녹차추출물 성분 catechin이 자외선에 의해 손상된 피부에 미치는 영향 (Protective Effect of Green Tea Extract, Catechin on UVB-Induced Skin Damage)

  • 녹차의 주요 성분인 catechin은 항돌연변이, 항종양, 항균, 항바이러스 활성 및 콜레스테를 저하, 혈압 상승 억제, 해독, 방사선 차단 작용 등과 같은 다양한 생물학적 작용들이 보고되고 있다. 최근 환경 오염의 급증으로 인한 오존층의 파괴로 지표에 도달하는 자외선이 증가됨에 따라 피부노출 위험이 증가하고 있다. 따라서 피부암의 발생이 증가하는 추세이며 역학적 및 실험실적 연구에서 자외선 조사가 가장 큰 원인이라고 보고되고 있다. 이에 본 연구는 catechin이 UVB에 의해서 손상된 피부조직에 미치는 영향을 연구하였고 광발암과정에서 catechin의 억제효과가 apoptosis에 어떤 영향을 주어 작용하는지를 in vivo hairless mouse skin에서 연구하였다. 녹차잎에서 추출한 catechin을 hairless mouse를 대조군, catechin을 처리한 군, 자외선을 처리한 군, catechin과 자외선을 처리한 군으로 나누었으며 catechin의 농도는 3mg/mouse로 피부에 도포한 후 자외선을 100 mJ/$ extrm{cm}^2$ 조사하였다. 자외선에 의한 피부조직에 미치는 catechin의 영향을 H&E stain을 실시하여 확인하였고, apoptosis에 미치는 catechin의 보호효과를 알아보기 위해 DNA laddering 형성 여부와 TUNEL assay로 확인하였으며 western blot을 이용하여 apoptoic activity와 관련된 단백질인 p53연구하였다. 또한 자외선에 의한 세포증식에 미치는 catechin을 영향을 bromodeoxyuridine(BrdU) 면역염색방법을 이용하여 살펴보았다. 본 연구결과 자외선에 의하여 피부는 진피층의 교원섬유의 증가가 정상피부보다 2~3배 비후되었고, 염증세포의 침윤과 섬유화세포 등이 관탈되었으며 표피층은 두꺼워졌으나 상피세포가 소실되어 1~2층으로 되었다. UVB조사와 catechin을 2주간 처리한 후의 mouse의 피부는 표피층이 9~12층으로 이루어져 있었고, 세포의 핵은 과염색성을 보이며 커져있어 손상된 표피층 상피세포가 재생되고 있음을 확인하였다. Apoptosis에 미치는 결과에서 UVB 조사군은 apoptosis의 대표적 특징인 DNA fragmentation이 관찰되었고, UVB와 catechin을 처리한 군에서는 생성되지 않아 뚜렷한 보호효과를 나타내었다. TUNEL assay를 실시한 결과 UVB군과 catechin 군을 비교하였을 때 UVB+catechin군에서 apoptotic cells이 유의성 있는 차이(p<0.05)를 보였다. Apoptosis 관련 단백질인 p53발현을 살펴본 결과 대조군과 비교해볼 때 UVB군과 UVB와 catechin을 처리한 군간에서는 차이가 없었다. 자외선에 의한 세포증식에 미치는 catechin의 영향을 살펴본 결과 BrdU에서는 UVB군과 UVB+catechin군을 비교하였을 때 UVB+catechin군에서 BrdU 세포수가 유의성 있는(p<0.05) 감소를 보였다. 본 연구 결과에 의해 녹차의 주요성분인 catechin은 자외선에 의해 유도되는 세포증식 및 apoptosis를 억제하고, 세포손상, 세포증식 및 apoptosis를 억제시킴을 확인하였다.

