Low-Voltage Activated $Ca^{2+}$ Current Carried via T-Type Channels in the Mouse Egg

  • Most of voltage operated $Ca^{2+}$ channels can be divided into three types (T-, N-, and L-type), according to the electrical and pharmacological properties. Their distribution is closely related to cell specific functions. Properties of the voltage activated $Ca^{2+}$ current in mouse eggs were examined to classify channel types and to deduce the function by using whole cell voltage clamp technique. $Ca^{2+}$ currents appeared below -40 mV and reached a maximum at -15 mV (half maximum was -31 mV), then decayed rapidly (inactivation time constant ${\tau}=28.2{\pm}9.59$ ms at -10 mV within 50 ms after the onset of step depolarization. Activation and inactivation of the $Ca^{2+}$ channel was steeply dependent on voltage, in a relatively low range of $-70\;mV{\sim}-10 mV,$ half maximum of activation was -31 mV and that of inactivation was -39 mV, respectively. This current was not decreased significantly by nifedipine, a specific dihydropyridine $Ca^{2+}$ channel blocker in the range of $1\;{\mu}M\;to\;100{\mu}M.$ The inhibitory effect of $Ni^{2+}\;on\;Ca^{2+}$ current was greater than that of $Cd^{2+}.$ The conductance of $Ba^{2+}$ through the channel was equal to or lower than that of $Ca^{2+}$ These results implied that $Ca^{2+}$ current activated at a lower voltage in the mouse egg is carried via a $Ca^{2+}$ channel with similar properties that of the T-type channel.

1H, 15N and 13C Backbone Assignments and Secondary Structures of C-ter100 Domain of Vibrio Extracellular Metalloprotease Derived from Vibrio vulnificus

  • Vibrio extracellular metalloprotease (vEP), secreted from Vibrio vulnificus, shows various proteolytic function such as prothrombin activation and fibrinolytic activities. Premature form of vEP has an N-terminal (nPP) and a C-terminal (C-ter100) region. The nPP and C-ter100 regions are autocleaved for the matured metalloprotease activity. It has been proposed that two regions play a key role in regulating enzymatic activity of vEP. Especially, C-ter100 has a regulatory function on proteolytic activity of vEP. C-ter100 domain has been cloned into the E. coli expression vectors, pET32a and pGEX 4T-1 with TEV protease cleavage site and purified using gel-filtration chromatography followed by affinity chromatography. To understand how C-ter100 modulates proteolytic activity of vEP, structural studies were performed by heteronuclar multi-dimensional NMR spectroscopy. Backbone $^1H$, $^{15}N$ and $^{13}C$ resonances were assigned by data from standard triple resonance and HCCH-TOCSY experiments. The secondary structures of vEP C-ter100 were determined by TALOS+ and CSI software based on hydrogen/deuterium exchange. NMR data show that C-ter100 of vEP forms a ${\beta}$-barrel structure consisting of eight ${\beta}$-strands.

Preparation and oxygen control of Ti-6Al-4V alloys by recycling dental pure Ti scraps (치과용(齒科用) 순(純) 타이타늄 스크랩을 재활용(再活用)한 Ti-6Al-4V 합금(合金)의 제조(製造) 및 산소(酸素) 제어(制御))

  • The Ti-6Al-4V alloys were prepared by recycling of dental Ti pure scraps using vacuum arc melting process, and their physical properties were evaluated the Ti-6Al-4V alloys with different oxygen concentrations. For the preparation of Ti-6Al-4V alloys, Ti pure scraps used for dental implant were utilized as a raw material, and their different oxygen concentrations were ranged from G1 to G4 grade in ASTM standards. It was confirmed that the weight loss of Al in the composition of Ti-6Al-4V alloy could be controlled under the Ar pressure of 875 torr during the melting of alloy. The oxygen concentrations of the Ti-6Al-4V alloys were ranged from 1170 to 3340 ppm. The vickers hardness change of the Ti-6Al-4V alloys showed a similar behavior with that of pure Ti. As a result, we confirmed a possibility of preparation of Ti-6Al-4V alloy by recycling of dental Ti scraps using vacuum arc melting process in this study.

By applying the Nehari manifold method and the concentration-compactness principle, we obtain the existence and concentration of ground state solutions when the parameter ${\gamma}$ is sufficiently large.

Location and Adsorbate Interactions of V(IV) Species in VH-SAPO-34 Studied by EPR and Electron Spin-Echo Modulation Spectroscopies

  • Vanadium-doped H-SAPO-34 samples were prepared by a high-temperature solid-state reaction between SAPO-34 and the paramagnetic V(Ⅳ) species and characterized carefully by EPR and Electron Spin-Echo Modulation(ESEM) studies. The paramagnetic vanadium species generated in both V$_2$O$\_$5/ and VOSO$\_$4/ of SAPO-34 have the same narrow range of g value fur vanadium species assigned to VO$\^$2+/ inferred from the isotropic EPR spectrum at 293 K. The EPR and ESEM data indicate that the V(Ⅳ) species exist as a vanadyl ion either as [V(Ⅳ)]O$\^$2+/ or V$\^$4+/. The [V(Ⅳ)]O$\^$2+/ species seems to be more probable because SAPO-34 having a low negative framework charged and more positively charged species like V$\^$4+/can not be easily stabilized. Tetravalent vanadium ion in vadium-doped H- SAPO-34 can only be observed at the temperature lower than 77 K, while the vanadyl ion, VO$\^$2+/in the activated sample of VH-SAPO-34 can produce the ion even at room temperature. After the adsorption of methanol, ethanol, propanol or ethene to the VH-SAPO-34, only one molecule coordinate to [V(Ⅳ)]O$\^$2+/ was observed in EPR and ESEM spectra.

