Comparative analysis of active components and antioxidant activities of Brussels sprouts (Brassica oleracea var. gemmifera) and cabbage (Brassica oleracea var. capitata)

  • The applicability of Brussels sprouts, which are widely cultivated in Jeju, as a functional biomaterial in the cosmetics and food industries is investigated. The active ingredients (total phenols and flavonoids, β-carotene, vitamin C, free sugar, minerals, glucosinolates, and isothiocyanates) and antioxidant activities of 70% ethanol extracts of Brussels sprouts and cabbage were analyzed. The total phenol, flavonoid, vitamin C, and β-carotene contents of Brussels sprouts were approximately 36%, 2.5 times, 2.3 times, and 65% higher than those of cabbage, respectively. The total free sugar content of Brussels sprouts was 58%-72% lower than that of cabbage. The K content was the highest among the other minerals, and there was little difference between the two samples. The Na content was approximately three times higher in cabbage than in Brussels sprouts. The total glucosinolate content of Brussels sprouts was 34.5 mol SE/g DW, twice that of cabbage. The sinigrin content of Brussels sprouts was thrice (10.06 μmol/g DW) that of cabbage. The isothiocyanate (sulforaphane, I3C) content in Brussels sprouts was double that of cabbage. The antioxidant activity of Brussels sprout extract was 2.5 to 2.8 times higher in 1,1-Diphenyl-2-picrylhydrazyl radical scavenging capacity and 3.3 to 3.6 times higher in 2,2'-Azino-bis(3-ethybenzothiazoline-6-sulfonic acid) radical scavenging capacity than those of cabbage extract. These are important basic data for the study of glucosinolates and isothiocyanates, which have anticancer activity, as well as antioxidant-related substances of Brussels sprout, which has high potential for use as a biomaterial in functional foods and cosmetics.

Determination of Phytochemical Contents and Biological Activities from the Fruits of Elaeagnus multiflora

  • The purpose of this study was to investigate the phytochemical contents and biological activities, including $\alpha$-glucosidase inhibitory activity and antioxidant activity, from the fruits of Elaeagnus multiflora using different solvent systems. The 50% ethanol extract of this species showed the most potent $\alpha$-glucosidase inibitory activity (88.5%) at a concentration of 1 mg/mL. Moreover, this extract also displayed the strongest antioxidant activity in each assay, showing 96.3% in DPPH scavenging activity, 98.2% in ABTS scavenging activity, and 2.5% in reducing power. The highest total phytochemical contents, including flavan-3-ols, phenolic acids, and flavonoids, were observed in the 50% ethanol extract. Interestingly, flavan-3-ols (3.150 mg/g) were detected at a significantly higher total content than those of phenolic acids (0.380 mg/g) and flavonoids (not detected). Additionally, the contents of individual phytochemicals showed remarkable differences, especially the epicatechin gallate (2.008 mg/g) and gallic acid (0.099 mg/g), which were the predominant constituents of each phytochemical type. Therefore, our results suggest that the 50% ethanol extract of E. multiflora fruits has strong biological activities, which are correlated with high phytochemical contents.

Antioxidant Activity of Deodeok (Codonopsis lanceolata) Sprout (새싹 더덕의 항산화 활성)

  • The purpose of this study was to determine the content of polyphenols and flavonoids, vitamin C, and antioxidant activity for the extract from the Deodeok sprout. To accomplish this, the Deodeok sprout whole (CLS-W), above ground part (leaf, stem, CLS-L), and root (CLS-R) were individually extracted using 70% ethanol. The highest levels of total polyphenols and total flavonoids were observed in the Deodeok sprout extract CLS-L2. Similarly, antioxidant activities resulting in radical scavenging activities increased significantly in the extract of CLS-L2. In conclusion, these results indicate that Deodeok sprouts can be used as a viable, new natural antioxidant source.