Protection of the Flowers of Prunus persica Extract from Ultraviolet B-Induced Damage of Normal Human Keratinocytes

  • For an attempt to develop safe materials protecting solar ultraviolet (UV)-induced skin damage, plant extracts were evaluated for their inhibitory activities of free radical generation and arachidonic acid/metabolites release from UVB-irradiated normal human keratinocytes. From the results of these screening procedures, the ethanol extract of the flowers of Prunus persica (Ku-35) was selected for further study. It was found that Ku-35 (100-1,000 ${u}g/m\ell$) inhibited the amount of $^{14}C$-arachidonic acid/metabolites release from UVB-irradiated keratinocytes. It was also demonstrated that Ku-35 possessed the protective activity against UV-induced cytotoxicity of keratinocytes and fibroblasts. In addition, Ku-35 was revealed to protect UVB-induced erythema formation using guinea pigs in preliminary in vivo study. All these results indicate that the flowers of P. persica extract may be beneficial for protecting UV-induced skin damage when topically applied.

Anti-wrinkling effects of "L-Skin Care" and molecular mechanisms on hairless mouse skin caused by chronic ultraviolet B irradiation.

  • Background: Naturally occurring antioxidants were used to regulate the skin damage caused by ultraviolet (UV) radiation because several antioxidants have demonstrated that they can inhibit wrinkle formation through prevention of matrix metalloproteinases (MMPs) and/or increase of collagen synthesis. We examined the effect of oral administration of the antioxidant mixture ("L-Skin Care") on UVB-induced wrinkle formation. In addition, we investigated the possible molecular mechanisms of photoprotection against UVB through inhibition of collagen-degrading MMP activity or through enhancing of pro collagen synthesis in mouse dorsal skin. Methods: Female SKH-l hairless mice were orally administrated "L-Skin Care" (test group) or vehicle (control group) for 10 weeks with UVB irradiation by three times a week. The intensity of irradiation was gradually increased from 30 to $180mJ/cm^2$. Microtopographic and histological assessments of the dorsal skins were carried out at the end of 10 weeks to evaluate wrinkle formation. Western blot analysis and EMSA were also carried out to investigate the changes in the balance of collagen synthesis and collagen degradation. Results: Our "L-Skin Care" significantly reduced UVB-induced wrinkle formation, accompanied by significant reduction of epidermal thickness, and UVB-induced hyperplasia, acanthosis and hyperkeratosis. Oral administration of "L-Skin Care" significantly prevented UVB-induced expressions of MMPs, mitogen-activated protein (MAP) kinases and activation of activator protein (AP)-1 transcriptional factor in addition to enhanced type I procollagen and transforming growth factor-$\beta$ (TGF-$\beta$) expression. Conclusion: Oral administration of "L-Skin Care" significantly inhibited wrinkle formation caused by chronic UVB irradiation through significant inhibition of UVB-induced MMP activity accompanied with enhancement of collagen synthesis.

Hairless 마우스에서 대추 열수추출물의 피부내 염증 및 장벽 손상 완화 효과 (Alleviative Effects of Jujube Water Extract on the Inflammation and Barrier Damage in Hairless Mice Skin)

  • To investigate the alleviative effects of jujube water extract (JWE) on the inflammation and skin barrier damage, both the irradiation of UVB and the application of squalene monohydroperoxide (Sq-OOH) were applied to the back skin of experimental animals for 4 weeks. And at the same time experimental materials were applied topically. Six weeks female SKH-1 hairless mice were divided into five groups (five animals for each group) including normal (N; saline), control (C; UVB+Sq-OOH+saline), vehicle control (VC; UVB+Sq-OOH+vehicle), positive control (PC; UVB+Sq-OOH+0.01% retinoic acid) and experimental (E; UVB+Sq-OOH+JWE) groups. The skin erythema index in the E group was significantly low compared to the C group (p<0.05). Lipid (p<0.05) and water (p<0.01) capacities in the E group were significantly high compared to the C group. In comparison with the C group, E group showed a relatively well preserved lipid lamellae in the epidermis and a relatively much less infiltration of mast cells in the dermis or hypodermis. As for the both absolute and relative weights of the spleen, PC group were significantly higher than the other groups. These results suggest that JWE have a considerably inhibitory effect on the inflammation and the skin barrier damage induced by UVB irradiation and Sq-OOH application.