  • For $0<p{\leq}{\infty}$ and a convex body $K$ in $\mathbb{R}^n$, Lutwak, Yang and Zhang defined the concept of dual $L_p$-centroid body ${\Gamma}_{-p}K$ and $L_p$-John ellipsoid $E_pK$. In this paper, we prove the following two results: (i) For any origin-symmetric convex body $K$, there exist an ellipsoid $E$ and a parallelotope $P$ such that for $1{\leq}p{\leq}2$ and $0<q{\leq}{\infty}$, $E_qE{\supseteq}{\Gamma}_{-p}K{\supseteq}(nc_{n-2,p})^{-\frac{1}{p}}E_qP$ and $V(E)=V(K)=V(P)$; For $2{\leq}p{\leq}{\infty}$ and $0<q{\leq}{\infty}$, $2^{-1}{\omega_n}^{\frac{1}{n}}E_qE{\subseteq}{\Gamma}_{-p}K{\subseteq}{2\omega_n}^{-\frac{1}{n}}(nc_{n-2,p})^{-\frac{1}{p}}E_qP$ and $V(E)=V(K)=V(P)$. (ii) For any convex body $K$ whose John point is at the origin, there exists a simplex $T$ such that for $1{\leq}p{\leq}{\infty}$ and $0<q{\leq}{\infty}$, ${\alpha}n(nc_{n-2,p})^{-\frac{1}{p}}E_qT{\supseteq}{\Gamma}_{-p}K{\supseteq}(nc_{n-2,p})^{-\frac{1}{p}}E_qT$ and $V(K)=V(T)$.

  • VANET technologies provide real-time traffic information for mitigating traffic jam and preventing traffic accidents, as well as in-vehicle infotainment service through Telematics/Intelligent Transportation System (ITS). Due to the rapid increasement of various requirements, the vehicle communication with a limited resource and the fixed frame architecture of the conventional techniques is limited to provide an efficient communication service. Therefore, a new flexible operation depending on the surrounding situation information is required that needs an adaptive design of the network architecture and protocol for efficiently predicting, distributing and sharing the context-aware information. In this paper, Vehicle-to-Infrastructure (V2I) based on communication between vehicle and a Road Side Units (RSU) and Vehicle-to-Vehicle (V2V) based on communication between vehicles are effectively combined in a new MAC architecture and V2I and V2V vehicles collaborate in management. As a result, many vehicles and RSU can use more efficiently the resource and send data rapidly. The simulation results show that the proposed method can achieve high resource utilization in accordance. Also we can find out the optimal transmission relay time and 2nd relay vehicle selection probability value to spread out V2V/V2I collaborative schedule message rapidly.

pH-mediated Regulation of Pacemaker Activity in Cultured Interstitial Cells of Cajal

  • Interstitial cells of Cajal (ICCs) are pacemakers in gastrointestinal tracts, regulating rhythmicity by activating nonselective cation channels (NSCCs). In the present study, we investigated the general characteristics and pH-mediated regulation of pacemaker activity in cultured interstitial cells of Cajal. Under voltage clamp mode and at the holding potential of -60 mV, the I-V relationships and difference current showed that there was no reversal potential and voltage-independent inward current. Also, when the holding potentials were changed from +20 mV to -80 mV with intervals of 20 mV, there was little difference in inward current. In pacemaker activity, the resting membrane potential (RMP) was depolarized (In pH 5.5, $23{\pm}1.5$ mV depolarized) and the amplitude was decreased by a decrease of the extracellular pH. However, in case of increase of extracellular pH, the RMP was slightly hyperpolarized and the amplitude was decreased a little. The melastatin type transient receptor potential (TRPM) channel 7 has been suggested to be required for intestinal pacemaking activity. TRPM7 produced large outward currents and small inward currents by voltage ramps, ranging from +100 to -100 mV from a holding potential of -60 mV. The inward current of TRPM7 was dramatically increased by a decrease in the extracellular pH. At pH 4.0, the average inward current amplitude measured at -100 mV was increased by about 7 fold, compared with the current amplitude at pH 7.4. Changes in the outward current (measured at +100 mV) were much smaller than those of the inward current. These results indicate that the resting membrane potential of pacemaking activity might be depolarized by external acidic pH through TRPM7 that is required for intestinal pacemaking activity.

Asn-Linked Glycosylation Contributes to Surface Expression and Voltage-Dependent Gating of Cav1.2 Ca2+ Channel

  • The Cav1.2 Ca2+ channel is essential for cardiac and smooth muscle contractility and many physiological functions. We mutated single, double, and quadruple sites of the four potential Asn (N)-glycosylation sites in the rabbit Cav1.2 into Gln (Q) to explore the effects of Nglycosylation. When a single mutant (N124Q, N299Q, N1359Q, or N1410Q) or Cav1.2/WT was expressed in Xenopus oocytes, the biophysical properties of single mutants were not significantly different from Cav1.2/WT. In comparison, the double mutant N124,299Q showed a positive shift in voltage-dependent gating. Furthermore, the quadruple mutant (QM; N124,299,1359,1410Q) showed a positive shift in voltage-dependent gating as well as a reduction of current. We tagged EGFP to the QM, double mutants, and Cav1.2/WT to chase the mechanisms underlying the reduced currents of QM. The surface fluorescence intensity of QM was weaker than that of Cav1.2/WT, suggesting that the reduced current of QM arises from its lower surface expression than Cav1.2/WT. Tunicamycin treatment of oocytes expressing Cav1.2/WT mimicked the effects of the quadruple mutations. These findings suggest that Nglycosylation contributes to the surface expression and voltage-dependent gating of Cav1.2.
