TRAIL in Combination with Subtoxic 5-FU Effectively Inhibit Cell Proliferation and Induce Apoptosis in Cholangiocarcinoma Cells

  • In the past decade, the incidence and mortality rates of cholangiocarcinoma (CCA) have been increasing worldwide. The relatively low responsiveness of CCA to conventional chemotherapy leads to poor overall survival. Recently, tumor necrosis factor-related apoptosis-inducing ligand (TRAIL or Apo2L) has emerged as the most promising anti-cancer therapeutic agent since it is able to selectively induce apoptosis of tumor cells but not normal cells. In this study, we aimed to investigate the therapeutic effect of TRAIL in CCA cell lines (M213, M214 and KKU100) compared with the immortal biliary cell line, MMNK1, either alone or in combination with a subtoxic dose of 5-fluorouracil (5-FU). We found that recombinant human TRAIL (rhTRAIL) was a potential agent which significantly inhibited cell proliferation and mediated caspase activities (caspases 8, 9 and 3/7) and apoptosis of CCA cells. The combined treatment of rhTRAIL and 5-FU effectively enhanced inhibition of CCA cell growth with a smaller effect on MMNK1. Our finding suggests TRAIL to be a novel anti-cancer therapeutic agent and advantage of its combination with a conventional chemotherapeutic drug for effective treatment of CCA.

miR-485 Acts as a Tumor Suppressor by Inhibiting Cell Growth and Migration in Breast Carcinoma T47D Cells

  • MicroRNAs (miRNAs) are small, non-coding RNAs (18-25 nucleotides) that post-transcriptionally modulate gene expression by negatively regulating the stability or translational efficiency of their target mRNAs. In this context, the present study aimed to evaluate the in vitro effects of miR-485 mimics in breast carcinoma T47D cells. Forty-eight hours after T47D cells were transfected with miR-485 mimics, an MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay was utilized to determine the effects on cell viability. Colony formation and cell migration assays were adopted to determine whether miR-485 affects the proliferation rates and cell migration of breast carcinoma T47D cells. Our results showed that ectopic expression of miR-485 resulted in a significant decrease in cell growth, cell colony formation, and cell migration. These findings suggest that miR-485 might play an important role in breast cancer by suppressing cell proliferation and migration.

Effect of Phytolaccae Radix on the Proliferation of Transplanted-L1210 cells in Mice (상륙이 생쥐에 이식된 L1210 세포의 증식에 미치는 영향)

  • Cellular death by apoptosis is an active process, depending on gene transcription and protein synthesis. It was reported that nitric oxide can induce apoptosis in several cancer cell-lines. We studied effects of Phytolacca esculentum van Houtt (Phytolaccaceae) Radix water extract (PRE) on the proliferation of transplanted-L1210 cells in mice. When PRE (500 mg/kg) was administered orally once a day for 7 days after transplantation of L1210 cells to mice, DNA fragmentation of transplanted-L1210 cells was induced and mitochondrial transmembrane potential of those cells was reduced. Additionally, DNA fragmentation of L1210 cells was induced by the treatment of PRE in vitro. Also, DNA fragmentation of L1210 cells was enhanced by co-culture with the peritoneal macrophages obtained from PRE-administered mice and was partly inhibited by L-NMMA in vitro. PRE enhanced the production of nitric oxide and tumor necrosis factor-α from peritoneal rnacrophages. These results suggest that PRE induces apoptosis of transplanted-L1210 cells via directive action on L1210 cells and stimulation of nitric oxide and tumor neaosis factor-α from macrophages.

