Anti-metastatic and Immunomodulating Activity of Water-Soluble Components from Five Mushroom Extracts

  • Objectives : This experiment was conducted to evaluate the inhibitory effects against lung metastasis and promotion of splenocytes by water-soluble components from five mushrooms extracts (WEFM): Garnoderma frondosa, Corious versicolor, Codyceps militaris, Hericium erinaceus and Lentinula edodes. Methods : Colon 26-L5 carcinoma cells were injected through the tail vein to induce lung metastatic cancer. Changes in weight of lung were observed and cytokine level was analyzed to evaluate immunological changes. Results : Oral administration of WEFM resulted in a significant inhibition of lung metastasis after intravenous injection of colon 26-L5 cells in a dose-dependent manner. There was also a significant increase in T cell and B cell mitogenic stimuli and production of IFN-g by splenocytes stimulated with Con A compared to untreated controls. Conclusion : WEFM may have anti-tumor activities via Th1-type dominant immune responses.

Effect of Black Garlic Extract on Cytokine Generation of Mouse Spleen Cells (흑마늘(Black garlic) 추출물이 마우스 비장세포의 Cytokine 생성에 미치는 영향)

  • The effect of black garlic extract on the activation of spleen cells from a C57BL6 mouse was investigated to examine immune activities of of fermented black garlic containing a variety of bioactive substances. xtract obtained from the concentration of commercial Namhae black garlic was used for the analysis of immune activities. Treatment with the extract increased the expression of interleukin-2 (IL-2) cytokine. The simultaneous administration of the extract plus lipopolysaccharide (LPS) increased the expression of IL-2, tumor necrosis factor (TNF)-${\alpha}$, and interferon (IFN)-${\gamma}$ compared with that of a control group. This result suggests that cellular immunity can be induced by macrophages, resulting in the expression of T lymphocytes and T helper type 1 (Th1) cells. In addition, treatment with the extract increased the late response of IL-6 cytokines, and the extract plus LPS augmented the expression of IL-4 and IL-6 compared with that of an LPS-treated group. Meanwhile, the extract plus LPS decreased the late response of IL-10, suggesting that humoral immunity can be activated by stimulating B lymphocytes, suppressing cellular immunity, and effectively modulating the conversion into humoral immune responses. These findings demonstrate that the black garlic extract activates Th1 and Th2 cells by stimulating T lymphocytes in mouse spleen cells and leads to immunomodulation by activating cellular and humoral immune responses of the immune system.

Effects of Aqueous Extract from Lonicera Japonica Flower on Trimellitic Anhydride-induced Contact Hypersensitivity in BALB/c Mice (수용성 금은화 추출물이 Trimellitic Anhydride 유도 마우스 접촉성 과민반응에 미치는 영향)

  • Objectives : Trimellitic anhydride (TMA), a sensitizer that induces occupational asthma and atopic dermatitis, is widely used industrially to make epoxy and alkyd resins, plasticizers, high temperature polymer, and surfactants. The aim of this study was to investigative the effects of aqueous extracts of Lonicera japonica Flower(LJFAE) on TMA-induced contact hypersensitivity (CHS) in Balb/c mice. Methods : The dried flowers of L. japonica were extracted with distilled water at $100^{\circ}C$ for 7 h. The extract was freeze-dried following filteration through 0.45 ${\mu}m$ filter. Mice were orally administrated with or without LJFE of a different doses(25-100 mg/kg) for 28 days. In the challenge period, mice were externally applied at difference doses of LJFAE one time per day 30 min before TMA treatment. We examined the effects of LJFAE on the the serum levels of IgE and prostagladin E2 (PGE2), the Thl/Th2 cytokine production of spleen cells, ear swelling responses, and the leukocyte infiltration induced by TMA. Results : The orally and externally administration of LJFAE dose-dependently reduced the serum levels of IgE and PGE2 production as well as ear swelling responses and leukocyte infiltration in TMA-induced Balb/c mice. Furthermore, the levels of Thl (TNF-${\alpha}$, IFN-${\gammer}$, IL-2)/Th2 (IL-4, IL-5, IL-13) cytokine production from spleen cells stimulated with anti-CD3 and CD28 mAbs was markedly suppressed by the orally and externally treatment with LJFAE in a concentration dependent manner. Conclusions : These results suggest that LJFAE suppresses the inflammatory mediators and regulates the Thl/Th2 cytokines. Therefore, these properties may contribute to the strong anti-CHS response effect of LJFAE.

