those in leaf. The oxalic acid, citric acid and malic acid contents of leaf was 45.4, 3.6, 2.7mg/100g-f.w., respectively. And the oxalic acid, citric acid and malic acid contents of root was 34.6, 2.1, 1.6mg/100g-f.w., respectively. Total free organic acid content of leaf was higher than that of root. The major free amino acids of dandelion were aspartic acid, serine, asparagine, glutamic acid, glycine, valine, isoleucine and content of glutamic acid was highest in free amino acids. The contents of vitamin A in leaf and root of dandelion was 135.4 and 34.1$\mu\textrm{g}$/100g-f.w., respectively. The contents of vitamin C in leaf and root of dandelion was 67.4 and 4.6 mg/100g-f.w., respectively.

Physiological activities of poly(amino acid)'s derivatives with β-sheet structure on the skin (베타시트 구조가 도입된 폴리아미노산 유도체의 피부활성에 관한 연구)

  • In this study, a synthesized poly(amino acid) self-assembly grafted with valine molecules was investigated on the skin activity of skin growth factors. The amphiphilic grafted poly(amino acid) derivatives were successfully synthesized by varying of degree of substitution(DS) and polymerization (DP) with valine molecules forming a β-sheet structure. Then, the pro-collagen biosynthesis of EGF(epidermal growth factor) was improved by 20%, and the inhibitory ability of tyrosinase activity was increased by 6.5 times by self-assembling of EGF with the poly(amino acid)s having β-sheet structures. This strategy of preparing protein self-assembly with poly(amino acid) derivatives will help improve the stability of protein growth factors and use it in medicals as well as cosmeceuticals through skin improvement.

Effect of dietary supplementation of β-mannanase on growth performance, carcass characteristics, excreta microflora, blood constituents, and nutrient ileal digestibility in broiler chickens

  • Objective: The aim of the present study was to investigate the effects of dietary supplementation of β-mannanase on growth performance, carcass characteristics, excreta microflora, blood constituents, and nutrient digestibility in broiler chickens. Methods: A total of 680 one-d-old Ross 308 (as hatched) broiler chickens were used in a 35-d growth assay. Chicks were sorted into pens with 17 birds/pen and 10 pens/treatment. Treatment diets were contained either 44% or 48% crude protein (CP) soybean meal (SBM) with or without β-mannanase. Results: Using SBM containing 48% CP led to an improvement (p<0.05) in feed conversion ratio (FCR) from d 1 to 14. Addition of β-mannanase to the diets significantly improved body weight gain (BWG) and FCR from d 1 to 14. During overall experimental period, BWG was affected (p<0.05) by CP level of SBM and inclusion of β-mannanase, but FCR and feed intake were not affected. Carcass characteristics were not influenced by treatment diets. The results showed that digestibility of dry matter (DM), nitrogen (N), and energy was not affected by CP level of SBM and/or inclusion of β-mannanase. Among essential amino acids (EAA) apparent digestibility of valine, methionine, and leucine improved (p<0.05) by the addition of β-mannanase to the diets. The results demonstrated that ileal digestibility of DM, N, and energy was not affected by treatment diets. Among EAA, the ileal digestibility of valine and arginine was higher (p<0.05) in the diets containing 48% CP SBM and/or β-mannanase. Excreta Lactobacillus count increased (p<0.05) by the addition of β-mannanase to the diets. Blood urea nitrogen, creatinine, and total protein level were not affected by treatments. Conclusion: Feeding chickens with diets containing 44% CP SBM resulted in detrimental effects on growth performance and digestibility of nutrients, but addition of β-mannanase to the 44% CP diet improved the growth performance of chickens without any effects on carcass characteristics.

Studies on the Chemical Composition of Grape Juices in Korea (한국산(韓國産) 포도의 과즙성분(果汁成分)에 관(關)한 연구(硏究))

  • The chemical composition of the of grape juices produced in Korea in 1969 were analyzed and the amino acids and sugars in these juices were detected by means of paper partition chromatography. The results are summarized as follows: 1. Less sugar and amino-N and more acid were contained in Korean grape juices than those of foreign's, and tannin content was not so different as foreign's. 2. More sugar, amino-N and tannin and less acid were contained in the juices of Campbell Early obtained in Anyang district than in Taejon and Pohang districts. 3. Twenty-one amino acids detected in grape juices were distributed as following frequency. Aspartic acid, serine, glycine, asparagine, lysine, arginine, threonine, alanine ${\gamma}-amino$ butyric acid, valine, leucine, proline (in 11 varieties), glutamine, tyrosine(10), cystine(9), glutamic acid, Hydroxyproline(8), isoleucine(4), phenylalanine, unknown(3). 4. Alanine was mostly abundant in all varieties and ${\gamma}-amino$ butyric acid was next and the decreasing order were arginine, valine, leucine, proline, glutamine, threonine, etc. in the amount. 5. Number of amino acid detected in grape juices of each varieties were 20 sorts in Delaware(Anyang), 19 in Black Hamburg, and Schuyler, 18 in Campbell Early(three districts), Delaware(seedless, Taejon) and Alden, 17 in Niagara, 16 in Muscat Hamburg, and 15 in Golden Muscat. 6. Number of essential amino acids contained in Delaware(Anyang) and Black Hamburg were 6 sorts and in Campbell Early (Anyang), Niagara and Muscat Hamburg were 5 and in others 4. 7. The same number of amino acid were detected in the juices of Campbell Early obtained from three districts, but hydroxyproline was detected in that of Anyang only, while isoleucine was appeared in those of Taejon and Pohang. 8. Glucose, fructose were detected in all grape juices.

