녹두의 하배축에서 분리한 Soluble Acid Invertase의 정제와 특성 (Purification and Characterizationof Soluble Acid Invertase from the Hypocotyls of Mung Bean (Phaseolus radiatus L.))

  • The soluble acid invertase ($\beta$-D-fructofuranoside fructohydrolase, EC 3.2.1.26) was isolated and characterized from the hypocotyls of mung bean (Phaseolus radiatus L.). The enzyme was purified to apparent homogeneity by consecutive step using diethylaminoethyl (DEAE)-cellulose anion exchange, Concanavalin (Con) A affinity and Sephacryl S-300 chromatography. The overall purification was about 148-fold with a yield of about 15%. The finally purified enzyme exhibited a specific activity of about 139 $\mu$mol of glucose produced mg-1 protein min-1 at pH 5.0 and appeared to be a single protein by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and nondenaturing PAGE. The enzyme had the native molecular weight of 70 kD and subunit molecular weight of 70 kD as estimated by Sephadex G-200 chromatography and SDS-PAGE, respectively, suggesting that the enzyme was composed of a monomeric protein. On the other hand, the enzyme appeared to be a glycoprotein containing N-linked high mannose oligosaccharide chain on the basis of its ability to bind to the immobilized C on A. The enzyme had a Km for sucrose of 1.8 mM at pH 5.0 and maximum activity around pH 5.0. The enzyme showed highest enzyme activity with sucrose as substrate, but the activity was slightly measured with raffinose and cellobise. No activity was measured with maltose and lactose. These results indicate the soluble acid invertase is a $\beta$-fructofuranosidase.

펜톤산화에 따른 복합폐수의 처리효율연구 (Treatment Efficiency of Complex Wastewater by Fenton's Oxidation Condition)

  • In order to treat the wastewater containing organic compound, pre-treatment system connected with MSP(molecular separation process) was investigated. With the aim of selecting an optimum process of Fenton's oxidation, removal efficiency of each process in the optimum reaction condition was recommended. The $Fe/H_{2}O_{2}$(ferric sulfate to hydrogen peroxide)reagent is referred to as the Fenton's regent, which produces hydroxyl radicals by the interaction of Fe with $H_{2}O_{2}$. The powerful oxidizing ability and extreme kinetic reactively of the hydroxyl radical was well established. Increasing dosage of $Fe/H_{2}O_{2}$ increased removal efficiency as molar ratio of $Fe/H_{2}O_{2}$ between 0.2 and 2.5. Optimum dosage of molar ratio was 1. The removal efficiency for reaction condition was increased as pH decreased when the molar ratio of $Fe/H_{2}O_{2}$ was 1.7. Fenton's oxidation was most efficient in the reaction time 35 min for complex wastewater. Also, coagulation aid experiments using kaolin resulted in 3% of kaolin dosage.

The use of natural remedies to treat osteoarthritis

  • Osteoarthritis (OA) is the leading medical condition for which patients use alternative treatments including the natural remedies. The aim of this review is to describe the dietary supplements and herbal remedies most commonly used in patients with osteoarthritis with an emphasis on the efficacy and safety of these natural remedies. Glucosamine and chondroitin sulfate, two of the molecular building blocks found in articular cartilage, are the most commonly used remedies in OA treatment. Most clinical researches suggest that glucosamine and chondroitin show efficacy in reducing or improving symptoms and their ability to arrest progression of the disease or regenerate damaged cartilage. Patented formulations of both remedies are recommended by several therapeutic guidelines for use as first line background OA treatment. Reliable evidence that the combination is more effective than either agent alone is however still lacking. Several other herbs or remedies are promoted for treating osteoarthritis such as S-adenosylmethionine, methylsulfonylmethane, Harpagophytum procumbens (devil's claw), Curcuma longa (turmeric), Zingiber officinale (ginger), and capsaicin but there is no reliable evidence on long-term efficacy or safety. The clinical usefulness of these remedies is therefore rather limited currently.

Purification and Properties of Novel Calcium-binding Proteins from Streptomyces coelicolor

  • Two novel calcium-binding proteins, named CAB-I and CAB-II, have been isolated from Streptomyces coelicolor. Purification of the calcium-binding proteins involved heat treatment, fractionation with ammonium sulfate, acid treatment, anion exchange and hydrophobic interaction column chromatography, FPLC gel filtration, and preparative isoelectric focusing. A chelex competitive assay and 45Ca autoradiography verified the calcium-binding ability of the proteins. The major band CAB-II has an apparent molecular weight of 26,000 determined by SDS-polyacrylamide gel electrophoresis and 340,000 determined by gel filtration. The isoelectric point of this molecule showed the acidic nature of the molecule. N-terminal amino acid sequence analysis shows homology to rat Ca2+/calmodulin-dependent protein kinase-II (CAB-II) and yeast phosphoprotein phosphatase (CAB-I).

