Effect of Bulnesia sarmienti Ethanol Extract on Plasma Levels of Glucose and Lipid in Streptozotocin Induced Diabetic Rats (Bulnesia sarmienti 에탄올 추출물이 streptozotocin 유발 당뇨 흰쥐의 혈당 및 혈장 지질수준에 미치는 영향)

  • This study examined the effects of Bulnesia sarmienti ethanol extract on lipid metabolism in streptozotocin (STZ)-induced diabetic rats. Sprague Dawley rats were induced with diabetes mellitus by injecting STZ (45 mg/kg) into the tail vein, and were then divided into 5 groups: normal, STZ-control, and three experimental diabetic groups. The rats were fed a basal diet along with B. sarmienti ethanol extract for 8 weeks. According to the results, after 8 weeks of consuming the experimental diets, the body weights and food efficiency ratios of the B. sarmienti ethanol extract-fed groups were not different from those of the control group. However, the concentrations of plasma total-cholesterol, LDL-cholesterol, and triglycerides were significantly lower in the B. sarmienti ethanol extract groups as compared to those of the control group. Although a blood glucose control effect was not observed after the eight weeks of B. sarmienti ethanol extract treatment in this type-l diabetes model, it was discovered that the experimental diets were highly effective at lowering levels of serum lipids, including triglycerides, total-cholesterol, and LDL-cholesterol.

Triglyceride Composition of Perilla Oil (들깨기름의 Triglyceride 조성(組成)에 관한 연구)

  • The triglyceride composition of perilla oil was investigated by high performance liquid chromatography (HPLC) in combination with gas liquid chromatography (GLC). The triglycerides were separated from perilla oil by thin layer chromatography (TLC), and fractionated into five groups on the basis of their partition numbers by reverse phase HPLC on a column packed with ${\mu}-Bondapak\;C_{18}$ using methanol-chloroform mixture as a solvent. Each of these collected fractions gave one to three peaks in the GLC chromatograms according to the acyl carbon number of the triglyceride, and fatty acid composition of the triglyceride was also analyzed by GLC. The results indicate that the perilla oil consists of fifteen kinds of triglycerides, and the major triglycerides in perilla oil were as follows: 68.0% of $(C_{18:3},\;C_{18:3},\;C_{18:3})$, 6.7% of $(C_{18:2},\;C_{18:3},\;C_{18:3})$, 5.9% of $(C_{18:1},\;C_{18:3},\;C_{18:3})$, 4.3% of $(C_{16:0},\;C_{18:3},\;C_{18:3})$, 3.8% of $(C_{18:1},\;C_{18:2},\;C_{18:3})$, 3.2% of $(C_{18:1},\;C_{18:1},\;C_{18:3})$, 2.0% of $(C_{16:0},\;C_{18:2},\;C_{18:3})$, 1.5% of ($C_{18:2},\;C_{18:2},\;C_{18:3})$, 1.0% of $(C_{16:0},\;C_{18:1},\;C_{18:3})$.

  • In order to define triglyceride compositions in fat and oil triglycerides were separated by thin layer chromatography (TLC) from corn oil, and the separated triglycerides were graduated according to each partition number(PN) by high performance liquid chromatography (HPLC) using column of ${\mu}-Bondapack\;C_{18}$ and each graduation was graduated again according to acylcarbon number by gas liquid chromatography(GLC). Fatty acid compositions were analyzed by GLC after their graduation were methylated in according to PN. The triglyceride compositions were estimated by synthesizing the above three results. The estimated triglycerides consisted of 36 kinds in corn oil. The major triglyceride compositions of sample oil were as follows: 21.5%$(C_{18:2},\;C_{18:2},\;C_{18:1})$, 17.4%$(C_{18:1},\;C_{18:2},\;C_{18:1})$, 15.4%$(C_{18:1},\;C_{18:2},\;C_{16:0})$, 11.1%$(C_{16:0},\;C_{18:2},\;C_{18:2})$, 9.0%$(C_{18:1},\;C_{18:1},\;C_{18:1})$, 8.0%$(C_{18:2},\;C_18:2},\;C_{18:2})$, 5.7%$(C_{18:1},\;C_{18:1},\;C_{16:0})$, 2.2%$(C_{16:0},\;C_{16:0},\;C_{18:2})$, 1.6%$(C_{18:2},\;C_{18:2},\;C_{18:2})$, 1.1%$C_{18:2},\;C_{18:0},\;C_{16:0})$, 1.1%$(C_{16:0},\;C_{16:0},\;C_{18:1})$.

