Development of Detection Methods for Cellulolytic Activity of Auricularia auriculajudae

  • To obtain basic information on the detection of cellulolytic activity in Auricularia auricula-judae, the influences of dye reagent, pH, and temperature were assessed. Chromogenic dye (congo red, phenol red, remazol brilliant blue, and trypan blue) was individually incorporated into a medium containing either carboxymethyl-cellulose, Avicel, or D-cellobiose as a polysaccharide carbon substrate. The other assessments utilized pHs ranging from 4.5 to 8.0 and temperatures from $15\sim35^{\circ}C$. Overall, when A. auricula-judae species were transferred onto media contained Congo red and adjusted pH 7.0 and then incubated at $25^{\circ}C$ for 5 days, the clear zone indicative of cellulolytic activity was more pronounced.

와송의 수종 암세포에 대한 항암작용 연구 (Anti-cancer Effects of Orostachyos Herba on some Kinds of Cancer Cells)

  • Background: Cancer reseach is done in earnest world-wide, because cancer is one of most threatening diseases to humans. Orostachyos Herba is a widely used herb that has long been in use in Korea as an anti-inflammatory and anti-cancer therapy. The purpose of this study is to verify any anti-cancer effects on stomach and liver cancer in vitro. Materials & Methods: AGS and KATO III stomach cancer cells and Hep3B and HepG2 liver cancer cells, all obtained from Korean Cell Line Bank, were used. The boiled extract of Orostachyos Herba(20 and 40 microliters) were injected into cultures and observed at 0 hours, and at 24-hour intervals up to 96 hours. The destruction of stomach and liver cancer cells was measured through Trypan blue exclusion testing. The suppression on viability of stomach and liver cancer cells was observed, and anti-cancer mechanisms was examined by analyzing the cell cycle. Results: In morphologic change, AGS, KATO III, HepG2 and Hep3B showed some of the withdrawn and floating appearance that is typical in cellular imparment. AGS, KATO III, HepG2 and Hep3B showed more destruction of stomach cancer cells in each test group than in the control group to a statistically significant degree. Analysis of the cell cycle after introduction of Orostachyos Herba showed very little inhibition of divisions of all cell lines. Conclusions: This experiment suggests that Orostachyos Herba has some anti-tumor effects on stomach and liver cancer cells. Progressive research on Orostachyos Herba and it's anti-tumor effects will be needed to determine its practicability as a cancer treatment.

  • 5-Fluorouracil (5-FU) is an antimetabolic of the pyrimidine derivatives that is used in chemotherapy for the treatment of several types of cancer. 5-FU have poor oral absorption and short biological half-time and strong side effects. Microneedle introduced to find a solution of problems. Microneedle device with roll was manufactured for transdermal delivery of various drugs. 5-FU was mixed in non-ionic surfactant such as tween 20 and tween 80. Camscope was used to analysis the permeation magnitude of treated skin by microneedle and trypan blue staining. The 5-FU solution with surfactant measured by ZETA-potential analysis system for stability of solution. The skin permeation rate of 5-FU determined by HPLC. We confirmed that cross treated skin was dyed more deeply than parallel treated skin through trypan blue staining. The results indicate that skin permeation rate of 5-FU was increased with the treatment types and treatment times.

구강암 세포주에서 실망초 추출물의 항암효능 (Anticancer effect of methanol extract of Erigeron bonariensis on oral cancer cells)

  • Objectives: Erigeron bonariensis is a type of Erigeron found throughout the tropical and subtropical areas as one of the perennial plants or pioneer plants. It is known to show detoxifying, antipyretic, and anticancer effects for various cancers. However, there are no reports on the anticancer effect of E. bonariensis on oral cancer cells. In the present study, we investigated the effects of the methanol extract of Erigeron bonariensis (MEEB) on the inhibition of cell growth and induction of apoptosis in mucoepidermoid carcinoma (MEC) cell lines, including the MC3 and YD15 oral cancer cells. Methods: MC3 Cells were treated by dimethyl sulfoxide (DMSO) or methanol extracts of 20 various natural products 20 ㎍/mL for 48 hours and cell viability were analyzed as Trypan blue exclusion assay. The effects of MEEB treatment on the cell viability of MC3 and YD15 cells, for 48 h, were analyzed by Trypan blue exclusion assay. The anticancer efficacy and apoptosis of oral cancer cell lines were analyzed by western blot analysis. The statistical significance of differences between groups was analyzed by Student's two-tailed t-test. A value of P<0.05 compared to the vehicle control was considered statistically significant. Results: Among 20 different naturally derived products, MEEB significantly inhibited cell viability and increased cleaved poly [ADP-ribose] polymerase 1 (PARP) protein in the MC3 and YD15 cells in a concentration-dependent manner. Conclusions: These results suggest that MEEB can be used as a natural anticancer drug for the treatment of human oral cancer.

