Vibrio harveyi, one of the major causal agent of vibriosis, affects a diverse range of marine vertebrates and invertebrates over a wide geographical area. The organism is synonymous with Vibrio carchariae, which is also known as a fish pathogen. The aims of this study were to investigate the characteristics of the pathogenic non-luminous V. harveyi and the luminous V. harveyi. And V. harveyi isolates were examined the pathogenicity to the black rockfish, Sebastes schlegeli. Both strains of V. harveyi showed haemolytic activity, and the survival rate of non-luminous V. harveyi FR 2 was higher than other strains in the skin, gut mucus and fresh serum of olive flounder, Paralichthys olivaceus and black rockfish, Sebastes schlegeli, respectively. The virulence of non-luminous V. harveyi FR 2 was higher than that of luminous V. harveyi VIB 391 in the intraperitoneally infected black rockfish. In conclusion, the present study revealed that the pathogenicity of V. harveyi FR 2 isolated from marine fish was higher than that of V. harveyi VIB 391 isolated from shrimp for black rockfish. It was suggested that the pathogenicity of V. harveyi on the black rockfish was related with bacterial luminescense.
Vibrio harveyi was a significant pathogenic agent and cause a high mortality of cultured fish and shrimp in the aquaculture industry. In this study, we have investigated biochemical, physiological, serological and immunological characteristics of V. harveyi isolated from marine cultured fish. The phenotypes of V. harveyi were differentiated with their own biochemical characteristics and colors of colony on the TCBS agar. V. harveyi were classified into more than four serogroups by agglutination test. Most isolates were classified into a group A which is categorized with the same band pattern generated by western blotting. Group A was characterized by a major protein, which is ranged from 26 and 34 kDa in size, and has a virulence to oliver flounder more than reference strain KCCM40866. Oliver flounders vaccined with FKC of V. harveyi C05011 were highly resistant to infection by other strains of group A.
In 1999, Vibrio harveyi infection occurred among cultured marine finfishes including olive flounder (Paralichthys olivaceus), black rockfish (Sebastes schlegeli), and turbot (Scophtalmus maximus) in the province of Gyeongsang, Korea. We examined the various microbiological characteristics of this V. harveyi strains. V. harveyi was grown well in the 3% NaCl at 30℃. The swarming activity appeared to be an one of the characteristic properties of the V. harveyi was the highest in the 2% NaCl concentration at TSA medium. These cells were elongated and had the several lateral flagella which is not waved and shorter than polar flagella. It was not luminescent in the all isolated strains. Whole cell of the V. harveyi had a major protein of 50 kDa and presented various band around of 40 kDa in strains.

The aims of this study were to investigate the characteristics of extracellular products (ECPs) from an isolate of the pathogenic non‐luminous V. harveyi and the luminous V. harveyi. And ECPs of V. harveyi were examined the pathogenicity to the black rockfish, Sebastes schlegeli, and histopathological traits of internal organs injected by ECPs. Four strains of V. harveyi cultures produced ECPs showing various enzymatic activites (caseinase, gelatinase, phospholipase, lipase, haemolysin). The ECPs showed strong cytotoxicity on macrophages of black rockfish, Sebastes schlegeli and olive flounder, Paralichthys olivaceus. Especially, the virulence of ECPs from the isolate of V. harveyi FR 2 was higher in the intraperitoneally injected black rockfish. Also, the ECPs of V. harveyi FR 2 caused the expansion of sinusoids in the liver, the activation of ellipsoid in the spleen and the sloughing of the epidermal cell in the intestine. It was suggested that the ECPs from V. harveyi play an important role in the pathogenicity process of the V. harveyi.

This study was conducted to select the media for the formalin killed vaccine (FKC) production of Vibrio harveyi and its application for olive flounder, Paralichthys olivaceus. For this, we have investigated the immune effects of Vibrio harveyi FKC vaccines grown in 3 different media i.e. Tryptic Soy Broth (TSB), TSB containing 2-2'-dipyridyl (TSB-D), Brain Heart Infusion Broth (BHIB) on the production of agglutinating antibody and protection against Vibrio harveyi in olive flounder. Additionally, a dual vaccine was prepared by combining Streptococcus parauberis vaccine to V. harveyi vaccine and its efficacy was also analyzed with the determination of optimal administration dosage. Consequently, olive flounder immunized with FKC grown in TSB-D showed the same protection with the vaccine grown in BHIB and the optimal dose of the vaccine was 10mg/kg of body weight. Indeed the dual vaccine showed higher agglutination titer and protection than control fish. The optimal dose for dual vaccine was 10mg/kg body weight for each of two vaccines.
Vibrio harveyi is one of the most serious pathogens causing vibriosis in larval and grow-out shrimp culture. This study was performed to investigate the bactericidal effect of a monopersulfate compound against V. harveyi and its toxicity in Litopenaeus vannamei. The monopersulfate compound was prepared at 0, 0.15, 0.3, 0.6, 1.2, and 2.4 ppm for the bactericidal efficacy study, and then V. harveyi was added at a rate of 1×106 CFU/mL. Subsequently, five shrimps/dose were added to the water bath. The other groups of shrimp were exposed to monopersulfate, but not to the bacterium. None of the shrimps exposed to any of the monopersulfate treatment doses without bacteria died, and no changes in their movement were detected for 7 days. However, shrimps exposed to bacteria without monopersulfate showed decreased movement and lethargy, but no death. The total number of other bacteria and V. harveyi at the different concentrations decreased significantly by 4-6 log values compared to that in the bacterial control group. The monopersulfate compound completely inhibited V. harveyi growth beginning 1 h after exposure to 2.4 ppm. These results show that the monopersulfate compound is an efficient disinfectant against naturally occurring marine bacteria and V. harveyi without being toxic to shrimp.