  • Extensive phytochemical investigation of the methanol extract from the whole plant of Youngia japonica (Asteraceae) led us to the isolation of a new guaiane-type sesquiterpene (1), together with three related guaianolides, youngiajaponicoside A (2), crepiside H (3) and crepeside E (4). The chemical structure of 1 was elucidated by the aid of spectroscopic analyses including 2D-NMR experiments (COSY, HMBC, HMQC and ROESY). The isolated components (1-4) were evaluated for the inhibitory effect on the proliferation of four cultured human tumor cell lines such as A549, SK-OV-3, SK-MEL-2 and HCT-15, in vitro.

miR-29a suppresses growth and invasion of gastric cancer cells in vitro by targeting VEGF-A

  • Increasing data shows miR-29a is a key regulator of oncogenic processes. It is significantly down-regulated in some kind of human tumors and possibly functionally linked to cellular proliferation, survival and migration. However, the mechanism remains unclear. In this study, we report miR-29a is significantly under-expressed in gastric cancer compared to the healthy donor. The microvessel density is negatively related to miR-29a expression in gastric cancer tissues. The ectopic expression of miR-29a significantly inhibits proliferation and invasion of gastric cancer cells. Furthermore, western blot combined with the luciferase reporter assays demonstrate that vascular endothelial growth factor A (VEGF-A) is direct target of miR-29a. This is the first time miR-29a was found to suppress the tumor microvessel density in gastric cancer by targeting VEGF-A. Taken together, these results suggest that miR-29a is a tumor suppressor in gastric cancer. Restoration of miR-29a in gastric cancer may be a promising therapeutic approach.

Effects of Samultang on Immune Function during the late stage of Pregnancy in BALB/c mice (사물탕이 임신 말기 생쥐의 면역능에 미치는 영향)

  • The purpose of this research was to investigate effects of Samultang water extract (SMT) on cytokine production from immune cells during the late stage of pregnancy in BALB/c mice. SMT(500 mg/kg) was administered p.o. once a day for 7 days, and then thymocytes and peritoneal macrophages were separated. At the late stage of pregnant mice, the proliferation of thymocytes and the production of ${\gamma}-interferon$ in thymocytes were decreased as compared with normal group, but the production of interleukin-2 and interleukin-4 was increased. The production of tumor necrosis $factor-{\alpha}$, nitric oxide and phagocytic activity in peritoneal macrophage was increased as compared with normal group. At the late stage of pregnant mice administered with SMT, the production of interleukin-2 in thymocytes was decreased as compared with a pregnant group, but the proliferation of thymocytes, the production of ${\gamma}-interferon$ and interleukin-4 was increased. The production of tumor necrosis $factor-{\alpha}$ and nitric oxide in peritoneal macrophages were decreased as compared with a pregnant group, but phagocytic activity were increased. These results suggest that SMT has the regulative action on immune function of thymocytes and peritoneal macrophages at the late stage of pregnant mice.

Ubiquitination of p53 is Involved in Troglitazone Induced Apoptosis in Cervical Cancer Cells

  • Peroxisome proliferator-activated receptor gamma (PPAR-${\gamma}$), a ligand-dependent nuclear transcription factor, has been found to widely exist in tumor tissues and plays an important role in affecting tumor cell growth. In this study, we investigated the effect of PPAR-${\gamma}$ on aspects of the cervical cancer malignant phenotype, such as cell proliferation and apoptosis. Cell growth assay, Western blotting, Annexin V and flow cytometry analysis consistently showed that treatment with troglitazone (TGZ, a PPAR-${\gamma}$ agonist) led to dose-dependent inhibition of cervical cancer cell growth through apoptosis, whereas T0070907 (another PPAR-${\gamma}$ antagonist) had no effect on Hela cell proliferation and apoptosis. Furthermore, we also detected the protein expression of p53, p21 and Mdm2 to explain the underlying mechanism of PPAR-${\gamma}$ on cellular apoptosis. Our work, finally, demonstrated the existence of the TGZ-PPAR-${\gamma}$-p53 signaling pathway to be a critical regulator of cell apoptosis. These results suggested that PPAR-${\gamma}$ may be a potential therapeutic target for cervical cancer.

