Process Development for Alcohol Production by Extractive Fermentation( I ) - Effect of Phase Forming Polymer - (추출발효에 의한 알콜제조 공정개발(I) - 수성이상계 구성 고분자의 영향 -)

  • Ethanol fermentations of inulin by K.fraglis in aqueous two-phase system of PEG/Dextran were performed not only to investigate the characteristics of partition of ethanol, sugar, and cell in the upper phase and the bottom phase but also to compare the fermentation properties with those of single phase system. In the range of 1 to 3 wt% of Dextran, ethanol fermentability and ethanol productivity reduced according to the increase of the molecular weight of PEG. But the cell yield and the cell productivity showed the opposite trend. In the region of 6 to 10 wt% of PEG, the increase of the concentration of PEG, caused the minute decrease of ethanol productivity but the remarkable augmentation of cell productivity. According to the increase of the molecular weight of PEG, the partition coefficient of inulin slightly decreased. But with the increment of the concentration of Dextran, the partition coefficient and the partition yield of inulin in the bottom Phase represented the trends of increase.

Quality Characteristics of High Acidity Apple Vinegar Manufactured Using Two Stage Fermentation (2단계 발효에 의한 고산도 사과식초의 품질특성)

  • This study investigated the manufacturing conditions of apple vinegar with high acetic acid content following fermentation according to alcohol concentration without any nutrients. We compared and analyzed the quality characteristics of high acetic acid fermentation by varying the initial alcohol content (6%, 7%, 8%, and 9%). In the results, it was possible to manufacture high acetic acid vinegar with 12% titratable acidity and an alcohol content of 6% and 7%. Lower initial alcohol content was associated with higher yield due to a shorter lag phase. For quality characteristics of the high acetic acid apple vinegar, pH was 2.91~3.20, titratable acidity was 12.0%, and organic acid consisted of acetic acid, malic acid, critic acid and oxalic acids. Based on the results, high acetic acid apple vinegar was produced using a two stage fermentation process after alcohol fermentation but the further research is needed to reduce the time of fermentation in fed-batch culture for industrial use.

Optimization of Anaerobic Process by Enzyme Treatment of High Concentration Organic Substances in Food Wastewater

  • Purpose: Since 2013, marine dumping of wastewater has been banned, and research on eco-friendly and efficient land treatment has emerged. This study compared and tested changes in biogas production and anaerobic process efficiency depending on whether or not enzyme pretreatment was performed during anaerobic digestion from single-phase and two-phase to medium-temperature. Research design, data and methodology: The total sugar, direct sugar, pH, and acidity before and after fermentation were analyzed by G/C by anaerobic fermentation of the liquor wastewater, food wastewater 1, and food wastewater 2 at 30℃ for 67 hours, and the amount of methane gas generated was analyzed by balloon volume. Results: It was found that stable organic acid concentration and pH were found in the enzyme-treated food wastewater 2, and the amount of methane gas generated was also increased. Conclusions: When anaerobic digestion of the liquor wastewater and the food wastewater together, the performance of enzyme pretreatment resulted in increased digestive efficiency. It will be the basic data that can contribute to carbon neutrality and greenhouse gas reduction by increasing the production of biogas.

Highly Branched Glucooligosaccharide and Mannitol Production by Mixed Cultrue Fermentation of Leuconostoc mesenteroides and Lipomyces starkeyi

  • The influence of process conditions on highly branched glucooligosaccharides production by mixed culture of Leuconostoc mesenteroides ATCC 13146 and Lipomyces starkeyi ATCC 74054 was studied. We divided the batch culture fermentations into two groups according to inoculation method. One-point inoculation was performed by coinoculation of L. mesenteroides and L. starkeyi at the ration of 10 to 1, and two-point inoculation by L. mesenteroides inoculation first and L. starkeyi inoculation after L. mesenteroides grew to the end of the log phase of growth. Two-point inoculation improved the yield of oligosaccharide by 1.5 to 20 fold more than one-point inoculation. In this process, the highest yield of oligosaccharides (48% of theoretical yield) and productivity (0.85 g/l/h) were obtained with starch as an initial substrate for L. starkeyi growth. The estimated composition of the end product consisted of 31.5% oligosaccharides, 17.6% dextran, and 46.5% mannitol.

