A disease of young Holstein calves characterized by recurrent pneumonia, ulcerative and granulomatous stomatitis, enteritis with bacterial overgrowth, periodontitis, delayed wound healing, persistent neutrophilia and death at an early age had been originally described in 1983 and again in 1987. Most of these calves had stunted growth and a persistent, progressive neutrophilia (often exceeding 100,000/ml). By investigation of pedigrees, all of the affected calves have now been traced to a common sire and confirmed by polymerase chain reaction (PCR) diagnostic DNA testing to be homozygous carriers of a defective allele for bovine CD18. Neutrophils from these calves have several functional deficits and, most importantly, fail to adhere in a ${\beta}_2$-integrin dependent manner. The ${\beta}_2$-integrins represent a family of glycoproteins which participate in various leukocyte adhesion reactions during host defense. The presence or absence of ${\beta}_2$-integrin molecules can be demonstrated on the surface of neutrophils, monocytes and lymphocytes from normal or affected calves using specific monoclonal antibodies and flow cytometry, or by colloidal gold immunolabeling and scanning electron microscopy in backscatter mode. Deficiency of the ${\beta}_2$-integrins on all leukocyte types in Holstein calves is analogous to leukocyte adhesion deficiency (LAD) seen in humans. Neutrophils in bovine (BLAD) and human LAD patients are unable to adhere to the endothelial lining of the cardiovascular system thus interrupting egression of neutrophils into infected tissues. Other leukocytes, while still deficient in expression of the ${\beta}_2$-integrins, are still able to efficiently egress from the blood stream due to interactions of other adhesion molecules that are not as highly expressed on neutrophils. Both BLAD cattle and LAD children (who do not receive bone marrow transplants) often die at an early age as a result of the failure of neutrophils to extravasate into infected tissues. In 1991, Shuster, et $al^{27}$, identified two point mutations within the alleles encoding bovine CD18 in a Holstein calf afflicted with leukocyte adhesion deficiency. One mutation causes an aspartic acid to glycine substitution at amino acid 128 (D128G) in an extracellular region of this adhesion glycoprotein that is highly conserved (> 95% identity) between humans, cattle and mice. The other mutation is silent. Numerous calves with clinical symptoms of leukocyte adhesion deficiency have since been tested and all have been found homozygous for the D128G allele. In addition, calves homozygous far the D128G allele have been identified during widespread DNA testing in the United States. All cattle with the mutant allele are related to one bull, who through artificial insemination (A.I.), sired many calves in the 1950's and 1960's. The carrier frequency of the D128G CD18 allele among U.S. Holstein cattle had reached approximately 15% among active A.I. bulls and 8% among cows. By 1993, the organization of the dairy industry and the diagnostic test developed to genotype cattle, enabled virtually complete eradication of bovine leukocyte adhesion deficiency among current and future A.I. bulls.
Field resistance is defined as the resistance that allows effective control of a parasite under natural field condition and is durable when exposed to new races of that parasite. To identify the genes for field resistance to rice blast, quantitative trait loci (QTLs) conferring the resistance for races and blast nursery screening in japonica rice cultivars were detected and mapped using SSR markers. QTL analysis was carried out in 190 RILs population from the cross between Suweon365 (moderately resistant) and Chucheong (highly susceptible). Twelve QTLs against nine blast races inoculated were detected on chromosomes 1, 2, 4, 6, 7, 11 and 12. They explained from 5.1% to 34.9% of total phenotypic variation. Eight QTLs against blast nursery screening in four regions for three years were detected on chromosomes 1, 2, 4, 11 and 12. The phenotypic variation explained by each QTL ranged from 4.3% to 37.7%. Three chromosome segment substitution lines (CSSLs) of $BC_2F_6$ by backcross method were developed to transfer the QTLs into the susceptible cultivar Chucheong as a recurrent parent. A CSSL4-1 containing two QTLs qLB6.2 and qLB7 against blast races showed to the reaction of 6 to 7 at blast nursery in two regions for two years. The CSSL4-2 and CSSL93 containing QTLs, qLB11.2 and qLB12.1 of the resistance against leaf blast in blast nursery screening, respectively, had enhanced the resistance for blast nursery screening across two regions and in two years.
Oxygen reduction in an alkaline fuel cell was studied by using perovskite type oxides as an oxygen electrode catalyst. The high surface area catalysts were prepared by malic acid method and had a formula of $La_{0.6}Sr_{0.4}Co_{1-x}Fe_xO_3$(x=0.00, 0.01, 0.10, 0.20, 0.35 and 0.50). From the result of XRD pattern and specific surface area due to the amount of Fe substitution and the consumption of ammonia-water, the complex formation of Fe ion with $NH_3$ was the main factor for both the phase stability of perovskite and the increase of specific surface area. Multi-step calcination was necessary to give a single phase of perovskite in catalyst precursor. The crystal structure of the catalysts was simple cubic perovskite, which was verified from the XRD patterns of the catalysts. The activity of oxygen reduction was monitored by the techniques of cyclic voltammetry, static voltage-current method, and current interruption method. The activity(current density) of oxygen reduction showed its minimum at x=0.01 and its maximum between 0.20 and 0.35 of x-value in $La_{0.6}Sr_{0.4}Co_{1-x}Fe_xO_3$. This tendency was independent of the change of surface area.