Fermented noodles with degraded gluten (FNDG) improved digestion and gut motility in enteritis-induced mice

  • Gluten proteins are key to developing a unique quality of flour because they confer viscosity, cohesiveness, and elasticity in the dough. However, gluten can impede digestion and absorption in gluten-sensitive individuals. In this study, enteritis was induced in mice with dextran sulfate sodium (DSS) salt. The mice later received a diet consisting of 3%, 12%, or 30% fermented noodles with degraded gluten (FNDG) or 30% normal noodle flour for 8 weeks. FNDG did not alter the growth performance or body composition. However, FNDG resulted in increased amylase activity in a dose-dependent manner (p<0.001), and it also improved the digestive capacity. FNDG at 30% concentration increased the level of gastrin (p<0.01) implying increased gut motility. The serotonin receptor levels were significantly reduced by FNDG at 12% (p<0.05) and 30% (p<0.01) concentrations. These findings indicate that a diet containing FNDG could help in the recovery from intestinal inflammation with improving digestive ability and gut motility. Overall, the inclusion of degraded gluten in the diet was found to enhance digestion, gut motility, and absorption in mice.

Physicochemical Properties and Antioxidant Effects of Fucoidans Degraded by Hydrogen Peroxide under Electron Beam at Various Irradiation Doses

  • Fucoidans were degraded by hydrogen peroxide under the electron beam (2.5 MeV) with various radiation doses (5 kGy, 10 kGy, 15 kGy, and 20 kGy) at room temperature. The degradation property was analyzed with a gel permeation chromatography (GPC-MALLS) method. An average molecular weight of fucoidan decreased from 99,956 at the irradiation dose of 0 kGy to 6,725 at the irradiation dose of 20 kGy. The solution viscosity of fucoidans showed a similar pattern to the molecular weight change. The number of chain breaks per molecule (N) increased with increasing the irradiation dose and concentration of hydrogen peroxide. The radiation yield of scission value markedly increased with increasing the irradiation dose up to 15 kGy. Also a 10% hydrogen peroxide concentration was more efficient than that of 5%. The structures of degraded fucoidan samples were studied with Fourier transform infrared spectroscopy (FT-IR). The results showed that the degradation process did not significantly change the chemical structure or the content of sulfate group. The sulfur content of each sample was determined with an Elemental Analyzer. With increasing the concentration of hydrogen peroxide, the ratios of sulfur/carbon, hydrogen/carbon, and nitrogen/carbon slightly decreased. The antioxidant activities of fucoidans were investigated based on hydroxyl radical scavenging activities. The ability of fucoidan to inhibit the hydroxyl radical scavenging activity was depended on its molecular weight.

된장에서 분리된 유산균의 결합력에 의한 Heterocyclic Amines 제거 (Heterocyclic Amines Removal by Binding Ability of Lactic Acid Bacteria Isolated from Soybean Paste)

  • 단백질이 풍부한 식품을 고온 하에서 조리하는 과정 중에 주로 발생되는 돌연변이원 heterocyclic amines (HCAs)에 대한 유산균의 결합력 및 제거능을 조사하였다. 당 발효능 및 16S rRNA 염기서열 분석을 통해 동정된 19종의 유산균 중 Lactobacillus acidophilus D11, Enterococcus faecium D12, Pediococcus acidilactici D19, L. acidophilus D38, Lactobacillus sakei D44, Enterococcus faecalis D66 및 Lactobacillus plantarum D70의 세포이나 배양 상등액은 3-amino-1,4-dimethyl-5H-pyrido[4,3-b]indole (Trp-P-1)과 3-amino-1-methyl-5Hpyrido[4,3-b] indole (Trp-P-2)에 의한 Salmonella typhimurium TA98 및 TA100의 돌연변이 유발을 억제할 수 있었다. HCAs에 대한 유산균 세포의 결합력은 cell wall, exopolysaccharide 및 peptidoglycan 보다 높게 나타났다. 한편, 이들의 결합력은 단백질 분해효소, 가열, sodium metaperiodate 및 산 처리에 의해 유의하게 감소되었으므로 세포벽에 존재하는 당이나 단백질 성분이 이들 HCAs을 결합시키는데 중요한 역할을 하는 것으로 확인되었다. 또한 E. faecium D12, L. acidophilus D38 및 E. faecalis D66의 결합력은 SDS나 금속이온에 의해 감소되었으므로 이들세포와 돌연변이원 사이에는 이온 결합이나 소수성 결합이 작용하는 것으로 추정되었다. 한편, HCAs 결합력이 높은 L. acidophilus D38과 L. plantarum D70은 장관 상피세포에 대한 부착력이 낮으므로 돌연변이원을 세포에 결합시켜 체외로 배출함으로써 독성물질을 제거하는데 효과적인 것으로 확인되었다.

