The novel gene LRP15 is regulated by DNA methylation and confers increased efficiency of DNA repair of ultraviolet-induced DNA damage

  • LRP15 is a novel gene cloned from lymphocytic cells, and its function is still unknown. Bioinformatic data showed that LRP15 might be regulated by DNA methylation and had an important role in DNA repair. In this study, we investigate whether the expression of LRP15 is regulated by DNA methylation, and whether overexpression of LRP15 increases efficiency of DNA repair of UV-induced DNA damage in HeLa cells. The results showed (1) the promoter of LRP15 was hypermethylated in HeLa cells, resulting a silence of its expression. Gene expression was restored by a demethylating agent, 5-aza-2'-deoxycytidine, but not by a histone deacetylase inhibitor, trichostatin A; (2) overexpression of LRP15 inhibited HeLa cell proliferation, and the numbers of cells in the G2/M phase of the cell cycle in cells transfected with LRP15 increased about 10% compared with controls; (3) cyclin B1 level was much lower in cells overexpressing LRP15 than in control cells; and (4) after exposure to UV radiation, the LRP15-positive cells showed shorter comet tails compared with the LRP15-negative cells. From these results we conclude that the expression of LRP15 is controlled by methylation in its promoter in HeLa cells, and LRP15 confers resistance to UV damage and accelerates the DNA repair rate.

  • High bandwidth RF such as Bluetooth, GPRS, EDGE, 3GSM, HSDPA is papular in the mobile phone market. A non-conducting metal coating process requires an e-beam deposition of metal, two steps of UV hard coating primer and top coating; however, it is inefficient. We navigate to the electron beam irradiation conditions(resin surface treatment conditions) in the PC/ABS resin injection process. By analyzing the experimental results, we find the optimum development conditions for the electro deposition pre-treatment process and mass production lines using the plasma generated electron beam source.

  • This paper reports on the results of weathering test, tracking test and salt-fog test of various kinds of cycloaliphatic epoxy systems. UV irradiation dramatically induced the loss of surface hydrophobicity due to the chain scission attack at the surface under UV irradiation. It could be seen that samples containing an UV absorbent/antioxidant and a silicone oil additive have a good performance in weathering test, while ATH(alumina tri-hydrate) filled ones have high resistance against tracking failure than others. Under salt fog chamber test, specimens mixed with silicone oil are able to suppress leakage current development. It was thought that silicone oil mixed into cycloaliphatic epoxy system could lead to lower the surface energy and to retain hydrophobic properties for a long time, which are desirable for outdoor use.

Indium Tin Oxide Based Reflector for Vertical UV LEDs (자외선 수직형 LED 제작을 위한 Indium Tin Oxide 기반 반사전극)

  • In this paper, we studied a p-type reflector based on indium tin oxide (ITO) for vertical-type ultraviolet light-emitting diodes (UV LEDs). We investigated the reflectance properties with different deposition methods. An ITO layer with a thickness of 50 nm was deposited by two different methods, sputtering and e-beam evaporation. From the measurement of the optical reflection, we obtained 70% reflectance at a wavelength of 382 nm by means of sputtering, while only 30% reflectance resulted when using the e-beam evaporation method. Also, the light output power of a $1mm{\times}1mm$ vertical chip created with the sputtering method recorded a twofold increase over a chip created with e-beam evaporation method. From the measurement of the root mean square (RMS), we obtained a RMS value 1.3 nm for the ITO layer using the sputtering method, while this value was 5.6 nm for the ITO layer when using the e-beam evaporation method. These decreases in the reflectance and light output power when using the e-beam evaporation method are thought to stem from the rough surface morphology of the ITO layer, which leads to diffused reflection and the absorption of light. However, the turn-on voltage and operation voltage of the two samples showed identical results of 2.42 V and 3.5 V, respectively. Given these results, we conclude that the two ITO layers created by different deposition methods showed no differences in the electric properties of the ohmic contact and series resistance.

