Effect of Follicular Fluid on Sperm Swim-up Separation with Sucrose Layer (난포액이 Sucrose 층을 이용한 정자의 Swim-up 분리에 미치는 효과)

  • To establish a system for sperm swim-up separation through sucrose layer, indiscreet sperm migration should sufficiently to block but movement of sperm shouldn't inhibit. Thus, the effects of sucrose levels in sucrose layer, incubation times and types of sucrose layer on sperm separation were examined. And the results obtained were as follows; 1. Layer of 10mM sucrose inhibited sperm swim-up migration through sucrose layer. 2. Incubation for 25 minutes without sucrose layer significantly increased sperm swim-up migration. However, incubation for 10 minutes to induce swim-up through sucrose layer significantly stimulated sperm migration and maintained sperm movement. 3. There was no significant difference between Type I and Type II in barrier effect of sucrose layer. However, sucrose layer of Type II with shorter distance of barrier was efficient for sampling. To elucidate a function of follicular fluid on sperm chemotaxis using in vitro system of sucrose layer of Type II and incubation for 10 minutes, the effects of dilution, heat treatment, and protein and lipid extracts of follicular fluid on sperm swim-up separation were examined. And the results obtained were as follows; 4. Follicular fluid stimulated sperm migration and movement, and significantly-attracted capacitated-sperm at 10% level. 5. Follicular fluid heated at 55$^{\circ}C$ for 30 minutes maintained the effect of follicular fluid stimulating sperm migration and movement. 6. Follicular protein stimulated sperm movement that was reduced by filtration of the protein. 7. Follicular lipid didn't significantly stimulate sperm migration and movement. 8. Both of follicular protein and lipid reduced the effect of follicular fluid stimulating sperm migration and movement. In conclusion, sucrose layer could be used for a barrier against indiscreet sperm migration by swim-up. And follicular fluid stimulated migration and movement of sperm and attracted capacitated-sperm through sucrose layer. Especially, heat-resistant protein of follicular fluid stimulated sperm migration.

Comparison of Sperm Motility, Recovery Rate, and Fertilization Rate using Three Different Sperm Preparation Methods: Swim-up, Percoll, Sit-Select (Swim-up, Percoll, Sil-Select를 이용한 정자처리법에 의한 정자회수율, 운동성 및 체외수정율의 비교분석)

  • It is well known that discard of seminal plasma from the semen and separation of motile sperm should be preceded before insemination for IUI or IVF. Till now, more than ten kinds of semen treatment methods have been developed. Of those, swim-up and Percoll methods have been used widely in ART laboratories as a routine semen treatment methods because of its advantages. However, there are reports that Percoll can make a genetic trouble because of its chemical structure and therefore the necessity has been arisen to substitute Percoll for other equivalent materials. This study was performed to evaluate the effects of three different sperm preparation methods (swim-up, Percoll and Sil-Select) on sperm motility, sperm recovery rate and fertilization rate. Also, the feasibility of using Sil-Select instead of Percoll in ART was evaluated. Each semen samples were divided into three fractions and motile sperm were recovered by swim-up, Percoll and Sil-Select gradient centrifugation methods. Normal and sub-normal criteria of fifteen semen samples and seventeen IVF cycles were included in these study. As results, no significant difference was found in sperm recovery rate in normal semen treated by a Swim-up, Percoll and Sil-Select method ($13.2{\times}10^6,\;17.5{\times}10^6\;and\;17.7{\times}10^6$ respectively). The initial sperm motility was 61.9% and this increased to 87.1%, 92.6% and 89.5% through Swim-up, Percoll and Sil-Select treatment, respectively. Higher motility was observed in Percoll and Sil-Select treated groups (81.5%, 79.2%, respectively) than swim-up group (66.8%) after incubation for 24hrs. In sub-normal group, sperm recovery rates were higher in Sil-Select group $(2.9{\times}10^6)$ than Percoll gradients group $(1.8{\times}10^6)$. In IVF cycles, the outcomes of fertilization using sperm treated by swim-up and Sil-Select group were similar (82.2%, 79.7% respectively). In conclusion, our results indicate that Sil-Select can be used as a substitute material for sperm preparation instead of Percoll.

  • Ongoing SWIM(System Wide Information Management) with the United States and European countries as the center is a part of the ASBU(Aviation System Block Upgrade) program improved performance of aeronautical data system in the International Civil Aviation Organization and a core technology of Integrated Aeronautical Data Management Network to elevate service through digitally aeronautical information management. Therefore, in this paper, we analyze SWIM architecture and network applied the concept of SOA(Service Oriented Architecture), and propose methods of implementation transforming applications operating established legacy aeronautical data system into integrated aeronautical data management network through adapter technology. This will allow development of middleware and application suitable for the next generation infrastructure network environment for efficient ATM(Air Traffic Management)and provide timely required information for users.

