Myogenesis is the formation process of multinucleated myofiber with a contractile capacity from muscle satellite cell (MSCs) during life. This process is tightly controlled by several transcription factors such as Pax3 and Pax7 (paired box protein 3 and 7), MEF2C (myocyte enhancer factor 2) and MRFs (myogenic regulatory factors) etc. On the contrary, myostatin (MSTN) is a transforming growth factor-${\beta}$ superfamily, which functions as a negative regulator of skeletal muscle development and growth. Carnosic acid (CA) is a major phenolic component in rosemary (Rosmarinus officinalis) and have been reported various biological activities such as anticancer, antioxidant, antimicrobial and therapeutic agents for amnesia, dementia, alzheimer's disease. This study was confirmed to effects of CA on muscle cell line and muscle tissue alteration of zebrafish by intramuscular injection or feeding methods. $10{\mu}M$ CA showed a non-cytotoxic on myoblast and a complete inhibition effect against myostatin activity on luciferase assay. In intramuscular injection experiment, the total protein and triglyceride amount of $10{\mu}M/kg$ of CA injected group increased by 11% and decreased by 13% compared to these of the no injected group. In histology analysis of muscle tissues by hematoxylin/eosin staining, the number of muscle fiber of $10{\mu}M/kg$ of CA injected group decreased by 29% and fiber area increased 40% compared to these of no injected group. In feeding experiment, the total protein and triglyceride amount no significance difference compared to these of the normal feeding group. In histology analysis, the number of muscle fiber of 1% CA fed group decreased by 35% and fiber area increased 56% compared to these of normal fed group. We identified that CA have an effect on hypertrophy of muscle fiber in adult zebrafish and the results of this study are considered as the basic data that can reveal the mechanisms of muscle formation via gene and protein level analysis.
Six triterpenoid compounds, friedelin (1), $3{\alpha}$-hydroxy-2-friedelanone (2), canophyllol (3), oleanolic aldehyde acetate (4), ursolic acid (5), and pachysandiol A (6) were isolated from the methylene chloride soluble fraction of the roots of A. japonica. The chemical structures of compounds 1-6 were determined by the basis of physico-chemical properties and spectroscopic methods such as 1D and 2D NMR. These compounds were isolated from this plant for the first time. For the isolated compounds (1-3), the inhibitory effect of IL-6 production in TNF-$\alpha$ stimulated MG-63 was examined. Among the isolates, $3{\alpha}$-hydroxy-2-friedelanone (2) showed potent inhibitory effect on IL-6 production in TNF-$3{\alpha}$ stimulated MG-63.

Objectives : The purpose of this study is to investigate whitening effects and anti-wrinkle effects of a few 80% ethanol extracted herbal medicines. Methods : In the first study, a few 80% ethanol extracted herbal medicines were screened for their inhibitory activities against the tyrosinase. In the second study, a few 80% ethanol extracted herbal medicines were screened for their inhibitory activities against elastase. Results : 1. We showed 28%, 27% and 19% inhibitions of mushroom tyrosinase at 500 $\mu$g/ml concentration of ASR, AIF and ABR extracts and they were showed higher anti-tyrosinase activity than arbutin's. We also could observe that the decreased mushroom tyrosinase activities in RR, CML, LR, AGR and TH extracts. 2. RR, AF and ABR (final concentrstion 1 mg/ml) were appeared 60%, 98%, 83% of inhibitions of elastase activity, and they were showed higher anti-elastase activity than that of ursolic acid. We also could observe that the decreased elastase activities in AIF, AR, LR and CML extracts. Conclusions : These results suggest that ASR, AIF and ABR extracts contribute to the anti-melanin activities and represent potential sources of whitening agent, and RR, AF and ABR extracts contribute to the anti-elastase activities and represent potential sources of anti-wrinkle agent. These results suggest that some herbal medicines could be strong potential sources of inhibition about anti-aging and whitening effects for the skin.
As control of swine consumptive disease is the key of success in pig farming business, the effect of Oldenlandidae Herba on prevention of swine diseases as an alternative medicine was investigated in-vivo. As it contains substances such as ${\beta}$-sitisterol, ursolic acid and 3-O-${\beta}$-D-giucoside, it is effective on suppression of colonic tumor in rats and also, is widely used as an preventative medicine of cancer and inflammation in Chinese medicine. In this experiment, sows and piglets on two different farms were treated with Oldenlandidae Herba, and they recoded weaning rate greater than 80%, which were higher than national average 70% (PSY 68%). Meanwhile, neither organ accumulation nor toxicity has been detected on the basis of hematology and serum chemistry. In conclusion, the extracts of Oldenlandidae Herba is expected to be an good alternative medicines for swine productivity.
Hexane fraction of Oldenlandiae diffusae Herba(ODH) which was being used for the treatment of caner in oriental medicine showed the best cytotoxicity against L1210 and A549 in the solvent fractions. Antitumor substance isolated from hexane fraction of ODH was identified as ursolic acid(UA) by photometric analysis. IC50 of UA against cancer cells as SNU-1, HCT15, XF498, SK-MEL2 and A549 was $13{\mu}g/m{\ell}$, $15{\mu}g/m{\ell}$, $12{\mu}g/m{\ell}$, $9{\mu}g/m{\ell}$ and $11{\mu}g/m{\ell}$ respectively. It significantly inhibited the metastasis to lungs and kidneys from pulmonary colonization assay and study on histological changes of organs and showed the enhancing effect on B cell dosage-dependently by FACS analysis. T/C % of UA against S-180 cells was 171 % and its cytotoxicity against SNU-1 iant was confirmed from the morphological changes by elctronic microscopes such as SEM and TEM that it induced undulated membrane 4 hr after UA treatment, and the breakdown of cell membrane and nucleus 24 hr after UA treatment.