Isolation of auxotrophs and drug resistant mutants of Lentinus edodes (표고버섯의 영양요구성 및 약물내성주의 분리)

  • Auxotrophs and drug resistant mutants from the mycelia of Lentinus edodes were obtained by UV irradiation at survival rates of $0.024{\sim}2.45%$ and ethidium bromide (EtBr) enrichment after UV irradiation. The mutation rate was 0.40%, and back mutation rate was $4.81{\times}10^{-4}{\sim}8.46{\times}10^{-4}$. Various amino acid-, nucleic acid-, and vitamin-requiring auxotrophs were isolated. The concentrations of several fungicides, antibiotics and amino acid analogues inhibiting the growth of L. edodes were determined. The MIC values for cycloheximide, benomyl, and p-fluorophenylalanine were 2, 2000, and 1000 ug/ml respectively. Five p-fluorophenylalanine-resistant mutants and eight benomyl-resistant mutants were selected by UV irradiation.

  • Modern plastics are of great importance in many practical applications and their performance can be enhanced by surface modification to improve their hardness, wear and chemical resistance. The object of this study is invention of functional hardening coating materials which are applied to plastic, such as polycarbonate, polymethylmethacylate and acrylonitrilebutadienestyrene. In order to invention of hardening coating materials, we synthesized new silicone acrylate oligomer and prepared coating films containing silicone acrylate oligomer (SAOE) on PC substrate. The curing of coatings is performed by UV-radiation. As results of experiments, The properties of coating films which are contained SAOE is greatly improved in comparison with coating films non-contained SAOE. Especially, when 1 wt% SAOE was introduced into the coating formulation, the UV hardening coatings obtained showed the best surface hardness and gloss.

Isoforms of Glucose 6-Phosphate Dehydrogenase in Deinococcus radiophilus

  • Glucose 6-phosphate dehydrogenase (G6PDH, EC 1.1.1.49) in Deinococcus radiophilus, an extraordinarily UV-resistant bacterium, was investigated to gain insight into its resistance as it was shown to be involved in a scavenging system of superoxide $(O_2^{-1})$ and peroxide $(O_2^{-2})$ generated by UV and oxidative stresses. D. radiophilus possesses two G6PDH isoforms: G6PDH-1 and G6PDH-2, both showing dual coenzyme specificity for NAD and NADP. Both enzymes were detected throughout the growth phase; however, the substantial increase in G6PDH-1 observed at stationary phase or as the results of external oxidative stress indicates that this enzyme is inducible under stressful environmental conditions. The G6PDH-1 and G6PDH-2 were purified 122- and 44-fold (using NADP as cofactor), respectively. The purified G6PDH-1 and G6PDH-2 had the specific activity of 2,890 and 1,033 U/mg protein (using NADP as cofactor) and 3,078 and 1,076 U/mg protein (using NAD as cofactor), respectively. The isoforms also evidenced distinct structures; G6PDH-1 was a tetramer of 35 kDa subunits, whereas G6PDH-2 was a dimer of 60kDa subunits. The pIs of G6PDH-1 and G6PDH-2 were 6.4 and 5.7, respectively. Both G6PDH-1 and G6PDH-2 were inhibited by both ATP and oleic acid, but G6PDH-1 was found to be more susceptible to oleic acid than G6PDH-2. The profound inhibition of both enzymes by ${\beta}-naphthoquinone-4-sulfonic$ acid suggests the involvement of lysine at their active sites. $Cu^{2+}$ was a potent inhibitor to G6PDH-2, but a lesser degree to G6PDH-1. Both G6PDH-1 and G6PDH-2 showed an optimum activity at pH 8.0 and $30^{\circ}C$.

