유근피 추출물의 피부개선효과 (Anti-Wrinkle Effect of Ulmus davidiana Extracts)

  • The bark of the root and stem of Ulmus davidiana var. japonica has been used as a traditional Korean medicine to treat inflammatory disorders. This plant reportedly shows antioxidant, anticancer, and anti-inflammatory effects. In this study, we investigated the protective effects of Ulmus davidiana var. japonica ethanolic extract (UDE) on UVB irradiation-induced wrinkle in hairless mice. We evaluated for their free radical-scavenging activities against 1,1-diphenyl-2-picrylhydrazyl (DPPH) and the anti-elastase activities, and for their anti-matrix metalloproteinase-1 (MMP-1) activity in human skin fibroblast cells. In the wrinkle measurement and image analysis of skin replicas, the results showed that UDE significantly inhibited wrinkle formation caused by chronic UVB irradiation. These results suggest that UDE has anti-wrinkle activity.

Anti-Obesity and Lipid Metabolism Effects of Ulmus davidiana var. japonica in Mice Fed a High-Fat Diet

  • The root bark of Ulmus davidiana var. japonica (Japanese elm) is used in Korea and other East Asian countries as a traditional herbal remedy to treat a variety of inflammatory diseases and ailments such as edema, gastric cancer and mastitis. For this study, we investigated the lipid metabolism and anti-obesity efficacy of ethyl alcohol extract of Ulmus davidiana var. japonica root bark (UDE). First, HPLC was performed to quantify the level of (+)-catechin, the active ingredient of UDE. In the following experiments, cultured 3T3-L1 pre-adipocytes and high-fat diet (HFD)-fed murine model were studied for anti-obesity efficacy by testing the lipid metabolism effects of UDE and (+)-catechin. In the test using 3T3-L1 pre-adipocytes, treatment with UDE inhibited adipocyte differentiation and significantly reduced the production of adipogenic genes and transcription factors PPARγ, C/EBPα and SREBP-1c. HFD-fed, obese mice were administered with UDE (200 mg/kg per day) and (+)-catechin (30 mg/kg per day) by oral gavage for 4 weeks. Weight gain, epididymal and abdominal adipose tissue mass were significantly reduced, and a change in adipocyte size was observed in the UDE and (+)-catechin treatment groups compared to the untreated control group (***p < 0.001). Significantly lower total cholesterol and triglyceride levels were detected in UDE-treated HFD mice compared to the control, revealing the efficacy of UDE. In addition, it was found that lipid accumulation in hepatocytes was also significantly reduced after administration of UDE. These results suggest that UDE has significant anti-obesity and lipid metabolism effects through inhibition of adipocyte differentiation and adipogenesis.

Suppressive effects of a water extract of Ulmus davidiana Planch (Ulmaceae) on collagen-induced arthritis in mice

  • Objective: Since Ulmus davidiana Planch (Ulmaceae) has been known to have anti-inflammatory and protective effects on damaged tissue, inflammation and bone among other functions, this study was undertaken to address whether the water extract of the bark of Ulmus davidiana Planch (Ulmaceae) (UD) could modulate the expression of inducible inflammatory cytokines in mice. The present study was also done in order to assess the therapeutic effects of UD in collagen-induced arthritis (CIA) in mice. Methods : DBA/1 mice were immunized with bovine type II collagen. After a second collagen immunization, mice were treated with UD orally at 100 mg/kg once a day for 3 weeks. Paws were evaluated macroscopically for redness, swelling and deformities. The levels of $TNF-{\alpha}\;and\;IL-1{\beta}$ in the ankle were examined. The severity of arthritis within the knee joints was evaluated by histological assessment of cartilage destruction and pannus formation. Results : Administration of UD significantly suppressed the progression of CIA and inhibited the production of $TNF-{\alpha}\;and\;IL-1{\beta}$ in the paws. The erosion of cartilage was dramatically reduced in mouse knees after treatment with UD. In the serum of UD-treated mice, the levels of IL-4 and IL-10, anti-inflammatory cytokines, were increased. Conclusion : From the results, it was concluded that administration of UD has therapeutic effects on CIA including protection of cartilage and RA for a potential therapy.

Effect of Alcohol Insoluble Residues from Stem and Root Barks of Elm (Ulmus davidiana) on Intestinal Characteristics in Rats

  • Sprague-Dawley rats (n=32) were fed a diet containing basal (control), cellulose (5%), or alcohol insoluble residue (AIR) (5%) extracted from the stem and root barks of elm (Ulmus davidiana var. japonica Nakai) for 4 weeks. The effects of the diets, on gastrointestinal functions and morphology were evaluated. The weight gains, food intake, and food efficiencies for the cellulose and AIR diet-fed groups were not significantly different from those of the AIR-free (basal) diet. The gastrointestinal transit times of the stem and root bark AIR diets were significantly reduced (p<0.01) compared to the basal diet, and were slower than those of the cellulose diet. The fecal weights of the stem and root bark AIR diets were significantly increased (p<0.01) up to 4-fold compared to those of the basal diet. The height of the mucosal villi, and mucosal and muscle layer thicknesses of the colon were greater and more developed in the stem and root bark AIR diets (p<0.01) than in the basal diet. The villus heights in the jejunum and the colon mucosal goblet cells were more developed in the order of cellulose > stem bark AIR > root bark AIR diets.