The Experimental Study on Inhibitory Effects of Wild Ginseng Pharmacopuncture Solution on Melanin Biosynthesis

  • Background: The purpose of this study was to investigate the effects of wild ginseng pharmacopuncture on melanin production in B16/F10 murine melanoma cells. Methods: To determine the effect of wild ginseng pharmacopuncture solution on B16/F10 cells, cytotoxicity was evaluated using the 3-(4,5-dimethylthiazol-2-yl)- 2,5-diphenyl-tetrazolium bromide (MTT) method. To observe B16/F10 cell growth, death, and morphological changes, Trypan blue solution was used. The Hosoi method was used to investigate the effect of wild ginseng pharmacopuncture solution on melanin production. The Martinez-Esparza method was used to investigate the effect of wild ginseng pharmacopuncture solution on tyrosinase activity. To determine the pathway involved in the melanogenesis in cells exposed to wild ginseng pharmacopuncture solution, a cell-free tyrosinase was used. Results: Following treatment with $200{\mu}L$ of wild ginseng solution, the cell survival rate was $76.32{\pm}2.45%$ which significantly decreased with higher concentrations (${\mu}L$) of wild ginseng (up to $200{\mu}L$). When $100{\mu}L$ of wild ginseng was used, the cell survival rate was $89.95{\pm}2.07%$. No morphological changes or abnormalities were observed in the B16/F10 murine melanoma cells as observed in the Trypan blue test. Melanin production was significantly reduced to $72.17{\pm}3.74%$ at $100{\mu}L$. Using $100{\mu}L$ of wild ginseng solution, tyrosinase activity was significantly decreased to $80.15{\pm}1.05%$. Wild ginseng pharmacopuncture solution reduced melanin production both directly and indirectly. Conclusion: This study suggests that wild ginseng pharmacopuncture solution may be effective in inhibiting melanin production. Further studies are needed to determine safe and effective clinical applications.

Our results will be useful to researchers who aim to study extracellular enzyme activity in Ganoderma lucidum.

Dioscorea bulbifera tuber extract causes sterility in mice

  • Dioscorea bulbifera tubers contain several phytochemicals of pharmaceutical value. They have been traditionally used for treating various ailments, including postmenopausal symptoms. In the present study, we analyzed the direct effects of Dioscorea tuber extract on mouse spermatozoa. Its contraceptive effect was also evaluated by an intravaginal application before copulation. Mouse spermatozoa were cultured in vitro with various concentrations of the extract. After culturing, the spermatozoa were stained with fluorescein isothiocyanate peanut agglutinin or Coomassie blue to study the acrosome reaction, stained with trypan blue to study the viability, or treated with a hypo-osmotic medium to study the membrane damage. Estrous female mice were intravaginally injected with the extract and copulated with males. The extract induced acrosome exocytosis, viability loss, and membrane damage in a concentration-dependent manner. Female mice treated with the extract showed complete loss of fertility. These observations indicate that the Dioscorea bulbifera tuber extract could be used as a topical contraceptive. Infertility could be due to the precocious acrosome exocytosis of the spermatozoa or membrane damage.

보존된 사람 동종 복재정맥 이식편혈관 내피세포의 생활성에 관한 연구 (Viability of Endothelial Cells in Preserved Human Saphenous Vein Allografts)

