Simultaneous Determination of Liquiritin, Nodakenin, Hesperidin and Glycyrrhizin in Bojungikgi-tang Using HPLC-PDA (HPLC-PDA를 이용한 보중익기탕 중 Liquiritin, Nodakenin, Hesperidin 및 Glycyrrhizin의 동시분석)

  • Bojungikgi-tang has been widely used for enhancement of physical fitness in Korea. The convenient, simple, and accurate high-performance liquid chromatography (HPLC) method was established for simultaneous determination of four marker compounds, liquiritin, nodakenin, hesperidin, and glycyrrhizin in Bojungikgi-tang (Buzhongyiqi-tang in Chinese, Hochuekkito in Japanese), a traditional Korean herbal prescription. The column for optimizing HPLC separation was used a Gemini $C_{18}$ column at column oven temperature of $40^{\circ}C$ with 1.0% (v/v) aqueous acetic acid (A) and 1.0% (v/v) acetic acid in acetonitirle (B) by gradient flow. The flow rate was 1.0 ml/min and the detector was a photodiode array (PDA) set at 254 nm, 280 nm, and 335 nm. Calibration curves of four components were acquired with $r^2$ values ${\geq}0.9999$. The recoveries were found to range 92.11~105.68% with relative standard deviations (RSDs, %) value less than 2.50%. The RSD values of intraand inter-day precision were 0.07~2.50% and 0.16~1.99%, respectively. The contents of liquiritin, nodakenin, hesperidin and glycyrrhizin in Bojungikgi-tang were 3.85~3.92 mg/g, 2.27~2.32 mg/g, 4.14~4.19 mg/g, and 3.39~3.45 mg/g, respectively. The established simultaneous analysis method will be effective for quality control of Bojungikgi-tang.

Evaluation of the Immune-Stimulating Activity of Samul-tang, a Traditional Korean Herbal Medicine, Standardized by HPLC-PDA

  • Objectives: We performed simultaneous determination of five constituents by HPLC in Samul-tang (SMT). Additionally, we investigated the immune-stimulatory potential of SMT on specific cellular and humoral immune responses in ovalbumin (OVA)-immunized mice. Methods: Reverse-phase chromatography using a Gemini C18 column operating at $40^{\circ}C$, and photodiode array (PDA) detection at 190-400 nm, were used for quantification of the five components of SMT. Mobile phase using a gradient flow consisted of two solvent systems. Solvent A was 1.0% (v/v) aqueous acetic acid and solvent B was acetonitrile with 1.0% (v/v) acetic acid. C57BL/6 mice were immunized intraperitoneally with OVA/alum ($100{\mu}g/200{\mu}g$) on days 1, 8, and 15. The extract of SMT (1000 mg/kg) was given to mice orally for 21 days (from day 1 to day 21). At day 22, OVA-, lipopolysaccharide (LPS)- and concanavalin A (Con A)-stimulated splenocyte proliferation and OVA-specific and total antibodies were measured in plasma. Results: Calibration curves were acquired with $r^2$>0.9999, and the relative standard deviation (RSD, %) for intra- and inter-day precision were both less than 3.5%. The recovery was in the range of 95.69-115.12%, with an RSD less than 6.0%. The contents of five components in SMT were 1.08-15.30 mg/g. SMT significantly enhanced Con A-induced splenocyte proliferation in OVA-immunized mice (p<0.01). Also, SMT significantly enhanced OVAspecific IgG, IgG1 and total IgM levels in plasma compared with the OVA-immunized group. Conclusions: The established method will be applied for the quantification of major components and immunestimulating activity in OVA-immunized mouse model of SMT.