Comparison of nutritional characteristics between wild and cultured juvenile black rockfish, Sebastes schlegeli (자연산 및 양식산 조피볼락 치어의 영양학적 특성 비교)

  • Cultured juvenile black rockfish, Sebastes schiegeli as a seed for sen ranching project were compared with the similar size of wild fish. Wild fish were analyzed and compared with cultured fish in body indices, and moisture, crude protein, lipid, ash, many kinds of amino acids and various minerals in the whole-body of juvenile rockfish from three different populations. There were significant (P<0.05) differences in the moisture, crude lipid, and ash in the whole-body of fish among three different populations; the moisture content of wild fish was higher than that of both cultured fish. The lipid content in wild and tank cultured fish was lowest and highest, respectively; The lipid content of fish cultured in embanked system was intermediate values. The condition factor (CE), hepatosomatic index (HSI), and intraperitoneal fat (IPE) of fish cultured in tank system and embanked system were significantly (P<0.05) higher than those of wild fish. But fish cultured in tank system produced similar intestinosomatic index (ISI) and muscle ratio (MR) values as the wild fish. There were significant differences in asparatic acid, glutamic acid, isoleucine, leucine, lysine, phenylalanine, threonine, valine of whole-body fish cultured by different methods; the wild fish was highest in these amino acids, followed by fish cultured in embanked system and then fish cultured in tank system. There were also significant differences in Ca, P, Mg, Mn, Zn of whole-body fish cultured by different methods: wild fish and fish cultured in tank system produced highest in Ca, P, Mg, Zn and lowest in Mn, respectively. There were significant differences in moisture, crude lipid, ash, many kinds of amino acids, several minerals, CE, HSI, UE, and ISI among the three juvenile populations; whole-body of wild fish showed higher in moisture, ash, asparatic acid, glutamic acid, isoleucine, leucine, Iysine, phenylalanine, threonine, valine, Ca, P, Mg, and Zn than fish cultured in embanked system, while they showed lower in the rest of the nutritious parameters studied.

Difference of Components Changes in Salt-Fermented Anchovy, Engraulis japonicus Sauce by Tank Size during Fermentation (숙성 발효조크기에 따른 멸치액젓의 성분비교)

  • To investigate difference of components changes in salt-fermented anchovy, Engaulis japonicus sauce during 18 months fermentation by tank size, various chemical properties were examined at 2$\~$3 months intervals. The contents of total and amino nitrogen, total ATP related compounds increased gradually during 18 months of fermentation, and showed higher content in salt-fermented anchor sauce produced by large tank scale (LTS) product than those of small tank scale (STS) product during fermentation. Hypoxanthine and uric acid were the most abundant in ATP related compounds, ranging from $81.1\%$ to $90.4\%$, The cross point of inosine (HxR) + hypoxanthine (Hx) and uric acid was faster in LTS with 10.3 months fermentation than in STS with 12.6 months fermentation. After 18month of fermentation, the LTS was rich in free amino acids, such as glutamic acid, alanine, aspartic acid, valine, Iysine in that order. On the other hand, the STS was rich in free amino acids, glutamic acid, aspartic acid, alanine, vsine, valine in that order. Absorbance at 453 nm were higher in STS than in STS, but was no difference the rate of increase during fermentation.

Quality Changes of a Fully Ripe Korean Native Pumpkin, Yangsan, during Long-term Storage, and High Temperature and Pressure Treatment (장기저장 및 고온고압 처리에 따른 한국재래종 호박 '양산'의 품질변화)

