This study investigated the effects of proline-serine (PS) and valine-serine (VS) dipeptides on melanogenesis in Mel-Ab cells. Proline-serine and VS significantly inhibited melanin synthesis in a concentration-dependent manner, though neither dipeptide directly inhibited tyrosinase activity in a cell-free system. Both PS and VS down-regulated the expression of microphthalmia-associated transcription factor (MITF) and tyrosinase. In a follow-up study also described here, the effects of these dipeptides on melanogenesis-related signal transduction were quantified. Specifically, PS and VS induced ERK phosphorylation, though they had no effect on phosphorylation of the cAMP response element binding protein (CREB). These data suggest that PS and VS inhibit melanogenesis through ERK phosphorylation and subsequent down-regulation of MITF and tyrosinase. Properties of these dipeptides are compatible with application as skin-whitening agents.
This study was performed to elucidate the inhibitory effects of Alveopora japonica extract on melanin synthesis by measuring the levels of cell viability, mRNA expression, tyrosinase activity, and melanin production in the B16F10 cell line. The effects of A. japonica extract on tyrosinase-related protein 1 (TYRP1), TYRP2, tyrosinase (TYR), and microphthalmia-associated transcription factor (MITF) mRNA expression levels and melanin content were determined. Quantitative real-time RT-PCR show that A. japonica extract decrease the mRNA expression levels of TYRP1, TYRP2, TYR, and MITF in B16F10 cell line, resulting in lower levels of melanin production compared to α-MSH-treated B16F10 cells. Tyrosinase activity assays reveal that A. japonica extract decrease melanin production in B16F10 cells. These results demonstrate the whitening effects of A. japonica extract on B16F10 cells; thus, A. japonica extract is a potent ingredient for skin whitening. Further research is needed on the mechanism of action of A. japonica extract. Such research will benefit not only cosmetics, but also the health food and medical industries.
This study was performed to clarify the whitening effects of anthricin on the B16F10 cell line. In order to elucidate the whitening effects of anthricin on the B16F10 cell line, cell viability, messenger ribonucleic acid (mRNA) expressions, tyrosinase activity assay, and melanin production assay were measured. The effects of anthricin on tyrosinase-related protein 1(TYRP1)/TYRP2/tyrosinase (TYR)/microphthalmia-associated transcription factor (MITF) mRNA expressions and melanin content were determined. Quantitative real-time RT-PCR showed that anthricin decreased the mRNA expression level of TYRP1/TYRP2/TYR/MITF genes and melanin production contents than α-MSH-treated B16F10 cells. The tyrosinase activity assay revealed that anthricin decreased the melanin production on the B16F10 cells. These data show that anthricin increases the whitening effects on the B16F10 cells; thus, anthricin is a potent ingredient for skin whitening. Thus, further research on the mechanism of action of anthricin for the development of not only cosmetics, but also healthy food and medicine should be investigated.

This study was carried out to investigate the effect of ethanolic extract Salicornia herbacea (SHEE) on melanogenesis and mechanism. The SHEE on ${\alpha}$-melanocyte stimulating hormone (${\alpha}$-MSH) induces melanogenesis in B16F10 melanoma cells. The effect of SHEE remarkably decreased protein expression of tyrosinase and tyrosinase relate protein (TRP)-2 increased extracellular signal related kinase (ERK) on ${\alpha}$-MSH 100 nM induced melanogenesis on B16F10 melanoma cells at dose-dependent manner. It has been reported that the activation of ERK reduce melanin synthesis by down-regulating micro-phthalmia-associated transcription fator (MITF). These results sugggest that possibility of extracted korean medicinal herbs as a functional ingredient for whitening cosmetic formula.
본 연구에서 오크라(Abelmoschus esculentus)에 대한 다양한 생리활성을 조사하여 기능성소재 응용가능성을 검토하였다. 오크라 추출물은 멜라노마 세포에 대하여 낮은 세포독성을 나타냈다. 세포독성이 거의 없는 농도에서 오크라 추출물 처리 시, 산화방지활성($ID_{50}$, $5.24{\mu}g/mL$), tyrosinase 활성저해($ID_{50}$, $102.12{\mu}g/mL$) 및 멜라닌 함량 저하($ID_{50}$, $17.85{\mu}g/mL$)를 보여주었다. 오크라 추출물 농도 의존적으로 tyrosinase 발현이 억제되었으며, 이는 오크라 추출물이 직접적인 tyrosinase 활성저해 및 세포 내 tyrosinase 발현을 억제시킴으로써 멜라닌 합성을 저해하는 것으로 판단된다. 또한, 오크라 추출물이 피부장벽 보호 지표물질로서 관련된 단백질인 involucrin 발현을 증가시키는 것을 확인하였다. 이와 같은 결과로 미루어 볼 때 오크라 추출물은 피부미백 소재 등 피부장벽 개선 효과를 지닌 기능성 화장품에 활용하기 위한 매우 효과적인 재료가 될 수 있다고 판단된다.
