Skin Anti-aging and Anti-wrinkle Effects of Pinus koreaiensis Seed Oil (해송자 오일의 피부 항노화 및 주름 개선 효과)

  • Objectives : The aim of this study was to investigated the anti-aging and anti-wrinkles effects of the pinus koreaiensis seed(PKS) oil. Methods : The anti-oxidant effect was performed by beta-carotene bleaching assay and the intracellular proteome was analyzed expression of each 15 proteins by 2-D electrophoresis. And fatty acid was analysed by gas chromatography. Anti-wrinkle effect was analyzing human skin by the PRIMOS system. Results : Fatty acid analysis of PKS oil has shown oleic acid was 49.7% and linoleic acid was 34.1%. And the antioxidant effect was about 125% compared with alpha-tocoperol(0.1%) by beta carotene bleaching assay. In 2D PAGE analysis, fifteen protein changes in five mechanisms which was collagen synthesis pathway, MMPs, ECM-cell interaction, cytokine, antioxidant enzymes were analyzed. In case of anti-wrinkle effect was proved in vivo by analyzing human skin by the PRIMOS system. The analysis results of eye wrinkles for 4 weeks showed an improvement effect of over 6%. Conclusions : In this study, the amount of protein change in the five mechanism through the cell experiment and the skin anti wrinkle efficacy by the human in vivo test were investigated. As a result pinus koreaiensis seed oil by supercritical extraction could be used as a anti-aging and anti-wrinkle substance for the skin.

Stability and antioxidant effect of rapeseed extract in oil-in-water emulsion

  • In this study, rapeseed extracts were obtained by supercritical carbon dioxide fluid extraction of defatted rapeseed to evaluate the stability and antioxidant activity of an oil-in-water (O/W) emulsion system. The oil-in-water emulsions were prepared from stripped soybean oil with different concentrations (0.3, 0.4, 0.5, and 0.6%) of rapeseed extract as an emulsifier. Their emulsion stability was compared to that of emulsions prepared with the commercial emulsifier, Tween 20 (Polysorbate 20, 0.2%). After stripping the soybean oil, the total tocopherol content was reduced from 51.4 g/100 g to 1.1 g/100 g. Emulsion stability and oxidative stability of emulsions prepared with Tween 20 and rapeseed extract as emulsifiers were evaluated. For 30 days droplet sizes of emulsions containing rapeseed extract (0.4, 0.5, and 0.6%) were not significantly different (p > 0.05). Similar results were obtained for emulsion stability (ES) and Turbiscan analysis, suggesting that the addition of rapeseed extract increased emulsion stability. The addition of rapeseed extract at more than 0.4% resulted in an emulsion stability comparable to the addition of 0.2% Tween 20. The antioxidative ability of rapeseed extract increased with the amount added in the emulsion. Moreover, the addition of 0.6% rapeseed extract resulted in the lowest emulsion peroxide values (10.3 mEq/L) among all treatments. Therefore, according to the stability of its antioxidative and physical stability properties, rapeseed extract from super critical extraction could be successfully applied to the food and cosmetic industries.

The antihyperlipidemic effect of alginate-free residue from sea tangle in hyperlipidemic rats

  • Background: In order to assess the high value-added use of the alginate-free residue of sea tangle, an animal study was performed to evaluate the functional activities and key compounds present. In the animal study, sea tangle and the alginate-free residue demonstrated good anti-hyperlipidemic and anti-arteriosclerotic abilities. Results: The functional compounds in the alginate-free residue of the sea tangle were effectively extracted by supercritical fluid extraction (SFE). The optimum extraction temperature and pressure were 40 ℃ and 6500 psi (M1) in the SFE, a better method in comparison to the conditions of 70 ℃ and 4500 psi (M2), respectively. The anti-atherosclerotic effects of the alginate-free residue of sea tangle (M1, M2) were investigated in Sprague-Dawley rats treated with poloxamer 407, Triton WR 1339, corn oil, and a high-fat diet. The M1 fraction reduced the serum lipid levels with greater efficacy than the M2 fraction. In the hyperlipidemic rats, treatment with M1 decreased the serum triglyceride (TG), total cholesterol (TC), and low-density lipoprotein-cholesterol (LDL-C) levels when compared to the levels in normal rats. Conclusion: Our results demonstrated that the alginate-free residue of sea tangle reduces serum TC, TG, and LDL-C. These results suggest that the alginate-free residue of sea tangle contains physiologically active components, such as fucosterol, that may exert beneficial effects in the prevention of atherosclerosis.