Study on the Mechanical Properties of Polyketone Fiber according to Dyeing and Finishing Process (폴리케톤 섬유의 염색 및 후가공 처리에 따른 기계적 물성에 관한 연구)

  • Polyketone fiber, a newly developed high strength fiber, has a tenacity and modulus similar to the p-aramid fiber, and can be used for reinforcing mechanical rubber goods(MRG), such as tires, hoses, and technical textiles. It will be expected for replacement of super fiber such as aramids and increasing the technical textile market share. This paper surveys the mechanical properties of polyketone fiber for technical textiles. For this purpose, dyed polyketone fabric is prepared, mechanical properties of coated and uncoated polyketone fabrics such as tensile strength, elongation and tear strength were examined before and after weather resistance test(temperature $63{\pm}3^{\circ}C$, humidity 60%, amount of power $0.35w/m^2$). The differences of mechanical properties between uncoated and coated fabrics for high functional technical textiles and composite materials are estimated through this study. The UV-stability of polyketone fabric showed obvious improvement after coating. After 168h(7day) of UV exposure, the coated fabric showed less deterioration in mechanical properties with the retained tensile strength and elongation at break greater than 22 and 17% of the uncoated polyketone fabrics values, respectively.

Hybrid Water Treatment of Tubular Ceramic MF and Photocatalyst Loaded Polyethersulfone Beads: Effect of Organic Matters, Adsorption and Photo-oxidation at Water Back-flushing (관형 세라믹 정밀여과와 광촉매 첨가 PES 구의 혼성 수처리: 물 역세척 시 유기물 및 흡착, 광산화의 영향)

  • The effect of humic acid (HA), and the roles of microfiltration (MF), PES (polyethersulfone) beads adsorption, and photo-oxidation were investigated in hybrid process of ceramic MF and PES beads loaded with titanium dioxide ($TiO_2$) photocatalyst for advanced drinking water treatment. The results of water and nitrogen back-flushing were compared in viewpoints of membrane fouling resistance ($R_f$), permeate flux (J), and total permeate volume ($V_T$). Because membrane fouling increased dramatically as increasing HA, Rf increased and J decreased, and finally $V_T$ was the highest at 2 mg/L HA. Average turbidity treatment efficiencies were almost same independent of HA concentration. Average organic matter treatment efficiency was the minimum 71.4% at 10 mg/L HA in water back-flushing, but those were almost constant in nitrogen back-flushing. The hybrid process of MF, PES beads, and UV (MF + $TiO_2$ + UV) have the lowest $R_f$, and the highest J and $V_T$ in both water and nitrogen back-flushing. The turbidity and organic matter treatment efficiencies were the maximum at MF + $TiO_2$ + UV independent of water and nitrogen back-flushing, and decreased sequently as simplifying the process to MF. However, adsorption performed the more important role than photo-oxidation in water back-flushing, and photo- oxidation was the more than adsorption in nitrogen back-flushing.

Evaluation of the mechanical properties and clinical efficacy of biphasic calcium phosphate-added collagen membrane in ridge preservation

  • Purpose: This study aimed to evaluate the biocompatibility and the mechanical properties of ultraviolet (UV) cross-linked and biphasic calcium phosphate (BCP)-added collagen membranes and to compare the clinical results of ridge preservation to those obtained using chemically cross-linked collagen membranes. Methods: The study comprised an in vitro test and a clinical trial for membrane evaluation. BCP-added collagen membranes with UV cross-linking were prepared. In the in vitro test, scanning electron microscopy, a collagenase assay, and a tensile strength test were performed. The clinical trial involved 14 patients undergoing a ridge preservation procedure. All participants were randomly divided into the test group, which received UV cross-linked membranes (n=7), and the control group, which received chemically cross-linked membranes (n=7). BCP bone substitutes were used for both the test group and the control group. Cone-beam computed tomography (CBCT) scans were performed and alginate impressions were taken 1 week and 3 months after surgery. The casts were scanned via an optical scanner to measure the volumetric changes. The results were analyzed using the nonparametric Mann-Whitney U test. Results: The fastest degradation rate was found in the collagen membranes without the addition of BCP. The highest enzyme resistance and the highest tensile strength were found when the collagen-to-BCP ratio was 1:1. There was no significant difference in dimensional changes in the 3-dimensional modeling or CBCT scans between the test and control groups in the clinical trial (P>0.05). Conclusions: The addition of BCP and UV cross-linking improved the biocompatibility and the mechanical strength of the membranes. Within the limits of the clinical trial, the sites grafted using BCP in combination with UV cross-linked and BCP-added collagen membranes (test group) did not show any statistically significant difference in terms of dimensional change compared with the control group.
