Inactivation of various bacteriophages by different ultraviolet technologies: Development of a reliable virus indicator system for water reuse

  • There is an urgent need to identify more reliable indicator systems for human pathogenic viruses in water reuse practice. In this study, we determined the response of different bacteriophages representing various bacteriophage groups to different ultraviolet (UV) technologies in real wastewater in order to identify more reliable bacteriophage indicator systems for UV disinfection in wastewater. Bacteriophage ${\varphi}X174$ PRD1, and MS2 in two different real wastewaters were irradiated with several doses of both low pressure (LP) and medium pressure (MP) UV irradiation through bench-scale UV collimated apparatus. The inactivation rate of ${\varphi}X174$ by both LP and MP UV was rapid and reached ${\sim}4{\log}_{10}$ within a UV dose of $20mJ/cm^2$. However, the inactivation rates of bacteriophage PRD1 and MS2 were much slower than the one for ${\varphi}X174$ and only ${\sim}1{\log}_{10}$ inactivation was achieved by the same UV dose of $20mJ/cm^2$. Overall, the results of this study suggest that bacteriophage MS2 could be a reliable indicator for human pathogenic viruses for both LP and MP UV disinfection in wastewater treatment processes and water reuse practice.

  • In this study, reliability assessment was carried out on UV(ultraviolet) indicators for the application of the indicator method on cultural heritages. As a result, some UV indicators(L, K, and M) proved to be unsuitable due to their low reactivity to UV and high reactivity to RH(relative humidity). On the other hand, indicator S showed considerable color changes and fading due to UV-B and UV-C according to the integral amount of UV irradiation. Indicator S showed the most distinct reaction to UV and a remarkable applicability to exhibition light, which are used in museums. Therefore, UV indicator S is an easy and useful monitoring tool for light environment management in cultural- property conservation facilities.

Applicability Investigation of E.coli, RNA and DNA Bacteriophages for Possible Indicator Microorganisms Based on the Inactivation Effectiveness by UV (UV 불활성화 효과에 의거한 E.coli, RNA 및 DNA 박테리오파지의 대체 지표 미생물로서의 적용성 검토)

  • This study compared UV and UV/$H_2O_2$ inactivation of E.coli, a possible indicator microorganism for fecal contamination of water, and $Q{\ss}$ phage, an indicator for pathogenic viruses. UV inactivation of $Q{\ss}$, T4 and lambda phages in actual secondary effluent was investigated, too. As a result, similar inactivation efficiency between $Q{\ss}$ phage and E.coli was observed during UV treatment, while $Q{\ss}$ phage showed higher resistance to UV/$H_2O_2$ than E.coli. $Q{\ss}$ phage resistance to UV or UV/$H_2O_2$ does not reflect those of all pathogenic viruses. However, the result tells that the use of E.coli inactivation efficiency in evaluating microbiological safety of water could not always ensure the sufficient safety from pathogenic viruses. Meanwhile, $Q{\ss}$ phage showed less resistance to UV than T4 and lambda phages, indicating that the use of $Q{\ss}$ phage as an indicator virus may bring insufficient disinfection effectiveness by causing the introduction of lower UV dose than required. Consequently, it can be thought that T4 or lambda phages would be more desirable indicators in ensuring the sufficient disinfection effectiveness for various pathogenic viruses.

Indicator microorganisms concentrations change after irrigation of wastewater treatment effluent in paddy field for water reuse (용수 재이용을 위한 하수처리수 관개 후 논에서의 지표성 미생물의 거동)

  • The objective of this research was to examine the significance of change to indicator microorganisms (TC, FC, and E. coli) in the paddy field and repair of UV disinfected secondary effluent. Average concentrations of Microorganisms were maintained by more than about 1,000 MPN/100mL in paddy field after irrigation. Microorganism repair was evaluated in relation to UV dose in photoreactivating light and dark. In addition, a significant inverse relationship was found between UV dose and repair of indicator microorganisms when the research of wastewater was conducted in the condition of low and high UV dose.