Distribution of the Genetic Resource and the Biomass of Root Bark of Ulmaceae Species

  • Stem and root of elm trees have used as traditional medical materials, but there is little information on the distribution and resources of habitats. Korean native growing Ulmus spp. (U. davidiana var. Japonica, U. parvifolia, U. davidiana, and U. macrocarpa) genetic resources studied through The National Forest Inventory of Korea data and field survey. The distributions of U. davidiana var. japonica according to elevation distributed evenly. Both U. parvifolia and U. davidiana were inhabited mostly at less than 200 m of altitude. Each Ulmaceae species widely were distributed nationwide, but a dominant species was different depending on locals. It observed that Ulmaceae inhabits mainly in steep slopes of 31-45 degrees. Most of the habitats regenerated by natural seeding and the most abundant species were a codominant tree. Distribution of trees in U davidiana var. japonica was 7 m-13 m, and in young U. parvifolia and U. macrocarpa, more than 25% of young trees less than 7 m observed. The distribution of the diameter of breast height of the U. davidiana var. japonica was 46.4% for 11-20 cm, 52.6% for 11-20 cm in U. parvifolia. The average T/R ratio was 0.83, and the mean weight ratio of root bark was 62%. As the results of this study, the domestic Ulmaceae biomassare very small. It is difficult to harvest in that the habitat on the slope. Thus, it is too hard to develop functional materials using biomass at present. Therefore, it is necessary to develop technology for the selection and propagation of elite trees of Ulmaceae.

Inhibitory Action of Ulmus Davidiana Planch Extract Solution to Osteoclast Cell Proliferation and Prostaglandin E2 Synthesis in Mice

  • Objectives : Ulmus davidiana Planch (UD) has long been known to have anti-inflammatory and protective effects on damaged tissue, inflammation and bone among other functions. Methods : This study was undertaken to address whether the water extract of the bark of UD could modulate proliferation of mouse osteoclasts in vitro and to investigate its effect on cyclooxygenase-2 (COX-2), which converts arachidonic acid to prostaglandin E2 (PGE2) and is highly expressed in osteoclasts. Mouse osteoclasts were tested in vitro for growth inhibition, proliferation cell nuclear antigen expression, and COX-2 activity and expression after treatment with UD extract. Results : Its effects were compared with those of indomethacin (a nonselective COX inhibitor) and celecoxib (a selective COX-2 inhibitor) by Cell viability assay, Cell cycle analysis, Immunohistochemical analysis of PCNA expression, Western blot analysis and PGE2 Enzyme immunoassay (EIA). UD demonstrated a strong growth inhibitory action in both tested osteoclasts cells. The IC50s were $10\;{\mu}g/ml$ for UD, $6\;{\mu}M$ for celecoxib and $42\;{\mu}M$ for indomethacin. UD, as well as celecoxib and indomethacin, suppressed proliferation cell nuclear antigen expression and PGE2 synthesis in osteoclasts. UD inhibited COX-2 expression, whereas celecoxib inhibited COX-2 activity directly. Conclusion : UD selectively and effectively inhibits osteoclasts cell growth in vitro. Inhibitory action of PGE2 synthesis via suppression of COX-2 expression may be responsible for its anti-inflammatory activity.

유근피(楡根皮)에 존재하는 matrix metalloproteinase-9 억제 물질의 분리 및 정제 (Separation and purification of substance having matrix metalloproteinase-9 inhibition effect in Ulmus davidiana Plancn. var. japonica Nakai)

  • 극성이 다른 여러 가지 용매를 사용하여 느릅나무의 뿌리 껍질(이하 유근피)을 추출하였다. MeOH 추출물이 SK-Hep-1에서 분리된 matrix metalloproteinase-9(MMP-9)의 zymography에서 저해능을 나타내었다. 분리 정제한 물질의 분자량은 GC-MS spectrum에서 $M^+=281$인 것으로 나타났고 MMP-9의 활성은 $314.7{\mu}g/g$에서 47% 억제되는 것으로 나타났다. 그리고 SK-Hep-1 세포주는 $31.47{\mu}g/g$에서 60%의 생존율을 나타났다.

유근피와 유백피 추출액의 유지에 대한 항산화 효과 (Antioxidant Effects of Ulmus davidiana Extracts on Various Oil)

