Reduction in Residual Pesticides and Quercetin Yields in Onion Peel Extracts by Washing (세척방법에 따른 양파껍질추출물의 Quercetin수율 및 잔류농약 제거효과)

  • This study was conducted to assess the removal of residual pesticides and to obtain high amounts of quercetin in onion peel extracts (OPEs) by 4 washing treatments. Washing is one of the standard processing steps in obtaining functional food ingredients from onion peel. After a first detergent wash (0.2% w/v) (DW) and hot air drying ($80^{\circ}C$, 24 hr) (B), 4 washing treatments were tested, including a second DW (C), ultrasonication ($50^{\circ}C$, 10 min) plus DW (D), 0.3% $H_2O_2$ (v/v) plus DW (E), and blanching ($95-97^{\circ}C$, 2 min) plus DW (F). This was followed by 60% (v/v) ethanol extraction and vacuum freeze drying of the OPE. The E treatment yielded 89.04% OPE and a quercetin content of 96.84% in the OPE compared with the B treatment, and had the highest efficiency of all treatments tested. The OPE was tested for the presence of 177 residual pesticides and three compounds were detected in all treatments: cyhalothirn, fluquinconazole and procymidone. Cyhalothirn and fluquinconazole levels were below the permitted levels for fresh onion, while procymidone was present in the high level range of 128.01~133.46 mg/kg in all samples. The E treatment was a better washing method than the others for removal of residual pesticides. It could reduce the level of residual pesticides without changing the functional properties of the OPE.

Bio-Derived Poly(${\gamma}$-Glutamic Acid) Nanogels as Controlled Anticancer Drug Delivery Carriers

  • We have developed a novel type of polymer nanogel loaded with anticancer drug based on bio-derived poly(${\gamma}$-glutamic acid) (${\gamma}$-PGA). ${\gamma}$-PGA is a highly anionic polymer that is synthesized naturally by microbial species, most prominently in various bacilli, and has been shown to have excellent biocompatibility. Thiolated ${\gamma}$-PGA was synthesized by covalent coupling between the carboxyl groups of ${\gamma}$-PGA and the primary amine group of cysteamine. Doxorubicin (Dox)-loaded ${\gamma}$-PGA nanogels were fabricated using the following steps: (1) an ionic nanocomplex was formed between thiolated ${\gamma}$-PGA as the negative charge component, and Dox as the positive charge component; (2) addition of poly(ethylene glycol) (PEG) induced hydrogen-bond interactions between thiol groups of thiolated ${\gamma}$-PGA and hydroxyl groups of PEG, resulting in the nanocomplex; and (3) disulfide crosslinked ${\gamma}$-PGA nanogels were fabricated by ultrasonication. The average size and surface charge of Dox-loaded disulfide cross-linked ${\gamma}$-PGA nanogels in aqueous solution were $136.3{\pm}37.6$ nm and $-32.5{\pm}5.3$ mV, respectively. The loading amount of Dox was approximately 38.7 ${\mu}g$ per mg of ${\gamma}$-PGA nanogel. The Dox-loaded disulfide cross-linked ${\gamma}$-PGA nanogels showed controlled drug release behavior in the presence of reducing agents, glutathione (GSH) (1-10 mM). Through fluorescence microscopy and FACS, the cellular uptake of ${\gamma}$-PGA nanogels into breast cancer cells (MCF-7) was analyzed. The cytotoxic effect was evaluated using the MTT assay and was determined to be dependent on both the concentration and treatment time of ${\gamma}$-PGA nanogels. The bio-derived ${\gamma}$-PGA nanogels are expected to be a well-designed delivery carrier for controlled drug delivery applications.

Analysis of Barbaloin in the Aloe vera Depending on the Various Extracting Conditions (추출조건(抽出條件)에 의(依)한 알로에 베라의 Barbaloin 분석)

  • Barbaloin in the Aloe vera depending on the various extracting conditions was analyzed by HPLC. The contents of the barbaloin extracted by the solvents increased in the order of methanol>ethanol>water extraction. In setting extraction, the contents of barbaloin extracted with methanol and ethanol were increased from four hours at $60^{\circ}C$ and then decreased. The contents of barbaloin extracted with water were different depending on the temperature and time. Increasing the extracting time and temperature, the contents of barbaloin were decreased in water extract. It was estimated that the barbaloin might be stable in organic solvent, but decomposed with hydrolysis in water.