  • Objectives : Oyaksungi-san(Wuyaoshunqisan) has been used for treatment of stroke and rheumatoid arthritis in Korea. In this study, a simple and accurate high-performance liquid chromatography(HPLC) method was established for simultaneous determination of six main components, liquiritin, ferulic acid, naringin, hesperidin, neohesperidin, and glycyrrhizin in Oyaksungi-san, a traditional Korean herbal prescription. Methods : The analytical column for separation of six constituents was used a Gemini $C_{18}$ column maintained at $40^{\circ}C$. The mobile phase consisted of two solvent systems, 1.0% (v/v) acetic acid in $H_2O$ (A) and 1.0% (v/v) acetic acid in acetonitrile (B) by gradient flow. The flow rate was 1.0 mL/min and the detector was a photodiode array (PDA) set at 254 nm for glycyrrhizin, 280 nm for liquiritin, naringin, hesperidin, and neohesperidin, and 320 nm for ferulic acid. Results : Calibration curves were acquired with $r^2$ values ${\geq}0.9998$. The results of recovery test were 91.58%-105.90% with a relative standard deviations (RSDs, %) value less than 2.0%. The values of RSD for intra- and inter-day precision were 0.03%-1.72% and 0.03%-1.63%, respectively. The contents of the six compounds in Oyaksungi-san were 0.33-9.30 mg/g. Conclusions : The newly established HPLC method will be helpful to improve quality control of Oyaksungi-san.

ROCHE MODEL AND ABSOLUTE DIMENSIONS OF THE ECLIPSING BINARY Y CAM WITH A PULSATING COMPONENT (맥동 식쌍성 Y Cam의 ROCHE 모형과 절대 물리량)

  • We performed CCD photometric observations of the eclipsing binary Y Cam with a pulsating component, for 16 nights from November 2000 to May 2001 using a V filter attached to the 61-cm reflector at Sobaeksan Optical Astronomy Observatory. Our V light curve and Broglia & Marin's (1974) BV ones were analyzed with Mode 2 for a detached Roche model as well as Model 5 for a semi-detached one in the Wilson-Devinney code. From the analysis, we found the following results: 1) It is hardly discernible from only light curve synthesis which model between the detached and semi-detached ones is a real Roche model of Y Cam. 2) There exist the third light of about 2% and 3% in B and V light curves of Y Cam, respectively. 3) The light curve solution is immune from the light variation due to the pulsation of the primary component of Y cam. However, the detached model rather than the semi-detached one has slightly better fitted to the observations which were get rid of the pulsation effect. 4) The absolute dimensions of Y Cam were calculated for two Roche models.

Skin safety of the UV Absorbers by Measurement Cytotoxicity High Functional Product with Water-in-Silicone System

  • Consumers have recently preferred to purchase extensive UV intercepting products, which are waterproof and free from side effects on skin. Testing Cytoroxicity in SR method, cell survivial ratio of UV-B interceptors decreased above 0.08W/V%, and so did that of UV-A interceptors above 0.06W/V%. Also, Patch-test of inorganic UV interceptors resulted in no skin irritation even below 10.0 and 11.25. UV interceptors in the sunlight showed yellowish discoloration in 5 to 14 days. In absorption curves, UV-B was most suitable for Octyl methoxycinnamate and UV-A for Butyl methoxy dibenzoylmethane. Fro this reason, Nylonpoly UVA/UVB the material of OMC and BMDM coated with Nylon & polyethylene, was used as the organic UV interceptor. And zinc oxide and titanium dioxide was used as inorganic UV ibterceptors. The appropriate mixture ratio of ZnO and TiO2 was 6 to 4.6% of ZnO, 4% of TiO2 and 5% of Nylonpoly UVA/UVB were all combined with our sunscreen cream. The SPF value of in-vivo applied to a guinea pig was 34.9 and that of in-vivo was 38.5. Cyclomerhicone and dimerthicone were used in water-in-Silicone system. Ceryl diverhicone and sorbitan sesquioleate were used as emulsifiers and MgSO4, 7H2O, Mg-stearate/Mg-Al-stearate copolymer as emulsification stabilizers. In practical application, each SPF duration of O/W type and W/S type containing sunscreen cream of the same content showed that W/S type of sunscreen cream was 5 times as durable as the other. This product is fit for using in swimming, climbing or skiing. This research is to minimize skin trouble used by UV interceptors and to make one with proper softness, skin safety and UV intercepting efficiency.

