Antimicrobial, Antioxidative, Elastase and Tyrosinase Inhibitory Effect of Supercritical and Hydrothermal Asparagopsis Armata Extract

  • In this paper, we present to evaluate physiological activity of Asparagopsis armata extraction. After extraction with Asparagopsis armata using hydrothermal and supercritical carbon dioxide, various physiological activities were examined. The total concentration of polyphenol compounds was determined to be 18.85 mg/g of hydrothermal Asparagopsis armata extraction and 14.74 mg/g of supercritical Asparagopsis armata extraction. In DPPH radical scavenging assay, ascorbic acid was used as positive antioxidant control. In ABTS radical scavenging assay, ascorbic acid was used as positive antioxidant control. The percentage of inhibition and IC50 were measured. The IC50 of Asparagopsis armata extraction is 261.44ppm and the IC50 of supercritical Asparagopsis armata extraction is 153.98 ppm. The elastase inhibitory assay showed concentration dependence and the IC50 of hydrothermal Asparagopsis armata extraction is 3387 ppm and the IC50 of supercritical Asparagopsis armata extraction is higher than 2500 ppm. In mushroom tyrosinase inhibition experiments, tyrosinase inhibition's IC50 of supercritical Asparagopsis armata extraction was 248.06. In the SOD-like experiments, the concentration-dependent results were showed and IC50 of hydrothermal Asparagopsis armata extraction is 845.29 ppm. In the antimicrobial experiments, maximum clear zones of supercritical Asparagopsis armata extraction represented 23.00 mm in Propionibacterium acnes. In the other hand, in experiments with the same conditions, hydrothermal Asparagopsis armata extraction had no effect in all strains.

Quality and characteristics of ginseng seed oil treated using different extraction methods

  • Ginseng seed oil was prepared using compressed, solvent, and supercritical fluid extraction methods of ginseng seeds, and the extraction yield, color, phenolic compounds, fatty acid contents, and phytosterol contents of the ginseng seed oil were analyzed. Yields were different depending on the roasting pretreatment and extraction method. Among the extraction methods, the yield of ginseng seed oil from supercritical fluid extraction under the conditions of 500 bar and $65^{\circ}C$ was the highest, at 17.48%. Color was not different based on the extraction method, but the b-value increased as the roasting time for compression extraction was increased. The b-values of ginseng seed oil following supercritical fluid extraction were 3.54 to 15.6 and those following compression extraction after roasting treatment at $200^{\circ}C$ for 30 min, were 20.49, which was the highest value. The result of the phenolic compounds composition showed the presence of gentisic acid, vanillic acid, ferulic acid, and cinnamic acid in the ginseng seed oil. No differences were detected in phenolic acid levels in ginseng seed oil extracted by compression extraction or solvent extraction, but vanillic acid tended to decrease as extraction pressure and temperature were increased for seed oil extracted by a supercritical fluid extraction method. The fatty acid composition of ginseng seed oil was not different based on the extraction method, and unsaturated fatty acids were >90% of all fatty acids, among which, oleic acid was the highest at 80%. Phytosterol analysis showed that ${\beta}$-sitosterol and stigmasterol were detected. The phytosterol content of ginseng seed oil following supercritical fluid extraction was 100.4 to 135.5 mg/100 g, and the phytosterol content following compression extraction and solvent extraction was 71.8 to 80.9 mg/100 g.

초임계유체 추출 기술 및 상업화 현황 (The Current Status of Supercritical Fluid Extraction Technology and Industrial Applications)

  • Because of their unique properties, supercritical fluids have been known as one of the most promising materials for the future technology. Supercritical fluid technologies have been widely applied to various operations such as extraction, impregnation, nano-particle generation, oxidation, reaction etc. Industrial applications, especially their successful usage of supercritical fluid, have been reviewed. A special case for the first successful industrial application of supercritical $CO_2$ extraction in Korea was reviewed. Its unique characteristics of enriched antioxidant, $'\grmma-tocopherol'$ enabled this industrial application in Korea in spite of its low market price. Also its size and operation conditions were known as world records.

The Improvement of skin barrier function and anti-obesity effect of Codonopsis lanceolata by supercritical carbon dioxide extraction

  • The root of Codonopsis lanceolata has been used in traditional medicine. This study was conducted to confirm the comparative effect of ethanol solvent extraction (CLE) and supercritical carbon dioxide extraction (CLS) of C. lanceolata roots. CLS had higher antioxidant than CLE. For supercritical co-solvent modified carbon dioxide extraction (CLS), it were extracted at 250 bar 50℃ 150 min at a flow rate of ethyl alcohol 3 mL/min for 90min. In addition, CLS inhibited the adipocyte differentiation of 3T3-L1 cells. When treated with the extract at a concentration of 100 ㎍/mL, the Wnt/β-catenin pathway reporter luciferase activity of HEK 293-TOP cells increased approximately by 3-folds compared to that of the untreated control group. Also, the treatment by CLS (50 ㎍/mL) showed a significant increase of involucrin expression. These results indicate that supercritical carbon dioxide extract of C. lanceolatamay serve as a cosmeceutical agent for improving skin barrier function and the treatment of obesity.

