복합레진의 미세누출에 관한 연구 (AN IN VITRO STUDY OF MICROLEAKAGE OF COMPOSITE RESINS)

  • 치과의 수복재로 널리 쓰이는 복합레진의 미세누출에 대한 할로겐광과 플라즈마광의 영향을 비교하고, 유동성 복합레진과 할로겐광으로도 중합시간이 20초로 단축된 복합레진 $Filtek^{TM}$ Z250이 미세누출면에서 기존에 널리 쓰이던 복합레진인 Z100을 대체할 수 있을지를 평가하며, 미세누출에 미치는 adhesive resin의 영향을 알아보고자 본 실험을 수행하였다. 발거된 치아에 5급 와동을 형성하고 레진의 충전은 각 군에 따라 시행하였다. 유동성복합레진 $Filtek^{TM}Flow$와 Z100, $Filtek^{TM}Z250$을 이용하였고, 할로겐광과 플라즈마광을 이용하여 중합하였다 충전이 완료된 시편은 보관, 연마, 열순환을 시행하고, 1% methylene blue를 침투시킨 후 협설로 절단하여 미세누출 정도를 관찰하여 다음과 같은 결론을 얻었다. 1. $Filtek^{TM}Flow$$Filtek^{TM}Z250$ 모두 광원에 따른 미세누출의 차이는 없었다(p>0.05). 2. 유동성 복합레진인 $Filtek^{TM}Flow$는 복합레진 $Filtek^{TM}Z250$과 Z100에 비해 통계적으로 유의성있는 큰 미세누출을 보여(p<0.05) 유동성 복합레진만으로 수복한다면 미세누출의 위험이 커질 것이다. 3. Adhesive resin을 도포한 것이 할로겐광을 이용한 경우와 플라즈마광을 이용한 경우 모두에서 통계적으로 유의하게 미세누출을 감소시켰다.(p<0.05)

Resazurin(alarmar Blue$^{TM}$)을 이용한 효적인 in vitro항비듬력 측정법 (The Effective in vitro Anti-dandruff Test Method with Resazurin(alarmar Blue$^{TM}$))

  • Resazurin(alarmar Blue$^{TM}$)을 이용하여 in vitro 적으로 P. ovale의 최소 발육저지농도 (Minimum Inhibitory Concentration , MIC)를 측정함으로써 여러 가지 조성의 항비듬제의 항비듬 효과를 측정하고자 하였다. 최적의 실험 조건을 결정하기 위한 기초 실험 결과, 약 2.6$\times$$10^{5}$ cfu/$m\ell$ 이상의 농도로 P. ovate가 주입되는 경우 alarmar Blue$^{TM}$ 흡광도의 변화를 정 하게 관찰할 수 있었고 alannar Blue$^{TM}$ 자체의 희석 배율이 1:1-1:4이며, 16시간과 24 시간 배양을 하여야 비교적 정량적인 결과를 얻을 수 있었다. alarmar Blue$^{TM}$ 가 균체의 증식에 미치는 영향을 고찰하여 본 결과 1:1의 희석 용액에서도 alarmar Blue$^{TM}$가 균체의 증식에 영향을 미치지 못함을 알 수 있었다. Zinc pyrithione과 Climbazole의 혼합계에서 항비듬 효과를 관찰하여 본 결과 일정 혼합비에서 효과적인 항비듬 효과를 관찰할 수 있었고, 현미경 관찰에 의한 MIC 측정법과,SDDM(Skin-Disk Diffusion Method)법에 의한 항비듬 효과와 비교하여 본 결과 유사한 경향성을 관찰할 수 있었다. 이상의 결과를 종합해볼 때 alarmar Blue$^{TM}$를 사용한 in vitro 항비듬 효과 측정법은 시료나 배지의 탁도에 크게영향을 받지 않으며, Micro-plate reader를 사용함으로써 많은 종류의 시료에 대한 결과를 마르고 정확하게 얻을 수 있다는 장점을 가지고 있으므로, 향후 효율적인 in-vitro 항비듬 평가 방법으로 활용이 기대된다.

