Sulfite 염에 의한 저염 명란젓의 보존 효과 (The Effects of Sulfite Salts on the Shelf-life of Low-salted Myungranjeot (Soused Roe of Alaska Pollack))

  • 저염 명란젓에 sulfite 염을 첨가하여 숙성 중에 일어나는 여러 가지 화학적 및 미생물 변화를 측정하여 저염 젓갈의 shelf-life에 미치는 결과를 요약하면 다음과 같다. Bisulfite 및 metasulfite 명란젓은 숙성초기에 pH가 급격하게 감소하였다가 그 후 서서히 감소하였으며 젖산 생성량도 증가하였다. 숙성이 진행됨에 따라 아미노태 질소량은 감소 경향을 나타내었으며 metasulfite 명란젓이 가장 높은 생성량을 나타내었다. Bisulfite와 metasulfite의 첨가는 숙성 후기의 VBN 및 TMA 생성 억제에 효과적이었으며, 숙성 초기의 TBA 생성을 억제하였다. Bisulfite 및 metasulfite는 fungi를 포함한 미생물의 성장을 현저하게 저해하였다. Sulfite 염 첨가시의 추정되는 shelf-life는 대조군, sulfate, bisulfite및 metasulfite인 경우 각각 16, 14, 20 및 24일이었다.

참다래 발효주와 참다래 리큐르 맛술의 관능적 특성 (Effect of Kiwi Wine and Kiwi Liqueur on Sensory Characteristics as Cooking Alcohol)

  • 참다래 술(발효주, 리큐르)을 요리용 맛술로 활용하고자 고등어조림과 쇠고기 연육에 처리하여 대한 관능효과를 조사하였다. 그 결과, 조효소 활성은 참다래 발효주와 참다래 리큐르 각각 91.02와 87.56%로 높게 나타났다. TMA함량은 무첨가구의 고등어조림에서 6.60 mg%로 나타났고, 발효주와 리큐르를 첨가하여 고등어조림을 조리하였을 때 각각 4.80과 5.79 mg%로 나타났다. 고등어조림의 관능검사에서는 참다래 발효주와 리큐르 첨가구간이 대조구에 비해 높은 기호특성을 보였고, 쇠고기의 연육효과에서도 참다래 발효주와 참다래 리큐르 첨가구간이 대조구에 비하여 우수한 연화작용을 하는 것으로 나타나 요리용 맛술로의 활용 가능성이 있는 것으로 나타났다.

Reliability Enhancement of Anisotropic Conductive Adhesives Flip Chip on Organic Substrates by Non-Conducting Filler Additions

  • Flip chip assembly on organic substrates using ACAs have received much attentions due to many advantages such as easier processing, good electrical performance, lower cost, and low temperature processing compatible with organic substrates. ACAs are generally composed of epoxy polymer resin and small amount of conductive fillers (less than 10 wt. %). As a result, ACAs have almost the same CTE values as an epoxy material itself which are higher than conventional underfill materials which contains lots of fillers. Therefore, it is necessary to lower the CTE value of ACAs to obtain more reliable flip chip assembly on organic substrates using ACAs. To modify the ACA composite materials with some amount of conductive fillers, non-conductive fillers were incorporated into ACAs. In this paper, we investigated the effect of fillers on the thermo-mechanical properties of modified ACA composite materials and the reliability of flip chip assembly on organic substrates using modified ACA composite materials. For the characterization of modified ACAs composites with different content of non-conducting fillers, dynamic scanning calorimeter (DSC), and thermo-gravimetric analyzer (TGA), dynamic mechanical analyzer (DMA), and thermo-mechanical analyzer (TMA) were utilized. As the non-conducting filler content increased, CTE values decreased and storage modulus at room temperature increased. In addition, the increase in tile content of filler brought about the increase of Tg$^{DSC}$ and Tg$^{TMA}$. However, the TGA behaviors stayed almost the same. Contact resistance changes were measured during reliability tests such as thermal cycling, high humidity and temperature, and high temperature at dry condition. It was observed that reliability results were significant affected by CTEs of ACA materials especially at the thermal cycling test. Results showed that flip chip assembly using modified ACA composites with lower CTEs and higher modulus by loading non-conducting fillers exhibited better contact resistance behavior than conventional ACAs without non-conducting fillers.ers.