Regional Stem Curve and Volume Function Model of Pinus densiflora in Kangwon-Province (강원도 지방 소나무의 지역(地域) 간곡선(幹曲線) 및 재적식(材積式) 모델)

  • Voume functions, which are usually expressed by the function of dbh and height, are estimated commonly through the regression analysis with the highest statistical accuracy considered. In Korea, general volume functions for each tree species were prepared by means of the regression analysis with the exponential function ($V=aD^bH^c$) having the dbh(D) and height(H) as independent variables. In this study, regional stem curve functions for the Pinus densiflora in Kangwon-province were derived and a regional volume function model, in which the stem volume can be directly estimated through the rotational integral of the regional stem curve functions, was prepared. The regional volume estimated by the prepared model was more accurate than the volume by the general volume table for the Pinus densiflora in Kangwon-province. Additionary, the form of stem curves derived by the regional stem curve functions showed difference from each other. The stem in Youngwol and Wonju taper down more fast in upper part than that in other regions. These various stem forms also led to the regional difference in volume estimates.

A Joining Method between HTS Double Pancake Coils (고온초전도 더블 팬케이크 코일들 사이의 접합 방법)

  • High temperature superconductor (HTS) winding coil is one of the key component in superconducting device fabrication. Double-pancake style coils are widely used for such application. High resistance between pancake coils greatly affects the machine design, operating condition and thus the stability. In order to reduce such resistance, experimentalists are looking for efficient and damage free coil connecting methods. In this respect, here we proposed parallel joining method to connect the coils. This is to do crossly joining with HTS tapes on two parallel HTS tapes. Joint samples between two parallel HTS tapes were prepared by using HTS tapes and current-voltage (I-V) characteristic curves were investigated at liquid nitrogen temperature i.e., 77.3 K. A 20 cm length joint connected between two parallel HTS tapes shows $32.5n{\Omega}$, for currents up to 250 A. A small HTS magnet, having two double pancake sub-coils connected together through new parallel joint method was fabricated and their current-voltage (I-V) characteristic curve was investigated. At 77.3K, critical current(Ic) of 97 A and resistance of $55n{\Omega}$ for currents upto 130 A were measured. At operating current 86 A lower than Ic, Joule heats generated in whole magnet and at joint region between sub-coils were 226 mW and 0.4 mW, respectively. Low Joule heat generation suggests that this joining method may be used to fabricate HTS magnet or windings.

Structural Phase Transition, Electronic Structure, and Magnetic Properties of Sol-gel-prepared Inverse-spinel Nickel-ferrites Thin Films

  • X-ray diffraction (XRD), X-ray photoelectron spectroscopy (XPS), and vibrating sample magnetometry (VSM) were used to investigate the influence of Ni ions on the structural, electronic, and magnetic properties of nickel-ferrites ($Ni_xFe_{3-x}O_4$). Spinel $Ni_xFe_{3-x}O_4$ ($x{\leq}0.96$) samples were prepared as polycrystalline thin films on $Al_2O_3$ (0001) substrates, using a sol-gel method. XRD patterns of the nickel-ferrites indicate that as the Ni composition increases (x > 0.3), a structural phase transition takes place from cubic to tetragonal lattice. The XPS results imply that the Ni ions in $Ni_xFe_{3-x}O_4$ substitute for the octahedral sites of the spinel lattice, mostly with the ionic valence of +2. The minority-spin d-electrons of the $Ni^{2+}$ ions are mainly distributed below the Fermi level ($E_F$), at around 3 eV; while those of the $Fe^{2+}$ ions are distributed closer to $E_F$ (~1 eV below $E_F$). The magnetic hysteresis curves of the $Ni_xFe_{3-x}O_4$ films measured by VSM show that as x increases, the saturation magnetization ($M_s$) linearly decreases. The decreasing trend is primarily attributable to the decrease in net spin magnetic moment, by the $Ni^{2+}$ ($2{\mu}_B$) substitution for octahedral $Fe^{2+}$ ($4{\mu}_B$) site.

