Contaminant transport in porous media is characterized by solving an advection-dispersion equation(ADE). The ADE can cover equilibrium phenomena of interest, which include sorption, decay, and chemical reactions. Among these phenomena, sorption mechanism is described by several types of sorption isotherm. If we assume the sorption isotherm as linear, the solution of ADE can be easily procured. However, if we consider the sorption isotherm as non-linear isotherm like a Dual Reactive Domain Model (DRDM), the resulting differential equation becomes non-linear. In this case, the solution of ADE cannot be easily acquired by an analytic method. In this paper, we present the numerical analysis of ADE using a DRDM. The results reveal that even if sorption data may be fitted well using linear or non-linear isotherm, the characteristics of contaminant transport of the two cases are different from each other. To be concrete, the retardation of linear isotherm has stronger effect than that of the DRDM. As the non-linearity of sorption isotherm increases, the difference of retardation effects of the two cases becomes larger. For a pulse source, the maximum concentration of the linear model is higher than that of the DRDM, but the plume of the DRDM moves faster than that of the linear model. Behaviors of contaminant transport using the DRDM are consistent with common features of a linear model. For instance, biodegradation effect becomes larger as time goes by The faster the seepage velocity is, the faster the plume of contaminant moves. The plume of the contaminant is distributed evenly over overall domain in the event of high dispersion coefficient.
Cis-dichlorodiammine platin${\mu}M$II (Cisplatin), an effective chemotherapeutic agent, induces acute renal failure by unknown mechanisms. To investigate direct toxic effects of cisplatin on the renal proximal tubular transport system, LLC-$PK_1$ cell line was selected as a cell model and the sugar transport activity was evaluated during a course of cisplatin treatment. Cells grown to confluence were treated with cisplatin for 60 min, washed, and then incubated for up to 5 days. At appropriate intervals, cells were tested for sugar transport activity using ${\alpha}-methyl-D-[^{14}C]glucopyranoside$ (AMG) as a model substrate. In cells treated with 100 ${\mu}M$ cisplatin, the AMG uptake was progressively impaired after 3 days. The viability of cells was not substantially changed with cisplatin of less than 100 ${\mu}M$, but it decreased markedly with 150 and 200 ${\mu}M$. In cisplatin-treated cells, the $Na^+$ -dependent AMG uptake was drastically inhibited with no change in the $Na^+$ -independent uptake. Kinetic analysis indicated that Vmax was suppressed, but Km was not altered. The $Na^+$ -dependent phlorizin binding was also decreased in cisplatin-treated cells. However, the AMG efflux from preloaded cells was not apparently retarded by cisplatin treatment. These data indicate that the cisplatin treatment impairs $Na^+$ -hexose cotransporters in LLC-$PK_1$ cells and suggest strongly that defects in transporter function at the luminal plasma membrane of the proximal tubular cells constitute an important pathogenic mechanism of cisplatin nephrotoxicity.
The effects of 5 novel hepatotrophic agents, dithiol malonate derivatives (DMDs; DMD1-DMD5), on the liver microsomal lipid peroxidation induced by carbon tetrachloride $(CCl_4)$ and the correlations with the changes of microsomal electron transport system were investigated. All DMDs were found to inhibit the lipid peroxidation induced by $CCl_4$ in mice and rats as well in vitro liver microsomal system. Therefore, each DMD seemed to have direct mode of action on liver microsomes to inhibit the lipid peroxidation. As an ex vivo study, the induced lipid peroxidation by $CCl_4$ and the changes in electron transport system were determined with liver microsomes obtained from rats chronically treated with DMDs for 7 days. The induced lipid peroxide contents in liver microsomal system were lower in DMD1, DMD2 and DMD3 treated group, but higher in DMD4 and DMD5 group when compared to the control group. Cyt. p.450 contents in the microsomes were decreased by the treatment with DMD1, DMD2 and DMD3, but increased significantly by DMD4 with great extent and by DMD5 with less extent. The cyt. p-450 isozymes induced by treatment of DMD4 and DMD5 were identified as 3-methylcholanthrene (MC) type. The NADPH cyt. -C reductase activities of the microsomes treated with DMD1, DMD2, DMD4 and DMD5 were increased in the range of around 20% to 50%, but decreased with DMD3, All DMDs increased dyt. $-b_5$ content and did not alter NAdH-cyt, $-b_5$ reductase activities in the microsomes. In summary, the 5 novel hepatotrophic agents (DMDs) markedly protected against lipid peroxidation induced by $CCl_4$ in vivo and in vitro possibly through the mechanism of direct action on the liver microsomes. The degree of inhibition produced by DMDs on lipid peroxidation induced by $CCl_4$ seemed to coincide rather with cyt. p-450 contents than with other components of liver microsomal electron transport system including NADPH-cyt, -C reductase.