Flavor Pattern and Sensory Properties of Meat Flavor Based on Maillard Reaction Products with Supercritical Fluid Extracted Lard Fractions (초임계 추출 Lard를 이용한 Maillard 반응생성물 유래 육류향미제의 향기패턴 및 관능적 특성)

  • We have investigated the effect of lard fraction extracted with supercritical carbon dioxide (SC-$CO_2$) on the flavor enhancement of maillard reaction product (MRP) based meat flavors. MRP based meat flavors were prepared with low glutamic acid (Glu) hydrolyzed wheat gluten (NaCl concentration: 7.61%(w/v)), ribose, cysteine, garlic juice powder, protease-digested Lentinus edodes powder and lard fractions extracted with SC-$CO_2$. Lard was extracted with SC-$CO_2$ at each of three temperatures (40, 60, and $80^{\circ}C$) and at each of four pressures (30, 40, 50, and 60 MPa). Obtained lard SC-$CO_2$ fractions and MRP based meat flavors with those fractions were analyzed for their total yield, aroma pattern by SMart nose system, and sensorial properties. The extraction yield had no difference as temperature increased from $40^{\circ}C$ to $60^{\circ}C$ and even decreased at $80^{\circ}C$. However, increase in pressure level at $40^{\circ}C$ drastically increased the extraction yield. The aroma patterns of raw lard and lard SC-$CO_2$ fractions with 30 MPa were significantly discriminated from those of SC-$CO_2$ lard fractions extracted with higher pressure by SMart nose system. Aroma pattern of MRP based meat flavors with higher pressure extracted lard fractions also showed significant difference through pattern analysis by the SMart nose system. The MRP based meat flavor with lard SC-$CO_2$ fractions at 50 and 60 MPa were described as less sulfuric, less pungent, and more balanced in roasted meat and sweet attributes from sensory evaluation.

Quality Characteristics of Ginseng Seed Oil Obtained by Different Extraction Methods (추출방법에 따른 인삼씨유의 품질특성)

  • This study aimed to determine the quality characteristics of ginseng seed oil as well as evaluate the efficacy of ginseng seed oil as a food resource. Ginseng seed oil was obtained by different extraction methods; from solvent extraction oil, supercritical fluid extraction oil, and screw pressed extraction oil. Total unsaturated fatty acids were present at 97.72~97.92%. Oleic acid (80.13~81.16%) was the highest, followed by linoleic acid (14.98~15.69%). The total phenol content (mg gallic acid equivalent/100 g oil) was higher in screw pressed extraction oil ($56.32{\pm}1.47$) compared to others. ${\gamma}$-Tocopherol was only present in ginseng seed oil and screw pressed extraction oil showed the highest levels of ${\gamma}$-tocopherol ($5.95{\pm}0.25$ mg/100g oil) among the tested samples. Screw pressed extraction oil showed the greatest oxidative stability with an induction time of 16.58 hours. Acid values and peroxide values of ginseng seed oil increased with increasing storage period. The total phenol and ${\gamma}$-tocopherol contents were higher in screw pressed extraction oil than in other ginseng seed oils, which suggests that screw pressed extraction oil has the greatest oxidative stability.

Aroma Characteristics of Applemint (Mentha rotundifolia(L.) Huds) with Different Extraction Methods (추출방법에 따른 애플민트의 향기특성)