  • Fish oil에 대한 유근피 및 유백피 추출물의 과산화물가를 측정한 결과 유근피는 모든 용매의 추출물에 대해 항산화 작용이 높게 나타났다. 0.1% 추출물 첨가구 모두 저장 6시간에는 30 meq/Kg을 보인 후 최종 18시간까지 40 meq/Kg이하의 과산화물가를 보여 저장 시간이 증가해도 산화 억제효과가 높게 나타났다. 0.05%의 경우 부탄올과 메탄올 추출물은 상업용 항산화제와 비교할 때 유사한 항산화 효과를 나타내었다. 유백피는 0.1% 첨가군에서 물, 부탄올 추출물이 가장 항산화 효과가 우수하였다. 금속이온 첨가에 따른 각 추출물의 항산화 효과는 유근피의 경우, 메탄올과 부탄올 추출물 0.1% 첨가군이 산화 억제효과가 높게 나타났다. 유백피는 에탄올 추출물이 가장 우수한 항산화 효과를 나타내었으며, 저장 시간 동안의 과산화물가의 변화는 거의 나타나지 않았다. Perilla oil에서는 천연 항산화제인 $\alpha$-tocopherol의 항산화능이 전혀 나타나지 않았으나, BHT는 항산화 효과가 나타났다. 유근피 0.2% 첨가군에서 메탄올, 부탄올 추출물군은 거의 산화가 진행되지 않아 가장 항산화력이 우수하였다. 유백피 역시 0.2% 메탄올 추출물에서 높은 항산화력을 나타났다. 금속이온 첨가에 의해서는 모든 추출물 중 유백피 0.2% 메탄올 추출물이 가장 우수하였다. Lard에서는 각 용매 추출물 농도별 (0.05, 0.1%) 첨가군에 따른 각 추출물간의 항산화 효과에 따른 차이는 크지 않았으며. 금속이온 첨가에서도 0.05, 0.1% 첨가에 따른 차이는 크지 않았다.

유근피내의 Matrix Metalloproteinase-9 활성 억제제에 관한 연구 (A Study of matrix metalloproteinase-9 inhibitor in root bark of ulmus davidiana planchon)

  • 70% 에탄올로 추출한 후 농축한 유근피 추출물을 여러 용매로 분획한 부분들의 MMP-9 활성 저해능 실험에서 ethyl acetate 분획분이 MMP-9 활성 저해능이 있는 것으로 나타났다. 이 분획분에서 MMP-9의 활성에 저해능을 나타내는 물질을 분리 정제하여 분석하였고 이 물질은 catechin 계열로 판명되었다. Zymography법에 의해 MMP-9의 활성 억제 효과를 관찰하였고 정제된 화합물은 MMP-9을 5 mM 농도에서 48%가 억제하였고 10 mM 에서는 43%를 억제하는 것으로 나타났다. 정상 세포주인 Chang 세포주와 인체 간암세포인 SK-Hep-1 세포주를 동일한 조건으로 배양한 후에 형태학적 변화를 관찰한 결과, 정상 세포주의 핵과 그 주변에서는 변화가 거의 관찰되지 않았으나 SK-Hep-1 세포주는 핵의 주위에 검은 반점이 관찰되었다. 세포의 증식과 살아있는 세포를 측정하기 위하여 MMP-9 억제물질인 정제된 추출물을 투여하고 MTT 검색법에 의해 ELISA reader로의 측정에서 정상세포주인 Chang 세포주에서는 1 mM에서 세포 활성이 오히려 증가하는 경향($130.77{\pm}13.71$)을 보인 반면, SK-Hep-1 세포주에서는 세포 증식이 상당히 억제된 것으로 나타났다.

삼칠근(三七根)의 형태(形態)에 관한 연구(硏究) (A Study on a Morphological Identification of Notoginseng Radix)

  • Objectives : In order to distinguish morphological characteristics of trunk bark and root bark of Ulmus davidiana var. japonica (Rehder) Nakai and the trunk bark and root bark of Hemiptelea davidii Planchon were sampled and compared in terms of their external and internal features with flour states according to their medical use, through microscopic examination. Methods : The slice of the tested material made by paraffin section technique was colored with Safranine Malachite Green contrast methods, and the flour of it was mounted by the liquid made by the same ratio of each of glycerin, acetic acid, and water, and then observed and photographed by olymphus-BHT. Results : 1. Internal Features 1) A large parenchymatous cell was observed in the phloem of the slice of both trunk bark and root bark of Ulmi Cortex, However, both of the trunk bark and root bark of Hemipteleae Cortex did not have parenchymatous cell in the phloem; instead, stone cells including much square crystal of calcium oxalate were distributed around fiber bundle, and the parenchymatous cell included much druse crystal of calcium oxalate. 2) In both the Ulmi Cortex and Hemipteleae Cortex, rhytidome was observed in trunk bark, but not in root bark, but in the parenchymatous cell of the root bark of the Ulmi Cortex contained starch grain. 2. Flour States 1) In the flour of root bark of the Ulmi Cortex, a large parenchymatous cell was observed. However, in the flour of trunk bark and root bark of Hemipteleae Cortex, no parenchymatous eel was found; instead, stone cell including square crystal of calcium oxalate and druse crystal of calcium oxalate were observed. 2) There was no remarkable difference between the trunk bark and root bark of Hemipteleae Cortex. However, starch grain was contained in the parenchymatous cell of the root bark of Ulmi Cortex but not in the trunk bark of it. Conclusions : There were some morphological differences in external, internal, and flour parts of Ulmi Cortex and Hemipteleae Cortex. In particular, there was a morphological difference in flour states between the trunk bark and root bark of Ulmi Cortex, it is possible to use microscope to distinguish their flour states.