Codium fragile (Suringar) Hariot, a green alga of the Codiales family, has been reported to have several bioactive properties, including antioxidant and anti-inflammatory properties. However, its antiobesity and whitening effects and their underlying mechanisms are unclear. This study aimed to evaluate the antiobesity and melanogenesis inhibitory effects of C. fragile using methanol extracts of C. fragile (MECF). The results of this study revealed that MECF inhibited the accumulation of lipid droplets and triacylglycerol in differentiated 3T3-L1 adipocytes, which was associated with the inhibition of the expression of adipogenesis-related transcription factors, such as peroxisome proliferator-activated receptor γ, CCAAT/enhancer-binding protein-α (C/EBPα), and C/EBPβ, which function as the key regulators of adipogenesis. Also, MECF reduced tyrosinase activity and melanin content in B16F10 cells as well as the expression of tyrosinase, tyrosinase-related protein-1 (TRP-1), TRP-2, and microphthalmia-related transcription factor in the presence of α-melanocyte-stimulating hormone. Taken together, our findings suggest that the extract of C. fragile could be considered a promising functional ingredient for the prevention and treatment of obesity and skin pigmentation in the food and cosmetic industry.
Background: Panax ginseng has been used to prolong longevity and is believed to be useful for improving skin complexion. Ginsenosides are the most active components isolated from ginseng, and ginsenoside Rg3 (G-Rg3) in particular has been demonstrated to possess antioxidative, antitumorigenic, and anti-inflammatory properties. The aim of this study was to examine the ability of G-Rg3 to inhibit melanogenesis. Methods: The effects of G-Rg3 on melanin contents and the protein levels of tyrosinase, microphthalmia-associated transcription factor (MITF), and tyrosinase-related protein 1 (TRP1) were evaluated. Melanogenesis-regulating signaling molecules such as Akt and extracellular signal-regulated kinase (ERK) were also examined to explore G-Rg3-induced antimelanogenic mechanisms. Results: G-Rg3 was found to significantly inhibit the synthesis of melanin in normal human epidermal melanocytes and B16F10 cells in a dose-dependent manner. The activity of cellular tyrosinase and the expression of MITF, tyrosinase, and TRP1 were all reduced, whereas ERK was strongly activated. PD98059 (a specific inhibitor of ERK) attenuated the G-Rg3-induced inhibition of melanin synthesis and tyrosinase activity. Conclusion: Taken together, these results showed that G-Rg3 induces the activation of ERK, which accounts for its antimelanogenic effects. G-Rg3 may be a promising safe skin-whitening agent, adding to the long list of uses of P. ginseng for the enhancement of skin beauty.
황칠나무는 한국의 남부 해안 및 제주도 등에서 자생하는 식물이다. 본 연구에서는 황칠나무 잎 추출물의 미백 활성의 원인을 규명하고자 연구를 수행하였다. ${\alpha}$-MSH로 자극된 B16 melanoma 세포에 대한 추출물의 멜라닌 생합성 저해 활성을 측정하였다. 추출물은 25 및 $50{\mu}g/mL$ 농도에서 유의적으로 멜라닌 함량을 감소시켰으며, 세포 내의 tyrosinase 활성뿐만 아니라 tyrosinase와 TRP-2의 단백질 발현도 상당히 저해하였다. 결과적으로, 황칠나무 잎 추출물은 세포 내의 tyrosinase활성과 멜라닌 생합성에 직접적으로 관련된 효소의 발현을 저해함으로써 미백효과를 나타내었다. 본 연구 결과는 황칠나무 잎 추출물이 새로운 미백 화장품의 원료로써 이용 가능함을 시사한다.
$\alpha$-MSH는 세포내 cAMP를 증폭시켜 멜라닌세포의 증식과 색소 증가에 관여한다. 본 연구에서는 $\alpha$-MSH로 자극한 B16F10 세포에서 세신추출물의 hypopigmenting 효과를 조사하고 그 억제기전에 대하여 조사하였다. 세신추출물은 $\alpha$-MSH에 의해 유도된 tyrosinase 활성과 멜라닌생성을 효과적으로 억제시켰으며, 이는 tyrosinase 발현을 조절하는 전사인자인 MITF의 발현억제와 연관성이 있었다. 즉 세신추출물은 MEK/ERK와 PI3K/Akt의 활성화를 통하여 MITF를 조절함으로서 $\alpha$-MSH에 의해 유도되는 tyrosinase, TRP-1, Dct 등 멜라닌생성관련 단백질을 억제함으로서 멜라닌생성을 저해하는 것으로 사료된다.
