미선나무의 저온 추출 공정에 의한 항주름 활성 증진 (Enhancement of Anti-wrinkle Activities of Abeliophyllum distichum Nakai through Low Temperature Extraction Process)

  • Skin anti-wrinkle activities of the stems and leaves of Abeliophyllum distichum Nakai were evaluated by the extracts obtained from various extraction processes such as using hot water at $100^{\circ}C$, 70% ethanol at $85^{\circ}C$, and 70% ethanol with ultrasonication at $60^{\circ}C$ The ultrasonicated extract showed 95.62% of the highest cell viability in addition of $0.3mg/m{\ell}$ of the extracts into the normal human fibroblast cell, CCD-986sk. For antioxidant activities, the extracts using ultrasonicated extract showed the highest DPPH free radical scavenging as 80.27%, followed by 75.88% and 62.44% for the extracts using ethanol extract and water extract. The ultrasonicated extract also showed the highest elastase inhibition activity as 25.32%, compared to ethanol extract and water extract based method at 22.01% and 12.88%, respectively. MMP-1 production was most effectively decreased down to $2908.1pg/m{\ell}$ with ultrasonicated extract while $6640.8pg/m{\ell}$ with water extract and $3609.3pg/m{\ell}$ with ethanol extract, in addition of $0.3mg/m{\ell}$. Collagen production was increased up to $154.7ng/m{\ell}$ in addition of ultrasonicated extract, and followed by $121.4ng/m{\ell}$ and $31.2ng/m{\ell}$ for ethanol extract and water extract, respectively. These results indicate that the ethanol extract should have skin anti-wrinkling activities and can be improved by the ultrasonication process that high energy input elute more amounts of bioactive substances eluting more amounts of bioactive substances from the high energy input of ultrasonication.

강원지역 폐옥수수대로부터 바이오에탄올 생산을 위한 전처리 방법 개발 (Pretreatment of Wasted Corn Stalk from Gangwon Province for Bioethanol Production)

  • 강원도에서 확보한 폐옥수수대는 글루칸 44.6 %, 자일란 23.8 %, 리그닌 23.8 %, 회분 4.5 %, 기타 8.1 %로 이루어져 있다. 묽은 황산을 이용한 전처리 공정을 통계적 방법인 완전요인배치으로 분석한 결과 온도가 가장 중요한 변수로 나타났으며 자일로스 수율 기준으로 처리 시간과 농도에 비해 각각 3.5배와 3.2배 큰 영향력을 보여주었다. 포도당 수율에 대해서도 온도의 영향력이 가장 큰 것으로 나타났지만 포도당 수율은 전체적으로 5 % 이내의 낮은 값을 나타내었다. 마이크로웨이브와 초음파 각각을 이용한 전처리 공정은 그 효과가 크지 않거나 미미하였으나 묽은 황산처리와 복합적으로 적용했을 때는 그 효과는 상당히 큰 것으로 나타났다. 특히 마이크로웨이브와 묽은 황산처리를 순차적으로 적용했을 때 자일로스 수율은 2배 넘게 향상되었다. 향후에는, 다양한 복합 전처리 공정에 대한 심도있는 연구와 더불어 전처리와 효소당화 공정의 최적화에 대한 연구를 수행할 계획이다.

반응표면분석법으로 최적화한 초음파와 저온 블랜칭의 병용처리 조건이 신선편이 양상추의 갈변과 품질에 미치는 영향 (Effects of Optimized Co-treatment Conditions with Ultrasound and Low-temperature Blanching Using the Response Surface Methodology on the Browning and Quality of Fresh-cut Lettuce)

