Profiling of Differentially Expressed Genes in Human Cervical Carcinoma

  • Using the DDRT-PCR, a series of differentially expressed genes in human primary cervical cancer was isolated. Among the 250 PCR amplimers, 88 gene fragments were confirmed by reverse Northern hybridization. Homology searches indicated that 26 out of 88 were previously known genes including calmodulin, human BBC1, histone H3.3, a series of ribosomal proteins (RPL19, RPS19, and RPS12), translation initiation factor (eIF-4AI), lactoferrin, integrin ${\alpha}6$, cell-surface antigens (CD9 and CD59), transcription factor (mbp-1), and mitochondrial proteins. Several unknown clones showed sequence homology with known genes. Furthermore, six of the unknown genes showed identical sequence with expressed sequence tags (EST) of unknown function. Differential expression patterns of identified genes were further examined and confirmed with multiple pairs of cervical cancer samples using Northern hybridization. Our profiling of differentially expressed genes may provide useful information about the underlying genetic alterations in human cervical carcinoma and diagnostic markers for this disease. The precise roles of these genes in cancer development remain to be elucidated.

경원소 적층 분석을 위한 탄성되튐-비행시간 측정시스템 (An ERD-TOF System for the Depth Profiling of Light Elements)

  • An ERD-TOF system is constructed for the nondestructive depth profiling of light elements in thin films in the range of several thousand angstroms. The particles, recoiled by 10 $MeV^{35}Cl$ projectiles, were detected by a Time-Of-Flight spectrometer composed of a MCP (Micro Channel Plate) and a SSB (Silicon Surface Barrier) detector. A two parameter data acquisition system composed of two PC's was constructed for registering simultaneous time and energy signals. A $Si_3N_4$/poly-Si/$SiO_2$/Si sample was anlayzed and the result is compared with RBS. The detection limit, maximum probable depth and depth resolution for light elements in silicon are about $4\times10^{14}atoms/\textrm{cm}^2$, 5, 000$\AA$ and 100$\AA$, respectively.

  • Recently, various bulk metallic glasses (BMG's) having good mechanical and chemical properties were developed. BMG's can easily be deformed in the supercooled liquid region, via viscous flow mechanism. By using the viscous flow, the very low pressure is needed to deform the materials. In this study, we investigated the structural transition and deformation behavior of Vitreloy 1 (Zr/sub 41.2/Ti/sub 13.8/Cu/sub 12.5/Ni/sub 10/Be/sub 22.5/) using TMA and DSC. We applied the results to the micro forming process. The forming condition was chosen based on the viscosity data from TMA, and Si wafer with micro patterns on the surface was used as a forming die. The deformed surface was analyzed by SEM and 3D Surface Profiling System. The alloy showed good replication of the patterns. Quantitative measurement of roughness was useful to evaluate the replication. Surface condition of the deformed surface was determined by the initial surface condition.

TiN 에 대한 W의 부착특성에 관한 연구(l) (Studies on the Adhesion of W to TiN(l))

  • 전면증착법에 의한 W공정에서 부착특성고양층으로 사용되는 TiN막에 대한 CVD W막의 부착특성을 인장법(pulling method)과 스크래치법(scratch method)을 사용하여 조사하고, 주사전자현미경과 반사도측정에 의한 표면거칠기측정, 응력측정 및 SIMS depth profiling 등에 의하여 그 원인을 분석하였다. 스퍼터링법으로 형성한 TiN막상에 바로 W막을 증착한 경우와 TiN막을 열처리한 후에 W막을 증착한 경우 간에 두 막간의 부착특성은 큰 차이를 보였다. 전자의 경우가 후자의 경우보다 부착특성이 더 우수한 것으로 나타났는데, 이것은 열처리하지 않은 TiN막이 열처리한 TiN막에 비해 표면이 더 거칠고, 응력수준이 더 낮으며, 열처리한 TiN막내에는 산소성분이 존재하는 반면, 열처리 하지 않은 TiN막내에는 산소성분이 거의 들어있지 않기 때문이다. 또한 TiN막 두께가 증가함에 따라 응력의 증가로 인하여 TiN막에 대한 W막의 부착강도가 저하되었다.

표면 웹기반 공개정보 수집을 위한 워크플로우 확장 연구 (A Study on the Expansion of Workflow for the Collection of Surface Web-based OSINT(Open Source Intelligence))

  • 전통적인 형사 사건에서 조사 대상에 관한 정보는 국가의 합법적 조직이 보유하고 있는 개인정보만이 제공되기 때문에 정보 수집에 한계가 있다. 일반 검색엔진으로 검색이 가능한 SNS와 포털사이트를 포함하는 표면 웹 기반 공개정보(OSINT)는 범죄수사에 사용할 수 있는 의미 있는 프로파일링에 활용할 수 있다. 한국형 공개정보 워크플로우를 사용하면 공개정보 기반의 효과적인 프로파일링이 가능하지만 "개인"의 경우에는 "성명"으로 시작되기 때문에 수집할 수 있는 공개정보가 제한적이고 동명이인의 정보가 수집되는 등의 신뢰성의 한계가 있다. 본 논문에서는 이러한 한계를 극복하기 위해 개인과 연관된 정보 즉, 등가정보를 정의하고 이를 기반으로 효율적이고 정확한 정보를 수집할 수 있도록 한다. 따라서, 공개정보에서 특정인과 연관된 정보 즉, 등가정보를 추출할 수 있는 개선된 워크플로우를 제시한다. 이때 인물의 인지도에 따라 서로 다른 워크플로우를 제시한다. 이를 통해 인물(개인)의 효과적인 프로파일링이 가능하여 수사 정보 수집의 신뢰도를 높인다. 본 연구를 통해 향후에는 해당 워크플로우를 인공지능 기술을 이용하여 수집된 정보의 분석과정을 자동화할 수 있는 시스템을 개발함으로써 범죄 수사에 있어서 공개 정보 활용을 위한 기틀을 마련하고 수사 방식 다양화에 기여할 수 있을 것이다.