Expression of Ki-67, p53 and VEGF in Pediatric Neuroblastoma

  • Background: Neuroblastoma (NB), is a neuroectodermal tumor derived from neural crest cells, and it is the second most common pediatric malignant tumor. The biological and clinical behavior of NB is very heterogeneous. This study was conducted to evaluate the expression of Ki-67, p53 and VEGF markers in tissues obtained from NB patients with different histologic types and stage. Materials and Methods: Tissue microarray (TMA) blocks were constructed from paraffin blocks of the NB tissues. Immunohistochemical staining was performed on TMA sections to detect the expression of Ki-67, p53 and VEGF markers. The association between the expression of these markers and clinicopathological parameters were then analyzed. Results: We had 18 patients with NB, one patient with ganglioneuroblastoma (GNB) and one with ganglioneuroma. Ki-67 was expressed in 13 (65%) tumors, and negatively correlated with age, prognosis, histologic type and stage of NB (all p<0.05). High and moderate expression of VEGF was found in 5% (1/20) and 65% (13/20) of the tumors, respectively; and it was positively correlated with age, prognosis and histologic types (all p<0.05) and negatively correlated with MKI (mitosis-karyorrhexis index). p53 expression was observed in 10% (2/20) of the tumors, which showed a relative correlation with MKI (p value=0.07). Conclusions: VEGF as a candidate for anti-angiogenic targeted therapy was correlated with the development and progression of NB; therefore, VEGF along with Ki-67 can serve as a valuable marker for the prognosis of this tumor type.

ALDH1 in Combination with CD44 as Putative Cancer Stem Cell Markers are Correlated with Poor Prognosis in Urothelial Carcinoma of the Urinary Bladder

  • Background: The aldehyde dehydrogenase 1 family member A1 (ALDH1A1) is one of the promising markers for identifying cancer stem cells in many cancer types, along with other markers including CD44. The aim of the present study was to evaluate the expression and clinical significance of putative cancer stem cell markers, CD44 and ALDH1A1, in a series of urothelial carcinomas of urinary bladder (UCUB) by tissue microarray (TMA). Materials and Methods: A total of 159 Urothelial Carcinomas (UC) including 96 (60%) low grade and 63 (40%) high grade carcinomas were immunohistochemically examined for the expression of CD44 and ALDH1A1. Correlations of the relative expression of these markers with clinicopathological parameters were also assessed. Results: High level expression of ALDH1A1 was found in 16% (25/159) of bladder UC which was significantly correlated with increased tumor size (p value=0.002), high grade (p value<0.001), pathologic stage (T1, p value=0.007 and T2, p value<0.001) and increased rate of recurrence (p value=0.013). A high level of CD44 expression was found in 43% (68/159) of cases, being positively correlated with histologic grade (p value=0.032) and recurrence (p value=0.039). Conclusions: Taken together, our results showed that ALDH1 was concurrently expressed in a fraction of CD44+ tumors and its expression correlated with poor prognosis in UCs. ALDH1A1 could be an ideal marker for targeted therapy of UCs in combination with conventional therapies, particularly in patients with high grade carcinomas. These findings indicate that cells expressing ALDH1A1 along with CD44 can be a potential therapeutic target in bladder carcinomas.

Application of Bioinformatics for the Functional Genomics Analysis of Prostate Cancer Therapy

  • Prostate cancer initially responds and regresses in response to androgen depletion therapy, but most human prostate cancers will eventually recur, and re-grow as an androgen independent tumor. Once these tumors become hormone refractory, they usually are incurable leading to death for the patient. Little is known about the molecular details of how prostate cancer cells regress following androgen ablation and which genes are involved in the androgen independent growth following the development of resistance to therapy. Such knowledge would reveal putative drug targets useful in the rational therapeutic design to prevent therapy resistance and control androgen independent growth. The application of genome scale technologies have permitted new insights into the molecular mechanisms associated with these processes. Specifically, we have applied functional genomics using high density cDNA microarray analysis for parallel gene expression analysis of prostate cancer in an experimental xenograft system during androgen withdrawal therapy, and following therapy resistance, The large amount of expression data generated posed a formidable bioinformatics challenge. A novel template based gene clustering algorithm was developed and applied to the data to discover the genes that respond to androgen ablation. The data show restoration of expression of androgen dependent genes in the recurrent tumors and other signaling genes. Together, the discovered genes appear to be involved in prostate cancer cell growth and therapy resistance in this system. We have also developed and applied tissue microarray (TMA) technology for high throughput molecular analysis of hundreds to thousands of clinical specimens simultaneously. TMA analysis was used for rapid clinical translation of candidate genes discovered by cDNA microarray analysis to determine their clinical utility as diagnostic, prognostic, and therapeutic targets. Finally, we have developed a bioinformatic approach to combine pharmacogenomic data on the efficacy and specificity of various drugs to target the discovered prostate cancer growth associated candidate genes in an attempt to improve current therapeutics.