  • Solvent extraction, soxhlet method, and supercritical fluid extracion were considered, respectively, as the method of choice for the recovery of DHA from lyophilized Thraustochitrium sp., and the results of corresponding extraction were compared. Supercritical fluid extraction seems to be the most appropriate process with respect to time, process simplicity, and extractant intoxicity.

Introduction of Modifying Solvents to Carbon Dioxide in Supercritical Extractions

  • A simple apparatus for adding a modifying solvent to supercritical CO2 extractant was described. Small, fixed volumes (typically 100 μL) of liquid modifying solvents were delivered during the extraction process by use of an in-line high pressure loop injector and an air pump. Without disconnecting the extraction cell from the supercritical fluid extraction system, the modifying solvent was repeatedly delivered. The solvent modification device was optimized during the extraction of carbaryl and bis(acetylacetonato) copper(Ⅱ). Extraction recoveries from spiked filter paper and soil samples ranged between 22% and 109%, depending on the analyte and matrix components. The addition of polar modifying solvents were necessary to improve the extractability of the nonpolar CO2.

아임계와 초임계유체로써 폐타이어 분해와 추출에 미치는 용매의 영향 (Effect of Solvents as Subcritical and Supercritical Fluid on Decomposition and Extraction of Used Automotive Tire)

  • 폐타이어에서 보강실과 고무로 배합된 부분(side wall)을 아임계와 초임계 상태의 세가지 용매, 물, 28% 암모니아수, 암모니아를 사용하여 용매의 분해와 추출능을 비교하였다. 6mm 입방체로 만든 폐타이어의 초임계물에 의한 분해와 추출속도는 140 kJ/mol의 활성화에너지를 갖는 1차 속도식을 잘 만족하였다. 초임계상태에서 28% 암모니아용액은 압력이 감소할수록 초기추출에서 초임계물보다 추출능이 높게 나타났다. 이 현상은 초임계물에 용해되어 있는 암모니아의 영향 때문이라고 추측된다.

  • 토종콩에 포함된 daidzein과 genistein을 초임계 $CO_2$와 아임계/초임계 $H_2O$를 이용하여 추출하였다. 추출액은 역상 고성능 액체 코토마토그래피(RP-HPLC)를 사용하여 분석하였다. 초임계 유체의 온도와 압력, 에탄올 농도의 변화에 따라서 비배당체의 체류시간, 체류인자, 컬럼효율, 컬럼의 선택도, 분리도를 비교하였다. 초임계 $CO_2$를 사용하였을 때, daidzein과 genistein의 추출특성은 온도와 에탄올농도의 영향을 주로 받았다. 아임계/초임계 $H_2O$를 사용하였을 때는 초임계 상태의 $400^{\circ}C$, 250 bar에서의 추출효율이 가장 우수하였다. 일반적으로 초임계 $CO_2$를 사용하는 경우, 초임계 $H_2O$에 비해서 약 10배 이상 비배당체의 추출수율이 증가하였다.

초임계 이산화탄소와 유기용매를 이용한 Chlorella vulgaris 오일의 추출 (Extraction of Oil from Chlorella vulgaris Using Supercritical Carbon Dioxide and Organic Solvent)

  • Three different types of extraction processes, which used supercritical carbon dioxide ($SCCO_2$) and organic solvent, were attempted to improve the extraction yield of oil from Chlorella vulgaris: cosolvent-modified $SCCO_2$ extraction, $SCCO_2$ extraction with ultrasonic sample treatment in organic solvent, and static extraction with organic solvent followed by dynamic $SCCO_2$ extraction. Among these, the last $SCCO_2$ extraction process was found to be most effective in the extraction of oil. Compared with pure $SCCO_2$ extraction, the extraction yield of oil was observed to increase about 7 times.

에탄올 보조용매 초임계$CO_2$를 이용한 대두 Daidzein 추출 (Ethanol Modified Supercritical$CO_2$ Extraction of Daidzein from Soybean)

  • 순수한 초임계 이산화탄소로는 대두박으로 부터 daidzein을 추출할 수 없었다. 보조용매로서 에탄올을 사용하면 daidzein을 추출이 가능하였다. 에탄올을 보조용매로 사용한 경우 온도가 증가할수록 daidzein의 추출 효율도 증가하였고, 압력이 증가할수록 추출 효율 역시 증가하였다. 에탄올을 보조용매로 사용하는 경우에 보조용매의 농도가 증가할수록 추출 효율도 비레하여 증가하였고 19% 이상에서는 추출 효율이 완만하게 증가하였다. 보조용매 농도가 15% 이상부터는 보조용매 농도가 증가할수록 daidzein함량은 감소하는 경향을 나타내었다. 보조용매 농도가 15%에서 93% 추출 효율을 나타냈다. 추출시간에 따른 추출 효율의 변화를 연구한 결과, 초기에는 추출 효율이 급격하게 증가하다가 후반부터는 완만하게 증가하는 즉, 포화현상을 나타내었다. 추출시간에 따른 추출된 daidzein함량의 변화는 뚜렷한 경향은 없지만, 대체적으로 추출시간이 증가할수록 감소하는 경향을 나타내었다.