TM-25659-Induced Activation of FGF21 Level Decreases Insulin Resistance and Inflammation in Skeletal Muscle via GCN2 Pathways

  • The TAZ activator 2-butyl-5-methyl-6-(pyridine-3-yl)-3-[2'-(1H-tetrazole-5-yl)-biphenyl-4-ylmethyl]-3H-imidazo[4,5-b]pyridine] (TM-25659) inhibits adipocyte differentiation by interacting with peroxisome proliferator-activated receptor gamma. 1 TM-25659 was previously shown to decrease weight gain in a high fat (HF) diet-induced obesity (DIO) mouse model. However, the fundamental mechanisms underlying the effects of TM-25659 remain unknown. Therefore, we investigated the effects of TM-25659 on skeletal muscle functions in C2 myotubes and C57BL/6J mice. We studied the molecular mechanisms underlying the contribution of TM-25659 to palmitate (PA)-induced insulin resistance in C2 myotubes. TM-25659 improved PA-induced insulin resistance and inflammation in C2 myotubes. In addition, TM-25659 increased FGF21 mRNA expression, protein levels, and FGF21 secretion in C2 myotubes via activation of GCN2 pathways (GCN2-$phosphoelF2{\alpha}$-ATF4 and FGF21). This beneficial effect of TM-25659 was diminished by FGF21 siRNA. C57BL/6J mice were fed a HF diet for 30 weeks. The HF-diet group was randomly divided into two groups for the next 14 days: the HF-diet and HF-diet + TM-25659 groups. The HF diet + TM-25659-treated mice showed improvements in their fasting blood glucose levels, insulin sensitivity, insulin-stimulated Akt phosphorylation, and inflammation, but neither body weight nor food intake was affected. The HF diet + TM-25659-treated mice also exhibited increased expression of both FGF21 mRNA and protein. These data indicate that TM-25659 may be beneficial for treating insulin resistance by inducing FGF21 in models of PA-induced insulin resistance and HF diet-induced insulin resistance.

Clinical implementation of PerFRACTIONTM for pre-treatment patient-specific quality assurance

  • This study is to assess the clinical use of commercial PerFRACTIONTM for patient-specific quality assurance of volumetric-modulated arc therapy. Forty-six pretreatment verification plans for patients treated using a TrueBeam STx linear accelerator for lesions in various treatment sites such as brain, head and neck (H&N), prostate, and lung were included in this study. All pretreatment verification plans were generated using the Eclipse treatment planning system (TPS). Dose distributions obtained from electronic portal imaging device (EPID), ArcCHECKTM, and two-dimensional (2D)/three-dimensional (3D) PerFRACTIONTM were then compared with the dose distribution calculated from the Eclipse TPS. In addition, the correlation between the plan complexity (the modulation complexity score and the leaf travel modulation complexity score) and the gamma passing rates (GPRs) of each quality assurance (QA) system was evaluated by calculating Spearman's rank correlation coefficient (rs) with the corresponding p-values. The gamma passing rates of 46 patients analyzed with the 2D/3D PerFRACTIONTM using the 2%/2 mm and 3%/3 mm criteria showed almost similar trends to those analyzed with the Portal dose imaging prediction (PDIP) and ArcCHECKTM except for those analyzed with ArcCHECKTM using the 2%/2 mm criterion. Most of weak or moderate correlations between GPRs and plan complexity were observed for all QA systems. The trend of mean rs between GPRs using PDIP and 2D/3D PerFRACTIONTM for both criteria and plan complexity indices as in the GPRs analysis was significantly similar for brain, prostate, and lung cases with lower complexity compared to H&N case. Furthermore, the trend of mean rs for 2D/3D PerFRACTIONTM for H&N case with high complexity was similar to that of ArcCHECKTM and slightly lower correlation was observed than that of PDIP. This work showed that the performance of 2D/3D PerFRACTIONTM for pretreatment patient-specific QA was almost comparable to that of PDIP, although there was small difference from ArcCHECKTM for some cases. Thus, we found that the PerFRACTIONTM is a suitable QA system for pretreatment patient-specific QA in a variety of treatment sites.