Recently, the demands for the effective and safe depigmentating and anti-aging agents of the skin have increased due to the medical, pharmaceutical and cosmetic reasons. The aim of this study is to search Galla Rhois (GR) inhibitory activity against aging process of skin in vitro. GR was screened for their inhibitory activity against elastase. GR (final concentration 1 mg/ml) appeared over 40% of inhibition of elastase activity. So we investigated anti wrinkle effects of GR look through MMP-1 inhibition activity. Also Extracts of GR showed higher anti-tyrosinase activity than arbutin (final concentration 1 mg/ml). These results suggest that GR could be potential sources of anti-aging and whitening effects for the skin. Objective : Galla Rhois extracts, Elastase (pancreatic from porcine pancreas), N-Succinyl-(Ala)3-p-nitroanilide, Ursolic acid, mushroom tyrosinase, L-tyrosine, arbutin, Amicon Ultra, Hand Hold UV lamp, MMP-1 primary antibody, MMP-1 secondary antibody Result : 1. GR appeared over 40% of elastase inhibitory activity. 2. GR had MMP-1 inhibition activity. 3. GR showed higher anti-tyrosinase activity than arbutin. Conclusion : According to above results, it is supposed that Galla Rhois could be strong potential sources of anti-aging and whitening effects for the skin.

Four compounds were isolated from Salvia plebeia aerial parts. Silica gel open column chromatography with a gradient elution system was used to isolate and purify these compounds. Nuclear magnetic resonance spectroscopy and mass spectroscopy were used for structural elucidation and identification, while electronic circular dichroism was used to confirm the absolute configuration. The structures were determined to be 𝛽-sitosterol (1), (-)-1S,5S,8S,10R-1-acetoxy-8-hydroxy-2-oxoeudesman-3,7(11)-dien-8,12-olide (2), ursolic acid (3), and N-methylhydroxylamine (4). Compounds 2 and 4 were isolated for the first time from this plant. Compound 2 was quantitatively analyzed via HPLC/UV. The results showed that the methanol extract of S. plebeia had a higher content of compound 2 (1.20 mg/g) than the ethanol extract (0.55 mg/g). This study could be used as a preliminary step in conducting HPLC/UV analysis of sesquiterpenoids in S. plebeia extract to assess their bioavailability and potency.

