Tuberculosis (TB) is infectious disease caused by Mycobacterium tuberculosis (MTB) infection. It is known that not only the property of microorganism but also the genetic susceptibility of infected patients is controlled. Interleukin 2 (IL-2) is a cytokine belonging to type 1 T helper (Th1) activity. In addition, IL-2, when infected with MTB, binds IL-2 receptor and promotes T cell replication and is involved in granuloma formation. The aim of this study was to investigate the genetic polymorphisms of the IL-2 receptor gene in tuberculosis patients and normal individuals. We analyzed 22 SNPs in three genes using the genotype data of 443 tuberculosis cases and 3,228 healthy controls from the Korea Association Resource for their correlation with tuberculosis case. IL2RA, IL2RB, and IL2RG genes were genotyped of 16, 4, and 2 SNPs, respectively. Among three genes, only IL2RA gene polymorphisms showed statistically significant association with tuberculosis case. 6 SNPs with high significance were identified in the IL2RA gene. In addition, the linkage disequilibrium (LD) structure of IL2RA gene was confirmed. SNP imputation of IL2RA gene was performed, it was confirmed that more SNPs were significant between case and control. If we look at the results of IL2RA gene analysis above, we can see that genetic polymorphism in the gene expressing $IL-2R{\alpha}$ will regulate the expression level of $IL-2R{\alpha}$, and the change in the immune system involved in $IL-2R{\alpha}$. In this study, genetic polymorphism that may affect host immunity suggests that susceptibility to tuberculosis may be controlled.

This study was performed to investigate immune changes by comparing the proportion and function of immune cells in the blood under high-temperature period and convalescence temperature period in Holstein dairy cows. The experiment was conducted using Holstein dairy cows of five animals per group (60 ± 20 months old, 175 ± 78 non-day) from the National Institute of Animal Science at high-temperature period (THI: 76 ± 1.2) and convalescence temperature period (THI: 66 ± 1.3). Complete blood count results showed no change in the number of immune cells between groups. In the analysis using Flow Cytometry of PBMCs, no significant differences were observed among B cells, Helper T cells, cytotoxic T cells, and γδ T cells between groups. However, there was an increase in Th17 cells producing IL-17a, while Th1 cells decreased during the convalescence temperature period. The results of gene expression analysis using qRT-PCR in PBMCs revealed an increase in IL-10 during the convalescence temperature period, while a decrease in HSP70 and HSP90 was observed. In conclusion, the increased expression of IL-10 and the decrease in HSP expression suggest the possibility of a weak recovery from heat stress. However, the lack of observed changes in B cells, T cells, and other immune cells indicates incomplete recovery from heat stress during the convalescence temperature period.
Objective : This study was undertaken to investigate effects of Opuntia ficus-indica (OP)extract on an asthma murine model. Methods : The total number of cells and eosinophils in BALF and the infiltration of inflammatory cells into lung tissues were determined by hematoxylin and eosin staining. Results : The number of OVA-induced total cells and eosinophils, a phenomenon of asthma, were decreased by treatment of the animals with OP extract (200 mg/kg) (respectively, p < 0.001 and p<0.01). Furthermore, we showed that OP extract reduced the increased immune cell infiltration induced by ovalbumin (p < 0.01). The levels of interleukin (IL)-4 in BAL fluid were measured by enzyme-linked immunosorbent assay, because the eosinophilia is associated with a T helper (Th) 2 response including IL-4. The level of OVA-induced IL-4 was decreased by OP extract in BAL fluid (p < 0.05). We investigated whether OP extract reduced nitrite (NO) production on lipopolysaccharide (LPS)-stimulated RAW 264.7 sells, because asthma is an inflammatory disease. The level of LPS-induced NO production was decreased by OP extract (50, 100 and 200 mg/ml) on RAW 264.7 cells (respectively, p < 0.05, p<0.01 and p<0.05). Conclusions : Our results indicate that OP extract has an inhibitory effect on lung eosinophilia of asthma and suggest that OP extract is a therapeutic candidate in the treatment of inflammatory disease, including asthma.
Zinc plays an important role in immunobiological responses, while excessive zinc attenuates immune functions in a dose-dependent manner. Zinc excess has been reported to increase levels of plasma prostaglandin E$_2$ (PGE$_2$), which is known to inhibit production of Th (helper T) 1-associated cytokines and to induce inflammatory responses. Thus, this study was investigated the effects of indomethacin, a potent inhibitor of PGE$_2$ synthesis, on the proinflammatory cytokine and lymphokine production in ICR mice exposed to excessive zinc. Indomethacin at doses of 5 mg/kg was administered i.p. 30 minutes before zinc chloride (Zn) 30 mg/kg orally daily for 10 days. Excessive Zn remarkedly increased tumor necrosis factor (TNF)-$\alpha$ and interleukin (IL)-1$\beta$ levels in both serum and splenic supernatants compared with those in controls, while indomethacin significantly reduced the excessive Zn-induced levels of IL-1$\beta$. In serum, excessive Zn significantly decreased the levels of IL-2 and interferon (IFN)-${\gamma}$ compared with those in controls, whereas indomethacin significantly enhanced the excessive Zn-decreased levels of IFN-${\gamma}$ but did not affect the Zn-decreased levels of serum IL-2. In splenic supernatants, All of excessive Zn, indomethacin, and combination of Zn and indomethacin significantly enhanced IL-2 levels compared with those in controls, but indomethacin didn't affect the Zn-induced production of IL-2. These data, therefore, suggest that indomethacin significantly attenuated the in vivo and ex vivo IL-1$\beta$ production increased by excessive zinc and remarkedly enhanced the in vivo excessive zinc-suppressed production of IFN-${\gamma}$ but not IL-2.