Effect of Alcohols and Carboxylic Acids on Extraction Characteristics for 1,3-Propanediol by Aqueous Two Phases Systems (수상이성분계에 의한 1,3-프로판디올 추출특성에 대한 알콜과 카르복실산의 영향)

  • 1,3-Propandiol is a promising chemical which can be produced from fermentation of glycerol because the application of 1,3-propanediol is mainly in the production of bio-polytrimethylene terephthalate (bio-PTT). However, the cost of downstream processes in the biological production of 1,3-propanediol can make a high portion in the total production cost due to the large amount of water and the by-produced carboxylic acids such as succinic, lactic and acetic acids in 1,3-propanediol fermentation broth. In this study, aqueous two-phases systems composed of hydrophilic alcohols and phosphate salts were applied to the recovery of 1,3-propanediol from its artificial aqueous solution. Formation of aqueous biphases in hydrophilic alcohols and phosphate salts was due to the salting-out effect of salts in bottom phase, thereby 1,3-propanediol in bottom phase was moved into top phase. Extraction efficiency for 1,3-propanediol was proportional to the polarity of hydrophilic alcohols and the basicity of salts and the maximum value of extraction efficiency was more than 98%. In the aqueous two-phases systems after extraction, there was no carboxylic acid in top phase. Therefore, it was concluded that the aqueous two-phases systems composed of hydrophilic alcohols and phosphate salts were effective for the selective recovery of 1,3-propanediol without any coextraction of carboxylic acids.

Anti-lipopolysaccharide antibody mitigates ruminal lipopolysaccharide release without acute-phase inflammation or liver transcriptomic responses in Holstein bulls

  • Anti-lipopolysaccharide (LPS) antibody administration has the potential benefits of neutralizing and consequently controlling rumen-derived LPS during subacute ruminal acidosis. Four Holstein bulls were used in this crossover study with a 2-week wash-out period. Anti-LPS antibody (0 or 4 g) was administered once daily for 14 days. Significantly lower ruminal LPS and higher 1-h mean ruminal pH were identified in the 4 g group. However, blood metabolites, acute-phase proteins, cytokines, and hepatic transcriptomes were not different between the two groups. Therefore, anti-LPS antibody administration mitigated ruminal LPS release and pH depression without accompanying responses in acute-phase inflammation or hepatic transcriptomic expression.

  • In order to search for new antibiotics from anaerobic bacteria, a large number of samples from domestic soil were collected and processed by apropriate methods. A potential strain, Streptococcus sp. An-21-1, was found to produce antimicrobial compounds. The Results were as follows; 1. During fermentation, the bacteria grew rapidly up to 20hr, thereafter entered the death phase. The optimal temperature and pH for the bacterial growth were $37^{\circ}C$ and pH 7.0, respectively. 2. Antibiotics were purified from culture broth by solvent extraction, silica gel column chromatography and Sepadex L.H 20 column. 3. Physicochemical properties of Ap-1 and Ap-2 were similar ; Their melting points were between $234-237^{\circ}C$. Color reactions of ninhydrin, 2,7-dichlorofluorescein, 4-dimethylaminobenzaldehyde, Dragendroffs reagent and 20% $H_2SO_4$, were positive. Therefore, we assumed that these antibiotics have amine group, immine group, alkaloid, and lipid components. These were stable to heat. UV spectrophotometry showed two peaks at 210 nm and 260 nm. From above results, we assumed these antibiotics are belong to the peptide antibiotic family.