Effects of Egg White and Ion Exchange Resin Pretreatment on Separation of Egg White Lysozyme (난백 및 이온교환수지의 전처리 조건이 난백 Lysozyme의 추출에 미치는 영향)

  • Pretreatment of egg white and ion exchange resins was attempted to separate lysozyme from egg white efficiently. Apparent viscosity of egg white could be decreased to 3cp by homogenization for 30 minutes at 2, 000rpm and ultrasonication for 45 minutes. The result of testing adsorption capacity of lysozyme was as follows; CM-Sephadex C-25 >Duolite C464>Amberlite C-50>Dowex MSC-1>Amberlite IRC-50>Amberlite IRC-84. Although CM-Sephadex C-25 showed highest adsorption capacity of lysozyme, egg white could not eluted easily. Duolite Cf64 was selected based on relatively high lysozyme adsorption and good egg white eluting property for separation of egg white lysozyme. Na$^{+}$ form of Duolite C-464 was most effective on adsorption of Iysozyme. To separate lysozyme from egg white efficiently rinse buffer and eluting solution were selected 0.1M sodium phosphate buffer at pH 6.5 and 10% ammonium sulfate respectively. After separating lysozyme from egg white, foaming power of egg white was decreased to 85.3%. Color of egg white gel was not changed while hardness of egg white gel was decreased by 30% after separating lysozyme. However, elasticity of egg white gel was increased by 13% in lysozyme-separated egg white.

Blastogenic responses of splenic Iymphocytes to Naegleria fowleri Iysates and T-cell mitogen in mice with primary amoebie meningoencephalitis. (실험적 뇌막수염에 있어 Naegleria fowleri 항원에 대한 세포매개성 면역 반응)

  • This study was to observe the changes of blastogenic responses of splenic Iymphocytes to T-cell mitogens, N. fcwleri Iysate and concanaualin A, and serum antibody titer during the course of experimental PAM in mice. Naegleria fcwleri, strain 0359, was cultured in the CGVS medium axenically and inoculated intranasally with $7{\times}10^4$ trophozoites for the development of experimental PAM in mice. The amoebae were subjected to ultrasonication and centrifuged at 20,000g for 60 minutes, and filtered through $0.2{\mu\textrm{m}}$ filter membrane. The supernatant, N. fcwleri Iysate, was used as T-cell mitogen, and antigen for ELISA. The serum antibody was examined by ELISA using peroxidase conjugate. Two hundred ${\mi}l$ of $10^6$ splenocytes in RPMI 1640 containing 0% fetal calf serum were added to each well of a microtiter plate. To each well was added T-cell mitogens, $100{\mu}g/ml$ of N. fowleri Iysate or $4{\mu}g/ml$ of con. A, and the plates were incubated for 42 hours at $37^{\circ}C$ in 5% $CO_2$ incubator. Cultures were pulsed with of $methyl-(^3H)-thymidine$ 6 hour before harvesting. The mean blastogenic response of the splenocytes to N. fewleri Iysate was reduced, whereas that to con. A was also reduced up to on day 11 after infection. Both of these results were statistically significant compared with those of uninfected control group. The serum antibody titers were increased gradually up to day 15. The results indicated that there was an impairment of the blastogenic response of splenocytes to N. fowleri Iysate during the acute course of experimental PAM in mice.

Evaluation of Growth Inhibition for Microcystis aeruginosa with Different Frequency of Ultrasonic Devices (초음파 장치의 주파수 변화에 따른 Microcystis aeruginosa의 성장억제 평가)

  • The growth inhibition effects of M. aeruginosa were verified using large volume (7.2 L) of algae samples and ultrasonication (high frequency of 1.6 MHz vs. low frequency of 23 kHz) in lab-scale experiment. The chlorophyll-a (chl-a) and cell number decreased gradually after 6 hr sonication with high frequency of 1.6 MHz whereas both decreased sharply after 6 hr sonication with low frequency of 23 kHz. Additionally, the first order degradation coefficient (k) values after sonication were greater than those during sonication. These results indicate that relatively low sonication energy per volume may affect the cell membrane and internal organs of M. aeruginosa in a slow and retarded manner and resulted in gradual decrease of cell numbers of M. aeruginosa. Based on the comparison of chl-a and cell number of M. aeruginosa after sonication, low frequency of 23 kHz is superior for growth inhibition of M. aeruginosa, since low frequency of 23 kHz easily penetrates the cell membrane and ruptures the internal organs including gas vesicles. As is evident in SEM and TEM images, ruptured cell membranes were clearly observed for low frequency of 23 kHz. Finally, the microcystin-LR in water is not detected and considered to be harmless in aquaculture systems.

Comparison of the stability between branched-chain amino acid (BCAA)-coated liposome and double emulsion (분지쇄아미노산(BCAA)이 포집된 더블에멀션과 리포좀의 안정성 비교)

  • This study was conducted to compare the stability between branched-chain amino acid (BCAA)-encapsulated liposome and double emulsion (DE). Liposome was produced by high-speed homogenization and ultrasonication whereas DE was prepared by homogenizing with surfactants. All samples were fixed at pH 4 and 7 and stored at 4, 25, and $40^{\circ}C$ for 5 days. Encapsulation efficiency and cumulative release rate were measured under $4^{\circ}C$ and at $25^{\circ}C$. The results showed that the size of BCAA-coated liposome was greater at pH 7 than at pH 4. The zeta-potential value of BCAA-coated liposome was greater at pH 4 than at pH 7. It was supposed that the negatively charged liposomes attracted the positively charged BCAAs at pH 4 resulting in the formation of the vesicle with smaller size. Particle size of DE was smaller than $100{\mu}m$. Encapsulation efficiencies of BCAA in DE or liposome were over 97%, approximately, and the cumulative release rates of them were below 30% for 5 days.