This study was undertaken to investigate the effect of indomethacin on the distribution of Langerhans cells and T-lymphocytes related with immune response of 4-Nitroquinoline-1-oxide induced carcinogenesis at the palate and tongue of albino rat. 54 Sprague-Dawley strain 10 weeks old albino rats, about 150gm weighted, divided into a normal group of 6 rats without treatment, a control group of 12 rats given indomethacin, a carcinogenesis group of 18 whose palatal mucosa were appiled with 4-Nitroquinoline-1-oxide three times a week, and experimental group of 18 rats were treated with indomethacin and whose palatal mucosa were applied 4-Nitroquinoline-1-oxide. All these 54 rats were subjected to be observed as being ATPase stained specimens, specimens for the observation of light and electron microscope, and T-lymphocyte stained specimens. The obtained results were summarized as follows; 1. In carcinogenesis group, proliferation of epithelial layer and rete peg were observed early period of the experiment and showed parakeratosis, individual cell keratinization, acanthosis, and lymphocyte infiltration from 13th week of the experiment on lightmicroscopically, while experimental group showed less reaction than that of carcinagenesis group. 2. The number of Langerhans cells in normal group rarely changed until 21st week of the experiment, while the Langerhans cells increased markedly from 3rd week of the experiment in control group. 3. The number of Langerhans cells were decreased markedly and persistantly until 21st week of the experiment both in carcinogenesis and experimental groups. 4. Appearance of the T-helper cells and T-suppressor cells were minimal and irregullar in number both in normal and control groups. Thus it is assumed that administration of indomethacin and distribution of Langerhans cells showed close relation. 5. In carcinogenesis and experimental groups, the number of the T-helper cells was apparently inereased than that of the T-suppressor cells, but increasing pattern in experimental group was less than in carcinogenesis group. These cells increased most in the 21st week, decreased from the 23rd week and the appearance of these cells were irregular in general throughout the experiment.
Asthma is a chronic inflammatory airway disease. There are many factors including genetic and environmental factors that influence asthma. The mitogen-activated protein kinase (MAPK) pathway is involved in maintaining the T helper cells 1 and 2 (Th1/Th2) balance and plays an important role in the development of asthma. In this study, the correlation between the NDFIP2 gene that regulates the MAPK pathway and asthma was analyzed. The genetic polymorphism of the NDFIP2 gene was analyzed between 193 asthma patients and 3,228 healthy controls in Korea. As a result, 4 single nucleotide polymorphisms (SNPs) showed a significant correlation (P<0.05) and high relative risk with asthma. Among them, rs2783122 of NDFIP2 showed a statistically significant association with asthma (P-value=9.76×10-6, odds ratio (OR)=1.67, 95% confidence interval (CI)=1.33~2.10). In the SNP imputation on the NDFIP2, 16 SNPs were discovered, and all of them showed significant correlation with asthma and high odds ratio. The genotype-based mRNA expression analysis revealed that the group of minor alleles of rs1408049 showed increased mRNA expression. Increased NDFIP2 expression causes the activation of the MAPK pathway, and this may influence the development of asthma. In conclusion, the polymorphisms of NDFIP2 are associated with asthma development and this can provide the basis for new guidelines for the management of asthma in the Korean population.
Background: The present study examines a hypothesis that short allergen-derived peptides may shift an Aspergillus fumigatus (Afu-) specific TH2 response towards a protective TH1. Five overlapping peptides (P1-P5) derived from Asp f1, a major allergen/antigen of Afu, were evaluated for prophylactic or therapeutic efficacy in BALB/c mice. Methods: To evaluate the prophylactic efficacy, peptides were intranasally administered to naive mice and challenged with Afu-allergens/antigens. For evaluation of therapeutic efficacy, the mice were sensitized with Afu-allergens/antigens followed by intranasal administration of peptides. The groups were compared for the levels of Afu-specific antibodies in sera and splenic cytokines evaluated by ELISA. Eosinophil peroxidase activity was examined in the lung cell suspensions and lung inflammation was assessed by histopathogy. Results: Peptides P1-, P2- and P3 decreased Afu-specific IgE (84.5~98.9%) and IgG antibodies (45.7~71.6%) in comparison with Afu-sensitized mice prophylactically. P1- and P2-treated ABPA mice showed decline in Afu-specific IgE (76.4~88%) and IgG antibodies (15~54%). Increased IgG2a/IgG1 and IFN-${\gamma}$/IL-4 ratios were observed. P1-P3 prophylactically and P1 therapeutically decreased IL-5 levels and eosinophil peroxidase activity. P1 decreased inflammatory cells' infiltration in lung tissue comparable to non-challenged control. Conclusion: Asp f1-derived peptide P1, prophylactically and therapeutically administered to Balb/c mice, is effective in regulating allergic response to allergens/antigens of Afu, and may be explored for immunotherapy of allergic aspergillosis in humans.

