Dose Dependent Effects of Panax Ginseng on the Reproductive Functions in Mice (농도별(濃度別) 인삼(人蔘) 투약(投藥)이 수컷 생쥐의 생식능력(生殖能力)에 미치는 영향(影響))

  • Purpose : This study was conducted to investigate the dose dependent effects of Panax Ginseng on the reproductive functions in mice. Methods : We used the 8-week-old mice, and administered 0.2ml extract solution of Panax Ginseng in the different concentration once a day for 60 days. The control group was administered 0.2ml normal saline in the same way and duration. We counted the total, motile and normal sperm number of the cauda epididymis and measured the activities of sperm hyaluronidase, peroxidase and catalase of the isolated testis tissues. And we observed histological changes of surgically isolated testis by histochemical methods. Results : All Panax Ginseng extract solution groups showed significantly dose dependent differences in the total number, the motility and normality of sperms compared with the control group, respectively. In the histological analysis of the testicular tissues, all Panax Ginseng extract solution groups showed the enlargement of testicular lobe diameter and apparent angiogenesis between seminiferous tubules. And the activity of typical sperm enzyme, hyaluronidase, was significantly increased in the Panax Ginseng extract solution groups compared to the control group. In the antioxidant activity analysis, the activities of peroxidase and catalase were significantly increased in the Panax Ginseng extract solution groups compared to the control group. Conclusion : This study shows that Panax Giseng has the beneficial effects on the concentration, morphology and motility of sperm, the activities of sperm hyaluronidase, testicular peroxidase and catalase. We can suggest that Panax Ginseng be useful for the treatment of male infertility.

  • The purpose of this study finds out the effect of red ginseng extract(1.0 g/kg) on bis(tributyltin)oxide(TBTO)(10, 20 and 40 mg/kg) which poisons against some organs like thyroid gland, liver, kidney, testis, ovary. Serum immunity and sex hormone activity of rats are examined by gastric tubing for 3 weeks. The weights of each in treated group were increased, especially liver in females and those of testis in males were signignificantly increased at 10, 20 and 40 mg/kg (P<0.05, P<0.01). In group treated with TBTO(10, 20 and 40 mg/kg) for females, the serum IgG level was significantly reduced in comparison with control group(P<0.05). The group of females which were poisonned by TBTO and rGe was significantly recovered in TBTO 10 mg/kg + rGe 1.0 g/kg and TBTO 20 mg/kg + rGe 1.0 g/kg. In case of sex hormone activity of each sex, the estradiol activity of females and testosterone activity of males were signignificantly decreased rather than the control group. And control group by bis(tributyltin) oxide and red ginseng extract is just increased estiradiol activity.

Influence of Ginseng upon Testicular Nucleic Acid Content in Rats (인삼이 흰쥐의 고환조직 핵산 함유량에 미치는 영향)

  • As a continuation of efforts to elucidate the influence of ginseng upon nucleic acid content of various tissues, a study was carried out which measured testicular RNA and DNA contents of rats following administration of ginseng. Thirty male rats $(body\;weight:\;180{\sim}230\;gm)$ were equally divided into a ginseng and a saline group. Once a day for 5 days they received subcutaneously 0.5 m1/100 gm body weight of ginseng extract solution (4 mg of ginseng alcohol extract in 1 ml of saline), and the same amount of satire, respectively. On the 5th experimental day, all animals were sacrificed 2 hours after the last medication and their testicular RNA and DNA contents were measured using the chemical method of Schmidt-Thannhauser-Schneider. Following results were obtained: 1. Testicular RNA and DNA contents were significantly higher in the ginseng group than in the saline group. 2. RNA/DNA ratio of the testis was lower in the ginseng group than in the saline group. However, the two groups did not differ significantly with regard to the RNA/DNA ratio. The ginseng is inferred to raise RNA and DNA contents of testis in rats.

Protective Effects of Blue Honeysuckle on Rat Hypothyroidism Induced by Propylthiouracil

  • The objective of the present study is to determine whether blue honeysuckle lyophilized concentrated powder (BH) has favorable effects on hypothyroidism and related reproductive organ damage. Hypothyroidism was induced by 9 subcutaneous administration of propylthiouracil (PTU) for 28 days. Levothyroxine (LT4)-treated group was intraperitoneally injected with LT4 for the same period, while for BH (125, 250, and 500 mg/kg) or Flos Lonicerae lyophilized aqueous extract (LF, 250 mg/kg)-treated groups, the test materials were orally administrated for 42 days: two weeks before PTU injection and during PTU administration. The changes in serum thyroid hormone levels, serum male sex hormone levels, and testis antioxidant defense system were observed by histopathology of the thyroid gland, epididymis, prostate, and testis. The oral administrations of 125, 250, and 500 mg/kg of BH showed favorable effects compared to LF on hypothyroidism and related damages of reproductive organs through augmentation of the antioxidant defense system in the testis. In conclusion, BH is a promising new potent thyroid gland protecting agent.

