Resveratrol regulates naïve CD 8+ T-cell proliferation by upregulating IFN-γ-induced tryptophanyl-tRNA synthetase expression

  • We found that resveratrol enhances interferon (IFN)-γ-induced tryptophanyl-tRNA-synthetase (TTS) expression in bone marrow-derived dendritic cells (BMDCs). Resveratrol-induced TTS expression is associated with glycogen synthase kinase-3β (GSK-3β) activity. In addition, we found that resveratrol regulates naive CD8+ T-cell polarization by modulating GSK-3β activity in IFN-γ-stimulated BMDCs, and that resveratol induces upregulation of TTS in CD8+ T-cells in the in vivo tumor environment. Taken together, resveratrol upregulates IFN-γ-induced TTS expression in a GSK-3β-dependent manner, and this TTS modulation is crucial for DC-mediated T-cell modulation. LMA와 FDTD를 이용한 유방암 진단용 알고리즘 연구 (Algorithm Study for Diagnosis the Breast Cancer Using LMA and FDTD)

  • 본 논문에서는 MT(Microwave Tomography)를 사용한 유방암 진단용 영상 재구성 알고리즘에 관하여 연구하였다. 사용된 유방암 진단용 시스템은 16개의 송수신 안테나로 구성되어 있으며, 신호 파형은 900 MHz 정현파를 사용하였다. 2차원 역산란 문제를 풀기 위하여 포워드 계산 방법으로 2차원 시간 영역 유한 차분법과 최척화 알고리즘으로 LMA(Levenberg-Marquardt Algorithm)를 이용하였다. 실제 유방암 환자의 MRI(Magnetic Resonance Imaging)로부터 얻은 수치 팬텀을 활용하여 영상 복원을 한 결과 유방암의 위치를 정확히 탐지함을 확인하였다.

소아암환자들을 위한 노래 만들기(Songwriting)의 음악치료적 적용 (Songwriting as Therapy with Pediatric Patients Undergoing Cancer Treatment)

  • 이 연구는 노래 만들기가 소아암을 치료받고 있는 과정중의 어린 환자들에게 어떻게 음악치료적인 목적으로 적용되어왔는지를 보여주는 연구이다. 본 연구자는 소아혈액종양학과에서 음악치료 인턴을 하면서 노래 만들기를 통하여 쉽게 표현하지 못했던 자신들의 생각과 느낌을 표현하는 것을 볼 수 있었다. 또한 노래 만들기를 통해 환자들과 음악치료사가 깊은 믿음을 바탕으로 건강한 치료적 관계가 형성되었으며, 그 관계는 환자가 병원환경에 좀 더 효과적으로 적응할 수 있도록 도와주었다. 본 연구의 사례연구에서는 환자 N이 소아 뇌종양을 치료를 받고 있는 자신의 삶에 대한 감정과 생각들을 노래 만들기를 통해 표현하였으며, 더 나아가 환자 N은 자신의 창조성과 내면의 힘(inner strength)을 자신이 만들어낸 음악을 통해 의식함으로써 자존감을 증진시키게 되었다.

직장암의 방사선 치료 시 3D-CRT, IMRT, Tomotheray를 이용한 치료계획 및 주변 정상장기 선량 비교 (Dosimetric Comparision for Rectal Cancer using 3D-CRT, IMRT, Tomotherapy)

  • 본 논문에서는 직장암 환자를 대상으로 일반적 치료방식인 3차원입체조형치료법과 선형가속기 기반의 IMRT, 그리고 토모테라피를 이용한 IMRT의 치료계획을 각각 수립하여 직장암 환자에 대한 최적의 치료법을 비교하고자 하였다. 치료법 비교 결과 종양조직에서는 처방선량의 95% 이상의 흡수선량을 만족시키고 있었으며 정상조직인 방광, 소장, 넙다리머리뼈의 정상조직의 합병증 발생율의 기준(V40, V30, V20, V10)을 만족하였다. 다만, 3가지의 치료법에서 선량분포측면에서 가장 효율적인 치료법은 Tomotherapy기반의 IMRT였으며 가장 낮은 효율을 보인 치료법은 3DCRT였다. 직장암의 방사선 치료시 환자의 자세재현성, 개인적인 신체환경 등을 고려하여 가장 적합한 치료방식을 적용한다면 환자의 예후와 삶의 질에 긍정적인 효과가 나타날 것으로 사료된다.