  • 핵전이 방법을 이용하여 효모에서 전분분해능이 향상된 새로운 우수 균주를 개발하고자 하였다. Saccharomycopsis fiburigera KCTC 7393과 Saccharomyces cerevisiae KCTC 7049에서 핵을 분리한 후 영양요구 돌연변이주인 S. cerevisiae의 안으로 전달시켜 잡종(MN-16)을 형성하여 전분 분해능이 증가된 잡종을 선별하였다. 세포 배양액으로 조효소 용액을 만든 후 ammonium sulfate 침전, DEE-Sephacel column chromatography, Sephacryl S-200 column chromatography 등의 정제과정을 통해 9.7%의 회수율로 약 10.6배 정제 된 효소를 얻을 수 있었다. 정제된 효소는 SDS-PAGE 전기영동을 통해 단일 band를 보여 주었으며, SDS-PAGE 와 Sephacryl S-200 column chromatography를 통해서 53kDa으로 나타났다. 정제효소의 최적 활성 온도는 40${\circ}C$이고, 안정성은 40~45${\circ}C$로 나타났다. 최적 활성 pH는 5.5이었고, pH 5.0~7.0 정도에서 pH안정성이 80%정도 유지되었다. 가용성 전분에 대한 $K_{m}$ 값은 2.5㎎/㎖이었다. 또한, 정제 효소의 금속 이온의 효과로 $Ca^{2+}, Co^{2+}, EDTA, Mg^{2+}, Mn^{2+}, Zn^{2+}$ 첨가시 활성이 촉진되었고, $Ca^{2+}$의 경우 가장 높은 반면 $Cu^{2+}, Fe^{2+}, Ni^{2+}$의 경우는 오히려 활성이 감소되었다.

  • 한국 전통 젓갈로부터 항균성 lipopeptide를 생산하는 JKK238 균주를 분리하여 16S rDNA및 세포벽의 지방산 조성 분석 등에 따라 Bacillus subtilis로 동정하였다. JKK238 균주를 3일간 배양한 TSB 액체배지의 $25\%$ ammonium sulfate 침전물이 대부분의 lipopeptide를 포함하고 이들이 다양한 식물병원성 곰팡이나 세균의 성장을 강력히 저해함을 알 수 있었다. 또한 FPLC에 의해 분리 및 정제된 각 lipopeptide의 Maldi-tof분석에서, 이 균주가 iturin A, fengycin, surfactin과 같은 세 종류의 lipopeptide의 isomer들을 동시에 생산하는 균주임을 밝혔다.

  • 질소원(窒素源)을 달리한 비료시용(肥料施用)이 대두(大豆)의 생육(生育), 수량(收量), 영양성분(營養成分) 및 단백질(蛋白質) 소화율(消化率) 등(等)에 미치는 비효(肥效)는 다음과 같다. 대두(大豆)의 생육상황(生育狀況)은 시비조건간(施肥條件間)에 대체로 유사(類似)하였으나 유안구(硫安區)와 무질소구(無窒素區)에 있어서 절수(節數)가 적었다. 근류수(根瘤數)와 무게는 유안구(硫安區)와 무질소구(無窒素區)가 낮았고 그 밖의 처리구(處理區)에 있어서는 차이가 보이지 않았다. 근류수(根瘤數) 당(當) 질소고정력(窒素固定力)은 생육초기(生育初期)와 개화기(開花期)에는 거의 비슷하였으나 pod 신장기(伸長期)에서는 개발복비구(開發複肥區)가 타처리구(他處理區)보다 비교적 높았다. 대두(大豆)의 100립중(粒重) 및 10a 당(當) 수량(收量)은 개발복비구(開發複肥區)가 타처리구(他處理區)보다 높았고 무질소구(無窒素區)는 질소원(窒素源) 비료(肥料)의 시용구(施用區)보다 대체로 낮았다. 또한 대두(大豆)의 조단백질(粗蛋白質) 함량은 질소원(窒素源) 비료(肥料)의 시용구(施用區)가 무질소구(無窒素區)에 비해 높았고 조지방(粗脂肪) 함량은 오히려 낮았다. 조회분(粗灰分) 함량은 시용구간(施用區間)에 대체로 비슷하였으나 조섬유(粗纖維) 함량은 기존복비구(旣存複肥區)가 타처리구(他處理區)에 비해 높았다. Ca, P, Na, K, Mg, Zn, Mn 및 Cu 함량은 시비조건간(施肥條件間)에 대체로 비슷하였으나 Fe 함량은 무질소구(無窒素區)가 질소원(窒素源) 비료(肥料)의 시용구(施用區)에 비(比)해 월등히 낮았으며, 단백질(蛋白質) 소화율(消化率)은 무질소구(無窒素區)에서 가장 높았다.
