In order to develop new immuno-stimulating ingredients from mature leaves of green tea, crude polysaccharides were isolated from pectinase digests of tea leaves (green tea enzyme digestion, GTE-0), after which their immuno-stimulating activities and chemical properties were examined. GTE-0 mainly contained neutral sugars (54.9%) such as glucose (14.2%), arabinose (12.2%), rhamnose (11.1%), and galacturonic acid (45.1%), which are characteristic of pectic polysaccharides. The anti-complementary activity of GTE-0 was similar to that of polysaccharide K (used as positive control). Number of morphologically activated macrophages was significantly increased in the GTE-0-treated group. GTE-0 significantly augmented $H_2O_2$ and reactive oxygen species production by murine peritoneal macrophage cells in a dose-dependent manner, whereas production of nitric oxide showed the highest activity at a dose of $100{\mu}g/mL$ among all tested concentrations. Murine peritoneal macrophages stimulated with GTE-0 showed enhanced production of various cytokines such as interleukin (IL)-6, IL-12, and tumor necrosis factors-${\alpha}$ in a dose-dependent manner. Further, GTE-0 induced higher phagocytic activity in a dose-dependent manner. In ex vivo assay for cytolytic activity of murine peritoneal macrophages, GTE-0-treated group showed significantly higher activity compared to the untreated group at an effector-to-target cell ratio of 20. The above results lead us to conclude that polysaccharides from leaves of green tea have a potent immuno-stimulating effect on murine peritoneal macrophage cells.

Environment-friendly agro-materials are are likely to be preferred to chemical insecticides recently. For this reason, many studies are conducted to develop environment-friendly insecticides containing natural materials. This study was also conducted so as to assess ecotoxicity for Emulsifiable concentrate (EC) containing 30% of garlic extract or two plant essential oils (Zanthoxylum, Lemongrass) expected to prevent from pests and be used for agro-materials. Target species used to assess acute toxicity were invertebrate (Daphina magna), fish (Oryzias latipes), honeybee (Apis mellifera L.) and earthworm (Eisenia fetida). The $EC_{50}$ values for of garlic extract 30% EC, Zanthoxylum oil 30% EC and lemongrass oil 30% EC to Daphina magna were 3.3, 10, and $10mg\;L^{-1}$, respectively. The category of garlic extract 30% EC was moderately toxic, while those of Zanthoxylum oil 30% EC and lemongrass oil 30% EC were slightly toxic according to standard of USEPA. $EC_{50}$ for both of Zanthoxylum oil 30% EC and lemongrass oil 30% EC were more than $10mg\;L^{-1}$ then they were considered as slightly toxicity. In case of acute toxicity test to fish, $LC_{50}$ of garlic extract 30% EC was $3.3mg\;L^{-1}$. Zanthoxylum oil 30% EC and lemongrass oil 30% EC indicated $LC_{50}$ > $10mg\;L^{-1}$. Classification of acute toxicity to all test substances was in Korea criteria. Acute contact and oral toxicity test to Honeybee were conducted. As a result, $LD_{50}$ of all test substances were more than 100 a.i. ${\mu}g\;bee^{-1}$ in the acute contact test while $LD_{50}$ of garlic extract 30% EC was 4.4 a.i. ${\mu}g\;bee^{-1}$ and $LD_{50}$ of Zanthoxylum oil 30% EC and lemongrass oil 30% EC were more than 100 a.i. ${\mu}g\;bee^{-1}$. In case of acute toxicity test to earthworm, $LC_{50}$ of garlic extract 30% EC, Zanthoxylum oil 30% EC and lemongrass oil 30% EC were 267, 592, and $430mg\;kg^{-1}$, respectively. In conclusion, if the safety for earthworm is confirmed, these substances are expected to be use for environment-friendly insecticide materials with low risk against ecosystem and contribute to developing environment-friendly agro-materials.
This study was conducted to assess the environmental risks of anthracnose resistant transgenic chili peppers with the PepEST gene on non-target organisms in the agroecosystem environments during the chili pepper growing seasons in 2006. We quantitatively collected arthropods assemblages living on leaves and flowers of chili peppers on June 20, July 25, and August 25 by using an insect vacuum collector to compare the patterns of arthropod community structures between non-transgenic chili peppers (nTR, WT512) and anthracnose resistant transgenic chili peppers (TR, line 68). We found the seasonal difference with the highest species richness and Shannon's diversity in July's sampling among the growing seasons (P<0.05) and each sampling season showed the different arthropod community composition. We also found there was no statistical difference between the two types of crops, nTR and TR, at each sampling time (P>0.05). The significance level of arthropod community showed that there were lots of seasonal difference of functional groups as well as taxa but only the herbivore group in the functional groups was significantly different for the types of plants (P<0.05). So, we further examined the herbivore groups to find any potential damage and identified the possibility of herbivorous damage from some herbivores, grasshoppers, aphids and thrips. Although we couldn't find any adverse effects from the environmental risk assessment between the arthropod community structures on two types of plants from our results, we should keep working for the environmental risk assessment because of the herbivorous potential risk possibility.

