DNA topoisomerases I (topo I) and II are essential enzymes that relax DNA supercoiling and relieve torsional strain during DNA processing. including replication. transcription. and repair. Topo I relaxes DNA by cleaving one strand of DNA by attacking a backbone phosphale with a catalytic lyrosine (Tyr723. human topo I). This enzyme has recently been investigated as a new target for antineoplastic drugs. Inhibitors to the enzyme intercalate between the DNA base pairs. interfering religation of cleaved DNA, therefore inhibit the activity of topo I. (omitted)
본 연구에서는 연약지반기초 및 암반사면 거동 계측 도구로서 스파이럴 볼트 변형률계의 적용 가능성을 살펴보고자 한다. 이 변형률계가 지중에 설치되었을 때, 지반에 대한 인발 저항성이 높아 지반 보강용으로 적용될 수 있을 뿐만 아니라, 유연성의 특징을 가지고 있기 때문에 지반의 거동 상태를 파악하는데 유용하게 이용될 수 있다. 연약지반기초에 대한 스파이럴 볼트의 변형률 계측결과, 변형률 측정 초기에는 안정한 상태를 보이다가 400일이 경과한 시점에서는 스파이럴 볼트 변형률계의 윗부분과 중간부분에서 큰 변화가 관찰되었다. 이러한 변화는 동일한 시기에 강수량 증가와 함께 빈번하게 발생한 진도 1~2의 지진에 의한 영향으로 지반이 이완되어 야기한 것으로 분석된다. 암반사면에 대한 스파이럴 볼트의 변형률 계측결과, 측정시작일로부터 50일 동안에 안정한 상태를 보였으며 50~160일 기간에는 매설길이 4.2 m 지점(P6)의 변형률 게이지에서 가장 큰 변형률을 보였다. 그러나 P6 변형률 게이지를 제외한 나머지 게이지는 특이한 변화를 보이지 않았다. 스파이럴 볼트 변형률계의 측정결과를 종합적으로 살펴보면 측정대상인 연약지반기초와 암반사면은 안정적인 것으로 판단된다. 본 연구에서 소개하는 스파이럴 볼트 변형률계는 연약지반기초 및 암반사면 거동의 모니터링과 동시에 대상지반의 보강에 적용될 수 있을 것으로 사료된다.
출아효모인 Sacharomyces cerevisiae S288C균주를 이용한 효모의 게놈이 완성된 후 S. cerevisiae는 다양한 연구 모델로 이용되어져 왔다. 현재까지 효모를 이용한 기능 유전체학 측면에서의 연구는 laboratory strainin인 S288C 균주 또는 그 유래의 균주들이다. 그러나 자연에서 분리된 효모 또는 산업적으로 이용되어지고 있는 S. cerevisiae의 유전학 측면에서의 연구는 낮은 포자형성률 및 형질전환률, 그리고 S288C 균주와의 게놈상의 상이성 때문에 거의 이루어지지 않고 있다. 여기서 우리 연구진은 자연에서 분리된 Saccharomyces cerevisiae KNU5377 균주를 이용하여 random spore analysis를 통해 MATa 및 $MAT{\alpha}$ 타입의 각각의 haploid cell을 분리 후 이미 보고된 KanMX module를 가지고 round PCR기법에 의한 short flanking homology 기법을 이용하여 전사조절인자인 HSF1 유전자가 치환된 변이주를 구축할 수 있었다. 덧붙여, 모든 유전자에 이 기법을 적용할 수는 없다는 것을 확인하였다. 앞으로 이 변이주를 통해 기능 유전체학적인 측면에서 이 유전자의 스트레스와의 관련성을 연구하고자 한다.
Cryparin은 Cryphonectria parasitica의 세포벽에 풍부한 소수성 단백질에 속한다. cryparin은 비록 하나의 유전자에 의해 발현되지만 액체배양 후 48시간이 지나면 발현된 전체 유전자중에서 22%를 차지할 정도의 높은 발현 양상을 나타낸다. 또한 cryparin은 RNA mycovirus인 Cryphonectria hypovirus 1의 감염에 의해 발현이 현저히 억제되는 유전자로 알려졌다. 이미 지난 실험(Kim et al., 1999)에서 상동염색체간의 재조합을 이용하여 cryparin 유전자를 항생제 hygromycin B 저항성 유전자로 치환한 치환체를 제조하였다. 발현율이 매우 높으면서도 virus에 의해 밀접하게 영향받는 cryparin 유전자의 promoter 분석을 위하여서는 대상이 되는 유전자 치환을 위한 vector만을 포함하며, 분석에 이용될 여러 유전자 운반체들이 어느 한곳에만 삽입되도록 하는 성질을 가진 균주의 개발이 필요하다. 그러나 지난번 실험의 결과 얻어진 cryparin 치환체는 치환용 vector외에도 무작위로 삽입된 vector가 존재하고 나아가 새로운 vector들이 어느 한곳에만 삽입되도록 하는 성질을 갖지 못하였다. 따라서 본 실험에서는 cryparin 유전자 치환체와 영양요구성 돌연변이체인 균주간의 교잡을 이용하여 분석 대상이 되는 유전자의 치환에 이용된 vector만을 포함하며, 분석에 이용될 여러 유전자 운반체들이 genome내의 어느 한곳에만 삽입되도록 하는 성질을 가진 균주를 제조하였다.