Galangin (3,5,7-Trihydroxyflavone) Shields Human Keratinocytes from Ultraviolet B-Induced Oxidative Stress

  • Most skin damage caused by ultraviolet B (UVB) radiation is owing to the generation of reactive oxygen species. Phytochemicals can act as antioxidants against UVB-induced oxidative stress. This study investigated the protective effects of the flavone galangin against UVB-induced oxidative damage in human keratinocytes. Galangin efficiently scavenged free radicals and reduced UVB-induced damage to cellular macromolecules, such as DNA, lipids, and proteins. Furthermore, galangin rescued cells undergoing apoptosis induced by UVB radiation via recovering mitochondrial polarization and down-regulating apoptotic proteins. These results showed that galangin protects human keratinocytes against UVB radiation-induced cellular damage and apoptosis via its antioxidant effects.

피부세포에서 옥돔 비늘로부터 추출한 펩타이드의 UVB에 대한 산화적 손상 및 광 노화 억제 (Peptides-derived from Scales of Branchiostegus japonicus Inhibit Ultraviolet B-induced Oxidative Damage and Photo-aging in Skin Cells)

  • 생체에서 가장 많은 비율로 분포하고 있는 콜라겐 펩타이드는 동물의 뼈와 해양 생물의 비늘에 많이 함유되어 있다. 콜라겐은 동물 생체의 여러 결합조직에서 구조 단백질로 흔하게 발견된다. 또한, 이들은 생물 의료 자재, 제약, 화장품, 식품 및 가죽 산업에 널리 이용된다. 각종 어류 비늘에서 추출된 펩타이드는 UVB 조사에 의해 유도된 피부의 손상 및 광 노화에 대한 보호 효과가 있었다. 그럼에도 불구하고, UVB 조사에 대한 옥돔 비늘 유래의 펩타이드 특성은 명확히 알려져 있지 않다. 이 연구에서는 옥돔 비늘 추출물에서 분리된 1 kDa 이상(HMP)과 1 kDa 이하(LMP)의 펩타이드를 이용하여 UVB 조사에 의해 유도된 피부 손상과 광 노화에 대한 효과를 연구하였다. 이들 펩타이드는 농도 의존적으로 DPPH 라디칼 소거능을 보였으며 LMP는 HaCaT 인간 피부세포에서 UVB 조사에 의해 유도된 세포 지질 과산화 산물인 8-isoprostane 생성을 억제하였다. 그리고 LMP와 HMP는 B16F10 마우스 흑색종 세포에서 tyrosinase 활성 및 melanin 함량을 감소시켰으며 또한 HaCaT 세포에서 UVB로 유도된 elastase 활성을 감소시켰고 matrix metalloproteinase-1의 활성을 감소시켰다. 이러한 결과는 옥돔 비늘에서 유래된 펩타이드가 미백효과, 항산화제 및 광 노화 억제제로서 유용한 물질이 될 것으로 기대된다.

Adenophora remotiflora protects human skin keratinocytes against UVB-induced photo-damage by regulating antioxidative activity and MMP-1 expression

  • BACKGROUND/OBJECTIVES: Chronic ultraviolet (UV) exposure-induced reactive oxygen species (ROS) are commonly involved in the pathogenesis of skin damage by activating the metalloproteinases (MMP) that break down type I collagen. Adenophora remotiflora (AR) is a perennial wild plant that inhabits Korea, China, and Japan. The present study investigated the protective effects of AR against UVB-induced photo-damage in keratinocytes. MATERIALS/METHODS: An in vitro cell-free system was used to examine the scavenging activity of 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical and nitric oxide (NO). The effect of AR on ROS formation, antioxidant enzymes, elastase, MMP-1 level, and mRNA expression of MMP-1 were determined in UVB-irradiated human keratinocyte HaCaT cells. RESULTS: AR demonstrated strong DPPH free radical and NO scavenging activity in a cell-free system exhibiting $IC_{50}$ values of 1.88 mg/mL and 6.77 mg/mL, respectively. AR pretreatment dose-dependently attenuated the production of UVB-induced intracellular ROS, and antioxidant enzymes (catalase and superoxide dismutase) were enhanced in HaCaT cells. Furthermore, pretreatment of AR prevented UVB-induced elastase and collagen degradation by inhibiting the MMP-1 protein level and mRNA expression. Accordingly, AR treatment elevated collagen content in UVB-irradiated HaCaT cells. CONCLUSION: The present study provides the first evidence of AR inhibiting UVB-induced ROS production and induction of MMP-1 as a result of augmentation of antioxidative activity in HaCaT human keratinocytes. These results suggest that AR might act as an effective inhibitor of UVB-modulated signaling pathways and might serve as a photo-protective agent.

