유아의 충동성 기질 및 사회극놀이 참여도와 자기규제간의 관계 (Relationships Between Children's Impulsivity, Sociodramatic Play and Children's self-regulation)

  • This study examined the relations between children's Impulsivity temperament, socio-dramatic play, and self-regulation of 4-year-old children. Subjects were 117 4-year-old children(59 boys and 58 girls) selected from three kindergartens. Each mother completed a questionnaire on her child's temperament. Naturalistic observations of total socio-dramatic play, complex socio-dramatic play, solitary dramatic play and of self-regulation in two classroom contexts - clean-up periods and group circle time - were conducted for obtaining data. Significance was analyzed by Pearson's correlation and multiple regression analysis. Children's Impulsivity was negatively related to self-regulation and to total and complex socio-dramatic play, but impulsivity was positively related to solitary dramatic play. Total and complex socio-dramatic play was positively related to self-regulation while solitary dramatic play was negatively related to self-regulation. Total and complex socio-dramatic play, and solitary dramatic play displayed a mediating role between impulsivity and self-regulation during clean-up periods.

Negative regulators in RANKL-induced osteoclastogenesis

  • Receptor activator of nuclear factor ${\kappa}B$ ligand (RANKL) induces osteoclast formation from hematopoietic cells via up-regulation of positive regulators, including $NF-{\kappa}B$, c-Fos, microphthalmia transcription factor (Mitf), PU.1, and nuclear factor of activated T cells (NFAT) c1. In addition to the positive regulation by these transcription factors, RANKL appears to regulate negative regulators such as MafB and inhibitors of differentiation (Ids). Ids and MafB are abundantly expressed in osteoclast precursors, bone marrowderived monocyte/macrophage lineage cells (BMMs). Expression levels of these genes are significantly reduced by RANKL during osteoclastogenesis. Overexpression of these genes in BMMs inhibits the formation of tartarate-resistant acid phosphatase (TRAP)-positive multinuclear osteoclasts by down-regulation of NFATc1 and osteoclast-associated receptor (OSCAR), which are important for osteoclast differentiation. Furthermore, reduced expression of these genes enhances osteoclastogenesis and increases expression of NFATc1 and OSCAR. Taken together, RANKL induces osteoclastogenesis via up-regulation of positive regulators as well as down-regulation of negative regulators.

  • The turnout system is typical railway facility to be in the way of conventional railway line's speed-up. The KNR(Korean National Railroad) :has a speed limit regulation(130km/h) for the passing the turnout to guarantee a safety. This research had been performed to confirm the propriety of this regulation by using experimental and analytical way before it was carried out. The reasonableness of the speed-limit regulation was judged base on the several experiments. It was proved that the regulation was not useless as it had been regarded and very important guide at least up to now.

프로테옴 분석에 의한 Bacillus subtilis PyrR 돌연변이체의 특성 (Characterization of a PyrR-deficient Mutant of Bacillus subtilis by a Proteomic Approach)

  • Bacillus subtilis의 pyrimidine biosynthetic (pyr) operon은 UMP의 de nove 생합성에 관여하는 enzyme들을 encode할 뿐만 아니라, 조절단백질인 PyrR도 encode한다. PyrR은 pyr mRNA-binding 조절 기능과 uracil phosphoribosyltransferase activity를 동시에 가지는 bifunctional 단백질이다. 본 연구에서는 Proteomic analysis를 이용하여 Uracil - 환경에서 DB104${\Delta}$pyrR의 단백질 패턴을 분석하여 단백질 레벨에서 PyrR 단백질의 실질적인 조절 양상을 관찰하였다. 두 균주의 세포질 단백질은 다양한 발현의 차이를 보였으며, Silver 염색된 2D-gel의 pI 4~10 사이에서는 1,300여개의 단백질이 검출되었으며, 단백질 발현 차이를 보이는 172개의 spot 중에서 42개의 단백질이 identification 되었다. 그 결과 pyr operon의 단백질(PyrAa, PyrAb, PyrB, PyrC, PyrD, and PyrF)이 모두 Up regulation이 이루어지고 있음을 확인할 수 있었으며, 이것은 단백질 레벨에서 Pyrimidine 생합성 과정이 PyrR에 의해서 정확히 Regulation 되어짐을 확인할 수 있었다. 또한 Pyrimidine 생합성의 Up regulation과 Down regulation 상태의 단백질의 패턴 양상도 분석할 수 있게 되었다. Pyrimidine의 생합성 과정은 DNA를 구성하는 기본적인 구성 요소를 생산하는 과정으로서 여러가지 Metabolism 가운데 중요한 위치를 차지하고 있다. 만약 Pyrimidine의 생합성 과정이 Over- expression된다면 다른 Metabolism의 균형에도 변화가 올 것이다. Proteomics Analysis에 이용한 DB104${\Delta}$pyrR 균주는 Pyrimidine 생합성의 조절에 관여하는 PyrR knock out 균주로서 Uracil - 환경에서는 전체적인 Pyrimidine 생합성 조절이 Up regulation이 되어지므로 Up regulation 동안 어떤 Metabolism에 영향을 주는지 관찰을 할 수 있게 되었다. 특히 Amino Acid Metabolism에 관계있는 단백질의 Up regulation이 이루어짐을 관찰할 수 있었으며 이것은 현재 각광을 받고 있는 단백질 산업에 응용함으로써 산업적으로 많은 기대를 할 수 있을 것으로 예상되어진다.