Effect of Naturally Derived Substances on Motion Parameters of In Vitro Non-Freezing Preserved Pig Sperm

  • Artificial insemination (AI) has been performed widely in swine industry using fresh liquid sperm instead of frozen type of sperm. However fresh sperm are not able to preserve more than three days with optimal motility and other sperm parameters for the successful fertilization, since in vitro stored sperm has an oxidative stress that resulted increase of abnormality and acrosome reation. To overcome these major problems, novel preservative formulation is needed to neutralize the oxidative stress and to provide suitable physiological environment for sperm in in vitro. In this study, naturally derived substances such as Poncirus trifoliate (Trifoliate orange), Garcinia mangostana (Mangosteen), pig placenta and testis extracts were tested as sperm preservative agents. Placenta extracts (PE), trifoliate orange extracts (TOE), testes extracts (TE) and mangosteen extracts (ME) were applied to analyze specific parameters for sperm motion characteristics individually and combinatorial. Each individual extract treatment can accelerate the sperm motility but noticeably TOE, TE and ME treatments exhibited the considerable and significant preservation of sperm motility. PE, TE and ME showed a significant (p<0.05) increase in ALH after one week. Further we evaluated the five different combinations of these extracts on sperm motility and its motion characteristics. Surprisingly even after one week ME, TOE and TE combination significantly preserved the sperm motility about 75%. It is noteworthy that unlike individual extract treatment, combination of ME, TOE and TE simultaneously protect the sperm motility and its motion characteristics. Taken together these data conclude that addition of ME, TOE and TE can be effective for preservation of pig sperm.

Effect of the Water Extract of Cultivated Wild Ginseng on the Toxicities Induced by Cyclophosphamide in Mice (산양삼이 Cyclophosphamide의 독성에 미치는 영향)

  • This study was investigated the protective effect of Cultivated Wild Ginseng(WG) on the toxicities induced by cyclophosphamide(CP) in mice. Twenty-four male BALB/c mice were divided into non-treated normal group(n=6), CP treated control group(n=6), CP+WG treated CP+WG group(n=6), WG treated WG group(n=6). CP(100 mg/kg of b.w., i.p) was injected at 0, 7 th, 14 th, 21 th, and 28 th day of the experiment respectively. WG(4.4 g/kg, i.p.) was administrated for 35days. Body and organ(heart, liver, kidney, testis) weight were measured. Histopathological examination on the organ(heart, liver, kidney, testis), morphometric analysis, and BrdU immunohistochemistry on the testis were performed. Body weight was decreased following CP administration. In contrast, such a decrease was significantly attenuated by WG administration. CPK and AST of CP+WG group were significantly decreased compared with CP group. Histopathologically, cross sectional area of testis and diameter of seminiferous tubule were significantly increased in CP+WG group compared with CP group. BrdU labelled cells in the seminiferous tubules were remarkably decreased in CP group. Whereas the number of seminiferous tubules labelled with BrdU in spermatogonia was increased by CP+WG administration. The obtained results suggest that WG has protective effect on CP-induced toxicity. This effect might be mediated through the supplementation of vital energy.

Effect of the Water Extract of Cultured Wild Ginseng on the Acute and Subacute Toxicities Induced by Doxorubicin in Mice (산양산삼이 Doxorubicin의 급성 및 아급성 독성에 미치는 영향)

