  • Various bacteria were isolated from Korean soil samples based on their capability inhibiting the growth of MRSA strains. Among them, strain YK5 with the highest activity was a Gram positive sporulative bacillus with motility. It did not produce indole and no acid was formed from mannitol by the bacterium. The 16S rRNA sequence of the strain showed $95{\sim}98%$ homology with those of Paenibacillus spp.. The bacterial isolate shared the highest homology with that of P. elgii (98%), but was named as Paenibacillus incheonensis YK5 due to differences in physiological properties. Butanol extract of the P. incheonensis YK5 culture grown in SST medium at $37^{\circ}C$ for 96 hr showed a broad antimicrobial activity against Gram-positive (MRSA and Streptococcus pneumoniae) and negative (Pseudomonas aeruginosa, Salmonella spp., Shigella spp., Escherichia coli, Klebsiella pneumoniae) pathogenic bacteria and fungi (Cryptococcus neoformans and Trichophyton). The antimicrobial activity in the crude extract was stable in a broad range of temperature and pH, $20{\sim}100^{\circ}C$ and $3.0{\sim}6.0$, respectively. Therefore, the antimicrobial activity of P. incheonesis YK5 had potential as a novel antibiotics for pathogens including MRSA.

  • The present study was conducted to examine the outbreak state of bovine dermatophytosis in 14 farms(4 dairy farms, 10 Korean indigenous cattle farms) in Gyeongbuk province from November 2000 to November 2001. The causative agents of dermatophytosis was identified by mycological examination. Antifungal susceptibility test of 26 isolates was performed by agar dilution method, using 5 antifungal drugs. Prevalence of bovine dermatophytosis was found to be 13.5%(90/665) in dairy cattle farms and 14.5%(220/1,520) in Korean indigenous cattle farms. The most common age at which this disease occurred was 2-12 months. This disease usually occurred from winter to spring and the occurrence subsequently decreased in the summer. But 4 Korean indigenous cattle farms with poorly hygienic status were occurred all the year round. The causative agent was identified as Trichophyton verrucosum exclusively in these case. Antifungal susceptibility test of T verrucosum (26 strains) was performed by agar dilution method, using 5 antifungal drugs including tolnaftate, griseofulvin, ketoconazole, amphotericin B and terbinafine. All isolates were highly sensitive to 5 antifungal drugs (geometric mean MICs 0.004∼0.032 $\mu\textrm{g}$/$m\ell$). The isolates were the most sensitive to especially tolnaftate.

  • An epidemiological study was performed on zoophilic dermatophytes. The number of patients with zoophilic dermatophytes diagnosed on clinical findings and culture at the Catholic Dermatological Clinic in Daegu City were 1,339 for 10 years from January 1995 to December 2004. Species of zoophilic dermatophytes isolated from 47,925 patients with dermatophytes were as follows; Trichophyton(T.) rubrum was isolated from 43,702 (91.2%), T. interdigitale 2,660 (5.6%), Epidermophyton(E.) floccosum 43 (0.1%), T. tonsurans 131 (0.3%), T. mentagrophytes 625 (1.3%), T. verrucosum 56 (0.2%), Microsporum(M.) canis 658 (1.4%), and M. gypseum 50 (0.1%). Zoophilic dermatophytes infections were chiefly Tinea facial in the cases of T. mentagrphytes and Tinea corporis in the cases of T. verrucosum, but Tinea capitis in the cases of M. canis. Number of patients with zoophilic dermatophytes by site and age were as follows; Most patients with infected scalps were young people under 15 years old. Infection of the trunk area (neck, chest, back) and extremities (arms, legs, hands, feet) were relatively high in those patients over 15 years old. M. canis were most frequently isolated in autumn and winter, and T. mentagrophyts and T. verrucosum were mostly in spring and winter.

  • Degradations of pine, yellow poplar and sweet gum by two fungi, Pycnoporus cinnabarinus and Trichophyton rubrum LSK-27 were investigated. P. cinabarinus degraded pine block samples much faster than T rub rum LSK-27, whereas P. cinnabarinus and T rubrum LSK-27 degraded yellow poplar and sweet gum at almost the same rate. In an effort to get a better understanding of how fungi degrade lignin in wood, contents of various functional groups were analyzed. After three-months of degradation of pine flour by these fungi, the following changes were observed: an increase in condensed phenolic OH group and carboxylic acid group content, a decrease in the guaiacyl phenolic OH content, and little change of aliphatic OH group content. Further studies in the degradation of pine flour by P. cinnabarinus indicated that the increase in condensed phenolic OH group content and the decrease in guaiacyl phenolic OH group content occurred in the first month of the degradation. The changes of functional group contents in the degradation of unbleached softwood kraft pulp by P. cinnabarinus had the same trends as those in the degradation of pine flour. That is, structural alteration of lignin due to the kraft pulping process had little effect on how P. cinnabarinus degraded lignin.

  • Xylanase (X) derived from Aurreobasidium pullulans and laccase-mediator system (LM) using Trichophyton sp. LKY-7 laccase (TrL) and N-hydroxy-2-pyridone analogue (NHP) as a mediator were applied in hardwood kraft pulp (HwKP) bleaching. The individual and the synergistic effects of X and LM stage were investigated in the enzymatic bleaching of HwKP. Also, the effects of subsequent alkaline extraction (E) and alkaline/hydrogen peroxide treatment (P) were examined. In X or LM treatment alone, an appreciable bleaching effect of HwKP was not observed, whereas subsequent E or P stage enhanced the increase of brightness and the decrease of kappa number. Especially, P stage significantly enhanced the bleaching effect of pulp. Bleaching of HwKP with XLM sequentially gave significantly higher pulp brightness and lower kappa number than that obtained after the treatment of HwKP with X+LM simultaneously. When HwKP was sequentially treated with XLM followed by P stage, the brightness increased by about 11% ISO and the kappa number decreased by about 3.6 in comparison with the initial pulp. Xylanase and laccase were strongly inactivated by NHP both in the absence and the presence of pulp.