Microtubule-based kinesin motor proteins are used for long-range vesicular transport. KIF5s (KIF5A, KIF5B and KIF5C) mediate the transport of various membranous vesicles along microtubules, but the mechanism behind how they recognize and bind to a specific cargo has not yet been completely elucidated. To identify the interaction protein for KIF5B, yeast two-hybrid screening was performed and a specific interaction with the unconventional myosin Myo9b, an actin-based vesicle transport motor, was found. The GTPase-activating protein (GAP) domain of Myo9s was essential for interaction with KIF5B in the yeast two-hybrid assay. Myo9b bound to the carboxyl-terminal region of KIF5B and to other KIF5 members. In addition, glutathione S-transferase (GST) pull-downs showed that Myo9s specifically interact to the complete Kinesin-I complex. An antibody to KIF5B specifically co-immunoprecipitated KIF5B associated with Myo9s from mouse brain extracts. These results suggest that kinesin-I motor protein interacts directly with actin-based motor proteins in the cell.
Catchments soil erosion, one of the most serious problems in the mountainous environment of the world, consists of a complex phenomenon involving the detachment of individual soil particles from the soil mass and their transport, storage and overland flow of rainfall, and infiltration. Sediment size distribution during erosion processes appear to depend on many factors such as rainfall characteristics, vegetation cover, hydraulic flow, soil properties and slope. This study involved laboratory flume experiments carried out under simulated rainfall in a 3.0 m long ${\times}$ 0.8 m wide ${\times}$ 0.7 m deep flume, set at $17^{\circ}$ slope. Five experimental cases, consisting of twelve experiments using three different sediments with two different rainfall conditions, are reported. The experiments consisted of detailed observations of particle size distribution of the out-flow sediment. Sediment water mixture out-flow hydrograph and sediment mass out-flow rate over time, moisture profiles at different points within the soil domain, and seepage outflow were also reported. Moisture profiles, seepage outflow, and movement of overland flow were clearly found to be controlled by water retention function and hydraulic function of the soil. The difference of grain size distribution of original soil bed and the out-flow sediment was found to be insignificant in the cases of uniform sediment used experiments. However, in the cases of non-uniform sediment used experiments the outflow sediment was found to be coarser than the original soil domain. The results indicated that the sediment transport mechanism is the combination of particle segregation, suspension/saltation and rolling along the travel distance.
In-situ photolysis is one of the most promising ways to clean up a soil contaminated with 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD). This study focuses on the mathematical description and model development of the convective upward transport of an organic solvent driven by evaporation and photodecomposition at the surface as the major transport mechanism in the clean up process. A finite-element-based numerical model was proposed to incorporate effects of multiphase flow on the distribution of each fluid, gravity as a driving force, and the use of van Genutchen equation for more accurate description of k-S-p relations. This paper presents results of extensive numerical calculations conducted to investigate the various parameters that play a role in the solvent migration through a laboratory-scale unsaturated soil column. The numerical results indicate that gravity affects significantly on the fluids distribution and evaporation for highly permeable soils. The soil texture has a profound influence on the fluid saturation profile during evaporation process. The amount of solvent convective motion increases with increasing evaporation rates and decreasing initial water saturation. Simulations conducted in this study have shown that the developed model is very useful in analyzing the effects of various parameters on the convective migration of an organic solvent in the soil environments.
This study was undertaken to examine the effect of oxidants on membrane transport function in renal epithelial cells. Hydrogen peroxide ($H_2O_2$) was used as a model oxidant and the membrane transport function was evaluated by measuring $Na^+$-dependent phosphate ($Na^+$-Pi) uptake in opossum kidney (OK) cells. $H_2O_2$ inhibited $Na^+$-Pi uptake in a dose-dependent manner. The oxidant also caused loss of cell viability in a dose-dependent fashion. However, the extent of inhibition of the uptake was larger than that in cell viability. $H_2O_2$ inhibited $Na^+$-dependent uptake without any effect on $Na^+$-independent uptake. $H_2O_2$-induced inhibition of $Na^+$-Pi uptake was prevented completely by catalase, dimethylthiourea, and deferoxamine, suggesting involvement of hydroxyl radical generated by an iron-dependent mechanism. In contrast, antioxidants Trolox, N,N'-diphenyl-p-phenylenediamine, and butylated hydroxyanisole did not affect the $H_2O_2$ inhibition. Kinetic analysis indicated that $H_2O_2$ decreased Vmax of $Na^+$-Pi uptake with no change in the Km value. Phosphonoformic acid binding assay did not show any difference between control and $H_2O_2$-treated cells. $H_2O_2$ also did not cause degradation of $Na^+$-Pi transporter protein. Reduction in $Na^+$-Pi uptake by $H_2O_2$ was associated with ATP depletion and direct inhibition of $Na^+$-$K^+$-ATPase activity. These results indicate that the effect of $H_2O_2$ on membrane transport function in OK cells is associated with reduction in functional $Na^+$-pump activity. In addition, the inhibitory effect of $H_2O_2$ was not associated with lipid peroxidation.