  • Triglycerides of cottonseed oil were separated by thin layer chromatography (TLC), and fractionated by high-performance liquid chromatography (HPLC) on the basis of partition numbers. From each fraction, it was fractionated again on the basis of acyl carbon numbers using gas liquid chromatography (GLC). The fatty acids of triglyceride for each partition number group were analyzed by GLC. From, these results, triglyceride constituents of cotton seed oil were estimated to be 37 kinds of triglycerides. The major triglycerides and their contents in cotton seed oil were as follows: 25.8%$(C_{16:0},\;C_{18:2},\;C_{18:2})$, 15.5%$(C_{18:2},\;C_{18:2},\;C_{18:2})$, 13.8%$(C_{16:0},\;C_{18:2},\;C_{16:0})$, 8.3%$(C_{18:2},\;C_{18:1},\;C_{18:2})$, 6.2%$(C_{18:2},\;C_{18:1},\;C_{18:1})$, 4.1%$(C_{18:1},\;C_{18:1},\;C_{14:0})$, 3.4%$(C_{16:0},\;C_{18:1},\;C_{16:0})$, 2.3%$(C_{18:1},\;C_{18:2},\;C_{16:0})$, 2.2%$(C_{18:1},\;C_{18:1},\;C_{18:1})$, 1.0%$(C_{14:0},\;C_{18:2},\;C_{18:1})$.

Hydrolysis of Triglycerides with Cold-Adapted Lipase of Psychrobacter sp. S3 Isolated from Intertidal Flat (갯벌에서 분리된 Psychrobacter sp. S3균 유래의 저온성 리파제에 의한 트리글리세리드의 가수분해 특성)

  • Lipase-producing bacteria (S3) were isolated from intertidal flat at Saemanguem. A isolated strain was identified as Psychrobacter species by physiological and fermentational characterization as well as 16S rRNA analysis. The strain was then named as Psychrobacter sp. S3. P. sp. S3 grew most rapidly at $30^{\circ}C$, but grew well even at $10^{\circ}C$ and its lipase activity was most high when cultivated at $20^{\circ}C$. Lipase S3 had optimum temperature of $30^{\circ}C$ for the hydrolysis of p-nitrophenyl caproate and had more than $80^{\circ}C$ activity even at $10^{\circ}C$. The activation energy was calculated to be 1.5 kcal/mol, which showed that it was a typical cold-adapted enzyme. It was an alkaline enzyme with optimum pH of $9.0\~9.5$. It could hydrolyze various length of triglycerides. Among them, it hydrolyzed most rapidly $C_4,\;C_{14},\; C_{16}-length$ triglycerides. When added to tributyrin-agarose gel, lipase S3 hydrolyzed tributyrin most rapidly at 30 and $40^{\circ}C$, but it could hydrolyze well even at $4^{\circ}C$.

The Effects of Dietary Patterns and Apolipoprotein E Phenotype on the Blood Lipid Profiles of Individuals from Cheju Area (제주지역 성인의 Apolipoprotein E Phenotype 분포와 식생활 및 혈청지질 농도의 관련인자 연구)