Cellular and Humoral Immune Responses to Sequential Periodontopathic Bacterial Immunization in Animal Model (상이한 치주병원균의 연속적 인공면역에 대한 세포성 및 체액성 면역반응에 대한 동물실험적 연구)

  • Antigen-specific T cell clones were obtained from mice immunized with Fusobacterium nucleatum ATCC 10953(F .nucleatum) and/or Porphyromonas gingi valis 381(P. gingivalis). 10 Balb/c mice per group were immunized with F. nucleatum followed by P. gingivalis, or with P. gingivalis alone by intraperitoneal injection of viable microorganisms. Spleen T cells were isolated and stimulated in vitro with viable P. gingivalis cells to establish P. gingivalisspecific T cell clones. T cell phenotypes and cytokine profiles were determined along with T cell responsiveness to F .nucleatum or P. gingivalis. Serum IgG antibody titers to F. nucleatum or P. gingivalis were also determined by ELISA. All the T cell clones derived from mice immunized with F. nucleatum followed by P. gingivalis demonstrated Th2 subsets, while those from mice immunized with P. gingivalis alone demonstrated Th1 subsets based on the flow cytometric analysis and cytokine profiles, All T cells clones from both groups were cross-reactive to both P. gingivalis and F. nucleatum antigens. Phenotypes of T cell clones were all positive for CD4. Mean post-immune serum IgG antibody levels to F. nucleatum or P . gingivalis were significantly higher than the pre-immune levels(p <0.01, respectively). There were no significant differences in the antibody titers between the two groups. It was concluded that P. gingivalis-specific T cells initially primed by cross-reactive F. nucleatum antigens were polarized to Th2 subsets, while T cells stimulated with P. gingivalis alone maintained the profile of Th1 subset.

Effects of Oral Administration of Phellinus linteus on the Productions of the Th1- and Th2-type Cytokines in Mice

  • Background: The mushroom Phellinus linteus (PL) has been shown to have the anti-tumor and immunostimulatory effects. We hypothesized that the hot water extract of PL (WEPL) exerts its significant immunostimulatory effect by inducing production of the Th1-derived cytokine interferon-${\gamma}$ (IFN-${\gamma}$) by T lymphocytes. Methods: T lymphocytes were isolated from the mice fed with 200 mg/kg of WEPL once a day for 4 weeks, and then stimulated with the mitogen concanavaline A (Con A). IFN-${\gamma}$ gene and intracellular protein expressions were analyzed by RT-PCR and flow cytometry, respectively. The production of IFN-${\gamma}$ was measured by enzyme-linked immunosorbent assay. Results: WEPL significantly enhanced the transcription of IFN-${\gamma}$ mRNA. The effect of WEPL on IFN-${\gamma}$ expression was further supported by a concomitant increase in the number of cells with intracellular IFN-${\gamma}$ protein as well as the secretion of IFN-${\gamma}$. However, WEPL did not modulate either gene expression or protein secretion of interleukin-4, a Th2-associated cytokine, by Con A-stimulated T lymphocytes. Conclusion: Our results demonstrate that one of the potentially beneficial anti-tumor and immunostimulatory effects of WEPL may be mediated through the enhancement of IFN-${\gamma}$ secretion by T lymphocytes.

Evaluation of Th1/Th2-Related Immune Response against Recombinant Proteins of Brucella abortus Infection in Mice

  • Brucellosis is a zoonotic disease caused by Brucella, a genus of gram-negative bacteria. Cytokines have key roles in the activation of innate and acquired immunities. Despite several research attempts to reveal the immune responses, the mechanism of Brucella infection remains unclear. Therefore, immune responses were analyzed in mice immunized with nine recombinant proteins. Cytokine production profiles were analyzed in the RAW 264.7 cells and naive splenocytes after stimulation with three recombinant proteins, metal-dependent hydrolase (r0628), bacterioferritin (rBfr), and thiamine transporter substrate-binding protein (rTbpA). Immune responses were analyzed by ELISA and ELISpot assay after immunization with proteins in mice. The production levels of NO, TNF-α, and IL-6 were time-dependently increased after having been stimulated with proteins in the RAW 264.7 cells. In naive splenocytes, the production of IFN-γ and IL-2 was increased after stimulation with the proteins. It was concluded that two recombinant proteins, r0628 and rTbpA, showed strong immunogenicity that was induced with Th1-related cytokines IFN-γ, IL-2, and TNF-α more than Th2-related cytokines IL-6, IL-4, and IL-5 in vitro. Conversely, a humoral immune response was activated by increasing the number of antigen-secreting cells specifically. Furthermore, these could be candidate diagnosis antigens for better understanding of brucellosis.