단일 공격과 달리 DDoS 공격들은 네트워크에 분산된 봇넷이 동시에 타겟 서버에 공격을 개시한다. 이 경우 타겟 서버에서는 정상적인 사용자의 편의를 고려해야 하기 때문에 DDoS로 간주되는 패킷에 대하여 접속거부 조치를 취하기 어려운 점이 있다. 이를 고려하여 본 논문에서는 사용자 네트워크단위로 DDoS 공격을 탐지하고 네트워크 관리자가 조치를 취할 수 있도록 하여 전체적으로 봇넷의 규모를 줄여서 타겟 서버의 부담을 줄일 수 있는 DDoS 봇넷 탐지 시스템을 구현 하였다. 본 논문에서 제안한 DDoS 봇넷 탐지 시스템은 공격 트래픽의 시간 단위 흐름을 분석하고 수집한 이상상태에 대한 데이터베이스를 바탕으로 공격을 탐지 하도록 프로그램을 구현하였다. 그리고 패킷들의 평균개수와 표준편차를 이용하여 현재 트래픽의 임계치(Threshold)를 계산하고 이 임계치를 이용하여 DDoS 공격 여부를 판단하였다. 공격의 대상이 되는 서버를 중심으로 이루어졌던 봇넷 탐지 단위를 DDoS 봇에 감지된 공격 모듈이 속한 네트워크 단위 탐지로 전환함으로써 DDoS 공격에 대한 적극적인 방어의 개념을 고려해 볼 수 있었다. 따라서 DDoS와 DoS 공격의 차이점이라 할 수 있는 대규모 트래픽 흐름을 사전에 네트워크 관리자가 차단함으로써 봇넷의 규모를 축소시킬 수 있다. 또한, 라우터 장비 이하의 네트워크 통신에서 트래픽 공격을 사전에 차단할 수 있다면 타겟 서버의 부담뿐만 아니라 WAN 통신에서 라우터의 네트워크 부하를 상당부분 감소시킬 수 있는 효과를 얻을 수 있을 것으로 기대한다.
Fed-batch cultures of Hansenula polymorpha were studied to develop an efficient biosystem to produce recombinant human serum albumin (HSA). To comply with this purpose, we used a high-purity oxygen-supplying strategy to increase the viable cell density in a bioreactor and enhance the production of target protein. A mutant strain, H. polymorpha GOT7, was utilized in this study as a host strain in both 5-l and 30-l scale fermentors. To supply high-purity oxygen into a bioreactor, nearly 100% high-purity oxygen from a commercial bomb or higher than 93% oxygen available in situ from a pressure swing adsorption (PSA) oxygen generator was employed. Under the optimal fermentation of H. polymorpha with highpurity oxygen, the final cell densities and produced HSA concentrations were 24.6 g/l and 5.1 g/l in the 5-l fermentor, and 24.8 g/l and 4.5 g/l in the 30-l fermentor, respectively. These were about 2-10 times higher than those obtained in air-based fed-batch fermentations. The discrepancies between the 5-l and 30-l fermentors with air supply were presumably due to the higher contribution of surface aeration over submerged aeration in the 5-l fermentor. This study, therefore, proved the positive effect of high-purity oxygen in enhancing viable cell density as well as target recombinant protein production in microbial fermentations.
Antimicrobial peptides (AMPs) are small but effective cationic peptides with variable length. In previous study, four hexapeptides were identified that showed antimicrobial activities against various phytopathogenic bacteria. KCM21, the most effective antimicrobial peptide, was selected for further analysis to understand its modes of action by monitoring inhibitory effects of various cations, time-dependent antimicrobial kinetics, and observing cell disruption by electron microscopy. The effects of KCM21 on Gram-negative strain, Pseudomonas syringae pv. tomato DC3000 and Gram-positive strain, Clavibacter michiganensis subsp. michiganensis were compared. Treatment with divalent cations such as $Ca^{2+}$ and $Mg^{2+}$ inhibited the bactericidal activities of KCM21 significantly against P. syringae pv. tomato DC3000. The bactericidal kinetic study showed that KCM21 killed both bacteria rapidly and the process was faster against C. michiganensis subsp. michiganensis. The electron microscopic analysis revealed that KCM21 induced the formation of micelles and blebs on the surface of P. syringae pv. tomato DC3000 cells, while it caused cell rupture against C. michiganensis subsp. michiganensis cells. The outer membrane alteration and higher sensitivity to $Ca^{2+}$ suggest that KCM21 interact with the outer membrane of P. syringae pv. tomato DC3000 cells during the process of killing, but not with C. michiganensis subsp. michiganensis cells that lack outer membrane. Considering that both strains had similar sensitivity to KCM21 in LB medium, outer membrane could not be the main target of KCM21, instead common compartments such as cytoplasmic membrane or internal macromolecules might be a possible target(s) of KCM21.
