미역(Undaria pinnatifida) 부위별로 추출한 다당의 이화학적 특성 (Chemical Composition and Rheological Properties of Polysaccharides Isolated from Different Parts of Brown Seaweed Undaria pinnatifida)

  • The chemical and rheological properties of fucoidan and alginate prepared from different parts of Undaria pinnatifida (sporophyll, frond, stipe) were investigated. The algal materials were extracted with HCl (pH 2.0, 3 h at 70℃) to prepare fucoidan, and the remaining solid was continuously re-extracted with Na2CO3 (pH 10.0, 70℃, 3 h) to prepare alginic acid. The fucoidan and alginic acid contents in the sporophyll, frond, and stipe were 11.14%, 3.84%, and 1.73% and 22.04%, 37.14%, and 31.74%, respectively. The content of fucoidan and alginate depends on the part extracted. The fucoidan extracted from the sporophyll mainly consists of fucose and galactose, but the fucoidan extracted from frond and stipe contains mannose in addition to fucose and galactose. Fourier-transform infrared spectroscopy analysis of fucoidan and alginate suggests the presence of sulfate groups (1261 and 840 cm-1) and carboxyl groups (1626 and 1419 cm-1), respectively. Alginate solutions (5%) had a low viscosity of 10.84-31.63 mPa·s. The activation energies of fucoidan and sodium alginate were 14.45-18.38 kJ/mol and 18.61-22.06 kJ/mol, respectively. The D-mannuronic acid/L-guluronic acid (M/G) ratios of alginate showed a relatively high (frond, 3.72; stipe, 2.88; and sporophyll, 1.80).

한국동해남부해역의 수온이 멸치와 미역의 생산량에 미치는 영향 (Effect of Temperature on Catches of Anchovy and Sea Mustard(Undaria pinnatifida) in Southern Part of East Sea of Korea)

  • 본 연구는 1969~2007년간 한국 전체의 어업생산통계자료에서 멸치 및 미역 생산량 중 1990~2007년간 동해남부해역의 변동 양상을 파악하다. 이를 위하여 국립수산과학원의 같은 기간의 207선 수온자료를 이용하였다. 표층수온의 저온화 시기가 4월이면 미역 생장에 호영향을 주어 미역의 생산량이 많고, 그 시기가 6월이면 멸치에 악영향을 주어 멸치의 생산량이 적게 나타났다. 반대로 표층수온의 고온화 시기가 4월이면 미역 생장에 악영향을 주어 미역의 생산량은 적었고, 그 시기가 6월이면 멸치에 호영향을 주어 멸치의 생산량이 많았다. 표층 수온이 고온화 되면 멸치 생산량은 증가하고, 표층수온이 저온화 되면 미역 생산량은 증가하는 것으로 나타났다.

미역 추출물을 이용한 견직물의 천연염색 (Natural Dyeing of Silk Fabrics with Undaria Pinnatifida Extracts)

  • Fabric made of 100% silk was dyed with Undaria pinnatifida extracts under different conditions such as varing colorant concentration, temperature, time and dyebath pH. The dye uptake (K/S, value), CIE L*,a*,b* and Munsell values of the dyed samples and mordanted samples were measured. Colorfastness, antibacterial property and UV protection property were also evaluated. As colorants concentration increased, the dye uptake increased progressively and maximum color strength was obtained at 100% V/V. Dye uptake also increased with increasing temperature and time, and the maximum color strength was obtained at $80^{\circ}C$ and 60min. With pre-mordanting, the color of the Al and Fe mordanted silk fabrics was yellow on the Munsell color system, while the Cu mordanted fabric was a greenish color. Al and Fe post-mordanted fabrics had a yellow tone, and the Cu post-mordanted fabric had a yellow greenish tone. The light fastness property of the Cu mordanted silk fabric was relatively good, The effect of Al and Fe mordanting on fastness was insignificant. The antibacterial activity of dyed and unmordanted silk fabric was excellent, whereas fabric dyed and mordanted with Undaria pinnatifida extract demonstrated superior ultraviolet protection.

