Antimutagenic and Antioxidative Effects of Methanol Extract of Pine Pollen (송화 메탄올 추출물의 항산화적 항돌연변이 효과)

  • This study was performed to investigate the antimutagenic and antioxidative activities of pine pollen with respect to the microbial mutation induced by various mutagens such as 1-NP, daunomycin, 2-NF, MNNG, NaN$_3$, 4NQO, 4-NOPD, AFB$_1$, Trp-P-1, 2-AF and oxidative mutagens such as t-BOOH, H$_2$O$_2$. Pine pollen, originally extracted with hexane, was reextracted with 70% methanol. The results obtained using the methanol extract, in terms of the antimutagenicity observed in relation to ten kinds of mutagens, showed that it exhibited 17.8, 82.2 and 80.9% inhibitory effects against daunomycin, AFB$_1$, and Trp-P-1, respectively, in Salmonella. typhimurium TA98 and a 72.3% inhibitory effect against AFB$_1$in S. tyPhimurium TA100. In terms of the antimutagenicity exhibited in relation to t-BOOH, a 72.3% inhibitory effect was observed, but no antimutagenicity was observed in relation to the other mutagens and strains. The methanol extract was further fractionated by chloroform, ethyl acetate, n-butanol. In S. typhimurium TA98, the chloroform(150 $\mu\textrm{g}$/plate) fraction showed strong antimutagenic effects of 55.6%, 93.7% and 93.5%, while the ethyl acetate(100 $\mu\textrm{g}$/plate) fraction showed 11.4%, 74.3% and 85.2% in relation to the mutagenicity induced by daunomycin, AFB$_1$and Trp-P-1, respectively. In S. typhimurium TA100, the chloroform and ethyl acetate fractions showed antimutagenic effects of 95.1% and 62.5%, respectively, on the mutagenicity induced by AFB$_1$. In S. typhimurium TA102, the chloroform fraction showed an antimutagenic effect of 93.6% on the mutagenicity induced by t-BOOH.

  • The intracellular invertase gene of alkalophilic and thermophilic Bacillus sp. TA-11 which was isolated from compost was cloned and expressed in E. coil HB101 using pUC19 as a vector. The invertase of the recombinant E. coli (pYC 17) was maximally produced when it was incubated at 37$^{\circ}C$ for 9 h in a SY medium containing 0.25% sucrose, 0.5% yeast extract, 0.1% each of $K_2HPO_4$ and $KH_2PO_4$, with an initial pH of 8.0. The final enzyme activity under the above condition was 47.7 U per ml of cell-free extract.

Production of $\beta$-Galactosidase from Alkalophilic, Thermophilic Baillus sp. TA-11 (호알칼리성, 고온성 Bacillus sp. TA-11에 의한 $\beta$-Galactosidase의 생산)

  • The conditions for ${\beta}$-galactosidase production from alkalophilic, thermophilic Bacillus sp. TA-11 were investigated. The maximal enzyme production was obtained when the strain was cultured at $50^{\circ}C$ for 5 days with fed-batch culture in the optimal medium containing 1.5% lactose, 0.6% yeast extract 0.15% $K_2HP0_4$and initial pH 9.5, and then final enzyme activity under the above conditions was 5200 unit/ml of cell free extract.

  • Alcoholic extract of Terminalia arjuna [TA] was evaluated far its hepatoprotective activity against carbon tetrachloride (CCl$_4$)-induced hepatic damage in rats. The hepatoprotective activity of TA was evaluated by measuring levels of serum marker enzymes like serum glutamate oxaloacetate transaminase (SGOT), serum glutamate pyruvate transaminase (SGPT), serum alkaline phosphatase (SALP). The serum levels of total proteins(TP), total albumins (TAL) and bilirubin (BILN) were also estimated. The histological studies were also carried out to support the above parameters. Silymarin (SM) was used as standard drug. Administration of TA (250 and 500 mg/kg/po) markedly prevented CCl$_4$-induced elevation of levels of SGOT, SGPT, SALP, TP, TAL and BILN. These biochemical observations were supplemented by histopathological examination of liver sections. Alcoholic extract of TA also shown significant in-vitro free radical scavenging activity against 1, 1-diphenyl-2-picryl hydrazyl (DPPH) and nitric oxide (NO) radicals. Thus, the present study provides a scientific rationale for the traditional use of this plant in the management of liver diseases.

Isolation and Characterization of Antitumor Agents from Xanthium strumarium L. (한국산 도꼬마리로부터 항암물질의 분리 및 특성)

  • In a mutagenicity test using the Salmonella typhimurium TA98 and TA100, the Xanthium strumarium L. extracts had not a mutagenicity. The extracts were assayed that antioxidative effect using a colony formation assay. The extracts showed protective effects against the cytotoxicity of H$_2$O$_2$ and increased the immunity induced by TNF and IL-1${\beta}$. The modulating effect of Xanthium strumarium L. extract on the induction of carcinogenesis by N-methyl-N'-nitro-N-nitrosoguanidin (MNNG), was investigated in Wistar rats. The GSH content was found to be reduced by MNNG treatment, but increased on adding extract. In addition the Xanthium strumarium L. extract increased p53 expression versus MNNG alone.

