목적: 단백뇨 질환의 실험모델인 puromycin aminonucleoside (PAN)에서 관찰할 수 있는 족세포의 병리학적 이상에 있어서 P-cadherin의 변화를 생체 외 족세포 배양실험을 통하여 알아보고자 하였다. 방법: PAN에 의한 족세포의 P-cadherin의 변화를 생체 외 배양실험을 통해 알아보고자 백서 사구체 상피세포(GEpC)를 배양하여 다양한 농도의 PAN을 투여하여 confocal 현미경을 통하여 P-cadherin의 분포를 관찰하였고, Western blotting 과 RT-PCR을 사용하여 P-cadherin 발현의 변화를 관찰하였다. 결과: 외곽세포질에 분포하는 P-cadherin은 단일세포 혹은 응집환경에서 PAN의 농도가 올라갈수록 내부로 응집되는 현상를 볼 수 있었다. Western 분석에서, P-cadherin 단백양은 PAN에 농도-의존적으로, 특히, 고농도인 50 mg/mL에서 24시간과 48시간이 노출 조건에서 각각 21.9% (P< 0.05)와 31.9% (P<0.01)의 의미 있는 감소소견을 보였다. RT-PCR에서도 48시간에서 50 mg/mL PAN을 첨가한 조건에서 23.5%의 의미 있는 감소를 보였다(P<0.05). 결론: PAN은 족세포에서 P-cadherin을 세포막으로부터 내부로의 응집을 유발하고, P-cadherin mRNA의 발현 감소와 단백수준에서 양의 감소를 초래함으로서, 단백뇨의 발생에 기여할 것이라 사료된다.
소르빈산 칼륨은 소르빈산의 일종으로 항세균 및 곰팡이 발육 저지 효과를 갖고 있다. 이러한 성질로 인하여 소르빈산 칼륨은 곰팡이와 진균에 대한 식품 방부제로서 사용되고 있다. 소르빈산 칼륨은 많은 양에 노출되었을 때도 체내에서 수분과 이산화탄소로 분해되는 특성으로 인하여 독성이 없는 것으로 알려져 있다. gap junction을 통한 세포 간 신호 전달(GJIC)는 생체의 성장과 분화에 있어서 조직의 항상성 유지에 본질적인 역할을 하고 있다. 본 연구는 WB-F344 랫드의 정상 간 상피 세포(WB 세포)에서 소르빈산 칼륨을 노출하였을 때 GJIC에 대하여 어떤 영향을 미치는지 알아보았다. 그 결과 소르빈산 칼륨에 노출에 의하여 WB세포의 GJIC가 농도 및 시간 의존적으로 현격하게 감소하는 것을 관찰하였다. 소르빈산 칼륨은 GJIC를 강력하게 억제하며, 이는 WB 세포에서 gap junction을 구성하는 connexin 43의 인산화와 병행하는 것을 확인하였다. 소르빈산 칼륨이 gap junction의 기능을 방해함으로 인해 소르빈산 칼륨에 의한 간독성이 유발될 수 있을 것이다.
본 연구는 양식넙치에 높은 폐사율과 성장저하를 일으키는 스쿠티카섬모충 Miamiensis avidus (syn. Philasterides dicentrarchi)의 성장과 병원성에 미치는 온도의 영향을 알아보았다. in vitro 배양조건에서 이분열에 걸리는 시간은 $5{^{\circ}C}$에서 61.82시간, $10{^{\circ}C}$에서 26.32시간, $15{^{\circ}C}$에서 21.14시간, $20{^{\circ}C}$에서 16.86시간 그리고 25℃에서 는 16.21시간이었다. 최고로 배양 가능한 충수는 $1.54-1.75{\times}10^{5}$/ ml의 범위로 $10{^{\circ}C}$이상의 모든 온도범위에서 유사했다. 넙치(평균 8.34 cm, 4.33g)마리당 $1{\times}10^{2}$/ ul, $1{\times}10^{6}$/ ul, $1{\times}10^{4}$/ ul 과 $1{\times}10^{5}$/ ul 으로 스쿠티카충을 복강에 감염시킨 후 $10{^{\circ}C}$, $15{^{\circ}C}$와 $20{^{\circ}C}$에서 사육하면서 폐사를 관찰하는 실험을 2회 실시한 결과, 수온 $10{^{\circ}C}$에서는 폐사가 현저히 낮았고 온도가 높아질수록 폐사가 증가하였다. 또한 감염농도가 높아질수록 폐사는 증가하였다. 그러므로 스쿠티카섬모충 M. avidus는 온도가 높을수록 증식이 잘 되며 또한 수온이 높아질수록 온도의존적으로 폐사가 증가하므로 폐사를 경감시키기 위해서는 고수온기의 수온관리가 중요한 것으로 사료된다.