Imaging of Tumor Cell Proliferation using Radiofluorinated Ethyluracil and Deoxyadenosine (Radiofluorinated Ethyluracil과 Deoxyadenosine을 이용한 종양세포 증식의 영상화에 대한 연구)

  • 목적 : 종양세포의 증식을 평가하기 위해 radiofluorinated ethyluracil (FEU)과 deoxyadenosine analogue(FAD)를 합성하여 종양의 영상화를 시도하였다. 대상 및 방법 : 5-(2-Fluoroethyl)uracil ([$^{18}F$]FEU)은 2, 4-dimethoxy-5-(2-hydroxyethyl) pyrimidine을 $K^{18}F$와 처리한 후 HBr로 가수분해하여 얻었으며 Fluorodeoxyadenosine은 adenosine의 triacetylated analogue를 $K^{18}F$와 처리하여 얻었다. 생물학적 조직분포는 유방암 세포(13762 NF, 100,000 cells per rat, im)를 쥐에 접종한 후 0.5, 1, 2 및 4시간에 주요장기를 적출하여 %ID/g을 측정하고 자가방사영상은 방사성의약품 투여 45분 후에 얻었다. PET 영상은 VX-2 종양을 접종한 가토를 이용하여 얻었다. In vitro cell proliferation assay는 사람의 말초단핵구를 이용하였다. 결 과 : In vitro assay상 ([$^{18}F$]FEU는 세포증식시 DNA/RNA에 결합함을 시사하였다. ([$^{18}F$]FAD와 ([$^{18}F$]FEU의 종양/비종양 방사능 섭취비는 시간경과에 따라 증가하였으며 ([$^{18}F$]FAD와 ([$^{18}F$]FEU를 이용한 자가방사영상과 ([$^{18}F$]FEU를 이용한 PET 영상에서 종양을 잘 관찰할 수 있었다. 결 론 : ([$^{18}F$]FAD 및 ([$^{18}F$]FEU를 이용하여 종양세포의 증식을 PET 영상에서 평가할 수 있으리라 사료된다.

Effects of Artemisiae Annuae Herba (AAH) on Cell Death of Ovarian Cancer in Vivo and in Vitro Study (개똥쑥이 난소암 세포의 사멸에 미치는 영향)

  • Objective: This study was designed to investigate the anti-cancer effects of AAH on ovarian cancer in vitro and by using allograft model in vivo. Methods: In this experiment, the effects of AAH on proliferation rates, cell morphology, cell death type, cell cycle, caspase activities and p38 mitogen-activated protein kinase (MAPK) pathway were investigated in A2780, human ovarian cell line. Results: AAH inhibited proliferation of A2780 cells in a dose dependent manner. In addition, AAH induced apoptosis but did not affect cell cycle of A2780 cells. AAH also effectively inhibited caspase 3 and caspase 9 activities respectively. In allograft tumor model, AAH reduced tumor volume and expanded life span in a dose dependent manner. Conclusion: It can be inferred that AAH can induce apoptosis in ovarian cancer cells and has possibility as an anticancer agent for ovarian cancer.

Anti-Cancer and Anti-Allergy Activities of Mycelia Extracts of Lentinus edodes Mushroom-Cultured Glycyrrhiza radix (감초로 배양한 표고버섯 균사체 추출물이 항암 효과 및 알레르기 억제 효과 검증)

  • This study investigated the effects of mycelia of Lentinus edodes mushroom-cultured Glycyrrihiza radix(LMG) on cancer cell lines and sarcoma 180(S-180), as well as on human mast cells. In an anti-cancer tests using Hep3B(hepatic cancer cell), MCF-7(breast cancer), and HeLa(uterine cancer) cells, LMG extract exhibited greater anti-proliferation effects than Glycyrrihiza glabra(GG) extract. LMG extract multiplication restraining effects were 60% that of ethanol at 3 mg/mL extract also displayed tumor suppressive effects in mice injected with S-180 cells. The growth-inhibition rates against tumor cells were 56% for LMG and 37% for GG. When LMG was added to human mast cells, the Intensity of RT-PCR products using primers($FC{\varepsilon}RI\;c-kit$) decreased. significantly compared with that of control. These results suggest that Lentinus edodes Mushroom-Cultured Glycyrrhiza glabra has an anti-proliferation effects against cancer cell lines(Hep3B, MCF-7 and HeLa) and S-180 tumors and will be also beneficial in treating allergic reactions.