High Quality Tissue Miniarray Technique Using a Conventional TV/Radio Telescopic Antenna

  • Background: The tissue microarray (TMA) is widely accepted as a fast and cost-effective research tool for in situ tissue analysis in modern pathology. However, the current automated and manual TMA techniques have some drawbacks restricting their productivity. Our study aimed to introduce an improved manual tissue miniarray (TmA) technique that is simple and readily applicable to a broad range of tissue samples. Materials and Methods: In this study, a conventional TV/radio telescopic antenna was used to punch tissue cores manually from donor paraffin embedded tissue blocks which were pre-incubated at $40^{\circ}C$. The cores were manually transferred, organized and attached to a standard block mould, and filled with liquid paraffin to construct TmA blocks without any use of recipient paraffin blocks. Results: By using a conventional TV/radio antenna, it was possible to construct TmA paraffin blocks with variable formats of array size and number ($2-mm{\times}42$, $2.5-mm{\times}30$, $3-mm{\times}24$, $4-mm{\times}20$ and $5-mm{\times}12$ cores). Up to $2-mm{\times}84$ cores could be mounted and stained on a standard microscopic slide by cutting two sections from two different blocks and mounting them beside each other. The technique was simple and caused minimal damage to the donor blocks. H&E and immunostained slides showed well-defined tissue morphology and array configuration. Conclusions: This technique is easy to reproduce, quick, inexpensive and creates uniform blocks with abundant tissues without specialized equipment. It was found to improve the stability of the cores within the paraffin block and facilitated no losses during cutting and immunostaining.

감마선 조사된 참굴의 빙장 기간중의 세균학 및 화학적 변화 (Microbiological and chemical changes in the Pacific oysters, Crassostrea gigas exposed to gamma radiation during ice storage)

  • 산란직전의 남해산 참굴(Crassostrea gigas)을 산채로 구입하여 통상조건하에서 탈각하여 얻은 굴육질을 polyethylene laminated aluminium pouch (Al+PE : 0.03+0.03mm) 속에 밀봉하여 살균선량(0.2Mrad) 및 멸균선량(2.0Mrad)의 감마선으조 조사하고, 한편, 100ppm의 염소수에 굴육질을 2분간 침지한 후 탈수하여 같은 방법으로 포장하여 35일간의 빙장기간중 세균학적 및 화학적 변화를 대조 굴(조사치 않고 염소수 처리하지 않은)의 경우와 비교하였다. 굴육질의 시초의 총균수는 g당 700-800에 달했고, 0.2 Mrad의 감마선 조사는 빙장 10일째의 저장 기간중의 총 균수를 검측할 수 있는 선 이하로 감소시켰으며 2.0 Mrad 조사는 20일째까지 거의 무균상태를 가져왔다. 감마선 조사는 또한 세균활동을 표시하는 대사물질인 TMA와 TCA soluble nitrogen의 축적을 현저히 억제시켰으며 빙장 후기부터 번식하기 시작하여 총균수가 g당 100,000을 초과케 된 35일째까지도 대조굴의 동일한 총균수 시기에 비하여 이 대사물의 축적억제가 현저했음은 감마선 조사의 효과가 총균수의 양적인 감소뿐만 아니라 동시에 TMA를 생산하고 단백질을 분해할 능력이 있는 세균제거 내지는 번식의 억제라는 선택적인 양면을 가지고 있음을 보여 준다. 염소수 처리한 굴육질의 경우 총균수의 양적인 감소효과는 현저하지 않했는데도 불구하고 감마선 조사 경우처럼 세균대사물질 축적의 철저한 억제를 가져 왔음은 주목할만 하다. 굴육질의 glycogen 함량은 사후 저장 기간 중 쉽사리 감소되지 않했으며 PH 측정으로 본 생굴의 신선도와의 관련성은 희박하며 조사와 염소수 처리굴의 경우 더욱 그러했다.

저전압 대회전을 위한 분리된 압전 구동기에 의한 미소거울 (Micromirrors Driven by Detached Piezoelectric Microactuators For Low-voltage and Wide-angle Rotation)

  • This paper presents a torsional micromirror detached from PZT actuators (TMD), whose rotational motion is achieved by push bars in the PZT actuators detached from the micromirror. The push bar mechanism is intended to reduce the bending, tensile and torsional constraints generated by the conventional bending bar mechanism, where the torsional micromirror is attached to the PZT actuators (TMA). We have designed, fabricated and tested prototypes of TMDs for single-axis and dual-axis rotation, respectively. The single-axis TMD generates the static rotational angle of $6.1^{\circ}$ at 16 VDC, which is 6 times larger than that of single-axis TMA, $0.9^{\circ}$. However, the rotational response curve of TMD shows hysteresis due to the static friction between the cover and the push bar in the PZT actuator. We have shown that 63.2% of the hysteresis is due to the static friction caused by the initial contact force of the PZT actuaor. Without the initial contact force, the rotational response curve of TMD shows linear voltage-angle characteristics. The dual-axis TMD generates the static rotational angles of $5.5^{\circ}$ and $4.7^{\circ}$ in x-axis and y-axis, respectively at 16 VDC. The measured resonant frequencies of dual-axis TMD are $2.1\pm0.1$ kHz in x-axis and $1.7\pm0.1$ kHz in y-axis. The dual-axis TMD shows stable operation without severe wear for 21.6 million cycles driven by 16 Vp-p sinusoidal wave signal at room temperature.
