Enzymatic synthesis of structured lipids containing conjugated linolenic acids extracted from pomegranate seed oil and their physicochemical characteristics (석류 종자유로부터 얻어진 Conjugated Linolenic Acid를 함유한 기능성 고체지의 효소적 합성 및 이화학적 특성 연구)

  • Lipozyme TL IM-catalyzed esterification was carried out to produce functional hard fat (structured lipid, SL) using palm stearin (PS) and hydrolysate of pomegranate seed oil (HPSO) of 1:6 molar ratio. HPSO contained conjugated linolenic acid (CLnA, about 80%). The reaction was performed at non-solvent system and solvent (n-hexane) system using Lipozyme TL IM (10% of total substrates, w/w) for 12, 24, and 72 hr in a shaking water bath ($55^{\circ}C$ and 185 rpm), respectively. SL synthesized in non-solvent system (NH-SL) and SL synthesized in n-hexane system (H-SL) were refined after deacidification, respectively. Their physicochemical properties were compared to obtain desirable functional hard fat. The content of CLnA in NH-SL increased from 34.38% to 40.63% with increasing reaction time. Similar results also observed in H-SL resulting in 36.81~45.83% of CLnA. In triacylglycerol (TAG) composition, the main molecules of LnLnLn (Ln=linolenic acid, PN=36) and the LnLnP (P=palmitic acid, PN=40) were newly synthesized in NH-SL and H-SL with increasing reaction time. After 72 hr reaction, iodine values of NH-SL (136.49) and H-SL (140.37) showed high values because of the high content of CLnA. Solid fat index (SFI) in NH-SL was higher than that in H-SL at each measured temperature. The predominant polymorphic forms of NH-SL and H-SL obtained after esterification for 72 hr were the desirable crystalline structure of the ${\beta}$' form.

Anti-aging Effects of the Extracts from Leaf. Stem, Fruit and Seed of Yew (Taxus cuspidata Sieb) by Solvent Extraction Method (용매추출법에 의한 주목의 잎, 줄기, 과실 추출물의 항 노화 효과)

  • Yew (Taxus cuspidata Sieb.) chose that grow as medicine, food, decorative plant in Korea's Kyong-Gi province surroundings. Extracts of yew extracted leaf of 250g and stems of 300g with 1,3-butylene glycol (l,3-BG), propylene glycol (PG) and water. As results, external appearance of leaf extract of yew was slightly brown clear extract. The pH was 5.3${\pm}$0.5, and specific gravity was 1.012${\pm}$0.05, and refractive index was l.375${\pm}$0.05. Also, appearance of stem's extract was slightly brown clear extract, and the pH was 5.4${\pm}$0.5, and specific gravity was 1.016${\pm}$0.05, and refractive index was 1.358${\pm}$0.05. Oil of yew separated from seeds, and extracted polysaccharide high purity from fruits. As a result, specific gravity of oil was 0.987, and obtained 40.0% of yield. Total polyphenols amount of yew extract is detected 0.563% in leaves, 0.325% in stems, whereas total tannins amount contained 0.054% and 0.037% each in leaves and stems. As effect in cosmetics, the anti-oxidative effect by DPPH method is 75.0% in leaves, and stems was 64.0%. Collagen synthesis rate was shown high activity by 54.16% in stem's extract, 33.18% in leaves' extract. Also, PPE-inhibitory activities were 13.7% and 23.5% each in leaves and stems. Anti-inflammatory effect of yew seed oil displayed superior effect of 41% than control. Polysaccharide's molecular weight that is gotten from fruits was 5${\times}$10$^4$-3${\times}$10$\^$5/ dalton, and got 20.0${\pm}$5% of yield.

A Study on the Effects of Taxus Extracts in Cosmetic Indurstry (화장품산업에서 주목추출물의 효능에 관한 연구)