Vibrio harveyi is a pathogenic marine bacterium causing systemic symptoms resulting in mass mortalities in fishes and shrimps in aquaculture. Outer membrane proteins(OMPs) are related to the pathogenicity and thus good targets for diagnosis and vaccination for Gram negative bacteria. Recently vaccination strategies using the OMPs have been suggested to control vibriosis in several fish species. In this study, we have isolated V. harveyi from diseased marine fishes from different regions of Korea and investigated genetic variations of four OMP genes including OmpK, OmpU, OmpV and OmpW. Consequently, OmpK and U genes could be divided into 3 subgroups of type I, II, III and type A, B, C, respectively, without any correlation with geographical regions and species while OmpV and W were highly homologous. OmpW gene of V. harveyi FP4138 was fully sequenced and predicted the deduced amino acid sequence to form ${\beta}-barrel$ with hydrophobic channel. Indeed, the immunogenicity of recombinant OmpW produced in Escherichia coli was assessed by vaccinating flounder. As a result, the high antibody response with antibody titer of $4.2{\pm}0.7$ and protection with relative percent survival of 60% against artificial infection of V. harveyi were demonstrated. This result indicates that OmpW is a virulence related factor and it can be a vaccine candidate to prevent a high mortality caused by V. harveyi infection in olive flounder, Paralichthys olivaceus.
The characteristics of the extracellular products (ECPs) of Vibrio harveyi grown under various conditions were studied. The extracellular products (ECPs) of Vibrio harveyi is well known as an important pathogenic factor. The optimal isolation condition of the V. harveyi ECPs was incubation with 1.5% NaCl added TSA medium at 25℃ for 24 h. The buffer system for ECPs isolation controlled as pH 8.0 displayed optimal condition. The major protein of ECPs isolated from the five V. harveyi strains originated from Korea (FF8, FF10, FR1, FR2 and FT1 strain) were revealed to serine protease.
Bacterial 16S rRNA gene sequences have been widely used for the studies on molecular phylogeny, evolutional history, and molecular detections. Bacterial genomes have multiple rRNA operons, of which gene sequences sometimes are variable. In the present study, heterogeneity of the Vibrio 16S rRNA gene sequences were investigated. Vibrio 16S rRNA sequences were obtained from GenBank databases, considering the completion of gene annotation of Vibrio genome sequences. These included V. cholerae, V. harveyi, V. parahaemolyticus, V. splendidus, and V. vulnificus. Chromosome 1 of the studied Vibrio had 7~10 copies of the 16S rRNA gene, and their intragenomic variations were less than 0.9% dissimilarity (more than 99.1% DNA similarity). Chromosome 2 had none or single 16S rRNA gene. Intragenomic 16S rRNA genotypes were detected at least 5 types (V. vulnificus #CMCP6) to 8 types (V. parahaemolyticus #RIMD 2210633, V. harveyi #ATCC BAA-1116). These suggest that Vibrio has high heterogeneity of the 16S rRNA gene sequences.

Recently, mass mortality of the young abalone Haliotis discus hannai has occurred in commercial seed production farms in Korea. The mortality rate was above 50% of the total cultured organisms in the farm, and the shell length of the moribund organisms was about 3cm. The mortal phenomenon was that the young abalones were weakly scattered on the bottom of the pond from the attachment matrix, or that they could not be moved back to their normal positions. The diseased farmed Pacific abalone had abdominal edema. From the edema in the moribund individuals, three bacterial strains were isolated and all the strains were identified as Vibrio harveyi. These strains were compared with thirty six strains isolated from the fish. The results was that the Vibrio harveyi from the fish were sorted into genogroup A or B; however, the three strains of the diseased farmed Pacific abalone were sorted into genogroup A and the new genogroup C. The identical mortality and pathological symptoms of the naturally infected organisms were reproduced by artificial infection with WA AG-1 and WA CS-5 strains. The $LD_{50}$ of WA AG-1 and WA CS-5 were each $1.0{\times}10^3cfu\;animal^{-1}$ and $1.7{\times}10^4cfu\;animal^{-1}$.