This study was conducted to determine whether invasive muskrats settle in the natural ecosystem of Korea. The study also aimed to provide information on the selection of target areas, basic behavioral characteristics, and habitat use-related data, all of which are essential for effective management strategy establishment and site management. To this end, a trend in international muskrat management was identified to examine the management direction according to the settlement stage. We also secured the location of 72 domestic farmers and classified five regions in Gyeonggi-do, Chungcheongnam-do, Gyeongsangbuk-do, and Sejong Special Self-governing City into regions with high potential for natural leakage to raise the need for a quick survey. We determined that muskrats successfully settled in the natural ecosystem of Korea, based on the fact that we consistently identified a muskrat habitat at the same point for three years, from 2016 to 2018, the age of the captured population was estimated to be 1.2 years, and there had been no additional make-up factors near the habitat since 2012. The muskrats that settled in Korea showed a small home range of 0.0027 ㎢ (MCP 95%) and were estimated to show regular mobility before Spring and Autumn. Also, the areas where the muskrat habitat was observed had a high level of habitat environment disturbance and their ecological and food intake characteristics reflected that disturbance. Muskrat traces were high in hydrophyte vegetation, which can be used as a hideout in open waters and where the growth of both floating leaved plants and floating plants are prevalent. Since international invasive muskrats have already settled in the domestic ecosystem and there is concern about their proliferation in the future, the government must consider a preemptive response and scientific management.
This study was conducted to assess the possibility of 5,8-dihydroxy-1,4-naphthoquinone (DHNQ) as a environmental friendly herbicide candidate. Foliar application of DHNQ showed excellent herbicidal effect to the 3 grasses and 5 broad-leaved weeds. Among them, Digitaria sanguinalis and Solanum nigrum were completely controlled by $250{\mu}g\;mL^{-1}$ of DHNQ with main symptoms of desiccation or burndown within 24 hours. Aeschynomene indica was also sensitive to DHNQ treatment. All of the eight weed species were controlled by 90~100% at a concentration of $1000{\mu}g\;mL^{-1}$. However, soil application of DHNQ to Digitaria sanguinalis did not show any herbicidal symptoms. DHNQ strongly inhibited KAPAS activities in vitro and the $IC_{50}$ was $4.4{\mu}M$. Cellular leakage from cucumber leaf squares treated with DHNQ increased depending on the concentrations increased from 6.25 to $100{\mu}M$ after 24 hours incubation with or without light. However, chlorophyll loss in cucumber leaf squares was negligible. Biotin supplements significantly rescued the inhibition of germination rate of Arabidopsis thaliana seeds previously inhibited by the DHNQ. According to above results, DHNQ is a good natural herbicide candidate having a new target KAPAS, which is involved in biotin biosynthesis pathway, with environmental friendly.

Bacteria are frequently contaminated during the collection and processing procedures of boar semen. Of the contaminants, Stenotrophomonas (S.) maltophilia is a Gram-negative bacterium that is widely distributed in a variety of habitats. Although PCR assays have been developed for the detection of S. maltophilia, they cross-react with some species of Xanthomonas. In this study, we designed a primer set for the detection of S. maltophilia in order to target the chiA (GenBank accession no. NC_010943) gene. The specific PCR products were amplified from S. maltophilia only, not from other tested strains that are frequently found in semen. The detection limit of the PCR was $1.5{\times}10^3$ CFU/ml with pure-cultured S. maltophilia and $1.5{\times}10^4$ CFU/ml with S. maltophilia spiked in semen. Twenty-six (5.9%) S. maltophilia were isolated from 440 semen samples. The PCR results exhibited 98.9% agreement with a comparison of S. maltophilia isolation. Also, the sensitivity and specificity of the PCR were 100% and 98.7%, respectively. In the antimicrobial susceptibility test, S. maltophilia isolates were highly susceptible to enrofloxacin and florfenicol, while the majority of them were resistant to amoxicillin/clavulanic acid, apramycin, ceftiofur, penicillin, and spectinomycin. These results indicated that the PCR using the chiA gene was proven to be reliable and effective for the detection of S. maltophilia with high levels of sensitivity and specificity.