Effects of Electrolyzed Alkaline Reduced Water on Echinostoma hortense Infection and Immune Response in C57BL/6 Mice (C57BL/6 생쥐에서 전해알칼리환원수가 호르텐스극구흡충 감염과 면역에 미치는 영향)

  • To examine the effect of the electrolyzed alkaline reduced water (ERW) on animal immunity, by employing Echinostoma hortense that is a parasite in the small intestine, the immune response of C57BL/6 was examined. To C57BL/6 mice, Echinostoma hortense metacercaria 15 per animal was in oculate dorsally, the worm was collected after 2 weeks, and the change of goblet cells and mast cells in the mucosa of small intestine was examined, and by using a protein chip, the change of cytokines in the serum was compared and observed. As a result, average 8.3 worms were collected from the C57BL/6 mice infected with E. hortense, and in the group fed with the ERW, average 10 worms were collected. In regard to the examination of the change of goblet cells, in the experimental group infected with E. hortense and fed with the ERW, average 4.3 worms per villus were detected, hence, it was found that the expression of goblet cells was low (p<0.001). Regarding the examination of the change of mast cells, similarly, in the group infected with E. hortense and fed with the ERW, average 11 worms per villus were detected, and it appears to be less than control group (p<0.001). Regarding the expression of cytokines in mouse serum, in comparison of the experiment group infected with E. hortense and control group, in the expression of the Th1 cytokines IL-6, IL-$1{\beta}$, IFN-${\gamma}$, TNF-${\alpha}$, and IL-2, and the Th2 cytokines IL-4, IL-5, IL-10, and IL-13, a significant difference was not detected. In our study, it was found that in the infection of E. hortense, the ERW mediates its effect on the number of goblet cells and mast cells in the intestinal mucosa, and simultaneously, the worm expulsion was delayed, and thus the conclusion that the ERW mediated its effect on the intestinal immunity of mice was obtained.

  • The efficacies of anti-atopic preparations were investigated in hairless mice suffering from 2,4-dinitro-chlorobenzene (DNCB)-induced atopic dermatitis-like lesion. Solid lipid nanoparticle (SLN) containing ceramide and astaxanthin was prepared by a melt-homogenization method using Aminsoft-CT 12 (Cocoyl glutamate) as an emulsifier. And then, the SLNs were coated with silk fibroin (SF) by taking advantage of an electrostatic interaction between the surface of SLNs and SF. SLNs were included in lotion (FL) and cream (FC) types of preparations. Anti-atopic efficacies of the preparations were investigated in terms of appearance of skin surface, spleen index, serum IgE level, and serum cytokine level. SLN-containing preparations suppressed IgE production and IL-4 expression, but promoted $IFN-{\gamma}$ expression.

  • Th cells, which orchestrate immune responses to various pathogens, differentiate from naive CD4 T cells into several subsets that stimulate and regulate immune responses against various types of pathogens, as well as a variety of immune-related diseases. Decades of research have revealed that the fate decision processes are controlled by cytokines, cytokine receptor signaling, and master transcription factors that drive the differentiation programs. Since the Th1 and Th2 paradigm was proposed, many subsets have been added to the list. In this review, I will summarize these events, including the fate decision processes, subset functions, transcriptional regulation, metabolic regulation, and plasticity and heterogeneity. I will also introduce current topics of interest.

Immunoregulatory Effect of Ginsenoside Rd against $CD4^+$ Th lymphocyte (인삼배당체 Rd의 $CD4^+$ Th 임파구에 대한 면역조절효과)

  • In this present study, we determined the immunoregulatory activity of ginsenoside Rd extract from Panax ginseng. To determine the activity, we tested Rd against $CD4^+$ Th cells in a murine model of type 1 diabetes, which involves Th1-dominant immunity. The type 1 diabetes was caused by streptozotocin (STZ) and the severity of the diabetes was evaluated by measuring the degree of hyperglycemia, a major symptom of diabetes. The data resulting from experiments showed that ginsenoside Rd induced a greater level of Th1 type cytokines [IFN-${\gamma}$ & IL-2] than Th2 type [IL-4 & IL-10] (P<0.05), which was determined by cytokine profile analysis. In the animal model of diabetes, the depletion of $CD4^+$ Th cells by a treatment of anti-CD4 mAb resulted in considerably lower values of blood-glucose levels than those of the mAb-untreated mice, which indicates that the Th1 immune response from $CD4^+$ Th cells are responsible for diabetes. Based on these observations, the effect of Rd on diabetes was examined in the same animal model. Results showed that Rd-treated mice groups had increased levels of blood glucose compared to Rd-untreated mice groups that were used as a negative control (P<0.05). In other words, Rd aggravated the diabetes via the Th1 immune response. In conclusion, ginsenoside Rd had an immunoregulatory activity of Th1-dominant immunity.