초거대 인공지능 정책 변동과정에 관한 연구 : 옹호연합모형을 중심으로 (A Study on the Process of Policy Change of Hyper-scale Artificial Intelligence: Focusing on the ACF)

  • Although artificial intelligence(AI) is a key technology in the digital transformation among the emerging technologies, there are concerns about the use of AI, so many countries have been trying to set up a proper regulation system. This study analyzes the cases of the regulation policies on AI in USA, EU and Korea with the aim to set up and improve proper AI policies and strategies in Korea. In USA, the establishment of the code of ethics for the use of AI is led by private sector. On the other side, Europe is strengthening competitiveness in the AI industry by consolidating regulations that are dispersed by EU members. Korea has also prepared and promoted policies for AI ethics, copyright and privacy protection at the national level and trying to change to a negative regulation system and improve regulations to close the gap between the leading countries and Korea in AI. Moreover, this study analyzed the course of policy changes of AI regulation policy centered on ACF(Advocacy Coalition Framework) model of Sabatier. Through this study, it proposes hyper-scale AI regulation policy recommendations for improving competitiveness and commercialization in Korea. This study is significant in that it can contribute to increasing the predictability of policy makers who have difficulties due to uncertainty and ambiguity in establishing regulatory policies caused by the emergence of hyper-scale artificial intelligence.

Effects of Soy-isoflavonoid on Molecular Markers Related to Apoptosis in Mature and Ovariectomized Female Rats, and Mammalian Tumor Cell Lines

  • Alteration of molecular markers related to apoptosis of in vivo normal system and in vitro cancerous system by soy-isoflavonoid with estrogen was investigated. Down-regulation of Bcl-2 was accompanied by decreased expression of COX-2 (cyclooxygenase-2) in mature female rats treated with soy-isoflavonoid and estrogen. In ovariectomized rat system, Bax was regulated by higher concentration of soy treatment. Bax up-regulation by soy-isoflavonoid genistein treatment was observed in MCF-7 mammary cancer cell system. Estrogen without soy induced similar pattern of Bax expression as soy-isoflavonoid in vivo, but exhibited opposite trend in vitro. These findings suggest soy-isoflavonoid may have potential to induce apoptosis at higher concentrations through up-regulation of Bax or down-regulation of Bcl-2 expressions depending on normal or cancerous state, and physiological status of rats.

L-glutamine:D-fructose-6-phosphate Aminotransferase as a Key Protein Linked to Multidrug Resistance in E. coli KD43162