The transcription of mRNA of avian influenza virus is regulated temporally during infection. Therefore, the measurement of transcript level in host cells should be performed before viral release from host cells because errors can occur in the analysis of the transcript levels if the viruses released from the infected cells re-infect cells. In this study, the timing of viral release was determined by measuring the level of viral RNA from viruses released from H9N2-infected chicken fibroblast cell line UMNSAH/DF-1 by semi-quantitative RT-PCR. The viral genomic RNA was isolated together with mouse total RNA which was added to the collected medium as carrier to monitor the viral RNA recovery and to use its GAPDH as an internal control for normalizing reverse transcription reaction as well as PCR reaction. It was found that viral release of H9N2 in the chicken fibroblast cell line UMNSAH/DF-1 took between 16 and 20 h after infection. We measured all 8 viral mRNA levels. Of the 8 transcripts, 7 species of viral mRNAs (each encoding HA, NA, PB1, PB2, NP, M, NS, respectively) except PA mRNA showed robust amplification, indicating these mRNA can be used as targets for amplification to measure transcript levels. These results altogether suggest that the method in this study can be used for screening antiviral materials against viral RNA polymerase as a therapeutic target.
The suppression of neuroinflammatory responses in microglial cells, well known as the main immune cells in the central nervous system (CNS), are considered a key target for improving the progression of neurodegenerative diseases such as Alzheimer's disease, Parkinson's disease, and Huntington's disease. Teleogryllus emma is widely consumed around the world for its broad-spectrum therapeutic effect. In a previous work, we performed transcriptome analysis on T. emma in order to obtain the diversity and activity of its antimicrobial peptides (AMPs). AMPs are found in a variety of species, from microorganisms to mammals. They have received much attention as candidates oftherapeutic drugs for the treatment of inflammation-associated diseases. In this study, we investigated the anti-neuroinflammatory effect of Teleogryllusine (VKWKRLNNNKVLQKIYFVKI-NH2) derived from T. emma on lipopolysaccharide (LPS) induced BV-2 microglia cells. Teleogryllusine significantly inhibited nitric oxide (NO) production without cytotoxicity, and reducing pro-inflammatory enzymes expression such as inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2). In addition, Telegryllusine also inhibited the expression of pro-inflammatory cytokines such as interleukin-6 (IL-6) and tumor necrosis factor alpha (TNF-α) through down-regulation of the mitogen-activated protein kinases (MAPKs) and nuclear factor kappa B (NF-κB) signaling pathway. These results suggest that T. emma-derived Teleogryllusine could be a good source of functional substances that prevent neuroinflammation and neurodegenerative diseases.

Target species used to assess acute toxicity were aquatic invertebrate (Daphina magna), fish (Oryzias latipes), honeybee (Apis mellifera L.) and earthworm (Eisenia fetida). The EC50 value, toxicological responses of thyme white, clove bud, and cassia to Daphina magna were 2.5, 2.8, and $6.9mg\;L^{-1}$ respectively and these values were moderately toxic according to standard of USEPA. $EC_{50}$ of Lavender and lemon eucalyptus were >$10mg\;L^{-1}$ then they were considered as slightly toxicity. In case of acute toxicity test to fish, $LC_{50}$ of thyme white and cassia were 6.7 and $7.5mg\;L^{-1}$ each other. The other plant essential oils indicated $LC_{50}$ >$10mg\;L^{-1}$. Acute contact and oral toxicity test to Honeybee were conducted. As a result, $LD_{50}$ of all essential oils were >$100{\mu}g$ a.i. $bee^{-1}$ in both of tests. In case of acute toxicity test to earthworm, $LC_{50}$ of thyme white, clove bud, cassia, lavender, and lemon eucalyptus were 149, 230, 743, 234, and $635mg\;kg^{-1}$, respectively. In conclusion, if the safety for earthworm is confirmed, 5 plant essential oils are expected to be use for environment-friendly insecticide materials with low risk against ecosystem and contribute to developing environment-friendly agro-materials.
The present study was conducted to confirm that a bivalve Ruditapes philippinarum can be used as a biomarker for the monitoring of the heavy metal pollution in the silt of the marine environment. The clams were collected from the silt of Cheonsu-bay, Buheung-ri, and Tan-island of the West Sea, Korea. To observe the normal structures of the target organs (hepatopancreas and gill), they were dissected out for the immunohistochemical study and the electron microscopy with TEM, SEM, and SEM-EDS. The immunohistochemical study showed that the interdiverticular connective tissues of the hepatopancreas, and the outer epithelium of the gill lamellae was strongly reacted to anti-metallothionein (MT), indicating the presence of MT, a metal-binding protein, involved in metal detoxifying process. According to the examinations under the TEM, the epithelial cells of the hepatopancreas of the clams collected from polluted area (Tan-island) showed certain changes such as swollen rER, swollen nuclear envelope and inclusion bodies in the nulcei. In the SEM-EDS analysis, tissue of the hepatopancreas showed relatively higher concentration of S, Zn, and Cd. These elements are supposed to be concerning with the MT-reaction in the hepatopancreas. Considering that the coastal bivalve R. philippinarum showed immediate subcellular responses to heavy metal pollution in the overall experiments conducted, this species might act as one of efficient biomarkers for the heavy metal contamination in the marine environment.