  • This study was carried out to examine the protective effect of Cultured Wild Ginseng(CWG) on the acute and subacute toxicities induced by doxorubicin(Doxo) in mice. Heart and liver weight was decreased following Doxo administration. In contrast, such a decrease was significantly attenuated by CWG administration. The value of serum CPK in Doxo group was increased compared with normal group. But the value of CWG group were decreased significantly compared with the values of Doxo group in the liver of the Doxo group, cloudy swelling of hepatic cells and narrowing of sinusoids were observed. Whereas in the CWG group, well oriented hepatic cell cords and sinusoids were observed. In the testis of the Doxo group, necrotic and degenerative changes of the seminiferous tubules, especially beneath testicular membrane were observed. But those lesions were alleviated in CWG group. Cross sectional area of testis and diameter of semineferous tubule were significantly increased in CWG group compared with Doxo group. Body weight was reduced in Doxo group compared with normal group. In contrast, such a decrease was significantly attenuated by CWG administration atwa5th day. Spermatogenetic cells in seminiferous tubules were necrotic and desquamated and the cellularity of seminiferous epithelia was reduced in Doxo group. But those lesions were attenuated by CWG administration. Cross sectional area of testis and diameter of seminiferous tubule were significantly increased in CWG group compared with Doxo group. In addition, the increase in lipid peroxidation(LPO) in testis was inaddition, the, iout such a increased was significantly inhibited in CWG group. BrdU labelled cells in the seminiferous tubules were remarkably decreased in Doxo group. Whereas the number of seminiferous tubules labelled with BrdU in spermatogonia was increased by CWG administration. The obtained results suggest that CWG has protective effect on doxorubicin-induced toxicity. This effect might be mediated through the supplementation of vital energy.

Effect of Cynomorii Herba on the Sperm Concentration, Motility and Spermatogenesis in Male Rat (쇄양(鎖陽)이 백서(白鼠) 정자(精子)의 수(數), 운동성(運動性) 및 형성(形成)에 미치는 영향(影響))

  • Purpose : This study was conducted to investigate the effects of Cynomorii Herba (鎖陽) on the reproductive competence in male mice. Methods : We used 8-week-old Spraque-Dawley rats and administered the extract powder of Cynomorii Herba to 5 rats and normal saline to control group once in a day for 28 days. Then we observed the changes of body weight before and after administration of Cynomorii Herba extracts and normal saline. We isolated their testis surgically and observed the weight of testis, epididymis, vascular gland and prostate. Also we examined the total, normal and motile sperm concentration, the concentration of testicular catalase, peroxidase and configuration of testicular tissue before and after administration of Cynomorii Herba extracts and normal saline. Results : We found that the concentration of total, normal, motile sperm in testis of Cynomorii Herba treated group shows significant difference compared with the control group. The body weight, the weight of testis, epididymis and the concentration of testicular catalase and peroxidase were higher in the Cynomorii Herba treated group, on the other hand the weight of vascular gland was lower in the Cynomorii Herba. But the contents of above statement showed no significant difference. The weight of prostate showed lower in the control group statistically than that of the Cynomorii Herba treated group. We observed the seminiferous tubules taken shape minutely and the number of normal sperm increased in Cynomorii Herba treated group's testicular tissue. Conclusion This study shows that Cynomorii Herba may have an effect on the concentration, morphology and motility of sperm, the important factor in male fertility.

Administration Duration Dependent Effects of Epimedium Herb Extract Solution on the Reproductive Capacities in the Mice (음양곽(淫羊藿)의 수컷 생쥐 생식능력(生殖能力)에 대한 투약기간별(投藥期間別) 효과(效果))

  • Purpose : This study was undertaken to evaluate the effects of the different administration duration of Epimedium Herb extract solution on the spermatogenic abilities such as concentration, motility and morphological normality of sperm from the testis and the activities of sperm hyaluronidase. Methods : We used the 2-month-old mice and administered the extract solution of Epimedium Herb 0.3 ml/g/day for 30, 60, 90 and 120 days. The control group was administered the normal saline as the same way. We examined the number of total, motile and normal sperm from the cauda epididymis, the activities of sperm hyaluronidase. Also we observed changes of isolated testis before and after administration of Epimedium Herb extract solutions in the mice. And we compared the testicular tissue especially seminiferous tubules with the control and treated group by histochemical methods. Results : The significant differences were observed in the concentration of total sperm and normality of spermatozoa of the Epimedium Herb extract solution administered groups compared to the control group in 60, 90 and 120 days groups. The significant differences were observed the motility of the Epimedium Herb extract solution administered groups compared to the control group in 60 days group. In the histological analysis of the testicular tissues, the enlargement of testicular lobe diameter and apparent vasculogenesis between testicular lobes were observed in the Epimedium Herb extract solution administered groups compared to the control group, respectively. Conclusion : This study shows that Epimedium Herb has the beneficial effect on the concentration, morphology and motility of sperm, the testicular tissues and the activities of sperm hyaluronidse in 60 days administration group. We can suggest that Epimedium Herb extract solution be useful for the treatment of male sexual dysfunction and infertility.

We can suggest that Morindae officinalis Radix extract solution be useful for the treatment of male sexual dysfunctions and infertility.