Extract of Ettlia sp. YC001 Exerts Photoprotective Effects against UVB Irradiation in Normal Human Dermal Fibroblasts

  • The identification of novel reagents that exert a biological ultraviolet (UV)-protective effect in skin cells represents an important strategy for preventing UV-induced skin aging. To this end, we investigated the potential protective effects of Ettlia sp. YC001 extracts against UV-induced cellular damage in normal human dermal fibroblasts (NHDFs). We generated four different extracts from Ettlia sp. YC001, and found that they exhibit low cytotoxicity in NHDFs. The ethyl acetate extract of Ettlia sp. YC001 markedly decreased UVB-induced cytotoxicity. Additionally, the ethyl acetate extract significantly inhibited the production of hydrogen peroxide-induced reactive oxygen species. Moreover, it inhibited UVB-induced thymine dimers, as confirmed by luciferase assay and thymine dimer dot-blot assay. Thus, the study findings suggest Ettlia sp. YC001 extract as a novel photoprotective reagent on UVB-induced cell dysfunctions in NHDFs.

  • 피부 손상은 주로 자외선, 열, 담배 등과 같은 환경적 요인으로부터 초래되는데, 이는 활성산소종의 과생성으로 인한 피부노화와 연관이 있는 것으로 알려져 있다. 돌배(Pyrus pyrifolia NAKAI)는 전 세계적으로 많이 소비되는 과일로써 항암, 항산화, 항염증효과가 알려져 있다. 본 연구에서는 돌배나무잎 추출물(Pyrus pyrifolia leaf extract, PPE)의 ultraviolet B (UVB)스트레스에 대한 피부 섬유아세포 보호효과를 검증하였다. Lactate dehydrogenase assay와 DCF-DA를 이용한 정성분석 실험은 PPE가 인간의 섬유아세포에서 UVB 스트레스에 의해 유발된 세포독성 및 과생성된 활성산소종을 농도 의존적으로 억제할 뿐만 아니라, 미토콘드리아 기능저하, 막전위 저하, 그리고 세포사멸과정의 핵심 인자인 caspase-3 활성도 유의하게 억제함을 보여주었다. 결론적으로, PPE는 UVB스트레스에 의해 과생성된 활성산소종을 억제시켰으며, 이로 인해 생기는 피부세포 사멸을 효과적으로 저해함을 확인하였다.

Oral Administration of Lactobacillus plantarum HY7714 Protects Hairless Mouse Against Ultraviolet B-Induced Photoaging

  • Ultraviolet (UV) irradiation alters multiple molecular pathways in the skin, thereby inducing skin damage, including photoaging. In recent years, probiotics have gained interest due to their beneficial effects on skin health, such as inhibiting atopic dermatitis and improving skin immunity or inflammation. However, little is known about the effects of probiotics on UVB-induced photoaging. In this study, we evaluated the effect of Lactobacillus plantarum HY7714 against UVB-induced photoaging in human dermal fibroblasts and hairless mice. The results showed that L. plantarum HY7714 treatment effectively rescued UVB-reduced procollagen expression through the inhibition of UVB-induced matrix metalloproteinase (MMP)-1 expression in human dermal fibroblasts. Data from a western blot showed that L. plantarum HY7714 inhibited the phosphorylation of Jun N-terminal kinase, thereby suppressing the UVB-induced phosphorylation and expression of c-Jun. Oral administration of L. plantarum HY7714 clearly inhibited the number, depth, and area of wrinkles in hairless mouse skin. Histological data showed that L. plantarum HY7714 significantly inhibited UVB-induced epidermal thickness in mice. Western blot and zymography data also revealed that L. plantarum HY7714 effectively inhibited MMP-13 expression as well as MMP-2 and -9 activities in dermal tissue. Collectively, these results provide further insight regarding the skin biological actions of L. plantarum HY7714, a potential skin anti-photoaging agent.