본 연구에서는 Comet assay를 이용해서 차 및 전통 약재로 사용하고 있는 한국산 겨우살이의 항유전독성을 측정하고자 하였다. 겨우살이 조추출물을 분자량에 따라 14,000 이상 분획(I), $8,000{\sim}14,000$의 분획(II), $3,500{\sim}8,000$ 분획(III) 그리고 3,500 이하 분획(IV)으로 분리하였다. 이중 분획 IV만이 비종양성 3T3 세포에서 농도에 따른 MNNG에 의한 DNA 손상을 감소시켰으며 겨우살이 lectin과 viscotoxin이 포함된 분획 I, III은 활성을 전혀 나타내지 않았다. 열처리에 의해서도 분획 IV는 시간이 지남에 따라서 활성이 약간 감소됨을 나타내었으나 여전히 유의적인 효과를 지니고 있었다. 겨우살이 분획 IV로부터 얻어진 유기용매 분획의 항유전독성 평가결과, 분획 IV의 항유전독성은 주로 수용성 물질에 의해 나타나는 것임을 알 수 있었다. Sephadex G-25 gel filtration chromatography에서, 겨우살이 분획 IV의 항유전독성에 기여하는 성분은 분자량 1,000 미만의 수용성 물질이었으며 이 저분자량 물질을 분리해내어 암 예방을 위한 기능성 식품소재로써 이용할 수 있는 가능성을 확인할 수 있었다.
국내에 서식하는 대표적인 무미양서류 중 두꺼비 (Bufo gargarizans), 청개구리 (Hyla japonica), 참개구리 (Rana nigromaculata) 및 무당개구리 (Bombina orientalis)를 대상으로 각 종의 배아 발달에 미치는 Pb의 효과를 FETAX를 활용하여 분석한 결과 4종의 배아 모두 Pb의 농도가 증가함에 따라 사망률, 기형율이 증가하고 다양한 기형 양상이 나타났으며 성장률은 감소하였다. 두꺼비, 청개구리, 참개구리, 무당개구리의 반수치사농도(half maximal lethal concentration; $LC_{50}$)는 각각 0.58, 0.49, 0.52, $0.54mg\;L^{-1}$를 나타내었고 반수영향농도 (half maximal effective concentration; $EC_{50}$)은 각각 0.35, 0.27, 0.30, $0.29mg\;L^{-1}$을 나타냈다. 기형성지수(teratogenic index; $TI=LC_{50}/EC_{50}$)는 두꺼비, 청개구리, 참개구리 및 무당개구리에서 각각 1.66, 1.81, 1.73, 1.86로 나타나 무당개구리가 가장 민감한 영향을 나타냈으며 다음으로 청개구리, 참개구리, 두꺼비 순으로 나타났다. 이는 서로 다른 종이 같은 농도의 오염물질에 노출되었다 하더라도 종에 따라 차이가 나타날 수 있음을 의미한다. 이상의 결과를 통해 Pb가 무미양서류 4종의 배아 발달에 기형성 물질로 작용함을 알 수 있었다.