  • 신선편이 양상추는 유통 중 갈변과 이취 및 무름현상이 발생되고, 미생물의 증식에 의해 안전성이 위협받는 등 원재료보다 품질 저하현상이 빠르다는 문제점이 있다. 또한, 건강지향적인 시대가 도래함에 따라, 선도 유지를 위한 화학적 처리를 거친 식품은 기피하는 실정이다. 이에 일반적인 블랜칭의 단점을 보완할 수 있는 저온블랜칭과, 다른 기술과 병용처리 시 synergy effect를 기대할 수 있는 초음파를 병용처리함으로써 신선편이 양상추의 품질특성 변화에 대해 알아보고자 하였다. 최적 병용처리조건은 예비실험을 토대로 확립하였으며, central composite design에 의한 반응표면분석법을 이용하였다. 즉, 독립변수를 ultrasonication time ($x_1$), blanching temperature ($x_2$) 및 blanching time ($x_3$), 종속변수를 ${\Delta}E$ value (y)로 하여 반응표면분석을 실시한 결과, 90초간 ultrasonication한 후 $45^{\circ}C$에서 90초간 blanching (US+LB)하는 것이 색도 변화가 가장 적은 것으로 나타났다. 최적병용처리구와 각각의 단일처리구 및 무처리구를 $10^{\circ}C$에서 9일간 저장하면서 실험을 실시한 결과, 전반적으로 US+LB 처리구가 유의적으로 높은 $L^*$값이 나타났으며, $a^*$, $b^*$, ${\Delta}E$ value, browning index, PPO 및 POD 활성, 총균수에서 유의적으로 낮은 것으로 나타났다(p<0.05). 또한, appearance, color, aroma, freshness, texture, overall acceptibility의 6가지 항목으로 관능평가를 실시한 결과, 모든 항목에서 US+LB 처리구가 가장 우수한 것으로 나타나, 이화학적 실험결과와도 일치하는 경향임을 알 수 있었다. 이로써, 초음파와 저온블랜칭의 병용처리는 신선편이 양상추의 품질 유지에 효과적인 방법인 것으로 확인되었다.

부영양 연못에서 초음파 작동에 따른 식물플랑크톤의 군집 변화 (Change of Phytoplankton Community by Ultrasonication in Eutrophic Ponds)

  • 초음파가 식물플랑크톤에 미치는 영향을 조사할 목적으로, 이화학적 그리고 생물학적으로 유사한 두 군데의 부영양화 연못에서 2003년 8월 18일부터 9월 30일까지 약 4일 간격으로 시료를 채취하여 실험을 실시하였다. 실험기간 동안 출현한 식물플랑크톤 군집은 규조류(Bacillariophyceae)가 거의 대부분을 차지하였으며, 남세균(Cyanophyceae)은 상대적으로 적은 비중을 차지하였다. 분자생물학적 기법중의 하나인 DGGE(denaturing gradient gel electrophoresis)를 이용하여 식물플랑크톤 군집 구조를 비교분석하였다. 16S rDNA의 DGGE profile 분석에 의하여 처리구 연못에서는 Oscillatoria acuminata를 비롯하여 CFB (Cytophaga-Flavobacterium-Bacteroides) group bacterium 등이 나타나는 것으로 확인되었으며, 초음파가 정지된 시기에 특이적으로 Pseudanabaena sp.가 나타나는 것을 확인하였다. 대조구 연못에서는 Synechococcus sp.와 Aphanizomenon flos-aquae가 출현하는 것을 확인하였다. 18S rDNA의 DGGE profile분석결과 처리구 연못에서는 Chlamydomonas rein. hardtii가 우점하는 것으로 확인되었으며, 초음파 처리에 크게 반응하지 않는 것으로 판단된다. 대조구 연못에서는 처리구 연못에서 나타난 C. reinhardtii를 포함하여 Pteromonas protracta가 출현하였다. 결과적으로 초음파는 진핵성 식물플랑크톤 보다는 원핵성 식물플랑크톤의 다양성에 더 큰 영향을 주는 것으로 판단되며, 일부 남세균(Pseudanabaena sp.)의 생장에 저해를 주는 것으로 판단된다.

Effect of ultrasonication, salt solution and liquid smoke treatment on germination of Setaria italica seeds

