펜톤 산화에 의한 하수 슬러지로부터 유기산 생성에 관한 연구 (A Study on the Production of VFAs from Sewage Sludge by Fenton's Oxidation)

  • 하수 슬러지를 펜톤 산화공정으로 처리하는 새로운 VFAs 생산 공정을 연구하였다. 과산화수소와 촉매인 $Fe^{2+}$의 적정농도를 실험하고, 최적 반응온도 및 반응시간에 대한 기초 자료를 얻고자 하였다. 기존의 과산화수소를 액상산화제로 사용한 공정보다 저농도의 $Fe^{2+}$를 촉매로 사용하는 펜톤 산화공정으로 과산화수소의 사용량을 대폭 감소시키고도 VFAs 생성량이 4배 정도 증가하는 결과를 얻었다. 과산화수소와 $Fe^{2+}$의 적정 농도는 각각 0.62 M과 0.007M 이었다. VFAs 생성반응은 초기 1분 이내에서 빠르게 진행되었으며 생성된 VFAs의 일부가 초산과 $CO_2$로 분해되는 연속반응의 특성이 나타났다. 경제성을 감안하면 반응온도는 $25^{\circ}C$, 반응시간은 10분 정도가 적절한 반응조건으로 생각된다. 초기 pH $3{\sim}6.3$에서 VFAs의 생성에 미치는 pH의 영향은 관찰되지 않았다.

Interactions between Entodinium caudatum and an amino acid-fermenting bacterial consortium: fermentation characteristics and protozoal population in vitro

  • Ruminal protozoa, especially entodiniomorphs, engulf other members of the rumen microbiome in large numbers; and they release oligopeptides and amino acids, which can be fermented to ammonia and volatile fatty acids (VFAs) by amino acid-fermenting bacteria (AAFB). Studies using defaunated (protozoa-free) sheep have demonstrated that ruminal protozoa considerably increase intraruminal nitrogen recycling but decrease nitrogen utilization efficiency in ruminants. However, direct interactions between ruminal protozoa and AAFB have not been demonstrated because of their inability to establish axenic cultures of any ruminal protozoan. Thus, this study was performed to evaluate the interaction between Entodinium caudatum, which is the most predominant rumen ciliate species, and an AAFB consortium in terms of feed degradation and ammonia production along with the microbial population shift of select bacterial species (Prevotella ruminicola, Clostridium aminophilum, and Peptostreptococcus anaerobius). From an Ent. caudatum culture that had been maintained by daily feeding and transfers every 3 or 4 days, the bacteria and methanogens loosely associated with Ent. caudatum cells were removed by filtration and washing. An AAFB consortium was established by repeated transfers and enrichment with casamino acids as the sole substrate. The cultures of Ent. caudatum alone (Ec) and AAFB alone (AAFB) and the co-culture of Ent. caudatum and AAFB (Ec + AAFB) were set up in three replicates and incubated at 39℃ for 72 h. The digestibility of dry matter (DM) and fiber (NDF), VFA profiles, ammonia concentrations, pH, and microscopic counts of Ent. caudatum were compared among the three cultures. The co-culture of AAFB and Ent. caudatum enhanced DM degradation, VFA production, and Ent. caudatum cell counts; conversely, it decreased acetate: propionate ratio although the total bacterial abundance was similar between Ec and the Ec + AAFB co-culture after 24 h incubation. The ammonia production and relative abundance of C. aminophilum and P. anaerobius did not differ between AAFB alone and the Ec + AAFB co-culture. Our results indicate that Ent. caudatum and AAFB could have a mutualistic interaction that benefited each other, but their interactions were complex and might not increase ammoniagenesis. Further research should examine how such interactions affect the population dynamics of AAFB.