Accelerating Effects of Ultrasonic Irradiation on Reaction Rates for the Asymmetric Ring Opening Reaction of Epoxides (초음파 조사에 의한 에폭사이드 비대칭 고리열림 반응의 속도 증진 효과)

  • In this study, effects of the ultrasonic irradiation during the reaction process were investigated for the enantioselective kinetic resolution (EKR) reaction of racemic epoxides in the presence of chiral cobalt salen catalysts, as compared to that of using the conventional mechanical stirring. In order to compare catalytic activities, the chiral cobalt salen complexes having $AlCl_3-$, $BF_3-$ and nitrobenzenesulfonic acid (NBSA) were synthesized and used as catalysts, and then three kinds of the racemic epoxides such as ephichlorohydrine (ECH), epoxy phenoxypropane (EPP) and propylene oxide (PO) were used as reactants. In addition, EKR reactions have been performed using the water and methanol as nucleophiles, respectively. The unique contribution of ultrasonic irradiation as a powerful mixing medium was evaluated in this study to improve the kinetics in comparison to the conventional mechanical agitation during EKR reactions. The reaction time to obtain the highest 99 ee% became shorten more than that of above 60%, when the ultrasonic irradiation was used. This result may be interpreted by the cavitation effect of ultrasound in the solution, generating a powerful shear force for the very violent mixing.

Variation of Pesticide Residues in Strawberries by Washing and Boiling Processes (딸기의 세척 및 가공 과정에 따른 농약 잔류량 변화)

  • BACKGROUND: As the demand for strawberries increases, people are paying attention to food safety in strawberry, especially pesticide residues. To remove the pesticides from strawberry, various washing and processing technique in households are additionally required. METHODS AND RESULTS: Strawberries were washed with water, detergent, alcohol, and ultrasonication and processed to strawberry jam. The mean reduction efficiency according to the washing solvent and method was found to be higher in the order of detergent (42.5%) > alcohol (41.7%) > water (41.3%) > ultrasoniation with alcohol (40.2%) > ultrasoniation with water (38.6%) > ultrasoniation with detergent (36.9%), but there was no significant difference among the treatments. The residue levels of pesticides during processing to jam decreased by 11.9-94.4% for etoxazole, fluopyram, procymidone, spiromesifen, and prochloraz, while the other pesticides were concentrated by boiling, or rather increased by 11.8-40.2%. However, when the residue levels were converted to residual amounts in consideration of the change in weight after processing, the residual amounts of the tested pesticides were reduced by 59.8-98.4% during processing. The processing factor (PF) were different for each pesticide, but PFs were < 1 for all washing solvents and methods, and 0.06-1.40 when processed into jam. CONCLUSION: To ensure the consumption of pesticide-free strawberry, the most efficient washing method is to immerse the strawberry in fresh water for few minutes, followed by rinsing them under running water.

Effect of Splenectomy on Development of Primary Amoebic Meningoencephalitis (비장적출(splenectomy)이 원발성 아메바성 뇌수막염의 발생에 미치는 영향)

  • To elucidate the effect of splenectomy on the development of experimental primary amoebic meningoencephalitis in mice, the death rate and survival time of mice infected intranasally with Naegleria fowleri trophozoites $5{\times}10^4$ cultivated in CGVS medium were compared according to the mouse age when splenectomy was done, and post-operation until experimental infection. Immunodigusion was undergone to detect the presence of serum antibod). due to N, fowleri infecttion in mice. Polyacrylamide gel electrophoresis was done to compare the protein fractions of mouse serum in each experimental groups. In experiment I, splenectomy was done 3 weeks and infection 4 weeks after birth, the death rate of control, sham operated and splenectomized group were 100%, 85% and 95%, and the mean survival time after infection 7.3 days, 7.5 days and 7.8 days, respectively. In experiment II, splenectomy was undergone 3 weeks and infection 6 weeks after birth, the death rate of of control, sham operated and splenectomized group were 95%, 95% and 95%, and the mean survival time after infection 12.1 days, 11.5 days and 11.5 days, respectively. In experiment III, splenectomy was done 5 weeks and infection 6 weeks after birth, the death rate of control, sham operated and splenectomized group were 95%, 90% and 95%, and the mean survival time after infection 8.1 days, 8.3 days and 8.5 days, respectively. By Ouchterlony immunodigusion, anti-JV. fowleri antibody in the serum of mouse with primary amoebic meningoencephalitis was detected against a N. fowleri antigen, which was prepared by ultrasonication of N, fowleri trophozoites, each reacting two lines of precipitation. The patterns of serum fractions by polyacrylamide gel electrophoresis were different between control and sham operated groups from splenectomized group in fraction II, III and V, the sera of which were collected after N. fowleri infection. This results may be summarized as that splenectomy has no effect on the development of primary amoebic meningoencephalitis in mice.