A Comparison of Antioxidant Effects among Non-fermented and Fermented Columbian Coffee, and Luwak Coffee Beans (발효 유무에 따른 콜롬비아 커피와 루왁커피의 항산화 활성 비교연구)

  • The purpose of this study was to investigate the antioxidant effects of non-fermented (CAC) and Monascus pilosus-fermented Columbia arabica coffee (FCAC), as well as Luwak coffee (LC) beans. The results indicated that total polyphenols content (mg/g of dry basis) was highest in CAC (70.69), followed by LC (62.07), and FCAC (41.38). However, the ratio of total flavonoids/polyphenols in FCAC was the highest. In terms of electron donating ability (%, coffee mg/mL), CAC was significantly higher than LC and FCAC. Regardless of fermentation, ferric reducing antioxidant powers were similar in CAC and FCAC and lowest in LC. LC also had the highest inhibitory activity against xanthine oxidase (XO). However FAAC had the highest inhibitory activity against aldehyde oxidase (AO), with nearly three times the levels found in CAC and LC. According to the above results, FCAC had a higher ratio of flavonoids/polyphenols and iron chelating activity than CAC. FCAC also had the highest AO inhibitory activity among the three experimental coffee beans. The results suggest that further studies are required to evaluate the bioactive components of various coffee beans so as to determine the potential benefits that coffee may have on preventing oxidative stress-related conditions.

Correlation between phytochemical contents in peel and flesh of cold-stored Fuji apple (저온 저장 후지 사과의 과피와 과육 중 파이토케미컬 함량의 상관관계)

  • This study determined the relationships between the total anthocyanin content in apple skin and the total polyphenols, flavonoids, proanthocyanidins, and soluble solids contents in the flesh of cold-stored Fuji apples. Total anthocyanin content in apple skin ranged from 0.130±0.005 mg CE/g fw to 0.262±0.028 mg CE/g fw, and the overall average was 0.200±0.008 mg CE/g fw. The total polyphenolic compounds in the flesh was ranged from a 4.283±0.141 mM GAE/g fw to 8.207±0.234 mM GAE/g fw, and the average content was 6.275±0.177 mM GAE/g fw. The total flavonoid content ranged from 4.510±0.080 mM QE/g fw to 2.467±0.458 mM QE/g fw, and the average total flavonoid content was about 3.586 mM QE/g fw. The total proanthocyanidin content was relatively high, ranging from 3.475±0.577 mM EE/g fw to 6.816±0.277 mM EE/g fw, and the soluble solid in the flesh was about 12 °Brix to 14 °Brix. The DPPH radical scavenging activity of extracts from apple flesh ranged from 66.36% to 94.99%, and the ascorbate equivalent concentration was 0.482 mM. The ABTS radical scavenging activity was 99.12% to 99.9%, indicating a higher inhibitory activity than the DPPH inhibitory activity, and the ascorbate equivalent concentration was 0.486 mM. The correlation between the total anthocyanin and total polyphenolic compounds was y = 15.192x + 3.2169 (R2=0.2748), but the concentration of total polyphenolic compounds increased when the total anthocyanin content was increased. The correlation equation of total anthocyanin with total flavonoids was y = 15.18x + 0.5555 (R2=0.6226), with total proanthocyanin was y = 14.918x + 2.3422 (R2=0.3372), and with soluble solid was y = 10.558x + 11.126 (R2=0.1925), indicating that the correlation of total anthocyanin with total flavonoid was higher than that with soluble solid.

Antioxidant Capacity and Bioactive Composition of a Single Serving Size of Regular Coffee Varieties Commercially Available in Korea (시판 커피 한 컵에 함유된 생리활성 성분과 항산화활성)

  • The major hydrophilic bioactive compounds (chlorogenic acid, caffeine, total phenolics, and flavonoids) and the antioxidant capacity in a single size were evaluated for regular coffee varieties commercially available in Korea. The content of total phenols (63.83-110.12 mg gallic acid equivalents) and flavonoids (35.27-69.27 mg catechin equivalents) were spectrophotometrically determined, and the content of chlorogenic acid (5.17-69.78 mg) and caffeine (74.38-146.32 mg) were determined using HPLC-UV. All varieties of regular coffee studied showed antioxidant potential (88.78-487.52 mg trolox equivalents/serving size), which was conferred by their concentrations of phenolic compounds, caffeine and chlorogenic acid. There were significant (p<0.01) correlations between the total phenolics ($r^2$=0.732) and flavonoids ($r^2$=0.8705) and the antioxidant capacity, suggesting that these components were likely a significant contributor to the antioxidant capacity of commercial regular coffee brews.