  • Lactobacillus plantarum KLAB21 isolated from kimchi has been shown to produce antimutagenic subtance(s) into the culture medium using Salmonella typhimurium TA100 and S. typhimurium TA98 (Rhee and Park, Korean J. Appl. Microbiol.. Biotechnol., 1999, in press). In this study, the effects of culture conditions were investigated to maximize the production of antimutagenic substance(s) against 4-nitro-O-phenylenediamine(NPD) by the strain KLAB21. Glucose(255) as a carbon source and yeast extract or bactopeptone(1%) as a nitrogen source showed the highest production of the antimutagenic substance(s). Optimal initial pH of the culture medium, culture temperature and shaking speed for the antimutagenic substance(s) production were pH 7.0, 37$^{\circ}C$ and 150rpm, respectively. Under the optimal conditions, the antimutagenic activity of L. plantarum KLAB21 culture supernatant against NPD on Salmonella typhimurium TA100 and S. typhimurium TA98 were 73.95% and 59.74%, respectively.

  • To investigate the antimutagenic effect of Platycodon grandiflorum DC, methanol extract of Platycodon grandiflorum DC was investigated. In Ames test, the methanol extract showed inhibitory effects of 80-90% on the mutagenicity induced by indirect mutagen of IQ(2-amino-3-methylimidazo[4,5-f]quinoline) and direct mutatgen of MNNG(N-methyl-N'-nitro-N-nitrosoguanidine) in Salmonella typhimurium TA 98 and TA 100. And then the methanol extract was further fractionated. Among the solvent extracted fractions from the methanol extract, the ethyl acetate fraction and butanol fraction exhibited the greatest antimutagenic effect suppressing the mutagenicity IQ and MNNG with inhibition rate of 99% and 98%.

Antimutagenic and Cytotoxic Effects of Kochujang Extracts Added Deep Sea Water Salt and Sea Tangle (해양심층수염 및 다시마분말 첨가 고추장추출물의 항돌연변이성 및 암세포 성장억제효과)

  • This study was performed to observe the antimutagenic and cytotoxic activities of methanol extract of kochujang added with sea tangle and deep sea water salts (SDK) and kochujang added with sea tangle (SK) using the Ames test and SRB assay, respectively. The direct antimutagenic effect of SDK and SK methanol extracts were examined by Ames test using Salmonella Typhimurium TA98 and TA100. In the Ames test, methanol extract of SDK and SK alone did not exhibit mutagenicity and most of the samples showed high antimutagenic effects against mutation induced by N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) and 4-nitroquinoline-1-oxide (4NQO). Methanol extract of SDK ($200{\mu}g$/plate) showed approximately 71.4% inhibitory effect on the mutagenesis induced by 4NQO against TA98 strain; whereas 56.1% and 83.6% inhibitions were observed on the mutagenensis induced by 4NQO and MNNG against TA100 strain. The cytotoxic effects of SDK and SK increased with increasing sample concentration against human cervical adenocarcinoma (HeLa), human hepatocellular carcinoma (Hep3B), human breast adenocarcinoma (MCF-7), human stomach adenocarcinoma (AGS), and human lung carcinoma (A549). The SDK at the concentration of 1 mg/ml showed cytotoxicities of 61.5%, 61.3%, 51.4%, 57.9% and 77.7% against HeLa, Hep3B, MCF-7, AGS and A549, respectively. In contrast 1 mg/ml treatment of SDK and SK methanol extract had only $2{\sim}38%$ cytotoxicity on human transformed primary embryonal kidney cell (293).

Antimutagenic Effect of Extract of Eleutherococcus senticosus Maxim (가시오갈피 추출물의 항돌연변이 효과)

  • Antimutagenic effect of Eleutherococcus senticosus Maxim. on the mutagenicity induced by 2-AF and Trp-P-1 was studied using the Ames test with Salmonella typhimurium TA98 and TA100. In S. typhimurium TA98, the methanol extract $(500\;{\mu}g/plate)$ of root, stem, and leaf of E. senticosus showed inhibitory effects of 72.8, 70.0, and 78.7% on the mutagenicity induced by 2-AF, and 69.2, 64.9, and 59.4% by Trp-P-1, respectively, whereas none was observed in S. typhimurium TA100. These results suggest that the methanol extract of E. senticosus inhibits a frame shift mutation. And then the methanol extract further fractionated by chloroform, butanol, and water. The chloroform fractions of root, stem, and leaf showed strong antimutagenic effects induced by 2-AF and Trp-P-1 in S. typhimurium TA98, whereas none was observed in the butanol and aqueous fractions. The chloroform fractions of root, stem, and leaf showed antimutagenic effects of $13{\sim]92%$ in a dose-dependent manner.

Effect of the Biological Activities of Ethanol Extract from Korean Traditional Kochujang Added with Sea Tangle (Laminaria longissima) (다시마 분말을 첨가한 전통고추장 에탄올 추출물의 생리활성 효과)

  • This study was carried out to investigate antimutagenic and anticancer effects of Korean traditional Kochujang added with sea tangle (Laminuia longissima). Most of the mineral content in Kochujang added with sea tangle was increased when compared to traditional Kochujang. In the Ames test, the inhibition rate of ethanol extract (200 $\mu\textrm{g}$/plate) of Kochujang added with 5% sea tangle in the S. typimurium TA100 strain showed 87.2% against the mutagenesis induced by MNNG. And also, the inhibition rate of ethanol extract (200 $\mu\textrm{g}$/plate) of Kochujang added with 5% sea tangle in the S. typhimurium TA98 and TA100 strains was 62.9% and 71.6%, respectively, against the mutagenesis induced by 4NQO. The suppression under the same condition against B($\alpha$)P and Trp-P-1 in the TA98 and TA100 strains showed 70.2% and 80.8%, and 62.9% and 73.1%, respectively. In the anticancer effects, the treatment of 1.0 mg/mL Kochujang added with 5% sea tangle showed strong cytotoxicity of 78.6% and 79.4% against KATOⅢ and HepG2, respectively.