USE OF SINGLE PRECURORS FOR THE PREP ARATION OF SILICON CARBIDE FILMS

  • Heteroepitaxial growth of cubic silicon carbide films on Si(001) and Si(111) substrates at temperatures 900-$1000^{\circ}C$ has been achieved by high vacuum chemical vapor deposition using the single precursor 1, 3-disilabutane without carrying out the carbonization process of the substrate surfaces. The deposition temperature range is much lowered compared with conventiontional chemical vapor deposition where separate sources for silicon and carbon are employed. The deposition procedure is quite simple and safe. The qualities of the films were found to be very good judging from the results obtained by various characterization techniques including reflection high energy electron diffraction, X-ray diffraction, X-ray pole figure analysis, Rutherford backscattering spectrometry, Auger depth profiling, and transmission electron diffraction.

Outbreak of Water Mass into the East Coast of Japan Evident in the Kuroshio Extension in June 2001

  • The trajectories of 8 autonomous profiling floats deployed in the Kuroshio Extension region in February 2001 are used to depict the circulation pattern at the surface and 2000db. The corresponding sea surface topography maps created from satellite altimeter and dynamic height climatology were compared with the tracks of nearly coincident floats and were found to agree well in most cases except for the period June 5 to 16 2001. It is shown that over the period the conspicuous breakaway of the floats from an expected path is possibly associated with the abnormal path of the Kuroshio Extension such as an outbreak event, as revealed by AVHRR infrared and SeaWiFS chlorophyll-a images and cruise data in cross sections.

Ambient Variable Pressure Field Emission Scanning Electron Microscopy for Trichome Profiling of Plectranthus tomentosa by Secondary Electron Imaging

  • Glandular and nonglandular trichomes on the leaf surface of Plectranthus tomentosa were investigated by variable pressure field emission scanning electron microscopy (VP-FESEM). The segments of the plant's leaves were directly mounted without any specimen preparation, and examined at ambient temperature using a variable pressure secondary electron (SE) detector under ca. 15 Pa. Foliar trichomes maintained their shapes and structures without severe surface collapse or charging. The adaxial leaf surface was abundantly covered with different types of trichome. Nonglandular trichomes consisted of a basal cell and a long (up to ca. $300{\mu}m$) stalk. Meanwhile, capitate glandular trichomes had a secretory head and a short or long stalk. Peltate glandular trichomes with globose secretory heads were observed in close contact with the leaf epidermis. Spherical projections on the secretory head showed the secretion process of glandular trichomes. In addition to the trichomes, oval stomata were distributed on the abaxial leaf surface. These results suggest that ambient VP-FESEM can be used to classify the dehydration-sensitive foliar trichomes of succulent plants by SE imaging. At the FESEM resolution, this approach facilitates the rapid and detailed morphological analysis of a variety of trichomes in diverse plant taxa with reduced labor and preparation.

Confocal Scanning Microscopy : a High-Resolution Nondestructive Surface Profiler

  • Confocal scanning microscopy is a measurement technique used to observe micrometer and sub-micrometer features due to its high resolution, nondestructive properties, and 3D surface profiling capabilities. The design, implementation, and performance test of a confocal scanning microscopy system are presented in this paper. A short-wavelength laser (405 nm) and an objective lens with a high numerical aperture (0.95) were used to achieve the desired high resolution, while the x- and y-axis scans were implemented using an acousto-optic deflector and galvanomirror, respectively. An objective lens with a piezo-actuator was used to scan the z-axis. A spatial resolution of less than 138 nm was achieved, along with successful 3D surface reconstructions.

아르곤 이온빔 조사로 형성된 주름진 PDMS 표면 경화층의 이질성 (Heterogeneity of hard skin layer in wrinkled PDMS surface fabricated by Ar ion beam irradiation)

  • Spatial distribution of binding state in depth direction is investigated in a hard skin layer on soft polydimethylsiloxane (PDMS) fabricated by Ar ion beam irradiations. The hard skin layer known as a silica-like homogenous layer was composed of two layers. Impinging Ar ions transfer energy to PDMS as a function of collisional energy transfer rate, which is the maximum at surface and decreases gradually as an ion penetrates. This formed the heterogeneous hard skin layer that consists of a top-most layer and an intermediate layer. XPS depth profiling showed the existence of the top-most layer and intermediate layer. In the top-most layer, scission and cross-linking were occurred simultaneously and Si-O bond showed dissociated status, SiOx (x = 1.25 - 1.5). Under the top-most layer, there was the intermediate layer in which cross-linking is mainly occurred and Si-O bond showed silica-like binding status, SiOx (x = 1.75 - 2). And theoretical analysis which calculates the collisional energy transfer and a displacement per atom explained the thickness variation of top-most layer according to Ar ion energy from 360 eV to 840 eV.