  • The preference for domestic cereal crop of Setaria Italica has been increased with the recent interest in healthy foods. However, the productivity of Korean domestic produce, which has been decreasing due to the lack of cultivation technology and the low rate of mechanization during cultivation. Increase of germination ability will have a positive effect on the cultivation by reducing the labor of the manpower consuming and weeding work. Therefore, red light, ultrasonication, liquid priming and liquid smoke treatment that are effective for the germination of the seeds evaluated. The seeds of 1.4mm or more were used for the experiment. The priming solution used in the experiment was 1% $KH_2PO_4$ (74mM). During the priming, the light treated seeds at 2000 lux for 15, 30, 60 and 120 minutes. Ultrasonicationd treatment was performed for 5, 10, and 20 minutes at exposures of 60%, 80%, and 100% of ultrasound up to 21.6 kHz during priming. Light or ultrasound treated seeds transferred to priming treatment at $15^{\circ}C$ for 24 hours. The treatment of the liquid smoke was divided into the treatment of the liquid smoke alone and the treatment of the liquid smoke with the priming. The liquid smoke alone was diluted with distilled water without priming solution and the treatment of the liquid smoke was diluted with the salt priming solution. Both treatments were performed at 0.0%, 0.5%, 1.0%, 5.0%, and 10.0% of the liquid smoke (pH7) concentration at $15^{\circ}C$ for 6 hours. After each treatment, the seeds were dried to moisture content ranged 5-8% at $25^{\circ}C$ for 24 hours. All treatments showed better results than the non-treated control. Light treatment for 120 minutes improved for germination percentage (GP), Germination uniformity (GU) and heath seed percentage (HS). Ultrasonication treatment was most effective when treated with ultrasound at 21.6 kHz for 5 minutes in all germination characteristics. Ten % of the liquid smoke increase in 92% GP, 1.8 days MGT, $54%{\cdot}day^{-1}GR$, 0.76 GU and 88% HS comparing to non-treated control (72% GP, 2.3 days MGT, $45%{\cdot}day^{-1}GR$, 1.48 GU, and 63 % HS). This study showed that it is possible to obtain high germination by adding liquid smoke treatment to the seeds supplied to the farmers. The efficacy of light, ultrasonication, inorganic salt priming, and liquid smoke treatment on the seeds found in the experiment will be a positive alternative to labor force problems in the cultivation by improving germination.

Enhanced Yield of Extraction from Gastrodia elata Blume by Ultrasonication and Enzyme Reaction

  • Gastrodia elata Blume (Chunma) belongs to Orchidaceae, which is a perennial parasitic herbaceous plant and grows in the woods of the central provinces of China, Korea and Japan. Recently, the constituents of the tubers of this plant have been investigated by researchers who have revealed the presence of phenolic compounds including gastrodin as a major constituent, together with 4-hydroxybenzaldehyde, 4-hydroxybenzyl alcohol, parishin, 4,4-dihydroxybenzyl sulfoxide, vanillin, vanillyl alcohol, beta-sitosterol, organic acids and polysaccharides, etc. In this study, we used ultrasonicator and two kinds of enzymes for enhancement of extraction yield. We also used electronic nose for the aroma pattern analysis of Chunma extracts. The concentrations of glucose and functional constituents (gastrodin, vanillin, 4-hydroxybenzaldehyde and 4-hydroxybenzyl alcohol) were measured by biochemistry analyzer and HPLC, respectively. Therefore, we showed that the yield of extraction was increased and discomfortable odor was reduced.

Cell Proliferation and Antioxidative Effects of Ultrasonic Coffee Extracts

  • Recently, coffee is the most popular beverage for modern people. A great number of substances are found in coffee beans and have been studied for many years such as aliphatic and aromatic compounds. However, studies on the physiological activity of coffee extracts are insufficient. This study was performed to determine the contents of caffeine and chlorogenic acid in coffee extracts according to the solvent and to investigate the physiological activity of coffee extracts. Coffee extracts were extracted by ultrasonication method with various types of solvents including distilled water, ethanol, and other organic solvents under $50^{\circ}C$ and $80^{\circ}C$. The contents of caffeine and chlorogenic acid in coffee extracts were determined by Liquid Chromatography-Mass Spectrometry (LC-MS). Also, cytotoxic and antioxidative effects of coffee extracts were evaluated with MTT and DPPH assays to analyze the physiological activity. As a result, it was confirmed that caffeine and chlorogenic acid contents were extracted in distilled water with the highest rate. Antioxidative activity was observed below 10-fold dilute of coffee extracts, however cytotoxicity was not observed. In conclusion, distilled water was the best solvent for extracting caffeine and chlorogenic acids from coffee bean with ultrasonication and these coffee extracts are less cytotoxic in human skin cell lines and have antioxidant effect.