Influence of Yeast Fermented Cassava Chip Protein (YEFECAP) and Roughage to Concentrate Ratio on Ruminal Fermentation and Microorganisms Using In vitro Gas Production Technique

  • The objective of this study was to determine the effects of protein sources and roughage (R) to concentrate (C) ratio on in vitro fermentation parameters using a gas production technique. The experimental design was a $2{\times}5$ factorial arrangement in a completely randomized design (CRD). Factor A was 2 levels of protein sources yeast fermented cassava chip protein (YEFECAP) and soybean meal (SBM) and factor B was 5 levels of roughage to concentrate (R:C) ratio at 80:20, 60:40, 40:60, 20:80, and 0:100, respectively. Rice straw was used as a roughage source. It was found that gas production from the insoluble fraction (b) of YEFECAP supplemented group was significantly higher (p<0.05) than those in SBM supplemented group. Moreover, the intercept value (a), gas production from the insoluble fraction (b), gas production rate constants for the insoluble fraction (c), potential extent of gas production (a+b) and cumulative gas production at 96 h were influenced (p<0.01) by R:C ratio. In addition, protein source had no effect (p>0.05) on ether in vitro digestibility of dry matter (IVDMD) and organic (IVOMD) while R:C ratio affected the IVDMD and IVOMD (p<0.01). Moreover, YEFECAP supplanted group showed a significantly increased (p<0.05) total VFA and $C_3$ while $C_2$, $C_2:C_3$ and $CH_4$ production were decreased when compared with SBM supplemented group. In addition, a decreasing R:C ratio had a significant effect (p<0.05) on increasing total VFA, $C_3$ and $NH_3$-N, but decreasing the $C_2$, $C_2:C_3$ and CH4 production (p<0.01). Furthermore, total bacteria, Fibrobacter succinogenes, Ruminococcus flavefaciens and Ruminococcus albus populations in YEFECAP supplemented group were significantly higher (p<0.05) than those in the SBM supplemented group while fungal zoospores, methanogens and protozoal population remained unchanged (p>0.05) as compared between the two sources of protein. Moreover, fungal zoospores and total bacteria population were significantly increased (p<0.01) while, F. succinogenes, R. flavefaciens, R. albus, methanogens and protozoal population were decreased (p<0.01) with decreasing R:C ratio. In conclusion, YEFECAP has a potential for use as a protein source for improving rumen fermentation efficiency in ruminants.

매립지 주입을 위한 음폐수 산발효 시 반응기 형태와 체류시간에 따른 특성 (Characteristics of Anaerobic Acid Fermentation with Food waste leachate by Reactor Type of Retention Time for Landfill Site Injection)

  • 본 연구에서는 매립가스 증대를 위한 음폐수의 원활한 주입을 위해 혐기성 산발효 전처리를 수행하였으며 이를 통해 점도 감소와 유기산 생산량을 확인하여 매립지 주입을 위한 최적조건을 확인하고자 하였다. 산발효 후 음폐수 가용화율은 약 15% 증가함을 보였고 체류시간의 변화와 반응기형태를 달리 한 결과 큰 차이를 보이지는 않았다. 반응기형태에 따라 점도변화를 확인한 결과 상향류식 반응기에서 $76.95{\pm}3.27%$로 완전혼합반응기에 비해 약 11.38% 높은 점도 저감효율을 보였으며, VFA생산에서는 체류시간을 3일에서 5일로 증가 시 2.01배(상향류식 반응기), 1.76배(완전혼합반응기) 높은 경향을 보였다. 이는 상향류식 반응기의 경우 고정층담체의 스크린 역할로 분자량이 작은 물질에 비해 큰 물질들이 상대적으로 반응기에 체류하는 시간이 길어져 효율이 높은 것으로 사료된다.

Effects of Monensin and Fish Oil on Conjugated Linoleic Acid Production by Rumen Microbes in Holstein Cows Fed Diets Supplemented with Soybean Oil and Sodium Bicarbonate