  • Taxus cuspidata Sieb selected cultivation as drug, food and decorative plant in Kyong-gi province in Korea. As a manufacturing method, there were extracted from 250g of dried-leaf and 300g of dried-stem with each 200g of BG, PG and water (to 100) mixing for 72 hour at 50$\pm$5$\^{C}$ and then they were filtered by 400-mesh filter. Appearance of extract of leaves was slight brown, pH=5.3$\pm$0.5, gravity was 1.012$\pm$0.05, and a reflective index was 1.375$\pm$0.05. And appearance of extract of stems was slightly dark brown, pH=5.4$\pm$0.5, gravity was 1.016$\pm$0.05, and a reflective index was 1.358$\pm$0.05. It was extracted oil from Taxus seed. Gravity was 0.922$\pm$0.05 and it should be obtained the 27.0$\pm$0.5% of yield. The molecular weight of polysaccharide was about 50,000 to 300,000 dalton and contained 5.0$\pm$1.2% of yield from Taxus fruit. The determinations of total polyphenols in measuring spectropotometer got 0.563% in leaves, and 0.325% in stems, whereas the quantitives of total tannins got 0.054% and 0.037%, respectively. As the effects in Cosmetics by DPPH-method, the antioxidative activities were very strong that the inhibitory ratio showed 75% in leaves and 64% in stems compared with 52% in greentea extract. These are more effective than other plant extracts. The increasing ratio of collagen synthesis rate on the activating fibroblast for extracts of Taxus cuspidata Sieb showed 54.16% (stems) and 33.18% (leaves), To improve the skin elasticity, PPE(porcine pancreatic elastase)-inhibitory activities were strongly effective as 13,7% (stems), 23.5% (leaves) and 66%(seed). Anti-inflammatory acitvity of seed oil was very the above 41% stronger than SG was 24% of anti-Inflammatory as a control sample.

Determination of $^{14}C$ in Environmental Samples Using $CO_2$ Absorption Method ($Co_2$ 흡수법에 의한 환경시료중 $^{14}C$ 정량)

  • A simple and precise method of $^{14}C$ was developed to analyze $^{14}C$ in the environment samples using a commercially available $^{14}CO_2$ absorbent and a liquid scintillation counter. An air sampler and a combustion system were developed to collect HTO and $^{14}CO_2$ in the air and the biological samples simultaneously. The collection yield of $^{14}CO_2$ by the air sampler was in the range of 73-89% . The yield of the combustion system was 97%. In preparing samples for counting, the optimum ratio of $CO_2$ absorbent to the scintillator for mixing was 1:1. No variation of the specific activity of $^{14}C$ in the counting sample was observed up to 70 days after preparation of the samples. The detection limit for$^{14}C$ was 0.025 Bq/gC, which is the level applicable to the natural level of $^{14}C$. The analytical result of $^{14}C$ obtained by the present method were within ${\pm}6%$ of the relative error from the one by the benzene synthesis. The specific activity of $^{14}C$ in the air collected at Taejon during the period of October 1996 ranged from 0.26 to 0.27 Bq/gC. The specific activity of $^{14}C$ in the air collected at 1km from the Wolsong nuclear power plant a 679 MWe PHWR, was $0.54{\pm}0.03$ Bq/gC. The ranges of specific activities of $^{14}C$ in the pine needles and the vegetations from the areas around the Wolsong nuclear power plant were 0.56-0.67 Bq/gC and 0.23-1.41 Bq/gC, respectively.

Effects of Ethylacetate Fraction of Plantago asiatica L. on Hypercholesterolemia Induced by High Cholesterol Diet in Rats (질경이(Plantago asiatica L.) 에틸아세테이트분획이 흰쥐의 고콜레스테롤혈증에 미치는 효과)

  • Plantain was extracted with ethanol and fractionated systemically with n-hexane, chloroform, ethylacetate, n-butanol and water to study inhibitory effect on cholesterol synthesis in vitro. To screen the effect, inhibitory activities on 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase obtained from Saccharomyces cerevisiae were examined using the five fractions of Plantain. The HMG-CoA reductase activity was inhibited most by ehylacetate fraction among the fractions, although the all five fractions had the effect To see the hypocholesterolemic effect of the ethylacetate fraction of Plantanin (PAE) in vivo, male Sprague-Dawley rats were received 5 types of diets for 6 weeks: normal diet group (NOR), high cholesterol diet group($1\%$ cholesterol and $0.25\%$ sodium cholate, CON), normal diet and PAE 70 mg/kg administered group(S1), high cholesterol diet and PAE 140 mg/kg administrated group(S2), and high cholesterol diet and PAE 140 mg/kg administered group(S3). Body weight gains of the CON were significantly increased compared to those of S1, S2 and S3. Activities of serum AST and ALT were tended to be increased in CON compared with NOR and reduced by the PAE administration. Concentrations of serum total cholesterol, free cholesterol, LDL-cholesterol, triglyceride and the atherogenic index were tended to be decreased in the PAE administered groups compared with the CON. HDL-cholesterol and phospholipid concentrations were significantly decreased in the CON and markedly increased by the PAE administered groups. Taten together, it is suggested that the ethylacetate fraction of Plantanin exerts antiatherosclerotic effect by reducing serum cholesterol concentrations in rats fed high cholesterol diets.