본 연구에서는, 수종별 벌채부산물의 압축 변형 특성을 규명함으로써 벌채부산물을 압축할 수 있는 장비의 개발을 위한 기초자료로 사용하고자 하였다. 만능재료시험기(Universial Testing Machine)를 이용한 압축재하 시험장치로 벌채부산물의 3회 반복 압축-변형 특성시험을 통해 수종별 벌채부산물의 목표밀도 압축에 필요한 소요압축력을 구하였다. 리기다소나무(Pinus rigida), 잣나무(Pinus koraiensis), 신갈나무(Quercus mongolica)의 반복 압축 시 응력-변형률 자료를 기반으로 지수함수 형태로 모델화한 것의 물리적 특성 값을 분석한 결과, 목표밀도 $350kg/m^3$과 $400kg/m^3$ 모두 벌채부산물 기계적 성질에 따른 응력계수는 압축횟수가 늘어나면서 줄어드는 경향을 나타냈으며, 반대로 변형율 계수는 늘어나는 경향을 나타냈다. 모델화를 통해 압축횟수가 증가할수록 압축에 요구되는 소요응력은 줄어들고, 변형율 변화에 비해 응력증가가 커지는 특성이 있으므로, 적절한 초기 압축력이 벌채부산물의 목표밀도 달성에 중요한 변수임을 확인할 수 있었다.
The harshness of legionellosis differs from mild Pontiac fever to potentially fatal Legionnaire's disease. The increasing development of drug resistance against legionellosis has led to explore new novel drug targets. It has been found that phosphoglucosamine mutase, phosphomannomutase, and phosphoglyceromutase enzymes can be used as the most probable therapeutic drug targets through extensive data mining. Phosphoglucosamine mutase is involved in amino sugar and nucleotide sugar metabolism. The purpose of this study was to predict the potential target of that specific drug. For this, the 3D structure of phosphoglucosamine mutase of Legionella pneumophila (strain Paris) was determined by means of homology modeling through Phyre2 and refined by ModRefiner. Then, the designed model was evaluated with a structure validation program, for instance, PROCHECK, ERRAT, Verify3D, and QMEAN, for further structural analysis. Secondary structural features were determined through self-optimized prediction method with alignment (SOPMA) and interacting networks by STRING. Consequently, we performed molecular docking studies. The analytical result of PROCHECK showed that 95.0% of the residues are in the most favored region, 4.50% are in the additional allowed region and 0.50% are in the generously allowed region of the Ramachandran plot. Verify3D graph value indicates a score of 0.71 and 89.791, 1.11 for ERRAT and QMEAN respectively. Arg419, Thr414, Ser412, and Thr9 were found to dock the substrate for the most favorable binding of S-mercaptocysteine. However, these findings from this current study will pave the way for further extensive investigation of this enzyme in wet lab experiments and in that way assist drug design against legionellosis.
Accurate and efficient microbial diagnosis is crucial for effective molecular diagnostics, especially in the field of human healthcare. The gold standard equipment widely employed for detecting specific microorganisms in molecular diagnosis is quantitative real-time polymerase chain reaction (qRT-PCR). However, its limitations in low metagenomic DNA yield samples necessitate exploring alternative approaches. Digital PCR, by quantifying the number of copies of the target sequence, provides absolute quantification results for the bacterial strain. In this study, we compared the diagnostic efficiency of qRT-PCR and digital PCR in detecting a particular bacterial strain (Staphylococcus aureus), focusing on skin-derived DNA samples. Experimentally, specific primer for S. aureus were designed at transcription elongation factor (greA) gene and the target amplicon were cloned and sequenced to validate efficiency of specificity to the greA gene of S. aureus. To quantify the absolute amount of microorganisms present on the skin, the variable region 5 (V5) of the 16S rRNA gene was used, and primers for S. aureus identification were used to relative their amount in the subject's skin. The findings demonstrate the absolute convenience and efficiency of digital PCR in microbial diagnostics. We suggest that the high sensitivity and precise quantification provided by digital PCR could be a promising tool for detecting specific microorganisms, especially in skin-derived DNA samples with low metagenomic DNA yields, and that further research and implementation is needed to improve medical practice and diagnosis.
