Phylogenetic and Morphological Comparison between Thamnaconus septentrionalis and T. modestus Collected in Southwest Seashore (서남해에서 채집된 말쥐치 (Thamnaconus modestus)와 유사종 (T. septentrionalis)의 형태 및 계통유전학적 비교)

  • Thamnaconus modestus, distributed in the Northwest Pacific, has high economic value and is used in various seafood. In this study, the morphological and genetic characteristics of T. modestus and T. septentrionalis were compared and analyzed. We observed the external and internal morphology of T. modestus, sketched skeletal elements, and analyzed phylogenetic evolutionary relationships using the cytochrome c oxidase subunit I (COI) gene on mitochondrial DNA compared to T. septentrionalis. The T. modestus observed in this study had blackish-brown patterns irregularly scattered on the gray-brown body, and the fins were blue-green. Genetic analysis results based on the COI sequences of T. modestus showed seven types of base sequence variation; however, the homology was more than 98.8%. In addition, as a result of comparison of the COI nucleotide sequences and phylogenetic analysis in Tetraodontiformes, two T. septentrionalis sequences (JN813099, MW485059) were similar to T. modestus with 99% homology, and the other two T. septentrionalis sequences (EF607583, KP267619) were similar to those of species belonging to another genus Thamnaconus with 95% homology with T. modestus. It was not easy to classify the species based on morphological characteristics, and phylogenetic analysis between T. modestus and T. septentrionalis confirmed the difference in classification. These results provide the external and internal morphology of T. modestus and will be used as important information for the taxonomic study of T. modestus and T. septentrionalis.

  • The feeding habits of black scraper, Thamnaconus modestus, were studied using 172 specimens collected in the coastal waters of Geomun-do, Korea. The size of the specimens ranged from 16.2 to 39.1 cm in standard length (SL). T. modestus was a omnivore that consumes mainly benthos. Of the benthos species nephtheidae was the most preferred prey. Its diet also includes, algae, thoracica, gastropoda, desmospongia. Small and medium size individuals (<21~33 cm, SL) fed mainly on nephtheidae and algae. Larger individuals (>33 cm, SL) fed mainly on thoracica. The proportion of nephtheidae decreased as body size increased (>33 cm, SL), whereas the consumption of thoracica gradually increased.

  • The maturation and spawning of black scraper, Thamnaconus modestus were studied using samples collected monthly from March, 2015 to February, 2016 in the coastal waters off Middle East Sea, Korea. The gonadosomatic index (GSI) of female was the highest in June. The spawning periods lasted from March to September based on histological observation of female gonad development. The percentage of sexually mature females estimated from a logistic function was over 50% for the size group 18.02 cm (total length, TL). The size of eggs spawned was between 0.40 to 0.58 mm. Fecundity varied between 185,648 and 9,747,250 eggs. The relationship between the fecundity and TL of the fish was expressed in the fecundity equation as $F=0.0297TL^{5.4835}$.

  • Filefish Thamnaconus modestus has seen a sharp decline in their catch, we must strive for continuous resource management. We investigated the maturation and spawning of T. modestus by trawl net and set net from January 2018 to November 2019, off Jeju Is. and in coastal waters of North Gyeongsang Province's Korea. We analyzed monthly change total length (cm), gonadosomatic index (GSI) and maturity stages, egg diameter (mm), the relations fecundity and total length. GSI for Jeju Is. was the highest in April (3.17, 2.43), and decreased from July to 1.73 in August and GyeongBuk's GSI was the highest in Apri l (1.86, 2.58), followed by the high in May and the sharp decline in September, which is 0.23. Using a histological method, the annual reproductive cycle of T. modestus can be divided into 5stage in females; the early growing stage (Jan.-Mar.), late growing stage (Apr.-May), Mature stage, spent stage (Oct.), Recover and resting stage (Nov.-Jan.).We estimate the TL at 50% maturity as 24.1cm for female, fecundity ranged from 560,044 eggs at 23.4cm total length to 1,580,387 eggs at 36.6.cm TL.

  • Annual reproductive cycle of the file fish, Thamnaconus modestus (Gunther), was histologically investigated. Samples were collected monthly in the coastal waters of Chungmun, south of Cheju Island, Korea from July 1997 to June 1999. In males and females of T. modestus GSI values reached the maximum in June and May, respectively. Reproductive cycle could be divided into the following successive stages: in females, growing stage (March to April), mature stage (April to May), spawning stage (May to June), degenerative and resting stage (July to February), and in males, growing stage (January to March), mature stage (April to May), spent stage (May to June), degenerative and resting stage (July to December), respectively. To clarify the spawning cycle of female in T. modestus, some were examined, that is, the weekly changes of GSI, detail developmental stages in the ovary and the weekly changes of sex steroid hormones ($E_2$ and T) levels in plasma during the spawning period. Throughout histological observation of the ovary during the spawning period, T. modestus belonged to an asynchronous and multiple spawner. Changes of plasma $E_2$ and T levels were similar to the changes of GSI and ovary maturity.

Diluents and Cryoprotectants for Cryopreservation of Filefish Thamnaconus modestus Sperm

  • The present study aimed to find the best diluent and cryoprotectant for sperm cryopreservation of filefish Thamnaconus modestus. Two kinds of artificial seminal plasma(ASP1, ASP2), 0.3 M glucose and marine fish Ringer's solution(MFRS) were employed as diluent. Dimethyl sulfoxide(DMSO) and methanol as cryoprotectant were selected for sperm cryopreservation. Sperm was diluted at the ratio of 1:3 with diluents containing cryoprotectants and adjusted for final concentration at 5%, 10%, 15% and 20%. Mixed milt was frozen at liquid nitrogen vapor after equilibration for 5 min. The highest motility($40.5{\pm}2.8%$) and swimming speed($81.5{\pm}4.1{\mu}m/s$) of frozen/thawed sperm were observed in ASP1 diluent containing 10% DMSO and in ASP2 containing 15% DMSO, respectively. Results showed that cryopreservation with ASP as diluent and DMSO as cryoprotectant could be adopted for long term storage of filefish sperm.