자연산 문치가자미, Pleuronectes yokohamae 피부의 육아종 (Papillomatosis on the skin of the wild marbled sole Pleuronectes yokohamae)

  • 충청남도 보령시 인근연안에서 어획된 문치가자미, Pleuronectes yokohamae 10개체 중에서 6개체가 피부에 육아종이 형성되었다. 기생충과 병원균으로 의심되는 세균은 증식하지 않았으며, 환부의 균질액을 CHSE-214 세포에 접종하였으나 CPE가 관찰되지 않았다. 환부를 광학현미경으로 관찰하였을 때 상피 세포의 증식으로 상피층은 정상개체와 비교하였을 때 유의적인 차이를 보이며 두꺼워졌고, 육아종 주름이 형성되었다. 비대되어진 상피세포는 핵융해 및 뚜렷하게 관찰되는 인이 특징이었으며, 세포질은 혼탁하였다. 이들 상피층에는 원형의 작은 핵과, 뚜렷한 인이 관찰되는 X-cell이 존재하였다. 진피층에는 다수의 신생 혈관들이 존재하고 있었다. 점액세포의 크기는 정상개체와 비교하였을때 유의적인 차이를 보이며 증가하였다. 본 연구에서는 병원체가 확인되지 않았지만 추가적인 실험을 통해서 원인체를 확인해야 할 것으로 생각된다.

Intestinal anti-inflammatory activity of Sasa quelpaertensis leaf extract by suppressing lipopolysaccharide-stimulated inflammatory mediators in intestinal epithelial Caco-2 cells co-cultured with RAW 264.7 macrophage cells

  • BACKGROUND/OBJECTIVES: Inflammatory bowel disease (IBD), including Crohn's disease and ulcerative colitis, involves chronic inflammation of the gastrointestinal tract. Previously, Sasa quelpaertensis leaves have been shown to mediate anti-inflammation and anti-cancer effects, although it remains unclear whether Sasa leaves are able to attenuate inflammation-related intestinal diseases. Therefore, the aim of this study was to investigate the anti-inflammatory effects of Sasa quelpaertensis leaf extract (SQE) using an in vitro co-culture model of the intestinal epithelial environment. MATERIALS/METHODS: An in vitro co-culture system was established that consisted of intestinal epithelial Caco-2 cells and RAW 264.7 macrophages. Treatment with lipopolysaccharide (LPS) was used to induce inflammation. RESULTS: Treatment with SQE significantly suppressed the secretion of LPS-induced nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$), IL-6, and IL-$1{\beta}$ in co-cultured RAW 264.7 macrophages. In addition, expressions of inducible nitric oxide synthase (iNOS), cyclooxygenase (COX)-2, and tumor necrosis factor (TNF)-${\alpha}$ were down-regulated in response to inhibition of $I{\kappa}B{\alpha}$ phosphorylation by SQE. Compared with two bioactive compounds that have previously been identified in SQE, tricin and P-coumaric acid, SQE exhibited the most effective anti-inflammatory properties. CONCLUSIONS: SQE exhibited intestinal anti-inflammatory activity by inhibiting various inflammatory mediators mediated through nuclear transcription factor kappa-B (NF-kB) activation. Thus, SQE has the potential to ameliorate inflammation-related diseases, including IBD, by limiting excessive production of pro-inflammatory mediators.

강녕탕(康寧湯)의 LPS 질내 주입 생쥐에 미치는 항염증(抗炎症) 효과 (Anti-inflammatory Effects of Gangyeong-Tang (康寧湯) on Mice injected LPS in Vagina)

  • Objective: We made the genital organs inflammatory mice model by vaginal injection of Lipopolysaccharide(LPS), and we intended to make study about anti-inflammatory effects of Gangyeong-Tang, among oral and rectal medication and Herbal-Acupuncture treatment. Method: The female ICR(20-30g) mice were used, the temperature was controlled within $22{\pm}0.5^{\circ}C$ and water and food was not limited. The environment was manipulated to simulate 12 hours of day and 12 hours of night. After LPS injection into vagina, we confirmed on-set of inflammation. 2 days before LPS injection, we started to medicate Gangyeong-Tang in Oral and Rectal and Aqua-acupucture treatment. After 3days from LPS injection, we mesured the White Blood Cell(WBC), Interrleukin-6(IL-6), Tumor Necrosis Factor-${\alpha}$(TNF-${\alpha}$) in blood which was collected from the Retro-orbital Plexus. Results: 1. We made the genital organs inflammatory mice model by vaginal injection of LPS successfully. 2. The number of WBC was decreased significantly as we medicated Gangyeong- Tang in Oral 1g/kg dose, 3g/kg dose and Rectal 1g/kg dose, rectal 3g/kg dose. 3. The concentration of IL-6 was decreased significantly as we medicated Gangyeong-Tang in all group of the Oral, Rectal, Herbal-Acupuncture treatment. 4. The concentration of TNF-${\alpha}$ was decreased significantly as we medicated Gangyeong-Tang in Oral 3g/kg dose and Rectal 1g/kg dose, Rectal 3g/kg dose group. Herbal-Acupuncture treatment group datas showed reductive tendency. Conclusion: As a result of this experiment, we made the genital organs inflammatory mice model by vaginal injection of LPS successfully and demonstrated anti-inflammatory effect of Gangyeong-Tang.