In vitro anti-oxidant activity of the leaves of Dillenia indica

  • The methanol extract of Dillenia indica was tested for antioxidant activity as determined by free radical scavenging of DPPH radical scavenging assay, reducing power, total antioxidant capacity measured by phosphomolybdenum method, total phenolic content and total flavonoids content determination assays. The extract showed significant activities in all antioxidant assays compared to the standard antioxidant in a dose dependent manner. In DPPH radical scavenging assay the $IC_{50}$ value of the extract was found to be 100.53${\mu}g/ml$ while ascorbic acid has the $IC_{50}$ value 58.92${\mu}g/ml$. Dillenia indica extract showed strong reducing power and total antioxidant capacity. Moreover, methanol extracts also possess high amount of phenolics and flavovonoids and expressed as gallic acid and rutin equivalent respectively. The remarkable activities exhibited in reactive oxygen species scavenging may attributed to the high amount of hydrophilic phenolics present in Dillenia indica.

Evaluation of Proanthocyanidin Contents in Total Polyphenolic Compounds of Pine (Pinus densiflora) Needle Extracts and Their Antioxidative Activities (적송잎 용매별 추출물의 총 폴리페놀성 성분에 대한 proanthocyanidin의 함량 분석 및 항산화 활성 검증)

  • Flavonoids in pine needles are known to be effective scavengers of free radical. Especially, proanthocyanidin, a kind of flavonoids possesses cardiovascular protection and antioxidative activities. Here, we evaluated proanthocyanidin contents in total polyphenolic compounds of pine needle extracts prepared by using hot water, ethanol, hexane or sub-supercritical $CO_2$. Analyses of total polyphenolic compounds and proanthocyanidin in each extracts indicated that hot water extract contained the highest concentrations, but sub-supercritical extract contained the lowest concentrations. On the other hand, evaluation of proanthocyanidin contents in total polyphenolic compounds in each extracts showed that sub-supercritical extract possessed the highest content, but hot water extract possessed the lowest content. These results indicate that extracts containing high concentrations of both total polyphenolic compounds and proanthocyanidin could be obtained by using hot water or ethanol extractions. Furthermore, extract containing high content of proanthocyanidin out of total polyphenolic compounds could be achieved by using sub-supercritical extraction. Measurement of antioxidative activities of extracts showed that hot water extract possessed the highest activity. In this study, we prepared extracts from pine needles by four different methods and evaluated the antioxidative compounds in extracts that could be used for effective components of functional food products.

Yield Analysis of Flavonoids in Acanthopanax divaricatus and A. koreanum Grown using Different Cultivation Methods

  • High-performance liquid chromatography was performed in order to analyze the changes in the flavonoid content (rutin, hyperin, afzelin, quercetin, and kaempferol) of Acanthopanax divaricatus and A. koreanum, in response to different cultivation methods (pinching height, planting time, and top dressing). The total flavonoid content of A. divaricatus and A. koreanum ranged from 0.201 to 0.690 mg/g with different pinching heights, 0.143 to 1.001 mg/g for different planting times, and 0.156 to 1.074 mg/g depending on the rate of fertilizer application. In both A. divaricatus and A. koreanum, the total flavonoid content in the upper section of the plant was greater than that in the lower section. These results demonstrate which cultivation methods maximize the flavonoid content of A. divaricatus and A. koreanum, and thus help to optimize flavonoid yields to improve production for nutraceutical, pharmaceutical, and cosmeceutical applications.

Total Polyphenols, Total Flavonoid Contents, and Antioxidant Activity of Korean Natural and Medicinal Plants (자생식물과 생약자원 추출물의 폴리페놀, 플라보노이드 함량 및 항산화 활성 탐색)

  • The purpose of this study was to investigate total polyphenols, total flavonoid, and DPPH radical scavenging activity in 26 species of Korean natural plants and 14 species of medicinal plant extracts. The plant containing the highest total polyphenols and total flavonoids were Plantago asiatica, and Lespedeza cuneata G. Don, respectively. The content of polyphenols and flavonoids in natural plants was the highest in Lespedeza cuneata G. Don, which also demonstrated high DPPH radical scavenging activity. In medicinal plant extract, Saururus chinensis (Lour.) Baill and Artemisiae Asiaticae Herba had the highest level of total polyphenols (194.60 and 203.92 mg/g extract, respectively) and DPPH radical scavenging activity. Diospyros kaki Thunb and Dryopteris crassirhizoma did not contain high levels of total polyphenols (112.98, 121.46 mg/g extract, respectively) and flavonoids (46.10, 58.69 mg/g extract respectively), but DPPH radical scavenging activity was relatively high. Further study regarding plants that exhibit higher total polyphenols, total flavonoid, and DPPH radical scavenging activity is needed.