  • The purpose of this study was to determine the relation between serum lipid profiles, apolipoprotein E phenotype, and dietary patterns in a cross-section of healthy individuals from Cheju-Do. Age, gender, anthropometric measurements, blood pressure, dietary consumption, drinking / smoking habits and menopausal status were surveyed. Total cholesterol, LDL-cholesterol, HDL-cholesterol, triglyceride, fasting blood glucose, and insulin levels were measured from overnight fasting blood. The study involved a total of 286 individuals(147 men and 139 women) between the ages of 20 and 60 years old. All of the subjects were recruited from a population of healthy individuals living in Cheju-Do. The results of the study are as follows : 1) Among the males, those in their 20's had the maximum food intake, while those in their 40's had the minimum food intake. For the females, food intake was the highest for those in their 30's. Energy and nutrient intakes were directly proportional to the amount of food intake. Men in their 30's were heavier than other men and women in their 40's were heavier than other women. The activity index for men in their 20's and 30's appeared to be lower than that of men above 40. The activity index of women in their 20's appeared to be lowest among all aged groups, and the index appeared to increase from the age of 30 onwards. 2) In terms of changes In serum constituents with age, men in their 40's appeared to have the highest levels of serum constituents such as lipids, glucose, and insulin. Men in their 50's showed the highest levels of serum LDL-cholesterol and glucose. Men in their 30's showed peak levels of serum triglycerides. On the other hand, women in their 50's appeared to have peak levels of serum total cholesterol, LDL-cholesterol, and triglycerides. There was no ch:ange with age in HDL-cholesterol and insulin levels for men and women. The percentage of the subjects had the following apo E phenotypes : E3/3, 91.3% ; E3/2, 5.4% ; E4/3, 2.5% ; E4/2, 0.7%. Lee's reserch with Korean female college students showed that the percentage of ApoE3/3, E3/2, E 4/2, E4/3, and E4/4 were 84.8%, 6.7%, 6.7%, 0.9%, 0.9%, respectively. The number of samples with ApoE mutation was so small that there was no statistical significance in the relation between apolipoprotein E phenotype and se겨m lipids. 3) To investigate the relati onship between weight and serum constituents, the subjects of this study were divided into three groups by BMI underweight, normal weight, and overweight. The serum constituents of men and women below the age 40 in the overweight groups belonged to the normal domain. On the other hand, serum cholesterol levels of both men and women above the age 40 in the overweight groups remained in the borderline-high region(above 200mg/dl), and the mean value of LDL-cholesterol(above 130mg/dl) and triglycerides of men were above normal. Fasting blood glucose levels also remained in the borderline-high region. Total cholesterol levels of women above the age 40 in the overweight group was in the borderline-high region. The effect of fenofibrate and 17β-estradiol on lipid metabolism in low-fat diet-fed OVX female mice (저지방식이를 섭취한 난소절제 암컷 쥐에서 지질대사에 대한 fenofibrate과 17β-estradiol의 영향)

  • It was studied whether fenofibrate alone or combinational treatments of fenofibrate and 17β-estradiol regulates serum lipid levels, and whether the effects of fenofibrate on serum lipid metabolism are affected by co-administration of 17β-estradiol in low-fat diet-fed ovariectomized female mice. Compared with low-fat diet-fed controls, mice treated with fenofibrate alone and mice treated with fenofibrate and 17β-estradiol didn't decrease body weight at 8 weeks. Fenofibrate alone or combinational treatments of fenofibrate and 17β-estradiol did not regulate serum levels of total cholesterol and HDL-cholesterol. Fenofibrate decreased plasma levels of LDL-cholesterol and triglycerides compared with controls. The combinational treatments of fenofibrate and 17β-estradiol showed more beneficial effects on triglycerides than fenofibrate alone. Therefore, the present study found that serum triglycerides reduced by fenofibtae treatment alone could be more improved by combinational treatments of fenofibrate and 17β-estradiol in low-fat diet-fed ovariectomized female mice.