Ecophysiological Characteristics of Chenopodiaceous Plants - An Approach through Inorganic and Organic Solutes - (명아주과 식물의 생리생태학적 특성 - 무기 및 유기용질을 통한 접근 -)

  • In order to clarify the ecophysiological characteristics of Chenopodiaceae which widely distribute on saline and arid habitats, we collected 10 chenopodiaceous plant species, examined their inorganic and organic solute patterns, and confirmed several common physiological characteristics. In spite of high soil Ca/sup 2+/ contents, chenopodiaceous plants had a little water-soluble Ca within cells, but contained high contents of acid-soluble Ca particularly as a result of Ca-oxalate formation. These plant species also showed accumulation of inorganic ions such as K/sup +/, NO₃/sup -/ and Cl/sup -/, and Na/sup +/especially in saline habitats instead of K/sup +/ Meanwhile, with respect to nitrogen metabolism they retained high N contents in leaves, but showed very low amino acid contents. Additionally, they contained very little proline known to act as a cytoplasmic osmolyte. To ascertain whether this physiological characteristics in the field also can be found under controlled conditions, 7 chenopodiaceous plants (Atriplex gmelini, Corispermum stauntonii, Salicornia herbacea, Suaeda aspayagoides, Suaeda japonica, Chenopodium album var. centrorubrum, C. serotinum) were selected and cultivated under salt treatments. As well as field-grown plants, selected plant species showed similar solute pattern in growth experiment. In summary, the family of Chenopodiaceae represents the following physiological properties; high storage capacity for inorganic ions (especially alkali cations, nitrate and chloride), oxalate synthesis to maintain lower soluble Ca contents within cytoplasm, and low contents of amino acids. In addition to some characteristics mentioned above, the physiological plasticities of Chenopodiaceae which can properly regulate their ion and solute pattern according to soil conditions may enable its representative to grow in dry sand dune and salt marsh habitats.

  • The increase in the frequency of occurrence of abnormal weather could include severe rainfall, which could cause rice submergence during the ripening stage. This experiment was conducted to clarify the effects of submergence during the ripening period on yield and quality of rice. The flooding treatment was conducted at 7 and 14 days after heading. Flooding conditions were created with two conditions, flag leaf exposed and overhead flooding, and each condition was divided into two conditions according to water quality-clear and muddy. Although the yield decrease was more severe at 7 days after heading because of the decrease in the ripening ratio, the head rice ratio was more affected at 14 days after heading because of the increase in the chalky kernel ratio. The maximum quantum yield (Fv/Fm), which indicates the photosynthetic efficiency, did not differ before and after the flooding treatment until flooding continued for 4 days. In addition, stem elongation occurred because of flooding as an avoidance mechanism in japonica rice. This phenomenon was expected to decrease the supply of assimilation products to the spikelet (sink). Overall, it was suggested that additional experiments should be conducted examining the change in the starch synthesis mechanism and transfer of assimilate products resulting from submergence, for development of cultivation techniques corresponding to submergence and breeding of varieties with submergence tolerance characteristics.

Effect of Sasa Borealis and White Lotus Roots and Leaves on Insulin Action and Secretion In Vitro (In vitro에서 조릿대, 연근과 연잎이 인슐린 작용 및 분비에 미치는 영향)

  • Anti-diabetic effects of extracts and fractions of Sasa borealis (SB), white lotus roots (LR) and leaves (LL), and their mixture were determined in 3T3-L1 adipocytes and Min6 cells by investigating insulin-sensitizing activity and glucose-stimulated insulin secretion, respectively. SB, LR, LL, and mixture of SB, LR, and LL (3 : 2 : 3) were extracted using 70% ethanol, and m mixture extract was fractionated by XAD-4 column chromatography with serial mixture solvents of methanol and water. Fractional extractions were utilized for anti-diabetic effect assay. SB and LR extracts increased insulin-stimulated glucose uptake, but not as much as mixture of SB, LR, and LL. Significant insulin-sensitizing activities of 20 and 80% methanol fractions of SB, LR, and LL mixture extract were observed in 3T3-L1 adipocytes, giving 0.5 or $5\;{\mu}g/mL$ each fraction with 0.2 nM insulin to attain glucose uptake level similar to that attained by 10 nM insulin alone. Similar to pioglitazone, peroxisome proliferators-activated $receptor-{\gamma}\;(PPAR-{\gamma})$ agonist, 20 and 80% methanol fractions increased adipocytes by stimulating differentiation from fibroblasts and triglyceride synthesis. LL extract and 20, 60, and 80% methanol fractions of the mixture suppressed ${\alpha}-amylase$ activity, but did not modulate insulin secretion capacity of Min6 cells in both low and high glucose media. These data suggest 20 and 80% methanol tractions contain potential insulin sensitizers with functions similar to that of $PPAR-{\gamma}$ agonist. Crude extract of SB, LR, and LL mixture possibly improves glucose utilization by enhancing insulin-stimulated glucose uptake and inhibiting carbohydrate digestion without affecting insulin secretion in vivo.