Neuroinflammation is known to be associated with brain injury in Alzheimer's disease (AD), and the inhibition of microglial activation, a key player in inflammatory response, is considerd as important target for AD. In this study, the ethanol extract of aerial parts of Forsythia velutina Nakai, a Korean native species, significantly inhibited nitric oxide (NO) production in LPS-stimulated BV2 microglial cells. Thus, the active principles in F. velutina aerial parts were isolated based on activity-guided isolation method. As a result, six compounds were isolated and their structures were elucidated based on NMR data and the comparison with the relevant references as arctigenin (1), matairesinol (2), rengyolone (3), ursolic acid (4), secoisolariciresinol (5), and arctiin (6). Among them, four compounds including arctigenin (1), matairesinol (2), secoisolariciresinol (5), and arctiin (6) significantly inhibited NO production in a dose-dependent manner. In particular, matairesinol (2) and secoisolariciresinol (5) reduced 60% of NO production compared to LPS-treated group. This inhibitory effects of matairesinol (2) and secoisolariciresinol (5) were accompanied with the reduced expression levels of iNOS and COX-2. These results suggest that the extract of F. velutina and its active compounds could be beneficial for neuroinflammatory diseases including AD.

We have previously screened 150 medicinal plants on the inhibition of elastase and found a significant inhibitory effects of the extracts of Areca catechu L. on the aging and inflammation against the skin tissues. To isolate and identify the compounds having biological activity, we was further purified by each of the solvent fractions, silica gel column chromatography, preparative TLC and reversed-Phase HPLC. Peak in HPLC, which coincided with the inhibitory activity against elastase, was identified as Phenolic substance using various colorimetric methods, UV, and IR. $IC_{50}$/ values of phenolic substance purified from Areca catechu were 26.9 $\mu\textrm{g}$/$m\ell$ for porcine pancreatic elastase (PPE) and 60.8 $\mu\textrm{g}$/$m\ell$ for human neutrophil elastase (HNE). This Phenolic substance showed more potent activity than those of reference compounds, oleanolic acid (76.5 $\mu\textrm{g}$/$m\ell$ for PPE, 219.2 $\mu\textrm{g}$/$m\ell$ for HNE) and ursolic acid (31.0 $\mu\textrm{g}$/$m\ell$ for PPE, 118.6 $\mu\textrm{g}$/$m\ell$ for HNE). According to the Lineweaver-Burk Plots, the inhibition against both PPE and HNE by this phenolic substance was competitive with substrate. Phenolic substance from Areca catechu exhibited high free radical scavenging effect ($SC_{50}$/ : 6 $\mu\textrm{g}$/$m\ell$) and inhibited effectively hyaluronidase activity ($IC_{50}$/: 210 $\mu\textrm{g}$/$m\ell$). These results suggest that the Phenolic substance Purified from Areca catechu showed anti-aging effect by protecting connective tissue proteins.

The chemical investigation of the methanolic crude extract of leaves of Diospyros iturensis gave us 15 known secondary metabolites identified as mixture of α-amyrenone (1) and β-amyrenone (2), β-amyrin (3), mixture of β-sitosterol (4) and stigmasterol (5), betulin (6), uvaol (7), betulinic acid (8), ursolic acid (9), corosolic acid (10), actinidic acid (11),11-O-p-hydroxybenzoylbergenin (12), bergenin (13) and mixture of stigmasterol glucoside (14) and β-sitosterol glucoside (15) respectively. The structures of secondary metabolites were elucidated with the help of NMR and mass spectral data and by comparison of their spectral data with literature. Among the fifteen isolated compounds, four compounds were identified for the first time in Diospyros genus. These included uvaol (7), corosolic acid (10), actinidic acid (11) and 11-O-p-hydroxybenzoylbergenin (12). Crude methanolic extract of leaves and four isolated compounds including betulin (6), betulinic acid (8), 11-O-p-hydroxybenzoylbergenin (12) and bergenin (13) were evaluated for their antiproliferative activity against two cancer cell lines CAL-27 and NCI-H460 by the MTT assay, antioxidant potential and inhibitory activity against the lipoxygenase and urease enzymes, respectively. The results indicated that the methanolic crude extract of leaves exhibited moderate antioxidant activity and was inactive against the two cancer cell lines. Betulin (6), 11-O-p-hydroxybenzoylbergenin (12) and bergenin (13) exhibited moderate antioxidant and lipoxygenase inhibition with IC50 = 65.8, 85.6, 82.5 μM and IC50 = 58.5, 95.2, 76.2 μM, respectively. Furthermore, 11-O-p-hydroxybenzoylbergenin (12) and bergenin (13) exhibited moderate urease inhibition activity with IC50 values of 45.6 μM and 49.8 μM, respectively.