Disinfection and Reactivation of Microorganisms after UV Irradiation for Agricultural Water Reuse of Biofilter Effluent

  • A pilot study was performed to examine the feasibility of UV disinfection system and the reactivation of indicator microorganisms (TC, FC, E. coli) after UV irradiation for agricultural reuse of reclaimed water. Photoreactivation and dark repair enable UV-inactivated microorganisms to recover and may reduce the efficacy of UV inactivation, which might be drawbacks of the UV disinfection method. The effluent of biofilter for 16-unit apartment house was used as input to the UV disinfection system, and average SS and BOD concentration were 3.8 and 5.7 mg/L, respectively, and the mean level of total coliform was in the range of $1.0\times10^4$ MPN/100mL. UV disinfection was found to be effective and it reduced mean concentration of indicator microorganisms (total coliform, fecal coliform, and E. coli) to less than 100 MPN/100mL within 60s exposure using 17, 25, and 40W lamps. Two UV doses of 6 and 16 mW$\cdot$s/$\textrm{km}^2$ were applied and microorganisms reactivation was monitored under the dark, photoreactivating light, and solar irradiation. Microorganisms reactivation was observed in the UV dose of 6 mW$\cdot$s/$\textrm{km}^2$, and numbers increased up to 5% at the photoreactivating light and 1% at the dark. However, microorganisms were inactivated rather than reactivated at the solar radiation and numbers decreased to non-detectible level about below 2 MPN/100mL in 4 hours. In the case of 16 mW$\cdot$s/$\textrm{km}^2$, microorganism reactivation was not observed indicating that UV dose might affect the reactivation process such as photoreactivation and dark repair. Therefore, concerns associated with microorganism reactivation could be controlled by sufficient UV dose application. Agricultural reuse of reclaimed water might be even less concerned due to exposure to the solar irradiation that could further inactivate microorganisms. The pilot study result is encouraging, however, sanitary concern in water reuse is so critical that more comprehensive investigation is recommended.

Analysis of Indicator Microorganism Concentration in the Rice Cultural Plot after Reclaimed Water Irrigation (하수처리수 관개후 벼재배 시험구에서 지표미생물 거동 분석)

  • A study was performed to examine the effects of UV-disinfected reclaimed water on microorganism concentration during rice culture. Four treatments were used and each one was triplicated to evaluate the changes of microorganism concentrations: stream water irrigation (STR), biofilter effluent irrigation (BE), UV-disinfected water irrigation with dose of 6 mW ${\cdot}$ s $cm{-2}$ (UV-6), and UV-disinfected water irrigation with dose of 16 mW ${\cdot}$ s $cm{-2}$ (UV-16). The indicator microorganisms of interest were total coliform (TC), fecal coliform (FC), and E. coli. The biofilter effluent from 16-unit apartment sewage treatment plant was used as reclaimed water and flowthrough type UV-disinfection system was used. Concentrations of indicator microorganisms in the treatment plots ranged from $10^2$ to $10^5$ MPN/100 mL during 24 hours after irrigation in May and June, where initial irrigation water for transplanting reparation was biofilter-effluent without UV-disinfection. It implies that initial irrigation using only non-disinfected reclaimed water for puddling in paddy field can be health-concerned because of more chance of farmer's physical contact with elevated concentration of microorganisms. The concentrations of microorganisms varied widely with rainfall, and treatments using UV-disinfected water irrigation showed significantly lower concentrations than others and their levels were within the range of paddy rice field with normal surface water irrigation. The mean concentrations of STR and BE during growing season were in the range of 4 ${\times}\;10^3$ MPN/100 mL for TC, and 2${\times}\;10^3$ MPN/100 mL for FC and E, Coli, While mean concentrations of UV-S and UV-lS were less than 1${\times}\;10^3$ MPN/100 mL for all the indicator microorganisms. Overall, UV-disinfection was thought to be feasible and practical alternative for agricultural reuse of secondary level effluent in Korea.