Objective: Omasum is an important site for the absorption of short chain fatty acids. The major route for the transport of acetate is via sodium hydrogen exchanger (NHE). However, a discrepancy in the symmetry of sodium and acetate transport has been previously reported, the mechanism of which is unclear. In this study, we investigated the possible role of carbonic anhydrase (CA) for this asymmetry. Methods: Omasal tissues were isolated from healthy sheep (N = 3) and divided into four groups; pH 7.4 and 6.4 alone and in combination with Ethoxzolamide. Electrophysiological measurements were made using Ussing chamber and the electrical measurements were made using computer controlled voltage clamp apparatus. Effect(s) of CA inhibitor on acetate and sodium transport flux rate of Na22 and 14C-acetate was measured in three different flux time periods. Data were presented as mean±standard deviation and level of significance was ascertained at p≤0.05. Results: Mucosal to serosal flux of Na (JmsNa) was greater than mucosal to serosal flux of acetate (JmsAc) when the pH was decreased from 7.4 to 6.4. However, the addition of CA inhibitor almost completely abolished this discrepancy (JmsNa ≈ JmsAc). Conclusion: The results of the present study suggest that the additional protons required to drive the NHE were provided by the CA enzyme in the isolated omasal epithelium. The findings of this study also suggest that the functions of CA may be exploited for better absorption in omasum.

Red algal fertilization is unusual and offers a different model to the mechanism of intracellular transport of nuclei and polyspermy blocking. A female carpogonium (egg) undergoes plasmogamy with many spermatia (sperm) simultaneously at the receptive structure, trichogyne, which often contains numerous male nuclei. The pattern of selective transport of a male nucleus to the female nucleus, located in the cell body of the carpogonium, remain largely unknown. We tracked the movement of spermatial nuclei and cell organelles in the trichogyne after plasmogamy using time-lapse videography and fluorescent probes. The fertilization process of Bostrychia moritziana is composed of five distinctive stages: 1) gamete-gamete binding; 2) mitosis in the attached spermatia; 3) formation of a fertilization channel; 4) migration of spermatial nuclei into the trichogyne; and 5) cutting off of the trichogyne cytoplasm from the rest of the cell after karyogamy. Our results showed that actin microfilaments were involved in the above steps of fertilization, microtubules are involved only in spermatial mitosis. Time-lapse videography showed that the first ("primary") nucleus which entered to trichogyne moved quickly to the base of carpogonium and fused with the female nucleus. The transport of the primary male nucleus to the egg nucleus was complete before its second nucleus migrated into the trichogyne. Male nuclei from other spermatia stopped directional movement soon after the first one entered the carpogonial base and oscillated near where they entered trichogyne. The cytoplasm of the trichogyne was cut off at a narrow neck connecting the trichogyne and carpogonial base after gamete nuclear fusion but gamete binding and plasmogamy continued on the trichogyne. Spermatial organelles, including mitochondria, entered the trichogyne together with the nuclei but did not show any directional movement and remained close to where they entered. These results suggest that polyspermy blocking in B. moritziana is achieved by the selective and rapid transport of the first nucleus entered trichogyne and the rupture of the trichogyne after gamete karyogamy.
Plastic mechanism analysis of structures subjected to large deformation has long been used in order to determine collapse mechanisms of steel structures, and the energy absorbed in plastic deformation during such collapses. In this paper the technique is applied to vehicle roof structures that undergo large plastic deformation as a result of rollover crashes. The components of such roof structures are typically steel spot-welded hat-type sections. Ten different deformation mechanisms are defined from investigations of real-world rollover crashes, and an analytical technique to determine the plastic collapse load and energy absorption of such mechanisms is determined. The procedure is presented in a generic manner, such that it may be applied to any vehicle structure undergoing a rollover induced collapse. The procedure is applied to an exemplar vehicle, in order to demonstrate its application in determining the energy absorbed in the deformation of the identified collapse mechanisms. The procedure will be useful to forensic crash reconstructionists, in order to accurately determine the initial travel velocity of a vehicle that has undergone a rollover and for which the post-crash vehicle deformation is known. It may also be used to perform analytical studies of the collapse resistance of vehicle roof structures for optimisation purposes, which is also demonstrated with an analysis of the effect of varying the geometric and material properties of the roof structure components of the exemplar vehicle.