Functional Identification of an 8-Oxoguanine Specific Endonuclease from Thermotoga maritima

  • To date, no 8-oxoguanine-specific endonuclease-coding gene has been identified in Thermotoga maritima of the order Thermotogales, although its entire genome has been deciphered. However, the hypothetical protein Tm1821 from T. maritima, has a helix-hairpin-helix motif that is considered to be important for DNA binding and catalytic activity. Here, Tm1821 was overexpressed in Escherichia coli and purified using Ni-NTA affinity chromatography, protease digestion, and gel filtration. Tm1821 protein was found to efficiently cleave an oligonucleotide duplex containing 8-oxoguanine, but Tm1821 had little effect on other substrates containing modified bases. Moreover, Tm1821 strongly preferred DNA duplexes containing an 8-oxoguanine:C pair among oligonucleotide duplexes containing 8-oxoguanine paired with four different bases (A, C, G, or T). Furthermore, Tm1821 showed AP lyase activity and Schiff base formation with 8-oxoguanine in the presence of $NaBH_4$, which suggests that it is a bifunctional DNA glycosylase. Tm1821 protein shares unique conserved amino acids and substrate specificity with an 8-oxoguanine DNA glycosylase from the hyperthermophilic archaeon. Thus, the DNA recognition and catalytic mechanisms of Tm1821 protein are likely to be similar to archaeal repair protein, although T. maritima is an eubacterium.

다이오드 레이저로 여기되는 Tm:YAG 레이저의 출력 특성연구 (Output characteristics of diode-laser-pumped Tm:YAG laser)

  • 다이오드 레이저 뒷면 여기 방식을 이용하여 발진 파장이 $2.02\mu\textrm{m}$인 중적외선 Tm:YAG 레이저를 제작하고, 출력 특성을 조사하였다. 레이저의 출력은 Tm:YAG 결정의 온도에 민감하게 변화하였으며, 레이저의 출력경이 곡면경인 경우에 평면적인 격우보다 더 큰 출력을 얻을 수 있었다. 곡면경을 사용하고 Tm:YAG 결정을 지지하는 구리 지지대의 온도를 $6^{\circ}C$로 낮추었을 때, 최고 1.2W의 출력을 얻을 수 있었다. 이 때, 문턱 여기 출력은 약 1.6W이었고, 기울기 효율(slope efficiency) 및 광학 효율(optical to optical efficiency)은 각각 29%와 20%이었다.

Thyme(Thymus vulgaris L.)과 Tarragon(Artemisia dracunculus L.) 물추출 첨가가 김치의 품질과 보존에 미치는 영향 (The Effects of Water Extracts from Thyme(Thymus vulgaris L.) and Tarragon (Artemisia dracuncculus L.) on Shelf-life and Quality of Kimchi)

  • The effects of the water extracts from thyme(TM) and tarragon(TG) on shelf-life and quality of kimchi were investigated by measuring the changes in pH, acidity, number of total microorganisms, number of Lactobacillii and Leuconostoc during fermentation at 1$0^{\circ}C$, and were tested for antimicrobial activities against Lactobacillus plantarum and Leuconostoc mesenteroides. TM and TG were extracted with water, ethyl ether, ethyl acetate and ethanol. Water, ethyl ether, ethyl acetate and ethanol extracts of TM showed antimicrobial activities against Lactobacillus plantarum and did not observed against Leuconostoc mesenteroides. On the other hand, water, ethyl acetate and ethanol extracts of TG showed antimicrobial activities against Leuconostoc mesenteroides and did not observed against Lactobacillus plantarum. The decrease of pH and the increase of acidity showed lower in kimchi prepared by adding water extracts from TM than in products from TG. The number of total microorganisms were also detected less in the kimchi prepared by adding water extracts from TM. And, the properties of barkless of kimchi measured instrumentally were higher for kimchi prepared by adding water extracts from TM, also maintaining good crispness. The optimal addition amounts of both TM and TG for good overall and spicy taste of kimchi were 0.03%. The results suggested the possible use of the extracts of TM and TG can be successfully used for the quality and extension of shelf-life of kimchi.

