  • A study on the acute toxicity in the Nile tilapia, Oreochromis niloticus fry showed that the 24hr-$LC_{50}$ was 18.6% for NaCl,152 ppm for formaline and 2.1 ppm for $KMnO_4$. The fry responded to narrow range of concentration of all the tested chemicals and their toxic effects were dose-dependent.

Distinct Repressive Properties of the Mammalian and Fish Orphan Nuclear Receptors SHP and DAX-1

  • It has been suggested that the structure and function of nuclear receptors are evolutionally conserved. Here, we compare the molecular functions of the nile tilapia (Oreochromis niloticus) small heterodimer partner (nSHP/NR0B2) and the Dosage-sensitive sex reversal AHC critical region on X chromosome gene 1 (nDAX-1/NR0B1) with those of human SHP and DAX-1 (hSHP and hDAX-1, respectively). We found that, upon transient cotransfection of human cells, nDAX-1 repressed the activity of tilapia SF-1 (nSF-1) but not that of human SF-1, although the physical interaction with human SF-1 was retained. Similarly, nSHP repressed the activity of nSF-1, whereas hSHP did not, pointing to divergent evolution of SHP/SF-1 in fish and human. We thus propose that the repressive functions of SHP and DAX-1 have been conserved in fish and mammals although with different transcriptional targets and mechanisms. These differences provide new insights into the physiological diversification of atypical orphan nuclear receptors during vertebrate evolution.

  • It has been very difficult to develop and evaluate efficient fish vaccines because fish immune cells have not been properly characterized. In this study, we investigated the cell-mediated immunological properties of B- and T-like cells in Nile tilapia (Oreochromis nilotica). Surface immunoglobulin negative ($slg^{-}$) cell population proliferated in response to mammalian T-cell mitogens PHA and Con A, while surface immunoglobulin positive ($slg^{+}$) cells responded to the B-cell mitogen LPS. The slg$^{[-10]}$ cells from hemocyanin (HC)-immunized Tilapia, compared to the non-immunized control, reacted more to PHA than to Con A. Unexpectedly, antigen (Ag)-specific response was observed in both $slg^{-}$ and $slg^{-}$cells. Regardless of HC immunization, whole leukocytes from 8 head kidney of fish showed natural killer (NK)cell activity. Especially, NK cell activity was much higher in slg$^{[-10]}$ cells than in slg$^{+}$cells, indicating the possibility that fish NK cells were not at least associated with slg$^{+}$ cell population and not activated by Ag. Further understanding of functional fish immune cells will help to evaluate and develop effective vaccines for fishes and to monitor the course of therapy In infected fishes.hes.

  • Changes of gill tissue and body composition of grey mullet (Mugil cephalus) and Nile tilapia (Oreochromis niloticus) by the manipulation of salinity were observed in a recirculating rearing system. Salinity was increased from $0\%_{\circ}$ to $33\%_{\circ}$for 1 day and remained far 15days, thereafter salinity was decreased from $33\%_{\circ}$ to $0\%_{\circ{$ for 1 day and remained for other 15 days. Any morphological differences of gill lamella in grey mullet were not observed in seawater and freshwater. However, on day 2 in exposure to seawater, Nile tilapia showed the edema and bloodclot in gill lamella. In the case of grey mullet, mitochondria and tubular system of chloride cell were more densely packed according to the salinity increase. The whole body of grey mullet showed no significant differences in moisture content during experimental period. However, moisture content in whole body of the Nile tilapia was signincantly decreased when exposed to seawater. Protein content in whole body of grey mullet showed no significant difference between beginning and day 15 of the experiment.

Accumulation of the Conjugated Linoleic Aacid (CLA) in Tilapia ( Tilapia nilotica) Fed Diets on Various Levels of CLA (CLA (Conjugated linoleic acid) 급이수준에 따른 역돔의 CLA 축적량)

  • Effects of conjugated linoleic acid (CLA), known as an effective anticarcinogen in several animal models, on the tilapia were investigated. The CLA was made from safflower oil by alkaline isomerization method. Isomers in CLA such as cis-9, trans-11 and trans-10, cis-12 occupied over $80\%$, and other isomers was below $20\%$. In experiment, 250 fishes (average weight is 32 g) were divided into 15 fishes per five treatment and triplicate group for 8 weeks: control, $1\%$ CLA, $2.5\%$ CLA, $5.0\%$ CLA, and $10\%$ CLA diets. Daily growth rate and feed coefficiency were measured every week. The most effective diet for the growth rate and feed coefficiency of tilapia was $1.0\%$ CLA diet group. Every two weeks, sampled and determined the contents of CLA in the muscle and liver, After 8 weeks, $1.0\%$ and $10.0\%$ of CLA fed group accumulated the CLA as 41.3 and 180.9 mg/g of fat in their muscle respectively, Also, n-9 and n-3 fatty acid (FA) compositions were almost not changed in the muscle and liver. But n-6 fatty acid was changed according to the contents of fed CLA. The $1.0\%$ CLA fed group was shown the highest contents of n-6 FA and the $10.0\%$ CLA group was shown the lowest contents of n-6 FA.

Combined effects of temperature and arsenic on hematological parameters of tilapia Oreochromis niloticus (틸라피아의 혈액성상에 미치는 온도와 비소의 복합적 영향)

  • Experiments were carried out to determine the hematological changes in freshwater tilapia Oreochromis niloticus, after water temperature (20, 25 and $30^{\circ}C$) and arsenic concentrations (200, and 400 ppb) exposure 20 days. The RBC count and hematocrit (Ht) were significantly increased in As 400 ppb and water temperature $30^{\circ}C$. Hemoglobin (Hb) was significantly increased under combined As (200 and 400 ppb) and water temperature $30^{\circ}C$. The tilapia exposed to water temperature $30^{\circ}C$ and As concentration 400 ppb increase significantly in calcium concentration. The high concentrations of total protein and glucose concentrations were observed at water temperature $30^{\circ}C$ and As concentration 400 ppb. A significant increment GOT and GPT activities in the tilapia serum noticed at water temperature (25 and $30^{\circ}C$) and As concentration (200, 400 ppb). These results indicate that tilapia can be affected by high water temperature ($30^{\circ}C$) and As concentration (400 ppb) in terms of inorganic matter, organic matter and enzyme activity in serum.