  • The aim of this study was to compare the results of morphological phenotypic examination and molecular biological method, randomly amplified polymorphic DNA (RAPD) using clinical isolates from Korean patients infected with dermatophytes. A total of 44 clinical isolates have been collected by Korean Collection Medical Fungi (KCMF) and those were subjected for the RAPD analysis. The results showed that the fungal strains that have RG1 band patterns were identified as Trichophyton rubrum by the neighbour-joining analysis. Additionally, other fungal strains that have RG3 band patterns were identified as Epidermophyton floccosum and RG5 band patterns were identified as Micorsporum gypseum. Even though molecular biological method such as RAPD are not necessary, it might be useful when some strains have similar morphological characteristics or when same species has phenotypic variations. In this study, therefore, we targeted internal transcribed spacer (ITS) regions of fungal rDNA, performed RAPD and then compared with morphological phenotypic examination.

  • A series of 6-[(N-Halophenyl)amino] -7-Bromo-5,8-quinolinediones (BQs) newly synthesized were tested for antifungal activities, in vitro, against Candida sp., Aspergillus niger and Trichophyton mentagrophytes. The MIC values were determined by the twofold dilution method. Among these derivatives, BQ4 and 7 showed more potent antifungal activities than fluconazole and ketoconazole. BQ 4 and 7 were compared with fluconazole in the treatment of established systemic infections with Candida albicans in normal rats. Intraperitoneally administered BQs for 7 days and 14 days reduced Candida albicans colony count in kidneys and livers as well as fluconazole. The therapeutic potential of BQs has been assessed by evaluating their activities (survival rate) against systemic infections in normal mice with Candida albicans. These compounds, particularly showed activities comparable with fluconazole. BQ4 and 7 had about EO$\_$50/, 0.05, 0.06mg/kg but fluconazole had ED$\_$50/, 6.0mg/kg respectively.

  • A series of 6-(N-arylamino)-7-alkylthio-5,8-quinolinedione derivatives(SQ1-12) were newly synthesized for the evaluation of antifungal activities. 6-(N-Arylamino)-7-chloro-5,8-quinolinediones (RCKs) were treated with Na$_2$S/(CH$_3$)$_2$SO$_2$ in EtOH In give SQs. RCKs were prepared by regioselective nucleophilic substitution of 6,7-dichloro-5,8-quinolinediones with arylamines. In the presence of CeCl$_3$, the N-arylamino groups were introduced at the 6-position of 5,8-quinolinedione ring by the regioselective substitution. These derivatives 1-12 were tested for antifungal and also antibacterial activities, in vitro, against Candida sp., Aspergillus niger and Trichophyton mentagrophytes. The MIC values were determined by the two-fold dilution method.

  • The actinomycete strain KH-6l4 possessed strong antifungal activity, especially antagonistic to the rice blast fungus, Pyricularia oryzae. Diaminopimelic acid (DAP) type and morphological and physiological characteristics, examined by scanning electron microscopy (SEM), indicated that KH-614 belonged to the genus Streptomyces. Antifungal agent produced by this strain was found to be most active, when the strain was cultured in the presence of glucose, polypeptone, and yeast extract (PY) medium for 6 days at $27^{\circ}C$. Based on the spectral report data, MS and NMR, the antifungal agent was identified as cyclo(L-leucyl-L-prolyl). According to the antimicrobial activity test measured by minimal inhibitory concentration (MIC), the cyclo(1eu-pro) exhibited the activity against Candida albicans IAM 4905, Mucor ramannianus IAM6218, Rhizoctonia solani IFO 6218, Aspergilus fumigatus ATCC 42202, Glomerella cingulata IFO 9767, Trichophton mentagrophytes ATCC 18749, and Trichophyton rubrum ATCC 44766, the order of MIC values were 50, 12.5, 5, 50, 25, 5, $5\;\mu\textrm{g}/ml$, respectively. Specifically, cyclo(1eu-pro) was one of the most effective elements against Pyricularia oryzae IFO 5994 with the MIC value of $2.5\;\mu\textrm{g}/ml$, thus indicating that cyclo(leu-pro) is a potential antifungal agent.

  • The anaerobically digestion and agricultural application of organic wastes was conducted using food wastes and cow dung. Twenty kilograms each of the feed stocks was added into two 30 liters-capacity batch digesters. The anaerobic digestion was carried out within a temperature range of $25-31^{\circ}C$ for a retention time of 51 days. The results showed a cumulative gas yield of 5.0 bars for food waste and no gas production for cow dung within the retention time. Bacteria such as Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, Proteus vulgaris and Clostridium sp were isolated. Fungi isolated included Aspergillus niger, Aspergillus nidulan, Trichophyton rubrum and Epidermophyton flocossum. The non-dispersive infrared (NDIR) analysis of the biogas produced confirmed that the gas consisted of $CH_4$, $CO_2$ and $H_2$. Statistical analysis revealed there was no significant correlation between temperature and biogas produced from the organic wastes (r= 0.177, p = 0.483).The organic wastes from the biogas production process stimulated maize growth when compared to control (soil without organic waste) and indicated maximum height. The study therefore reveals that food waste as potential substrates for biogas production has a moderate bio-fertilizer potential for improving plant growth and yield when added to soil.