  • Aroma was extracted from Applemint(Mentha rotundifolia(L.) Huds) with SDE(simultaneous distillation and extraction), SFE(supercritical fluid extraction) and headspace method and the compounds of aroma were tentatively identified with GC-MS. The functionality of aroma compounds were determined with GC-olfactometry. Total 67 compounds were identified. Among them, 39 compounds were determined from SDE, 42 from SFE and 16 from headspace extract. Many terpene compounds were extracted with SDE and headspace methods but hydrocarbones with SFE. The major constituents of aroma obtained from SDE and SFE, were piperitenone oxide, germacrene-D and trans sabinene hydrate, but those from headspace method were 3-octanol, 1,8-cineol, camphene and benzeneacetaldehyde. Results of sniffing test, determining characteristics and strength of aroma showed that the major constituents of SDE extract were refreshing sweet and apple-like(ethyl-2-methyl butanoate), sweet and fruity-like$({\alpha}-thujene)$, fresh mushroom-like(1-octen-3-ol, 3-octanol), and bitter herb-like$({\delta}-cadidene)$. Major constituents of aroma extracts obtained from headspace method were alcoholic, refreshing sweet and apple-like(ethyl 2-methyl butanoate), unpleasant chemical, and bitter herb and grassy-like(camphene).

  • N-vinyl caprolactam (NVCL), a kind of N-vinyl amide monomer, must be dissolved in continuous phase ($scCO_2$) for dispersion polymerization in supercritical carbon dioxide. Phase behavior of $CO_2$+NVCL is very important and necessary for determining initial polymerization condition and for monomer extraction from final polymer. There is the limitation of experimental method for obtaining pure properties of the monomer because of the possibility of polymerization. And N-methyl caprolactam (NMCL) is the useful solvent for the gas treating process. In the viewpoint of molecular thermodynamics, NVCL and NMCL have same functional group i.e. ${\varepsilon}$-caprolactam. In the case of NVCL, hydrogen of amide group is substituted with vinyl group and for NMCL, hydrogen of amide group is substituted with methyl group. We suggested modified group contribution method for this ${\varepsilon}$-caprolactam derivatives. This new group contribution parameter was applied to correlate $CO_2$ + N-vinyl caprolactam or N-methyl caprolactam system.

  • The study was done to determine the effect of storage property and qualities of soybean sauce and paste by adding different amount of pyroligneous liquor obtained by supercritical fluid extraction(SFE). Purified pyroligneous liquor obtained by SFE at $40^{\circ}C$/110bar contained ${\rho}$-cresol, o-cresol and m-cresol food sterilizers, but no toxic substances such as tar, scorched, furfuraland and monophenol. Thus pyroligneous liquor was suitable as natural food preservative. In case of soybean sauce, pyroligneous liquor was tested for the possibility of utilizing it as natural food preservative to prevent film formation on soybean sauce for the test period of 15days at $30^{\circ}C$. In case of paste, pyroligneous liquor was tested for the possibility of utilizing it as natural food preservative to inhibit browning on paste for the test period of 60days at $30^{\circ}C$. As a result, purified pyroligneous liquor offered a promising way of improving the quality and storage property of soybean sauce and paste .

Whitening Effects of Arctigenin Extracted from the Arctii Fructus (우방자로부터 추출한 악티게닌의 미백효과)

  • In this study, Arctigenin was obtained by using supercritical fluid extraction and bio-conversion process from Arctii Fructus. Arctigenin is efficacious in anti-inflammatory and anti-influenza. For this reason, Arctigenin is studied in various field. It was identified as 4-[(3,4-Dimethoxyphenyl)methyl)]dihydro-3-[(4-hydroxy-3-methoxyphenyl)methyl]-2(3H)-furanone (arctigenin) by FT-IR, $^1H$-NMR and the purity of it was 95.1 % by HPLC analysis. Arctigenin inhibited tyrosinase (up to $85.06{\pm}0.9%$ at $260{\mu}g/m{\ell}$ concentration) and melanin synthesis in a dose dependent manner (up to $51.1{\pm}3.7%$ at the concentration of $3.0{\mu}g/m{\ell}$). The results were better than arbutin. Therefore, it is expected that manufactured Arctigenin is useful for whitening cosmetics.

  • With petroleum reserves dwindling, interest has been increasing worldwide in Fischer-Tropsch synthesis (FT) as a method of producing synthetic liquid fuels and chemicals from coal, natural gas or biomass. In general, FT synthesis is operated through the gas phase fixed-bed reaction system. Recently, there are lots of study in supercritical fluid due to unique characteristics such as the quick diffusion of reactant gas, effective removal of reaction heat, and the in-situ extraction of high molecular weight hydrocarbon, such as wax. In this study, our major aim is to obtain a deeper insight into the effect of the type of support on the reaction performance over a supported cobalt catalyst in a fixed bed reactor.