Chlorothalonil이 미역(Undaria pinnatifida) 배우체의 생존 및 상대성장률에 미치는 영향 (The impact of chlorothalonil on female gametophyte survival rate and relative growth rate of Undaria pinnatifida)

  • TBT 사용이 금지된 이후, 방오도료의 방오능력을 보완하기 위하여 booster biocides를 추가하여 사용하고 있다. Booster biocides 중에서 chlorothalonil은 해양환경 내 지속적으로 유입되며 다양한 해양생물에게 심각한 독성영향을 미치고 있지만 해조류에 대한 연구는 제한적이 다. 따라서 우리나라의 주요 양식생물인 미역(Undaria pinnatifida)의 초기생활사에 chlorothalonil이 미치는 영향을 분석하였다. U. pinnatifida의 암배우체를 chlorothalonil(0, 0.03, 0.05, 0.10, 0.20, 0.40 mg L-1)에 노출하여 생존율과 상대성장률을 분석하였다. 암배우체 생존율의 무영향 농도(NOEC), 최소영향농도(LOEC), 반수치사농도(LC50)는 0.05, 0.10, 0.141 (0.121~0.166) mg L-1이었고 상대성장률의 NOEC, LOEC, 반수영향농도(EC50)은 0.10, 0.20, 0.124 (0.119~0.131) mg L-1로 분석되었다. 따라서, 미역의 암배우체는 0.05~0.10 mg L-1 이상의 농도에서 독성영향을 받기 시작할 것으로 판단된다. 본 연구의 결과는 chlorothalonil에 대한 U. pinnatifida 초기생활사의 건강도를 평가하기 위한 기준자료로 활용될 것으로 기대된다.

Undaria pinnatifida Fucoidan-Rich Extract Recovers Immunity of Immunosuppressed Mice

  • We investigated the immune restoration activity of Undaria pinnatifida fucoidan-rich extract in cyclophosphamide-induced immunosuppressed mice. C57BL/6 mice were intraperitoneally injected with 80 mg/kg of cyclophosphamide (CP) and orally administered with either drinking water (DW), red ginseng extract (RG), or one of three different doses of Undaria pinnatifida fucoidan-rich extract (DSU02 50, 100, and 150 mg/kg). After 14 days, liver, spleen, and whole blood were isolated from each animal. The frequencies of NK and CD3+, CD4+, and CD8+ T cells were significantly increased in splenocytes isolated from the DSU02 100 mg/kg and DSU02 150 mg/kg groups (NK1.1+, 5.4% or 4.9% vs 3.8%; CD3+, 39.3% or 37.9% vs 32.3%; CD4+, 22% or 20.2% vs 17.4%; CD8+, 12.7% or 11.6% vs 10.1%). NK cytotoxicity was enhanced in the DSU02-fed groups at all doses (CP-treated DW, 93.4%; RG, 107.2%; DSU02 50, 107.3%; DSU02 100, 107.3%; DSU02 150, 107.1%), and the proliferation of T cells (CD3+, CD4+, and CD8+) was also greater in the DSU02 100 mg/kg and DSU02 150 mg/kg administered groups compared with the unfed group. Plasma concentrations of TNF-α, IgM, and total IgG from the DSU02 150 mg/kg group were also significantly higher compared with the other groups (TNF-α: CP-treated DW - 21.5 pg/ml, DSU02 150 - 47.1 pg/ml; IgM: CP-treated DW - 82.9 ng/ml, DSU02 150 - 110.8 ng/ml; total IgG: CP-treated DW - 114.4 ng/ml, DSU02 150 - 162.7 ng/ml). We suggest that Undaria pinnatifida fucoidan-rich extract could be a promising candidate for a marine natural immune stimulator.

두부제조시 해조류 첨가 효과 (Effect of Seaweeds added in preparation of Tofu)

  • This study was performed to investigate some quality characteristics of 새려 prepared from soybean milk and various seaweed(Undaria pinnatifida, Laminaria japonica, Porpyra tenera, Enteromorpha sp., Codium sp.) pulps in the ration of 9:1(v:v) with 20% MgCl2. The yields of tofu containing Undaria pinnatifida, Laminaria japonica, or Enteromorpha sp. increased but porphyra tenera, Codium sp. decreased in comparison with tofu prepared from whole soybean milk. The protein content of tofu containing Undaria pinnatifida, Laminaria japonica, Porphyra tenera, or Codium sp. increased but Enteromprpha sp. decreased in comparison with the tofu prepared from whole soybean milk. The content of Ca in Undaria pinnatifida, Porphyra tenera added tofu was higher than that of the tofu prepared from whole soybean milk or other seaweeds added tofu. In sensory evaluation the texture, color, taste of tofu were favored with the addition of sea mustard(Undaria pinnatifida) pulp than that of the tofu prepared from whole soybean milk or tofu prepared other seaweed. Tofu prepared was possible with adding 0.5~1.5% sea mustard to soybean milk but the feasible added amount level was 1% of sea mustard. The yields, protein Ca, and K content of tofu were increased by the more adding amount of sea mustard tan tat of the tofu prepared from whole soybean milk. The hardness values of 1% sea mustard added tofu were decreased than that of the tofu prepared from whole soybean milk ; on the other hand, elasticity, cohensiveness, gumminess and brittleness of tofu with sea mustard increased. The L and a values of tofu were lower and b values were higher with the addition of 1% sea mustard. The content of histidine, tyrosine, leusine, and phenylalanine were decreased but the other amino acid were increased in tofu prepared from 1% sea mustard pulp added to soybean milk. The saturated fatty acid and monoene fatty acid content of tofu were increased and C18:2, C18:3(${\gamma}$), C18:3, C20:5 of polyene fatty acid were slightly decreased in tofu prepared from 1% sea mustard pulp added to soybean milk.