  • 관상동맥 우회술이나 말초 혈관 우회술 시 가장 많이 쓰이는 대체 혈관은 자가 복재정맥이나, 이러한 정맥을 사용할 수 없는 경우 동종 복재정맥이 이용되어 왔다. 1900년대 초부터 시작된 동종 정맥의 이식은 아직도 그 이용에 대하여 논란이 있으며 보존 방법에 따른 혈관 내피세포나 평활근의 생활성 평가에 있어도 큰 편차가 있다. 특히 혈관 내피세포의 항응고 기능이나 nitric oxide에 대한 연구가 이루어지면서 동종 정맥 이식에 있어도 이들의 생활성 및 항원성 여부에 관심이 모아지고 있다. 대상 및 방법: 본 연구는 사람의 복재정맥을 보존하였을 때 보존 방법 및 기간에 따른 혈관 내피세포의 생활성의 평가를 목적으로 하였다. 신선 복재정맥을 대조군으로 하였으며, 실험군 복재정맥은 RPMI (Roswell park memorial institute) 1640에 10% 우태혈청이 포함된 $4^{\circ}C$ 조직 배양액에 넣어 각각 1일(24시간), 3일, 5일, 7일, 14일간 냉장 보존(cold storage)하였다. 기간별 냉장 보존이 끝나면 반으로 나누어 이 상태의 정맥을 I군(I-1, I-3, I-5, I-7, I-14)으로 정하고 냉장 상태에서 실험하였으며, 나머지는 10% 우태혈청과 10% DMSO가 포함된 RPMI 1640용액에 넣고 $-196^{\circ}C$의 액화 질소 탱크에서 2주간 냉동 보존 후(cryopreservation), $37^{\circ}C$에서 급속 해동하여 냉장 보존된 기간에 따라 II군(II-1, II-3, II-5, II-7, II-14)으로 정하여 실험하였다. 각 군의 생활성을 알아보기 위하여 전자현미경을 이용한 내피세포의 관찰, 트롬보모듈린(thrombomodulin) 면역조직화학검사를 이용한 혈관 내피세포의 항응고기능, 효소 소화에 의한 정맥에서의 혈관 내피세포 분리 후 트리판 블루(trypan blue)를 이용한 세포 생존율 검사 등을 시행하였다. 결과: 전자현미경을 통한 세포의 형태학적 검사에서 Groups I-7, I-14, II-5, II-7, 그리고 II-14정맥군들이 Gundry score의 의미 있는 증가(p <0.05)를 보이는 형태학적 변화가 나타났다. 트롬보모듈린을 이용한 면역조직화학검사와 트리판 블루를 이용한 세포 생존율 검사에서는 냉장군은 7일째 생활성 저하를 나타냈으나 냉동 보존군에서는 보존 기간에 상관없이 모두 생활성이 저하된 것으로 관찰되었다. 결론: 사람의 복재정맥을 우태혈청이 포함된 $4^{\circ}C$의 RPMI 1640에 냉장 보존하였을 경우 냉장 7일째부터 형태학적, 기능적 생활성 저하를 관찰할 수 있었으나, $-196^{\circ}C$에 냉동 보존한 경우에는 냉장 5일 후 냉동 보존한 정맥에서도 형태학적인 생활성 저하를 관찰할 수 있었으며 혈관 내피의 항응고 기능을 나타내는 트롬보모듈린 면역조직화학검사에서는 냉동 보존한 대부분 정맥에서 기능적 생활성이 저하된 것으로 관찰되었다. 그러나 이러한 생활성 및 항응고 기능의 저하가 일시적인 현상인지의 여부와 이러한 기능 저하가 이식 혈관의 장기 개통률에 미치는 영향에 대하여 지속적 연구가 필요하다.

Flavonoid Fraction Purified from Rhus verniciflua Stokes Actively Inhibits Cell Growth Via Induction of Apoptosis in Mouse Tumorigenic Hepatocytes

  • Dietary flavonoids are currently receiving considerable attention in developing novel cancer-preventive approaches because of their potential capacities to actively induce apoptosis of cancer cells. In our previous report, a flavonoid fraction, which consisted mainly of protocatechuic acid, fustin, fisetin, sulfuretin, and butein and named RCMF (RVS chloroform-methanol fraction), was prepared from a crude acetone extract of Rhus verniciflua Stokes (RVS) that is traditionally used as food additive and herbal medicine. In this study, we evaluated the effects of the RCMF on cell proliferation and apoptosis using SV40-transformed tumorigenic hepatocytes, BNL SV A.8. Tritium uptake assay showing the proliferative capacity of the cells was strongly suppressed in the presence of RCMF. This anti-proliferative effect was further confirmed through trypan blue exclusion. RCMF-mediated suppression of cell growth was verified to be apoptotic, based on the increase in DNA fragmentation, low fluorescence intensity in nuclei after propidium iodide staining, and the appearance of DNA laddering. Collectively, this study demonstrated that RCMF can be approached as a potential agent that is capable of significantly inhibiting cell growth of hepatic cancer cells.