Optimal pH Profile in Rifamycin B Fermentation (리파마이신B 발효생산의 최대화를 위한 pH변화의 최적화)

  • The kinetic study of rifamycin B production in batch culture of Nocardia mediterranei was undertaken in part of our endeavor to optimize the fermentation condition. The growth parameters such as $\mu$$_{m}$ and Ks values for nitrogen source were evaluated by employing Monod equation. From the experiments, $\mu$$_{m}$ and Ks were 0.15hr$^{-1}$ and 8.35g/1, respectively. The growth kinetics in batch culture was found successfully interpreted by logistic law, i.e., the initial specific growth rate and the maximum cell mass concentration were determined as function of pH and both found to have maxima. For the production of rifamycin B, a non-growth associated production kinetics was employed and the specific productivity as a function of pH was found to have two maximum points. The yield coefficient and the specific productivity were calculated as mean values in production phase. Utilizing these experimental data as a function of pH, the optimal condition for the rifamycin B production was discussed with regad to the pH effect on the cell growth and production of the antibiotic. As a result, growth phase at pH 6.5 and production phase at pH 7.0 were found to be recommended.ded.

Efficient Production of ε-Poly-L-Lysine by Streptomyces ahygroscopicus Using One-Stage pH Control Fed-Batch Fermentation Coupled with Nutrient Feeding

  • ε-Poly-L-lysine (ε-PL) is a homopolymer of L-lysine molecules connected between the epsilon amino and alpha carboxyl groups. This polymer is currently used as a natural preservative in food. Insufficient biomass is a major problem in ε-PL fermentation. Here, to improve cell growth and ε-PL productivity, various nitrogen-rich nutrients were supplemented into flask cultures after 16 h cultivation, marking the onset of ε-PL biosynthesis. Yeast extract, soybean powder, corn powder, and beef extract significantly improved cell growth. In terms of ε-PL productivity, yeast extract at 0.5% (w/v) gave the maximum yield (2.24 g/l), 115.4% higher than the control (1.04 g/l), followed by soybean powder (1.86 g/l) at 1% (w/v) and corn powder (1.72 g/l) at 1% (w/v). However, supplementation with beef extract inhibited ε-PL production. The optimal time for supplementation for all nutrients examined was at 16 h cultivation. The kinetics of yeast-extract-supplemented cultures showed enhanced cell growth and production duration. Thus, the most commonly used two-stage pH control fed-batch fermentation method was modified by omitting the pH 5.0-controlled period, and coupling the procedure with nutrient feeding in the pH 3.9-controlled phase. Using this process, by continuously feeding 0.5 g/h of yeast extract, soybean powder, or corn powder into cultures in a 30 L fermenter, the final ε-PL titer reached 28.2 g/l, 23.7 g/l, and 21.4 g/l, respectively, 91.8%, 61.2%, and 45.6% higher than that of the control (14.7 g/l). This describes a promising option for the mass production of ε-PL.

Fermentation of Red Ginseng using CKDHC 0801 and CKDHC 0802 (CKDHC 0801과 CKDHC 0802 균주를 이용한 홍삼발효)

  • In this study, we isolated two species of bacteria for the powerful biotrasnformation of ginsenosides from Kimchi and human feces. Using biochemical tests and 16s rRNA sequencing, the selected strains were identified as Latobacillusplantarum (CKDHC0801) and Lactobacillussakei (CKDHC0802). Changes in cell growth and pH were examined in red ginseng. CKDHC 0801 and CKDHC 0802 reached their maximum growth phase after 24 hr and 48 hr, respectively, whereas the combined culture of CKDHC 0801 and CKDHC 0802 showed higher cell growth than bacterial strain alone. During fermentation of CKDHC 0801 and the combined culture, the pH values decreased from 5.2 to 4.2 after 24 hr, but CKDHC 0802 reached pH of 4.2 after 3day. The identities of ginsenosides were biotransferred from high molecular (Rg1 and Rb2) to low molecular (Rg3, Rg5, Rk1, PPD) by fermentation of both bacteria. Therefore, the results of this study demonstrate that CKDHC 0801 and CKDHC 0802 could be used to enhance to effects of red ginseng.