Statistical Analysis on Microcrack Length Distribution in Tertiary Crystalline Tuff (제3기 결정질 응회암에서 발달하는 미세균열의 길이 분포에 대한 통계적 분석)

  • The scaling properties on the length distribution of microcrack populations from Tertiary crystalline tuff are investigated. From the distribution charts showing length range with 15 directional angles and five groups(I~V), a systematic variation appears in the mean length with microcrack orientation. The distribution charts are distinguished by the bilaterally symmetrical pattern to nearly N-S direction. The whole domain of the length-cumulative frequency diagram for microcrack populations can be divided into three sections in terms of phases of the distribution of related curves. Especially, the linear middle section of each diagram of five groups represents a power-law distribution. The frequency ratio of linear middle sections of five groups ranges from 46.6% to 67.8%. Meanwhile, the slope of linear middle section of each group shows the order: group V($N60{\sim}90^{\circ}E$, -2.02) > group IV($N20{\sim}60^{\circ}E$, -1.55) > group I($N60{\sim}90^{\circ}W$, -1.48), group II($N10{\sim}60^{\circ}W$, -1.48) > group III($N10^{\circ}W{\sim}N20^{\circ}E$, -1.06). Five sub-populations(five groups) that closely follow the power-law length distribution show a wide range in exponents( -1.06 - -2.02). These differences in exponent among live groups emphasizes the importance of orientation effect. In addition, breaks in slope in the lower parts of the related curves represent the abrupt development of longer lengths, which is reflected in the decrease in the power-law exponent. Especially, such a distribution pattern can be seen from the diagram with $N10{\sim}20^{\circ}E,\;N10{\sim}20^{\circ}W$ and $N60{\sim}70^{\circ}W$ directional angles. These three directional angles correspond with main directions of faults developed around the study area. The distribution chart showing the individual characteristics of the length-cumulative frequency diagrams for 15 directional angles were made. By arraying above diagrams according to the categories of three groups(A, B and C), the differences in length-frequency distributions among these groups can be easily derived. The distribution chart illustrates the importance of analysing microcrack sets separately. From the related chart, the occurrence frequency of shorter microcracks shows the order: group A > group B > group C. These three types of distribution patterns could reveal important information on the processes occurred during microcrack growth.

The First Photometric Study of NSVS 1461538: A New W-subtype Contact Binary with a Low Mass Ratio and Moderate Fill-out Factor

  • New multiband BVRI light curves of NSVS 1461538 were obtained as a byproduct during the photometric observations of our program star PV Cas for three years from 2011 to 2013. The light curves indicate characteristics of a typical W-subtype W UMa eclipsing system, displaying a flat bottom at primary eclipse and the O'Connell effect, rather than those of an Algol/b Lyrae eclipsing variable classified by the northern sky variability survey (NSVS). A total of 35 times of minimum lights were determined from our observations (20 timings) and the SuperWASP measurements (15 ones). A period study with all the timings shows that the orbital period may vary in a sinusoidal manner with a period of about 5.6 yr and a small semi-amplitude of about 0.008 day. The cyclical period variation can be interpreted as a light-time effect due to a tertiary body with a minimum mass of 0.71 M. Simultaneous analysis of the multiband light curves using the 2003 version of the Wilson-Devinney binary model shows that NSVS 1461538 is a genuine W-subtype W UMa contact binary with the hotter primary component being less massive and the system shows a low mass ratio of q(mc/mh)=3.51, a high orbital inclination of 88.7°, a moderate fill-out factor of 30 %, and a temperature difference of ΔT=412 K. The O'Connell effect can be similarly explained by cool spots on either the hotter primary star or the cool secondary star. A small third-light corresponding to about 5 % and 2 % of the total systemic light in the B and V bandpasses, respectively, supports the third-body hypothesis proposed by the period study. Preliminary absolute dimensions of the system were derived and used to look into its evolutionary status with other W UMa binaries in the mass-radius and mass-luminosity diagrams. A possible evolution scenario of the system was also discussed in the context of the mass vs mass ratio diagram.