  • The present study was conducted with four ruminally canulated Holstein cows to observe the effects of monensin or fish oil on diet fermentation and production of conjugated linoleic acids (CLAs) in the rumen when fed diets supplemented with soybean oil and sodium bicarbonate. Cows of the control treatment were fed a basal diet (CON) consisting of 60% commercial concentrate and 40% chopped rye grass hay. Cows of other treatments were fed the same diet as CON, but the concentrate was supplemented with 7% of soybean oil and 0.5% of sodium bicarbonate (SO-B), SO-B supplemented with monensin (30 ppm, SO-BM) or concentrate supplemented with 6.3% of soybean oil, 0.5% of sodium-bicarbonate, 30 ppm of monensin and 0.7% of fish oil (SO-BMF). Dry matter (DM) intake of the cows was significantly (p<0.011) reduced by feeding the SO-BMF diet compared to the other diets which did not differ in DM intake. Whole tract digestibility of major dietary components was significantly (p<0.004-0.027) higher for SO-BMF than the other supplement-containing diets. Dietary supplements did not clearly affect rumen pH and ammonia concentrations compared to the CON diet. Significantly reduced (p<0.05) total VFA concentration was obtained by the addition of fish oil to the diet (SO-BMF) compared to other diets. No differences, however, were obtained in major VFA proportions as well as in total VFA between the supplemented diets. The SO-BM diet increased (p<0.01-0.05) the concentrations of trans-11 $C_{18:1}$ and linoleic acid in rumen fluid. Total CLA concentration was also increased by the feeding of SO-B and SO-BM diets during early fermentation times (up to 3 h) post-feeding. Cis-9, trans-11 CLA concentration in rumen fluid was highest (p<0.05) for SO-B up to 1 h while the highest (p<0.01) value for SO-BM occurred at 3 h post-feeding. An increased trans-10, cis-12 CLA concentration was obtained from the SO-B and SO-BM diets at 1 and 3 h post feeding compared to the other diets. Supplementation of oils with monensin and sodium bicarbonate increased (p<0.05) the proportions of $C_{18:1}$ and CLA in the plasma of cows, but the effect of monensin and/or fish oil was limited to trans-10, cis-12 CLA.

Effects of Different Dietary Levels of Mannanoligosaccharide on Growth Performance and Gut Development of Broiler Chickens

  • Different levels of dietary mannanoligosaccharide (Bio-MOS, Alltech Inc.) were evaluated for their efficacy on performance and gut development of broiler chickens during a 6-week experimental period. Experimental diets contained (g MOS/kg diet) a low (0.5 g during the entire period), medium (1 g during the entire period), high (2 g during the entire period), or step down (2 g in the first week; 1 g in the second and third week; 0.5 g in the last three weeks) level of MOS. Control diets included a negative and a positive control (zinc bacitracin, ZnB, 50 ppm and 30 ppm in the first and last three weeks, respectively). MOS supplementation improved the growth performance of young birds and the effects became less when the birds got older. The growth response of birds was more obvious at the high dosage level of MOS treatment than the other MOS treatments and the growth performance of birds fed on the high MOS diet was comparable to that of birds fed on the ZnB diet. Depending on the dosage level and the age of birds, MOS seemed to reduce the size of the liver and the relative length of the small intestine but did not affect the relative weight of the other visceral organs (proventriculus, gizzard, pancreas, bursa and spleen) and that of the small intestine. A numerical increase in the small intestine digestibility of nutrients was noticed in the young birds fed on the MOS diet(s), but not in the older ones. Medium and/or high MOS treatment also increased the villus height of the small intestine of birds at different ages. Similar results were observed on the ZnB treatment. However, MOS and ZnB affected caecal VFA profile in different ways. MOS increased, or tended to increase, whereas ZnB reduced individual VFA concentrations in the caeca.

Influence of Dietary Salinomycin on Feeding-induced Variations of Glucose Kinetics and Blood Volatile Fatty Acids and Insulin Concentrations in Sheep Fed a High-roughage Diet

  • This study was conducted to determine effects of salinomycin (SL) on feeding-induced changes in glucose kinetics and blood VFA concentrations in sheep fed a high-roughage diet. Four sheep were fed the diet with or without 20 mg/kg diet of SL once daily for 21 d. Glucose entry and utilization rates were determined during the prefeeding and 3 h postfeeding periods, using a [$^{13}C_6$]glucose dilution method and non-steady state equations. Ruminal characteristics and concentrations of blood VFA, plasma glucose and insulin were also measured during the same periods. A feeding-induced increase in ruminal total VFA concentration tended to be inhibited (p<0.10) with SL, although ruminal pH was unaffected (p>0.10) with SL or by feeding. Salinomycin decreased (p<0.05) acetate proportion and increased (p<0.05) propionate proportion in the rumen, but did not modify these changes in response to feeding (p>0.10). A feeding-induced increase in blood acetate concentration was attenuated (p<0.05) with SL. Salinomycin tended to increase (p<0.10) blood propionate concentration without modifying its response patterns to feeding (p>0.10). Plasma concentrations of glucose or insulin were unaffected (p>0.10) with SL. Salinomycin tended to enhance (p<0.10) glucose entry and utilization rates. Feeding also enhanced (p<0.01) both rates, whereas their interactive effect was not detected (p>0.10). We conclude that SL possibly enhances whole body glucose entry and utilization with an increase in blood propionate concentration in sheep given a high-roughage diet, although SL does not appear to affect their responses to feeding.