A Land Resources Survey of the Mula Area, S. E. Spain (동남(東南)스페인 Mula지역(地域)에서의 Land Resources Survey)

  • A land resources survey in the semi-arid area, Mula in S. E. Spain, of $400km^2$ is compiled. The basic aim of the project is to investigate the intrinsic qualities of the land resources of the area by means of applying an integrated method of natural resources survey mainly concerning with analysis and synthesis of land complexes, each of them represents an area or a group of areas with similar patterns of landforms, soils and vegetation, based on a geomorphological approach. The area is characterized by a linear arrangement of relief pattern with an asymmetric homoclinal repetition of slope attitudes elongating WSW-ENE, dipping steeply on the NW sides and gently on the SE sides, which have been resulted from the post-Alpine folding of the Triassic to Cretaceous limestone, the Eocene limestone, the Oligocene sandstone and the lower Miocene limestone and marl, and the post-lower Miocene faulting, tilting and subsequent differential erosion of the Miocene sedimentary formations. An integrated body of information in geology, landforms, soils and vegetation, which are significantry interrelated as an environmental complex, has been obtained. Using this data, 26 land complexes developing on the various situations of landforms, such as folded mountain ranges, tilted tablelands, bevelled cuestas, degraded hill-lands associating with enormous foots lopes, undulating terrains and terraced or flat plains, have been differentiated, mapped and described. The soils of the area are mostly light colored calcic lithosols which have been derived dominantly from the marly parent materials and developed into remarkable slope catenas in some places depending on the relief conditions. The land uses of the area are mainly characterized by the perennially irrigated cultivation of citrus orchards along the terraced alluvial deposits fringing the Segura and Mula River, and the dry-land cereal cultivation on gentler slopes. Pioneer dry-land cultivations within the shrubs on steeper slopes are restricted to the unchannelled tributary drainage floors. The availability of water is a fundamental controlling factor for existence of native and cultivated vegetation as a whole, and a number of active processes including sheet wash and gully erosion, especially on the scarp slopes, are the other important factors to be considered in conservation and management of the land in the area.

Study on the Genetic Characteristics of Waterlogging Tolerant Pepper (Capsicum annuum L.) for Breeding Tolerant Varieties against Flooding Stress (내습성 고추 품종 육성을 위한 선발계통의 유전적 특성 구명)

  • This study was conducted to select pepper lines that were tolerant to excessive water injury among the pepper germplasm and investigate the genetic characteristics of those lines to contribute to the breeding of pepper cultivars with stable productivity in abnormal weather. Each of the tolerant and susceptible lines went through immersion treatment, and differentially expressed genes between them were analyzed. The tolerant line showed increased expression of the CA02g26670 gene, which is involved in the CONSTANS protein pathway and regulation of flowering by day length, but it exhibited decreased expressions of CA01g21450, CA01g22480, CA01g34470, CA02g00370 and CA02g00380. The susceptible line showed increased gene expressions of CA02g09720, CA02g21290, CA03g16520, CA07g 02110, and CA12g17910, which are involved in the inhibition of proteolytic enzyme activity, DNA binding, inhibition of cell wall-degrading enzyme, and inhibition of nodulin, respectively. Meanwhile the expressions of CA02g02820, CA03g21390, CA06g17700 and CA07g18230 decreased in the susceptible line, in relation to calcium-ion binding, high temperature, synthesis of phosphocholine and cold stress, respectively. The expressions of genes related to apoptosis and peroxidase increased, while that of CA02g16990, which functions as a nucleoside transporter, decreased in both the tolerant and susceptible lines. Based on the different gene expressions between the tolerant and susceptible lines, further studies are needed on breeding abiotic stress-tolerant lines.