유전종양간호 관련 연구경향: 텍스트 네트워크 분석을 중심으로 (Research Trend of Genetics in Oncology Nursing: Based on Text Network Analysis)

  • 본 연구는 국내 외의 종양유전간호 관련 연구를 분석하여 연구동향을 파악하고자 하였다. 종양유전간호 관련 주제로 학술지에 게재된 논문들의 초록에서 제시한 핵심어들을 중심으로 한 텍스트 네트워크 분석을 실시하였다. 핵심어이자 중심성이 높은 주제어로 Nurse, Cancer, Genetic, Patient, Knowledge, Care, Genetic Test 등이 확인되었으며, 시기별 연구동향을 확인한 결과, 2003년 이후 Information, Care, Knowledge 등의 주제어를 포함한 연구들이 증가하였다. 간호학의 메타 패러다임으로 주제어를 분류한 결과, 건강, 간호, 인간, 환경 순으로 중심성이 높게 나타났다. 건강 영역 중 건강 위험 범주에서 Genetics, Risk, 건강 증진 범주에서 Genetic Test, Prevention 등이 가장 높은 빈도로 나타났다. 본 연구를 통해 종양유전간호 연구의 동향을 파악할 수 있으며, 유전성 암 환자들을 위한 간호 중재에 주축이 되는 간호사의 역할 및 중재프로그램 개발의 방향 설정에 활용될 수 있다는 점에서 의미가 있다.

학교보건수업 사례 분석을 통한 학교보건교육 모형 개발 일부 여자중학생을 중심으로 (The Model Development of School Health Education throung The Case Analysis of School Health Instruction -middle school girls-)

  • The purpose of this study was designed for the model development of curriculum of school health education including learning-teaching method, through analysis of results compared between a regular schedule health instruction and irregular health education. And this study is an attempt to give fundamental information for establishing of school health subject as the concept and scope of the school health subject is still not recognized and accepted satisfactorily in Korea. The data were collected by questionnaire from middle school girls and were carried out from 1989 to 1991. The major results obtained from this study were as follows: A Analysis of the case of school health. instruction 1. The responses of students about health eaducation : The positive rate of answers on the 'Health education is very important subject' was 91.2%, and on the 'I can maintain well healthful life : int of selfcare' was 87.1%. 2. The condition on knowledge, attitude and practice about health education: Education group had higher scores than comparison group about all most of questions, especially sex-education and drug abuse prevention education. 3. All the case of disease early founded out during the health instruction were children disease such as bone-tumor, lymphoma, hydrocephalus, and leukemia. B. Cytochalasin B Modulates Macrophage-Mediated Inflammatory Responses

  • The actin cytoskeleton plays an important role in macrophage-mediated inflammatory responses by modulating the activation of Src and subsequently inducing nuclear factor (NF)-${\kappa}B$ translocation. In spite of its critical functions, few papers have examined how the actin cytoskeleton can be regulated by the activation of toll-like receptor (TLR). Therefore, in this study, we further characterized the biological value of the actin cytoskeleton in the functional activation of macrophages using an actin cytoskeleton disruptor, cytochalasin B (Cyto B), and explored the actin cytoskeleton's involvement in morphological changes, cellular attachment, and signaling events. Cyto B strongly suppressed the TLR4-mediated mRNA expression of inflammatory genes such as cyclooxygenase (COX)-2, tumor necrosis factor (TNF)-${\alpha}$, and inducible nitric oxide (iNOS), without altering cell viability. This compound also strongly suppressed the morphological changes induced by lipopolysaccharide (LPS), a TLR4 ligand. Cyto B also remarkably suppressed NO production under non-adherent conditions but not in an adherent environment. Cyto B did not block the co-localization between surface glycoprotein myeloid differentiation protein-2 (MD2), a LPS signaling glycoprotein, and the actin cytoskeleton under LPS conditions. Interestingly, Cyto B and PP2, a Src inhibitor, enhanced the phagocytic uptake of fluorescein isothiocyanate (FITC)-dextran. Finally, it was found that Cyto B blocked the phosphorylation of vasodilator-stimulated phosphoprotein (VASP) at 1 min and the phosphorylation of heat shock protein 27 (HSP27) at 5 min. Therefore, our data suggest that the actin cytoskeleton may be one of the key components involved in the control of TLR4-mediated inflammatory responses in macrophages.