  • The mechanism of synthesis of the toacco mosaic virus(TMV) and the potato virus X(PVX) was investigated using the methods of ultraviolet light irradiation and serological analysis. In vitro irradiation of UV on the infected tobacco juice for 10 minutes caused the infectivity of TMV and PVX to decrease markedly on their respective local lesion indicator hosts, Nicotiana glutinosa L. and Gomphrena globosa L., indicating that UV destroys directly the infectivity of the virus particles. Ten minutes after the UV was irradiated on the leaves of the two indicator hosts before inoculation, the infectivity of TMV decreased as it was irradiated in vitro, whereas that of PVX increased by 26% as compared with the unirradiated control. When the two viruses were mix-inoculated in the common host of tobacco and the synthetic products were analyzed by serological methods for a two week infection period, it was found that both viruses were multiplying more rapidly and abundantly than they were singly inoculated into the same host species. Titers from mixed series were often two times as high as those of singly inoculated series. A mechanism of competition in the synthesis between the mixed viruses in the common host is postulated.

Investigation of Indicator Microorganism Concentrations after Reclaimed Water Irrigation in Paddy Rice Pots (재처리수 관개후 지표미생물의 농도변화 조사)

  • A study was performed to examine the effects of reclaimed-water irrigation on microorganism con-centration in ponded-water of paddy rice plots. Several treatments were used and each one was triplicated to evaluate the change of indicator microorganisms (total coliforms (TC), fecal coliforms FC), and E. coli) concentrations in 2003 and 2004 growing seasons. Their concentrations increased significantly right after irrigation, but decreased about $45\%$ in 24 hours. It implies that agricultural activities such as plowing and fertilizing should be practiced one or two days after irrigation considering health-risks. Treatments with UV-disinfected water irrigation demonstrated significantly lower concentrations than others including control plots where natural surface water was irrigated. The monitoring results from actual paddy rice fields and experimental paddy plots showed that concentrations of indicator microorganisms ranged from $10^2\;to\;10^5$ MPN/100mL. A comprehensive assessment of existing agricultural practices and a thorough monitoring in the field as well as treatment-plots are recommended to make more realistic national guidelines more applicable. UV-disinfected water irrigation reduced microorganism concentrations in paddy fields down to below the concentration of conventional paddy rice culture, and is thought to be an effective and feasible measure fur agricultural reuse of secondary effluent.

Printable Time Temperature Integrator Consisting of Oxygen Indicator and Cover Film with Various Oxygen Permeability (다양한 산소 투과도를 가진 커버필름과 산소지시물질로 제작된 인쇄형 TTI)

  • A printable time temperature integrator (TTI) consisting of oxygen indicator and cover films with various oxygen permeability was developed. The printing ink contained methylene blue (oxygen indicator) which changed in color during storage. $TiO_2$ and glycerol for UV-activation of TTI and zein and ethanol for printing performance were also contained in the printing ink. The cover film on the ink was employed to control the color change rate and temperature dependency (Arrhenius activation energy, $E_a$) by using the different films (PE, PET, OPP, and LLDPE). The film properties were varied by annealing. TTI was produced by silk screen printing. As a result, the color change rates were different for the cover films, being the highest in TTI with LLDPE, followed by OPP, PE, and PET. The rate decreased with increase in the cover film thickness. The $E_a$ was the highest in TTI with LLDPE, followed by OPP, PE, and PET. The $E_a$ did not change with the cover film thickness. The annealed PVC and PET film were lower in oxygen permeability than the unannealed ones, indicating the lower color change rate.

Investigation of Potential Photoreactivation of Pseudomonas aeruginosa after LP or MP UV Irradiation (저압 및 중압 자외선 조사에 의해 불활성화된 Pseudomonas aeruginosa의 광회복능 조사)

  • Recently, there is growing interest in ultraviolet (UV) irradiation as a disinfection technic in drinking water production due to its effectiveness to inactivate microorganisms such as Crytosporidium parvum without forming disinfection byproducts. However, UV disinfection is known for its drawback such as photoreactivation. Despite many works concerning the photoreactivation, most of works were focused on indicator or non pathogenic microorganisms. The objective of this study is to examine the photoreactivation of Pseudomonas aeruginosa which is an opportunistic pathogen as UV radiation by LP and MP UV lamp was applied. The result showed that P. aeruginosa had high photo repair efficiency regardless of the type of UV irradiation. Both of the effective log repair values of LP and MP UV irradiation were found approximately 2.6 log. In addition, photo repaired P. aeruginosa was not significantly different in forming biofilm in comparison with non treated P. aeruginosa.