Tm3+ 도핑에 따른 YTa7O19의 형광특성 (Luminescence Properties of YTa7O19 doped with Tm3+)

  • 본 연구에서는 고상반응에 의하여 $Tm^{3+}$를 도핑한 $YTa_7O_{19}$를 제조하고, 이들에 대한 빛발광 특성을 조사하였다. 시료의 특성평가에는 XRD, SEM 및 PL을 사용하였다. 359 nm의 여기하에서, $Tm^{3+}$를 도핑한 $YTa_7O_{19}$의 발광 스팩트럼은 455 nm에서 피크를 나타내며, 매우 좁은 발광밴드를 갖는 청색 형광체가 얻어졌다. $YTa_7O_{19}$의 최대발광강도는 $Tm^{3+}$의 농도를 0.12몰 도핑하였을 때 얻어졌으며, 이는 발광강도의 농도퀀칭과 관련이 있다.

대구 의생 김광진의 동서의학 절충 그리고 한의학 혁신 - 『의학승강법』을 중심으로 - (Kim Gwangjin's Integration of Medicine and Innovation of Traditional Medicine - Centering on the principle of Up and Down in Medicine -)

  • Objectives : In this paper, the main objective is to analyze and evaluate Kim Gwangjin(金光鎭, 1885-1940)'s academical achievements of integration of eastern with western medicine and innovation of traditional medicine(TM, 漢醫). Methods : This paper is composed of two parts. First, analyzing Kim's medical book, the principle of Up and Down in Medicine(醫學升降法) by the way of text analysis and interpretation faithfully. Second, adding historical perspectives about his achievements. Results : Kim opened a clinic and practiced TM in Daegu, a city of Youngnam(嶺南) area of Korea, in 1924, and published the book in 1936. While Kim acknowledged the scientific results by western medicine and criticized the typical explanation of five viscera or qi circuits of TM, he clarified the treatment range for TM and provided his own interpretations of pathophysiology through the ups and downs of qi, without conflicting between western medicine and TM. Moreover, he critically follows the TM discourses of the most noted practitioner, Lee Gyoojoon(李圭晙), in the Youngnam area of the time. Conclusions : In the 1930s, although western medicine has become the new core orientation and has forced out TM to the districts, within the abyss of the districts, TM still maintained its strong presence.

어깨가슴운동과 등뼈가동운동이 어깨돌림근띠 파열을 가진 환자들의 통증과 기능장애에 미치는 영향 (The Effect of Scapulothoracic Exercise and Thoracic Mobilization on Shoulder Pain and Function in Shoulder Partial Thickness Rotator Cuff Tear)

  • Purpose : The purpose of this study was to determine the effect of scapulothoracic exercise and thoracic mobilization on shoulder pain and function in individulas with shoulder partial- thickness rotator cuff tear. Methods : Forty-five subjects were recruited and randomly divided into 3 groups of 15 subjects each : Scapulothoracic exercise with Thoracic mobilization (STwTM), Scapulothoracic exercise (ST), and Thoracic mobilization (TM) groups. All subjects were assessed for pain and disability of shoulder, thoracic angle base on the Shoulder Pain and Disability Index (SPADI) and Thoracic kyphosis assessment (TKA) using Baseline Bubble Inclinometer. All groups performed 50 minutes of the intervention twice a week for a total of six weeks. These tests were evaluated Three times : prior to the start of the intervention, after three weeks and, after six weeks had passed. Results : The SPADI score showed that pain and dysfunction in the STwTM group decreased compared to those in the other two groups. The TKA score showed a decrease over time in the ST, TM, and STwTM groups, in increasing order. However, the TKA score decreased significantly in the STwTM group compared to that in the oher two groups. Conclusion : ST and TM are effective in improving pain and dysfunction in patients. However, STwTM may be more effective for patients.