  • The milt properties of filefish(Thamnaconus modestus) included physical properties of sperm and biochemical properties of seminal plasma. The physical properties of milt were $0.3{\pm}0.1\;mL{\cdot}fish^{-1}$ in sperm volume, $2.6{\pm}0.1{\times}10^7\;spermatozoa{\cdotg}mL^{-1}$ in sperm concentration and $73.3{\pm}6.7$ in spermatocrit. The biochemical properties of seminal plasma contained $9.8{\pm}0.9\;mmol{\cdot}L^{-1}$ potassium, $164.0{\pm}4.0\;mmol{\cdot}L^{-1}$ sodium, $151.0{\pm}1.2\;mmol{\cdot}L^{-1}$ chloride, $14.9{\pm}0.6\;mg{\cdot}dL^{-1}$ calcium, $7.2{\pm}0.1\;mg{\cdot}dL^{-1}$ magnesium, $1.0\;mg{\cdot}dL^{-1}$ glucose, $0.1\;g{\cdot}dL^{-1}$ total protein and $1.0\;mg{\cdot}dL^{-1}$ total lipid. The osmolality and pH of seminal plasma were $322.8{\pm}2.8\;mOsmol{\cdot}kg^{-1}$ and $7.7{\pm}0.1$, respectively. The spermatozoon of filefish consisted of three parts: head without acrosome, mid-piece with five mitochondria and flagellum with "9+2" pattern. The head of spermatozoon in longitudinal section was horseshoe-shaped, and $1.3{\sim}1.6\;{\mu}m$ long and $1.0{\sim}1.3\;{\mu}m$ wide.

Developmental Changes in Digestive Organ and Digestive Enzyme Activity of Filefish Thamnaconus modestus (말쥐치 Thamnaconus modestus 자치어의 성장에 따른 소화기관 및 소화효소 발달)

  • Larvae and juveniles of the filefish Thamnaconus modestus were reared for 64 days after hatching (DAH) in order to determine the activity of four enzymes (trypsin, pepsin-like enzyme, lipase, amylase) during ontogeny. Larvae were fed on rotifer Brachionus plicatilis from 2 to 26 DAH, Artemia nauplii from 10 to 64 DAH, and then gradually changed to pelleted feed from 40 DAH. Temperature was kept between $21.5{\sim}24.2^{\circ}C$ Activity of trypsin and lipase was found in larvae 4 DAH ($6.0{\pm}1.4unit$) and 6 DAH ($4.5{\pm}1.4unit$), respectively. The evolution of activity in both enzymes showed a profile marked by drastic increases between late larval and early juvenile stages. Pepsin-like enzyme activity was found at 10 DAH and drastically increased from 28 DAH, corresponding with the early juvenile stage of T. modestus. Interestingly, developmental changes in the pepsin-like enzyme activity coincided well with increases in the number of gastric glands. Amylase activity was found at 10 DAH and was maintained at a low level up to 28 DAH, followed by a drastic increase from 28 DAH to 40 DAH. It might be concluded that a drastic increase in trypsin and pepsin-like enzyme activities, and a corresponding increase in the number of gastric glands reflects a higher somatic growth of T. modestus during the early juvenile period.

  • This study is the first to report the biometric information between the length and weight relationships (LWR) and seasonal body condition factors (K) of Sebastes zonatus Chen & Barsukov, 1976 and Thamnaconus modestus (Günther, 1877) inhabiting the waters off Ulleung-do and Dokdo. The LWRs in spring and summer, and all seasons combined were highly correlated (r2>0.959), and the regression slopes of LWRs were significantly different between the spring and summer in both species. The body conditions of the two fish were significantly higher during the spring than during the summer, reflecting their fatness in relation to spawning. The results from this study contribute to the understanding of the biology of S. zonatus and T. modestus and provide useful data for the development of conservation and management plans for these species.

Egg Development and Morphological changes of Larvae of the File Fish, Thamnaconus modestus (말쥐치, Thamnaconus modestus의 난발생과정과 부화자어의 형태발달)

  • The egg development and morphological changes of larvae of the file fish, Thamnaconus modestus(Gunther), was investigated by artificial insemination. The fertilized egg was spherical in shape, colorless and adhesive with 0.63~0.67mm in diameter and yolk had 20~30 oil globules of various size (0.04~0.16 mm in diameter). The hatching occured at 53 hours after fertilization in $21.0\sim23.0^{\circ}C$ of water temperature. Hatched larvae measured 2.07~2.20 mm in total length with 5~6+17~18=22~24 myotomes. The 3~4 days after hatching larvae grew to the total length 2.62~2.77 mm, opened mouth and anus. The 5 days after hatching, total length of the larvae was 2.74~3.04 mm, the first dorsal fin was formed at the behind of head. After hatching 12~13 days larvae attained 3.78~3.99 mm in total length. This period formed rudimental dorsal fin and anal fin. Twenty days after hatching, larvae grew to 6.04~6.17mm in total length, at which time the dorsal, anal and caudal fin rays reached 19~20, 18~19 and 9~10, respectively.