  • We have studied quality changes of fully ripe fruit of Korean native pumpkin 'Yangsan' regarding the following parameters: pH, sugar content, weight, water content, contents of crude protein and amino acids during 60 days storage at room temperature. As the results, there was no changes in sugar contents according to the storage period, but pH was changing to a little acidic direction with slight decrease in weight and water content. Contents of total crude proteins and comprising amino acids were increased during the storage period. The main contents of amino acids of the Korean native pumpkin, Yangsan, were glutamic acid (15.5%), aspartic acid (10.1%), lysine (8.7%), valine (7.5%), leucine (7.1%) and alanine (6.6%), which were not highly influenced during storage period. Additionally we have investigated the content of free amino acids and color changes during processing of Yangsan under high temperature at $121^{\circ}C$ and high pressure at $1\;kg/cm^2$. In fully ripe fruits, a total of 29 kinds of free amino acids were detected including 8 kinds of essential amino acids (histidine, isoleucine, leucine, lysine, phenylalanine, methionine, threonine and valine). More than 35% of total free amino acids were aspartic acid (20.3%) and asparagine (15.4%); ornithine, citrullin, and arginine, which are related to Ornithine cycle, were also detected in fully ripe fruits. But when treated with high temperature and high pressure, glutamic acid and arginine were decreased rapidly whereas ammnonium chloride was relatively increased. Moreover "b" value as yellow color indicator was decreased from 17.45 to 9.14 while treated for 60 minutes with high temperature and pressure, caused by the degradation of ${\beta}-carotene$ and other yellowish pigments in Yangsan.

A Study on Flavorous Taste Components in Kimchis -On Free Amino Acids- (김치의 맛 성분(成分)에 관(關)한 연구(硏究) -유리(遊離) 아미노산(酸)에 관(關)하여 -)

  • Free amino acids and total free sugar of kimchi were identified. Free amino acids of kimchis were extracted by 80% ethanol and isolated by ion exchange chromatography. Identification and quantitative determination of individual free amino acids were performed by amino acid autoanalyzer. Free sugar of kimchis was extracted by aqueous ethanol and isolated by ion exchange chromatography. Quantitative determination of it was perfermed by spectrophotometer. The results are summarized as follows: 1. Lysine, histidine, arginine, tryptophan, aspartic acid, threonine, glutamic acid, proline, glycine, alanine, cysteine, valine, methionine, leucine, isoleucine, tyrosine and phenylalanine were found in all kimchis. 2. The change of free amino acid composition during fermentation of kimchis was not observed, but the amount of total free amino acids of fermented kimchi decreased as compared with those of raw kimchi. 3. In kimchi containing 10 ml of fermented anchovy solution/100 g of chinese cabbage, the amount of total free amino acids was more than that of fermented salt kimchi and the characteristic flavor of it was attributed to such amino acids as lysine, aspartic acid, glutamic acid, valine, methionine, isoleucine and leucine. 4. Large amount of free sugar in raw salt kimchi decreased during fermentation, but, after fermentation, significant difference of free sugar content between salt kimchi and kimchi containing fermented anchovy solution was not observed.

Biosynthesis of $\beta$-Lactam Antibiotics by Cell-free Extract from Lysobacter lactamgenus

  • Using cell-free extract of Lysobacter lactamgenus, enzymatic conversion of $\delta$-L-($\alpha$-aminoadiphyl)-L-cysteinyl-D-valine (ACV) the first substrate of $\beta$-lactam biosynthesis, into antibiotic compounds was attempted. In high performance liquid chromatographic (HPLC) analysis, the biosynthetic intermediates for cephalosporin antibiotics including isopenicillin N, deacetoxycephalosporin C, deacetylcephalosporin C and unknown cephem compound were detected in reaction mixtures. It implies that cephabacin compounds from L lactamgenus could be produced by biosynthetic routes through penicillin ring formation and its expansion to cephalosporin ring, likely as cephalosporin C from Cephalosporium or cephamycin C from Streptomyces. Among biosynthetic enzyme in cell-free extract, the ring formation activity (isopenicillin N synthetase activity) was separated in 50-60% of ammonium sulfate fraction, and ring expansion activity (deacetoxycephalosporin C synthetase activity) was found to be in 40-50% fraction. The partially purified isopenicillin N synthetase could convert as much as 90% ACV to isopenicillin N during 6-hour reaction.

Biological Activities and Partial Characterization of Beauveria bassiana Mycelium

  • Some biological activities of Beauveria bassiana were studied to elucidate pharmacological function of B. bassiana-infected larva of the silkworm. The mycelium consisted mainly of carbohydrate (65.8%), followed by protein (15.9%) and fat (8.3%). Glucose (68.8%), mannose (7.1%), and galactose (6.1%) were major components in carbohydrates. Ten amino acids including glutamine, threonine, valine, aspartic acid, alanine, leucine, serine, glycine, arginine, and isoleucine were found in protein as major amino acids. Various extracts were prepared from the freeze-dried mycelium of B. bassiana by systemic extraction and their biological activities were investigated. Among tested fractions, the hot-water extract (HW) contributed significantly to the anti-coagulant activity, anti-complementary activity, and stimulation of intestinal immune system. The methanol extract (ME) increased acetylcholinesterase (AChE) inhibition activity and reactive oxygen species (ROS) scavenging activity.