Characterization of Edwardsiella tarda specific phage isolated from fish farms on west coast of Korea (서해안 양식장에서 분리한 Edwardsiella tarda의 특이 bacteriophage 특성)

  • Edwardsiella tarda is a broadly infectious agent against freshwater and seawater fishes. In the present study, E. tarda specific phage was isolated from fish farms on the west coast of Korea, and the effect of environmental factors such as pH and water temperature on the phage activity was investigated. As a fish model, Nile tilapia (Oreochromis niloticus) was used and the interaction between E. tarda and phage was investigated. The phage activity, in pH test, appeared even higher in seawater than freshwater and was evenly constant up to $50^{\circ}C$. The phage and E. tarda were inoculated in tilapia and the phage activity and E. tarda viability were checked on the time intervals. As a result, the number of E. tarda in the group of E. tarda plus phage was constantly reduced to 24 hr post-inoculation compared to the control group without phage, whereas the phage activity was slightly increased in the experiment group. The results suggest that it might be possible to use phage to control the fish disease caused by E. tarda.

Oxygen Consumption, Operculum Movement Number and Ammonia Excretion of Cultured Tilapia (Oreochromis mossambicus) by Salinity Changes (염분 변화에 따른 자바틸라피아(Oreochromis mossambicus)의 산소소비, 아가미 호흡수 및 암모니아 배설)

  • The effects of salinity on oxygen consumption, operculum movement number and ammonia nitrogen excretion were examined in cultured tilapia, Oreochromis mossambicus (total length 10$\pm$1.0 cm, total weight 17.3$\pm$4.2 g). The fish were exposured to certain salinities (0, 3, 9, 15, 21, 27 and 32 psu) step by step with 3 psu$\cdot$d$^{-1}$ and changed rapidly from certain salinity to another salinity. The oxygen consumption of the fish which was transferred step by step showed increasing tendency in the range of 3 $\sim$ 27 psu, but markedly appeared very low value at 32 psu. The tolerance limit in tilapia by salinity change without acclimation was shown 23.3 psu from 96-h TL$_{50}$. The oxygen consumption, operculum movement and ammonia nitrogen excretion of fish which was transferred rapidly from certain salinity to each salinity (0, 9, 15, 21 and 32 psu) showed a changing point at 15 psu; they showed increasing and/or decreasing pattern before 15 psu, and showed decreasing pattern after 15 psu. From these results, it was concluded that the appropriate salinity without physiological change for Java tilapia was below 15 psu.

Characteristics and Antigenicity of Edwardsiella tarda Isolated from Tilapia (Oreochramis niloticus) (틸리피아에서 분리(分離)한 Edwardsie tarda의 성상(性狀)과 항원성(抗原性)에 대하여)

  • In March 1987, a bacterial disease occured among cultured tilapia (Oreochromis niloticus), in the aquarium of Fish Pathology Laboratory of National Fisheries University of Pusan. The diseased fish showed abdominal inflamation of ascites. The causative organisms isolated from the kidney were Gram negative rods. On SS agar colonies developed within 48 hours at $25^{\circ}C$. On the basis of the morphological and biochemical characteristics the organisms were identified as Edwardsiella tarda. They were sensitive to chloramphenicol, ampicilline and oxytetracycline. The pathogenicity was proved positive by intraperitoneal injection. Three kinds of antigen were prepared with E. tarda for the immune response in tilapia : i.e., by inactivating the strain with 0.4% formalin for 24 hrs at $25^{\circ}C$ ultrasonicating it 3 times with 5 watts for 30 seconds each time and filtrating it through millipore filter. The formalin killed antigen showed good efficacy at the injection concentration of over $10^7$ cells per fish of 50g. All the immunized tilapia showed only a little agglutination titer to the three antigens. The highest survival rate was recorded in challenged tilapia immunized with formalin killed cells.

Changes of Hepatic Microsomal Cytochrome P450 Monooxygenase System in Nile tilapia, Oreochromis niloticus Exposed to PCBs (PCBs노출에 의한 틸라피아, Oreochromis niloticus의 간 cytochrome P450 효소계의 변화)

  • The study was conducted to investigate the changes of hepatic cytochrome P450 monooxygenase system induced by dietary administration of polychlorinated biphenyls (PCBs) in the tilapia, Oreochromis niloticus. Tilapia were fed pellet with PCBs (Aroclor 1254) 0.05, 0.25 and 0.50 mg/kg body weight/day for 30 days. The dietary administration of PCBs (0.05 mg/kg) induced a significantly increased the concentration of cytochrome P450 and the activity of 7-ethoxyresorufin-O-deethylase (EROD) in the hepatopancreas at 30 days, while the augmentation of both responses was found at 20 days with a higher administration of PCBs-diet (0.25 mg/kg). However, hepatic 7-penthoxyresorufin-O-dealkylase (PROD) activity did not show any noticeable changes with the PCBs-diet 0.05-0.5 mg/kg range compared to control group during the experimental periods of 30 days in the tilapia. These results indicate that tilapia fed PCBs at the concentration of more than 0.05 mg/kg for 30 days are affected by PCBs in terms of cytochrome P450 concentration and EROD activity in the hepatopancreas.