  • A microarray study has been employed to understand changes of gene expression in E. coli KD43162 resistant to ampicillin, ampicillin-sulbactam, piperacillin, piperacillin-tazobactam, cefazolin, cefepime, aztreonam, imipenem, meropenem, gentamicin, tobramycin, ciprofloxacin, levofloxacin, moxifloxacin, fosfomycin, and trimethoprim-sulfamethoxazole except for amikacin using disk diffusion assay. Using Sodium dodecyl sulphate-polyacrylamide gel electrophoresis and MALDI-TOF MS analyses, 36 kDa of outer membrane proteins (OMPs) was found to be deleted in the multidrug resistant E. coli KD 43162. Microarray analysis was used to determine up- and down-regulated genes in relation to multidrug resistant E. coli KD43162. Among the up-regulated genes, these genes were corresponded to express the proteins as penicillin-binding proteins (PBPs), tartronate semialdehyde reductase, ethanolamine utilization protein, shikimate kinase I, allantoinase, predicted SAM-dependent methyltransferase, L-glutamine: D-fructose-6-phosphate aminotransferase (GFAT), phospho-glucosamine mutase, predicted N-acetylmannosamine kinase, and predicted N-acetylmannosamine-6-P epimerase. Up-regulation of PBPs, one of primary target sites of antibiotics, might be responsible for the multidrug resistance in E. coli with increasing amount of target sites. Up-regulation of GFAT enzyme may be related to the up-regulation of PBPs because GFAT produces N-acetylglucosamine, a precursor of peptidoglycans. One of GFAT inhibitors, azaserine, showed a potent inhibition on the growth of E. coli KD43162. In conclusion, up-regulation of PBPs and GFATs with the loss of 36 kDa OMP refers the multidrug resistance in E. coli KD 43162.

Single-cell RNA sequencing reveals the heterogeneity of adipose tissue-derived mesenchymal stem cells under chondrogenic induction

  • This study investigated how adipose tissue-derived mesenchymal stem cells (AT-MSCs) respond to chondrogenic induction using droplet-based single-cell RNA sequencing (scRNA-seq). We analyzed 37,219 high-quality transcripts from control cells and cells induced for 1 week (1W) and 2 weeks (2W). Four distinct cell clusters (0-3), undetectable by bulk analysis, exhibited varying proportions. Cluster 1 dominated in control and 1W cells, whereas clusters (3, 2, and 0) exclusively dominated in control, 1W, and 2W cells, respectively. Furthermore, heterogeneous chondrogenic markers expression within clusters emerged. Gene ontology (GO) enrichment analysis of differentially expressed genes unveiled cluster-specific variations in key biological processes (BP): (1) Cluster 1 exhibited up-regulation of GO-BP terms related to ribosome biogenesis and translational control, crucial for maintaining stem cell properties and homeostasis; (2) Additionally, cluster 1 showed up-regulation of GO-BP terms associated with mitochondrial oxidative metabolism; (3) Cluster 3 displayed up-regulation of GO-BP terms related to cell proliferation; (4) Clusters 0 and 2 demonstrated similar up-regulation of GO-BP terms linked to collagen fibril organization and supramolecular fiber organization. However, only cluster 0 showed a significant decrease in GO-BP terms related to ribosome production, implying a potential correlation between ribosome regulation and the differentiation stages of AT-MSCs. Overall, our findings highlight heterogeneous cell clusters with varying balances between proliferation and differentiation before, and after, chondrogenic stimulation. This provides enhanced insights into the single-cell dynamics of AT-MSCs during chondrogenic differentiation.

에스트로겐에 의한 암쥐의 심장조직의 COX-2, Mapkinases 및 관련된 Apoptotic Markers의 발현의 변화에 관한 연구 (Changes in the Expression Pattern of Cyclooxygenase-2, Mapkinases and Related Apoptotic Markers by Different Levels of Estrogen Supplementation in Mature or Ovariectomized Female Rat Heart)

  • The effects of different concentrations of estrogen supplementation to mature female rats or estrogen supplementation to ovariectomized rats on cyclooxygenase-2 (COX-2) expression, PGE$_2$ production and mapkinases expression were investigated in experimentally induced atherogenic rats with feeding a high fat. high cholesterol diet. In the first experiment using 48-week old mature rats, the supplementation of three different levels of estrogen was compared to the basal diet. The high concentration of estrogen supplementation induced the marked up-regulation of COX-2 protein and the increase in plasma PGE$_2$ production and this seems to be followed by the up-regulation of p38 among mapkinases. The regulation of bax showed in a reverse trend of COX-2 in heart tissues of mature female rats. In the second ex-perimental system, female Sprague-Dawley rats were bilaterally ovariectomized; sham-operated animals were used as controls. Three weeks later, the animals were supplied with basal diet to sham-operated control group and ovariectomized control group, and estrogen supplemented diet to ovariectomized group for an eight-week experimental period. In a group supplemented with a medium dose of estrogen, COX-2 expression was up-regulated. This up-regulation was accompanied by the elevated expression of pERK1/2. Bax was increased in estrogen-fed animals indicating bax might be involved in estrogen feeding state in ovariectomized rats. Further investigations on the relationship between COX-2 and biological activities such as vasodilation by estrogen are required in in vivo system of female rats at the various physiological states.