Radical Scavenging Activities of Undaria pinnatifida Extracts Fermented with Cordyceps militaris Mycelia

  • The present study was performed to investigate the various radical scavenging activities of fermented Undaria pinnatifida by the mycelia fermentation method. U. pinnatifida was fermented with Cordyceps militaris (C. militaris ) mycelia using solid culture and compared with unfermentated U. pinnatifida and C. militaris mycelia for antioxidant activities. The various radical scavenging activities of extracts from U. pinnatifida fermented with C. militaris mycelia (FUCM) were evaluated by electron spin resonance. The antioxidant activities of the FUCM extracts were assayed for ferric reducing antioxidant power, 2,2'-azinobis-(3-ethybenzothiazoline-6-sulfonic acid) radical scavenging activity, and oxygen radical absorption capacity. The free radical scavenging activity of FUCM extracts was higher than that of C. militaris mycelia or U. pinnatifida alone. FUCM extracts were significantly (p < 0.05) increased up to 35 times, 10 times, and 16 times that of U. pinnatifida extracts on DPPH, alkyl, and hydroxyl radical scavenging activities, respectively. These results indicate that FUCM extracts have different chemical ingredients from U. pinnatifida and could provide beneficial antioxidant activity.

감마선 조사에 의한 미역의 저장성 개선 (Improvements in Storage Stability of Undaria pinnatifida after Gamma Irradiation)

  • 감마선 조사가 미역의 저장성 개선에 미치는 영향을 평가하였다. 미생물 오염도 평가 결과 3 kGy 이하의 선량에서 미생물이 검출되지 않았다. 2주 저장한 후, 조사처리 하지 않은 미역의 미생물이 6 log CFU/g이 증가하였다. 그러나 5 kGy 조사 처리한 미역의 경우 미생물이 검출되지 않았다. 5 kGy의 감마선 조사를 적용하였을 때, 미역의 명도 값이 소폭 증가하였다. 미역의 경도 측정 결과 3 kGy 이하의 감마선 조사에 의해 영향을 미치지 않았으며, 저장기간에 따른 차이는 없는 것으로 나타났다. 이상의 결과를 종합하여 볼 때, 3 kGy 감마선 조사는 미역의 품질에 영향을 받지 않고, 저장성을 연장시킬 수 있는 효과적인 기술이라고 판단된다.

미역중의 Calcium 및 Iron 의 함량에 대하여 (A Study on the Calcium and Iron Content of the Undaria pinnatifida suringar)

  • 해운대 동백섬 근해에서 채취한 미역을(Undaria pinnatifida suringar) 유초와 성초로 구분하여 각각 부위별로 calcium과 iron의 함량을 정량화하였다. Calcium은 과망간산칼륨 적정법에 의하였으며, iron은 o-phenanthrolin 비색법에 의하여 정량하였다. 실험결과 유초나 성초에서 calcium은 잎부분에 가장 많이 함유되어 있는 경향이 였으며, 잎부분의 Iron 함유량은 줄기부분의 약 2배가 됨을 발견하였다.

Biosorption of Heavy Metal lons by Biomass of Marine Brown Algae in Cheju using Their Immobilization Techniques: Biosorption of Copper by Undaria pinnatifida

  • The biosorption performances of copper were investigated by the immobilized biomass of nonliving marine brown algae Undaria pinnatifida by each of the Ca-alginate method(Ca-ALG), Ba-alginate method(Ba-ALG), polyethylene glycol method(PEG), and carrageenan method (CARR). The copper removal performance increased but the copper uptake decreased as the biomass amount was increased. However, the copper uptake by the immobilized biomass increased with increasing initial copper concentration. Among the immobilization methods, the copper uptake decreased in the following sequence: Ca-ALG > Ba-ALG > PEG > CARR. The pattern of copper uptake by the immobilized biomass fitted the Langmuir isotherm better than the Freundlich isotherm. Desorption of deposited copper with 0.05 ~0.5M HCI, resulted in no changes of the copper uptake capacity of the immobilized biomass by the immobilization methods except for PEG, through five subsequent biosorption/desorption cycles. There was no damage to the immobilized biomass which retained its macroscopic appearance in repeated copper uptake/elution cycles.