Cordyceps sinensis has been clinically used for the treatment of recovering fatigue, promoting the production of body fluid and qi, resolving sputum and arresting cough. Recent studies showed that Cordyceps sinensis produced immune-modulatory, anticarcinogenic and antioxidative effects. But, there are lack of studies regarding the effects of Cordyceps sinensis on the asthma. So, this study was performed to investigate the oral administrated effects of this herb against the disease. Asthma was induced to Balb/c mouse by i.p. injection and aerosol immunization with ovalbumin, and the change of the eosinophil number in the bronchoalveolar avage fluid(BALF) was observed. Concentrations of Interleukine-4(IL-4), Interleukine-5(IL-5) in BALF and splenocyte were assessed by ELISA, ImmunoglobinG(IgG) and ImmunoglobinE(IgE) from serum were calculated by same method. We found that the effects of Cordyceps sinensis in asthma mouse was implicated in reductions of IL-4, IL-S released from type2 T helper (Th2) cell, and decreases of IgE from plasma cell. These findings suggest that Cordyceps sinensis can produce anti-asthmatic effect, which may play a role in allergen-induced asthma therapy.

The effect of black garlic extract on the activation of spleen cells from a C57BL6 mouse was investigated to examine immune activities of of fermented black garlic containing a variety of bioactive substances. xtract obtained from the concentration of commercial Namhae black garlic was used for the analysis of immune activities. Treatment with the extract increased the expression of interleukin-2 (IL-2) cytokine. The simultaneous administration of the extract plus lipopolysaccharide (LPS) increased the expression of IL-2, tumor necrosis factor (TNF)-${\alpha}$, and interferon (IFN)-${\gamma}$ compared with that of a control group. This result suggests that cellular immunity can be induced by macrophages, resulting in the expression of T lymphocytes and T helper type 1 (Th1) cells. In addition, treatment with the extract increased the late response of IL-6 cytokines, and the extract plus LPS augmented the expression of IL-4 and IL-6 compared with that of an LPS-treated group. Meanwhile, the extract plus LPS decreased the late response of IL-10, suggesting that humoral immunity can be activated by stimulating B lymphocytes, suppressing cellular immunity, and effectively modulating the conversion into humoral immune responses. These findings demonstrate that the black garlic extract activates Th1 and Th2 cells by stimulating T lymphocytes in mouse spleen cells and leads to immunomodulation by activating cellular and humoral immune responses of the immune system.

This study aims to evaluate the anti-inflammatory effect of Coptidis Rhizoma (CR) extract for atopic dermatitis through maintaining skin barrier and regulating Th2 cell differentiation. We divided NC/Nga mice into 3 groups as follows; atopy-like dermatitis induced group with CR treatment (CT, n=10), no treatment group(Ctrl), atopy-like dermatitis elicited group(AE). Atopy-like dermatitis was induced to NC/Nga mice by sensitizing with dermatophagoides farinae(DfE) on 7, 8, 9, 11, 12, and 13th week. After inducing atopic dermatitis, CR extract was administered 20 mg/kg daily for the experimental duration to the CT group. We measured the integrity of lipid layers in the epidermis and Th2 differentiation through immunohistochemical staining against filaggrin, loricrin, IL-4, and IL-13. We also measured the distribution of subcutaneous collagen fibers by the Masson's trichrome staining. Administration of CR significantly inhibited the reduction of lipid layers in the skin that caused atopy. The expression of IL-4, IL-13, each of which is a cytokine secreted by T helper type 2 (Th2) cells, was markedly suppressed in the CT group as compared with AE group (p<0.05). CR treatment also decreased the expression of iNOS, $p-I{\kappa}B$. Atopic dermatitis induced dermatological damage to skin, such as hyperplasia of epithelium, and capillary proliferation was significantly reduced by CR administration. CR effectively inhibited the thinning of the skin barrier and inflammatory responses in atopic dermatitis-induced mice. In particular, it showed anti-inflammatory effects by reducing the expression of IL-4 and IL-13, Th2 cell cytokines, which play a crucial role in development of atopic dermatitis. Therefore, CR can be a good candidate to ameliorate and treat atopic dermatitis.