Porous matrices of bioactive polymers such as polyglycolic acid (PGA) or polylactic acid (PLA) can be used as scaffolds in bone tissue growth during bone repair process. These polymers are highly porous and serve as a template for the growth and organization of new bone tissues. We evaluated the effect of PGA and PLA polymers on osteoblastic MC3T3-E1 cell extracellular mineralization. MC3T3-E1 cells were cultured in a time-dependent manner -1, 15, 25d as appropriate - for the period of bone formation stages in one of the five culture circumstances, such as normal osteogenic differentiation medium, PGA-plated, fetal bovine serum (FBS)-plated, PGA/FBS-coplated, and PLA-plated For the evaluation of bone formation, minerals (Ca, Mg, Mn) and alkaline phosphatase activity, a marker for osteoblast differentiation, were measured Alizarin Red staining was used for the measurement of extracellular matrix Ca deposit During the culture period, PGA-plated one was reabsorbed into the medium more easily and faster than the PLA-plated one. At day 15, at the middle stage of bone formation, cellular Ca and Mg levels showed higher tendency in PGA- or PLA-plated treatments compared to non-plated control and at day 25, at the early late stage of bone formation, all three cellular Ca, Mg or Mn levels showed higher tendency as in order of PGA-related treatments and PLA-plated treatments, compared to control even without significance. Medium Ca, Mg or Mn levels didn't show any consistent tendency. Cellular ALP activity was higher in the PGA- or PLA-plated treatments compare to normal osteogenic medium treatment PGA-plated and PGA/FBS-plated treatments showed better Ca deposits than other treatments by measurement of Alizarin Red staining, although PLA-plated treatment also showed reasonable Ca deposit. The results of the present study suggest that biodegradable material, PGA and also with less extent for PLA, can be used as a biomaterial for better extracellular matrix mineralization in osteoblastic MC3T3-E1 cells.
실험실에서 배양한 어린 포자체 (최대길이 0.63 cm)를 밀도 4단계로 조절하여 경남 일광연안에 양성하고, 1996년 1월부터 7월까지 생장과 생존을 조사하였다. 서로 다른 밀도로 양성한 포자체는 저밀도에서 높은 생존율을 보였다. 실험된 4개의 밀도집단 중 3개의 저밀도 집단에서 최종적으로 생존한 개체의 수는 34$\~$38 개체${\cdot}100 cm^{-2}$의 범위로 유사하였다. 그러나 가장 고밀도 집단에서는 사망률이 다른 집단에 비해 상대적으로 높아 7월 말에는 단지 21 개체${\cdot}100 cm^{-2}$ 개체만이 생존하였다. 양성 후 2개월 이전에는 각 밀도집단의 엽체의 크기는 초기 밀도가 높을 수록 작았으나, 집단간 밀도차이가 없어지는 3개월 이후로는 크기 차이가 없어졌다. 집단의 밀도가 높아질수록 엽장의 빈도분포가 오른쪽으로 심한 비대칭을 이루었다. 각 집단에서 가장 큰 10개의 크기는 언제나 저밀도 집단에서 높았다. 엽체의 크기가 클수록 상대성장률과 생존율이 높았다. 3월부터 7월까지 yield-den-sity 관계식의 기울기는 -0.060에서 -0.137의 범위였다. 이 결과들로부터 밀도가 다시마의 성장과 생존에 심각하게 영향을 미침을 알 수 있었으나, 밀도 종속적 성장과 사망유형은 육상식물의 경우와는 일치하지 않았다.
Objective: Ovarian follicular development, which dependent on the proliferation and differentiation of granulosa cells (GCs), is a complex biological process in which miRNA plays an important role. Our previous study showed that miR-458b-5p is associated with ovarian follicular development in chicken. The detailed function and molecular mechanism of miR-458b-5p in GCs is unclear. Methods: The luciferase reporter assay was used to verify the targeting relationship between miR-458b-5p and catenin beta-1 (CTNNB1), which is an important transcriptional regulatory factor of the Wnt/β-catenin pathway. The cell counting kit-8 (CCK-8) assay, flow cytometry with propidium iodide (PI) and annexin V-fluorescein isothiocyanate (FITC) labeling were applied to explore the effect of miR-458b-5p on proliferation, cell cycle and apoptosis of chicken GCs. Quantitative real-time polymerase chain reaction and Western blot were used to detect the mRNA and protein expression levels. Results: We demonstrated that the expression of miR-458b-5p and CTNNB1 showed the opposite relationship in GCs and theca cells of hierarchical follicles. The luciferase reporter assay confirmed that CTNNB1 is the direct target of miR-458b-5p. Using CCK-8 assay and flow cytometry with PI and Annexin V-FITC labeling, we observed that transfection with the miR-458b-5p mimics significantly reduced proliferation and has no effects on apoptosis of chicken GCs. In addition, miR-458b-5p decreased the mRNA and protein expression of CD44 molecule and matrix metallopeptidase 7, which are the downstream effectors of CTNNB1 in Wnt/β-Catenin pathway and play functional roles in cell proliferation. Conclusion: Taken together, the data indicate that miR-458b-5p regulates ovarian GCs proliferation through Wnt/β-catenin signaling pathway by targeting CTNNB1, suggesting that miR-458b-5p and its target gene CTNNB1 may potentially play a role in chicken ovarian follicular development.