Comparison of Faecal Microbial Community of Lantang, Bama, Erhualian, Meishan, Xiaomeishan, Duroc, Landrace, and Yorkshire Sows

  • The objective of this study was to investigate differences in the faecal microbial composition among Lantang, Bama, Erhualian, Meishan, Xiaomeishan, Duroc, Landrace, and Yorkshire sows and to explore the possible link of the pig breed with the gut microbial community. Among the sows, the Meishan, Landrace, Duroc, and Yorkshire sows were from the same breeding farm with the same feed. Fresh faeces were collected from three sows of each purebred breed for microbiota analysis and volatile fatty acid (VFA) determination. Denaturing gradient gel electrophoresis (DGGE) analysis revealed that samples from Bama, Erhualian, and Xiaomeishan sows, which from different farms, were generally grouped in one cluster, with similarity higher than 67.2%, and those from Duroc, Landrace, and Yorkshire sows were grouped in another cluster. Principal component analysis of the DGGE profile showed that samples from the foreign breeds and the samples from the Chinese indigenous breeds were scattered in two different groups, irrespective of the farm origin. Faecal VFA concentrations were significantly affected by the pig breed. The proportion of acetate was higher in the Bama sows than in the other breeds. The real-time PCR analysis showed that 16S rRNA gene copies of total bacteria, Firmicutes and Bacteroidetes were significantly higher in the Bama sows compared to Xiaomeishan and Duroc sows. Both Meishan and Erhualian sows had higher numbers of total bacteria, Firmicutes, Bacteroidetes and sulphate-reducing bacteria as compared to Duroc sows. The results suggest that the pig breed affects the composition of gut microbiota. The microbial composition is different with different breeds, especially between overseas breeds (lean type) and Chinese breeds (relatively obese type).

Changes in the ruminal fermentation and bacterial community structure by a sudden change to a high-concentrate diet in Korean domestic ruminants

  • Objective: To investigate changes in rumen fermentation characteristics and bacterial community by a sudden change to a high concentrate diet (HC) in Korean domestic ruminants. Methods: Major Korean domestic ruminants (each of four Hanwoo cows; $545.5{\pm}33.6kg$, Holstein cows; $516.3{\pm}42.7kg$, and Korean native goats; $19.1{\pm}1.4kg$) were used in this experiment. They were housed individually and were fed ad libitum with a same TMR (800 g/kg timothy hay and 200 g/kg concentrate mix) twice daily. After two-week feeding, only the concentrate mix was offered for one week in order to induce rapid rumen acidosis. The rumen fluid was collected from each animals twice (on week 2 and week 3) at 2 h after morning feeding using an oral stomach tube. Each collected rumen fluid was analyzed for pH, volatile fatty acid (VFA), and $NH_3-N$. In addition, differences in microbial community among ruminant species and between normal and an acidosis condition were assessed using two culture-independent 16S polymerase chain reaction (PCR)-based techniques (terminal restriction fragment length polymorphism and quantitative real-time PCR). Results: The HC decreased ruminal pH and altered relative concentrations of ruminal VFA (p<0.01). Total VFA concentration increased in Holstein cows only (p<0.01). Terminal restriction fragment length polymorphism and real-time quantitative PCR analysis using culture-independent 16S PCR-based techniques, revealed rumen bacterial diversity differed by species but not by HC (p<0.01); bacterial diversity was higher in Korean native goats than that in Holstein cows. HC changed the relative populations of rumen bacterial species. Specifically, the abundance of Fibrobacter succinogenes was decreased while Lactobacillus spp. and Megasphaera elsdenii were increased (p<0.01). Conclusion: The HC altered the relative populations, but not diversity, of the ruminal bacterial community, which differed by ruminant species.