Sperm DNA fragmentation and sex chromosome aneuploidy after swim-up versus density gradient centrifugation

  • Objective: The aim of this study was to compare the efficacy of swim-up and density gradient centrifugation (DGC) for reducing the amount of sperm with fragmented DNA, sex chromosome aneuploidy, and abnormal chromatin structure. Methods: Semen samples were obtained from 18 healthy male partners who attended infertility clinics for infertility investigations and were processed with swim-up and DGC. The percentages of sperm cells with fragmented DNA measured by the sperm chromatin dispersion test, normal sex chromosomes assessed by fluorescence in situ hybridization, and abnormal chromatin structure identified by toluidine blue staining were examined. Results: The percentage of sperm cells with fragmented DNA was significantly lower in the swim-up fraction (9.7%, p= 0.001) than in the unprocessed fraction (27.0%), but not in the DGC fraction (27.8%, p= 0.098). The percentage of sperm cells with normal X or Y chromosomes was comparable in the three fractions. The percentage of sperm cells with abnormal chromatin structure significantly decreased after DGC (from 15.7% to 10.3%, p= 0.002). The swim-up method also tended to reduce the percentage of sperm cells with abnormal chromatin structure, but the difference was not significant (from 15.7% to 11.6%, p= 0.316). Conclusion: The swim-up method is superior for enriching genetically competent sperm.

Effect of swimming exercise on anti-angiogenesis of white adipose tissue in high-fat diet-fed female ovariectomized mice (고지방식이를 섭취한 난소절제 암컷 쥐의 수영운동이 백색지방조직의 항혈관신생에 미치는 효과)

  • This study investigated whether swimming exercises improves obesity through regulation of angiogenesis in white adipose tissue. Female mice with high-fat diets were divided into sham-operated group (Sham), ovariectomized group (OVX), and swim-trained ovariectomized group (OVX + Swim). Compared to the Sham, OVX increased body weight, adipose tissue mass and size of adipocyte. However, these factors (: such as body weight, adipose tissue mass and size of adipocyte) of OVX + Swim decreased compared with OVX. Compared with the Sham, OVX increased the mRNA expression of angiogenic activator and MMPs and decreased the mRNA expression of angiogenic inhibitors in white adipose tissue. But OVX + Swim decreased the mRNA expression of angiogenic activator and MMPs and increased the mRNA expression of angiogenic inhibitors in white adipose tissue, compared with the OVX. Theses results suggested that swimming exercises the angiogenesis in white adipose tissue, resulting to improve obesity in high-fat diet-fed female OVX mice.

  • Objective: We tested the usefulness of swim-down technique using human follicular fluid (hFF) in sperm preparation. Methods: Semen samples were obtained from twelve male partners showing asthenozoospermia (sperm motility < 50%) at the time of routine andrologic evaluation in Seoul National University Bundang Hospital. After dividing into two aliquots, each samples were processed either by swim-down using 100% hFF or density gradient using SpermGrad. Sperm quality was assessed by computer-assisted semen analyzer (CASA). Results: Motility, Rapid motility, VCL (curvilinear velocity), ALH (amplitude of lateral head displacement), and hyperactivated sperms were significantly increased, and LIN (mean linearity) was decreased significantly after sperm preparation in both groups. Motility was significantly higher after swim-down using 100% hFF when compared with density gradient using SpermGrad ($81.2{\pm}4.7$ vs. $67.6{\pm}2.3$, p=0.02) The other parameters assessed by CASA were not different between the two methods. Conclusion: Swim-down method with 100% hFF may be a useful method in preparation of sperm from asthenozoospermia.

Effects of Different Type of Exercise on Blood Variables and Leptin Hormones in SD Rats (훈련방법의 차이가 흰쥐의 혈액성분과 랩틴농도에 미치는 영향)

  • The purpose of this study is to evaluate the different type of exercise training on the changes of blood variables and leptin in SD rats. For this study, SD rats were divided into three groups: control group (CG: n=10), swim trained group (SG: n=10), and treadmill trained group (TG: n=10). The animals were housed in a pathogen-free animal facility ($22-24^{\circ}C$, 50-60% relative humidity, 08:00-20:00 lighting hours) at D university animal center, Pusan, Korea). Food and water were available ad libitum. The trained rats underwent a 8-wk endurance swim training (5 times/wk) in water at $26-29^{\circ}C$ (SG) and treadmill training (5 times/wk) in DAEJONG treadmill for 60 min. All data were expressed as mean and standard deviation by using SPSS package program (ver 10.0). The result through the statistical analysis of this data were summarized as follows: 1. In the weight changes, there were significant differences among CG, SG and TG(p<.05) after regular swim and treadmill training. TG showed the lowest weight than the other groups. 2. In the epididymal & perirenal adipose tissue levels, there were significant differences among CG, SG and TG(p<.05) after regular swim and treadmill training. TG showed the lowest adipose tissue levels than the other groups. 3. In the triglyceride changes, For the SG and TG, there were significantly decreased after regular swim and treadmill training. TG showed the lowest triglyceride levels than the other groups. 4. In the insulin hormone, For the SG and TG, there were significantly decreased after regular swim and treadmill training. TG showed the lowest insulin levels than the other groups. 5. In the leptin changes, For the SG and TG, there were significantly decreased after regular swim and treadmill training. TG was the lowest than the other groups. Based on the results, Regular swim and treadmill training decrease body weight, epididymal & perirenal adipose tissue levels significantly, this is caused but by decreased triglycerides, insulin, and leptin hormone levels not by the other factors. Regular treadmill training decreased insulin hormone levels compare to swim training, however there was no direct insulin effect on the weight changes. and it might be the direct effect of leptin hormones.