Background: Gintonin is a ginseng-derived exogenous ligand of the G protein-coupled lysophosphatidic acid (LPA) receptor. We previously reported that gintonin stimulates gliotransmitter release in primary cortical astrocytes. Astrocytes play key roles in the functions of neurovascular systems. Although vascular endothelial growth factor (VEGF) is known to influence the normal growth and maintenance of cranial blood vessels and the nervous system, there is little information about the effect of gintonin on VEGF regulation in primary astrocytes, under normal and hypoxic conditions. Methods: Using primary cortical astrocytes of mice, the effects of gintonin on the release, expression, and distribution of VEGF were examined. We further investigated whether the gintonin-mediated VEGF release protects astrocytes from hypoxia. Results: Gintonin administration stimulated the release and expression of VEGF from astrocytes in a concentration- and time-dependent manner. The gintonin-mediated increase in the release of VEGF was inhibited by the LPA1/3 receptor antagonist, Ki16425; phospholipase C inhibitor, U73122; inositol 1,4,5- triphosphate receptor antagonist, 2-APB; and intracellular $Ca^{2+}$ chelator, BAPTA. Hypoxia further stimulated astrocytic VEGF release. Gintonin treatment stimulated additional VEGF release and restored cell viability that had decreased due to hypoxia, via the VEGF receptor pathway. Altogether, the regulation of VEGF release and expression and astrocytic protection mediated by gintonin under hypoxia are achieved via the LPA receptor-VEGF signaling pathways. Conclusion: The present study shows that the gintonin-mediated regulation of VEGF in cortical astrocytes might be neuroprotective against hypoxic insults and could explain the molecular basis of the beneficial effects of ginseng on the central nervous system.
Objectives : This study investigated cellular-protective effects of Nardotidis seu Sulculii Concha water extract (NSCE) against oxidative stress induced by arachidonic acid (AA)+iron or tert-butylhydroperoxide (tBHP). Methods : In vitro, MTT assay was assessed for cell viability, and immunoblotting analysis was performed to detect expression of AMP-activated kinase (AMPK) signaling pathway and autophagy related proteins. In vivo, mice were orally administrated with the aqueous extract of NSCE of 500 mg/kg for 3 days, and then injected with CCl4 0.5 mg/kg body weight to induce acute damage. The level of liver damage was measured by serum aspartate aminotransferase (AST), alanine aminotransferase (ALT) and lactate dehydrogenase (LDH) analysis. Results : Treatment with NSCE inhibited cell death induced by AA+iron and tBHP. NSCE induced the phosphorylation of AMPK, and this compound also induced the phosphorylation of LKB1, an upstream kinase of AMPK, and Acetyl-CoA carboxylase (ACC), a primary downstream target of AMPK. NSCE increased the protein levels of autophagic markers (LC3II and beclin-1) and decreased the phosphorylation of mammalian target of rapamycin (mTOR) and simultaneously increased the phosphorylation of unc-51-like kinase-1 (ULK-1) in time-dependent manner. Conclusions : NSCE has the ability 1) to protect cells against oxidative stress induced by AA+iron or tBHP. NSCE 2) to activate AMP-activated protein kinase (AMPK), and 3) to regulate autophagy, an important regulator in cell survival.