Inflammatory Cytokines and Dietary Factors in Korean Elderly with Chronic Disease (만성 질환 노인에서의 면역 성분 양상과 식이예방인자)

  • The purpose of this study was to investigate the association between inflammatory cytokines and chronic disease status in Korean elderly. The subjects were 248 elderly people aged over 65 years recruited from Health Center in Seoul. The subjects were classified into 3 groups based on their disease (diabetes, hypertension, and hyperlipidemia) status: subjects with one diagnosed disease of diabetes, hypertension, and hyperlipidemia fall into singular group (n=89), subjects with more than 2 disease into multiple group (n=39), and those with free of the diseases into normal (n=122). Anthropometric and biochemical characteristics, and dietary intakes were assessed. Dietary intakes were surveyed by 24-recall method. The means of IL-2, IL-6, MCP-1 and C3 were not differ among 3 groups. However, when subjects classified into tertiles of IL-6, MCP-1, TNF-$\alpha$ and C3 and frequencies of each fertile were compared, the multiple group showed significantly lower frequencies in lowest fertile than normal group (p<0.05), suggesting higher tendency of inflammatory responses. For hematological values, blood pressure, triglycerides, fasting blood glucose levels were highest in multiple group (p<0.05) compared to other 2 groups. BMI, body fat(kg), and triceps skinfold thickness were also significantly higher in multiple group than in 2 other groups(p<0.05). Moreover, the concentrations of IL-2, IL-6 and C3 were significantly correlated with hematologic values of fasting blood glucose, total cholesterol, LDL-cholesterol, and triglycerides or obesity factors such as triceps skinfold thickness, BMI, and body fat(%). Among singular and multiple group, the subjects with higher intakes for vitamins A, C, and E showed the higher level of IL-2 and the lower level of MCP-1, and C3. In conclusion, blood concentrations of triglycerides and proinflammatory cytokines, blood pressure, obesity parameters (BMI, body fat, triceps skinfold thickness) were higher in multiple group than in normal, but this result strongly suggest that the increasement of the vitamin A, C, and E intakes would modify the cytokine levels to reduce the inflammatory response in the elderly people with chronic diseases.

  • The lipid and fatty acid compositions and vitamin E content were analyzed in 48 human mature milk, 3 infant formulas (modified milk powder) and 8 market milk samples. The total lipid content in modified milk powder was similar those in human milk and market milk when total solids content of modified milk powder was corrected to that of human milk. In comparison with human milk, modified milk powder contained a lower proportion of triglycerides and higher proportions of phospholipids, free fatty acids and cholesterol esters. The ratios of phospholipids/triglycerides, total cholesterol / triglycerides and total tocopherol/total lipids in modified milk powder were significantly higher than those in human milk and market milk. The American recommendation for linoleic acid (0.7 IU/g) could be satisfied with human milk, modified milk powder and market milk. The proportions of short- and medium-chain even numbered saturates were higher and the proportions of long-chain derivatives of linoleic (${\omega}6$ series) and linolenic (${\omega}3$ series)acids were lower in modified milk powder and market milk than in human milk. It is concluded that in view of their levels in breast milk, the polyenoic derivatives of linoleic and linolenic acids must be taken into account when assessing infant foods.

Leisure Time Physical Activity and its Relationship to Coronary Risk Factors in Male Workers (성인남성 근로자의 여가시간 중 육체적 활동양상 및 관상동맥질환 위험인자들과의 관련성)

  • This study was attempted to observe leisure time physical activity pattern and its relationship to coronary risk factors (BMI, systolic blood pressure, diastolic blood pressure, serum total cholesterol, triglycerides, fasting blood sugar). Subjects participated in this study were 277 adult male workers of an industry in Kyungpook province, Korea. Energy expenditure were measured using modified Physical Activity History questionnaire of CARDIA study by interviewing. Serum total cholesterol, triglycerides and blood sugar level were tested with 5 ml fasting blood and height, weight and blood pressure were masured. Data on smoking and drinking habits and others were obtained. Geometric mean of leisure time physical energy expenditure were estimated as 212.80 kcal per week for study subjects and only 22.4% of them expended 2,000 kcal or more per week in leisure time physical activity. Taking walks or hikes and jogging or running were more frequent leisure time physical activities in study subjects. Statistically significant mean differences in total weekly leisure time physical activity for all coronary risk factors were not found among three groups. Because energy expenditure of leisure time was generally low in this subjects and most of them were healthy men, we did not found that leisure time physical activity was significantly associated with coronary risk factors.