Effects of Swim Training on Tuberculosis infection in the Mouse Model at Different Temperature (온도 차이에 따른 수영훈련이 결핵균 감염정도에 미치는 영향)

  • Tuberculosis is the leading infectious disease in the world. It is urgent to develop new vaccine and treating drugs. Besides vaccines, we want to know the effects of regular swim training on TB infection in the mouse model. This study was designed to examine the effects of regular swim training on lung and spleen TB counts and $INF-\gamma$ activity in the trained mice at different temperature. The trained mice underwent a 10-wk endurance swim training (5 times/wk) in water at $29\~33^{\circ}C$ (WWG) and $21\~23^{\circ}C$(CWG) for 60 min. And they were divided into 3 groups according to the regular swim training (CG; control, WWG; warm water group, and CWG; cold water group). Mice were challenged by aerosol infection with M. tuberculosis H37Rv using an inhalation device (Glas-Col, Terre Haute, Ind.) calibrated to deliver bacteria into lungs. Three weeks after immunization, the mice were challenged. Four weeks after challenge, the mice were sacrificed and the numbers of viable bacteria in lung and spleen were determined by plating serial dilution of whole organ homogenates on nutrient Middlebrook 7H11 agar (Difco, Detroit, MI). Colonies were counted after four weeks incubation at $37^{\circ}C$. All data were expressed as mean, standard deviation by using SPSS package program (win 10.0). The result through the statistical analysis of this data were summarized as follows; In the weight changes, there were significant differences among CG, WWG, and CWG following the swim training at different temperature, and CWG was the lowest. In the change of $INF-\gamma$ following the swim training, there were significant differences (p<.05) among CG, WWG, and CWG after stimulated with media and CFP. In MTB counts, there were significant differences (p<.05) between CG and WWG in the lung. And also there were significant differences (p<.05) among CG, WWG, and CWG. These results suggest that regular swim training suppress Th1 immune response caused by decreased $INF-\gamma$ level in the WWG, Also For the WWG, highly increased level of TB counts appear in the lung and spleen compare to CG.

A Survey and Analysis of Swim Zone Width and Beach Scale Factor for Gangwon Beaches (강원도 해수욕장의 유영폭 및 해빈단면 축척계수 조사 분석)

  • In the present study, a beach survey such as sand sizes, swim zone widths, and slopes of swash zone has been accomplished for 39 beaches along the Gangwon coastline. The data analysis presents that the swim zone width has a strong correlation with the sand size, showing that the swim zone width is reversely proportional to the 1.1 power of a grain size. The profile factor A has been analyzed using the beach profile form of $h=Ay^{2/3}$, where A was resulted to be proportional to the 0.77 power of a gram size.

Taurine-enriched Rotifers Improve the Growth and Swim Bladder Inflation of Yellowtail Seriola quinqueradiata Larvae (타우린 영양 강화 로티퍼 공급이 방어(Seriola quinqueradiata) 부화 자어의 성장 및 부레 팽창에 미치는 영향)

  • We investigated the effects of taurine-enriched rotifers on larval growth and swim bladder inflation of yellowtail Seriola quinqueradiata. Rotifers were enriched with a commercial taurine supplement at two levels (0 and 800 mg/L). The larvae (initial notochord length=3.98±0.24 mm) were fed the enriched rotifers in triplicate from two days post-hatch for five days. The average taurine contents of the taurine non-enriched and enriched rotifers were 0.35±0.01 and 4.77±0.05 mg/g dry matter, respectively. The weight gain and specific growth rate of the fish fed enriched rotifers with the taurine supplement at 800 mg/L significantly improved compared with those of fish fed rotifers without taurine enrichment (P<0.05). The swim bladder inflation rate of larvae fed taurine enriched rotifers significantly (P<0.05). The results of the present study indicate that yellowtail larvae benefit from taurine concentrations compared with those typically reported to feed on non-taurine supplemented rotifers. Furthermore, taurine-enriched food for fish larval effectively improved the growth performance and swim bladder inflation of yellowtail larvae.