The genetic analyses of Acanthopanax senticosus Harms from Korea, China and Russia, were made by comparing the internal transcribed spacer (ITS) sequences of the nuclear ribosomal DNA. The ITS region of A. senticosus was amplified by polymerase chain reaction (PCR) using the universal primers and then directly sequenced. The length of the ITS region including 162 bp 5.85 rRNA gene ranged from 608 bp (for Korean and Chinese) to 611 bp (for Russian). The G+C content of ITS region were 60.20% for Korean and Chinese plants and 60.06% for Russian plants. Sequence comparisons indicated that ITS regions of A. senticosus from Korea and China were identical, whereas the ITS sequence of A. senticosus from Russia showed 99.2% homology with the plants from Korea. Variation in sequences were attributable to 5 bp substitution such as transversion or insertion events. These results suggested that A. senticosus Harms from Korea and China were closely related in phylogenetic relationship compared to Russian. In addition, A. senticosus Harms were more similar to Kalopanax pictus than A. sessiliflorus in their ITS sequences.
[ $CdS_{0.46}Se_{0.54}$ ] single crystal was grown by a sublimation method. The crystal structure and the temperature dependence of carrier density and mobility of $CdS_{0.46}Se_{0.54}$ single crystal were studied. Heterojunction solar cells of $n-CdS_{0.46}Se_{0.54}/p-Cu_{2-X}S_{0.46}Se_{0.54}$ were fabricated by the substitution reaction. The spectral response, the J-V characteristics and the conversion efficiency of the $n-CdS_{0.46}Se_{0.54}/p-Cu_{2-X}S_{0.46}Se_{0.54}$ heterojunction solar cells were studied. The open-circuit voltage, short-circuit density, fill factor and conversion efficiency of $n-CdS_{0.46}Se_{0.54}/p-Cu_{2-X}S_{0.46}Se_{0.54}$ heterojunction solar cells under $80mW/cm^2$ illumination were found to be 0.48V, $21mA/cm^2$, 0.75 and 9.5%, respectively.
Strong alkaline electrolyzed water which is produced in cathode by electrolyzing the solution where electrolytes (NaCl, $K_2CO_3$ etc.) are added in diaphragm electrolytic cell, is eco-friendly and has cleaning effects. So, it is viewed as a substitution of chemical cleaner. In addition, strong alkaline electrolyzed water is being used by some Japanese automobile and precision parts manufacturing industries. When strong alkaline electrolyzed water is produced by using diaphragm electrolytic cell, it is necessarily produced at the anode side. Since strong acidic electrolyzed water produced is discarded when its utilization cannot be found, production efficiency of electrolyzed water is consequently decreased. Also, there is a weakness electrolytic efficiency is decreasing due to the pollution of diaphragm. In order to overcome this, non-diaphragm all-in-one electrolytic cell integrated with electrode reaction chamber and dilution chamber was applied. Strong alkaline electrolyzed water was produced for different composition of electrolytes, and their properties and characteristics were identified. In comparing the properties between strong alkaline electrolyzed water produced in diaphragm electrolytic cell and that produced in all-in-one electrolytic cell, the differences in ORP and chlorine concentration were found. In emulsification test to confirm surface-active capability, similar results were obtained and strong alkaline electrolyzed water produced in non-diaphragm all-in-one electrolytic cell was identified to be useable as a cleaner like strong alkaline electrolyzed water produced in diaphragm electrolytic cell. Strong alkaline electrolyzed water produced in non-diaphragm all-in-one electrolytic cell is thought to have sterilizing power because it has active chlorine which is different from strong alkaline electrolyzed water produced in diaphragm electrolytic cell.
Escherichia coli tryptophan synthase (TS) contains ${\alpha}_2{\beta}_2$, which catalyzes the final two steps in Trp biosynthesis. A molecular tunnel exists between the two active sites of ${\alpha}$ and ${\beta}$ subunits in TS. Via intersubunit communication, TS increases catalytic efficiency, including substrate channeling. The ${\beta}$ subunit of TS is composed of two domains, one of which, the COMM (communication) domain, plays an important role in intersubunit communication. The ${\alpha}$ subunit has a TIM barrel structure. This protein has functional regions at the C terminal of ${\beta}$ pleated sheets and in its loop regions. Three regions of the ${\alpha}$ subunit (${\alpha}L6$ [${\alpha}-loop$ L6], ${\alpha}L2$, and ${\alpha}L3$) are implicated in intersubunit communication. In the present study, conformational changes in ${\alpha}L6$ were monitored by measuring the sensitivity of mutant proteins in these regions to trypsin. The addition of a ${\alpha}$ subunit-specific ligand, D,L-${\alpha}$-glycerophosphate (GP), partially restored the sensitivity of mutant proteins to trypsin. In contrast, the addition of the ${\beta}$ subunit-specific ligand L-serine (Ser) resulted in varied sensitivity to trypsin, with an increase in PT53 (substitution of Pro with Thr at residue 53) and DG56, decrease in NS104 and wild type, and no change in GD51 and PH53. This finding may be related to several reaction intermediates formed under this condition. The addition of both GP and Ser led to a highly stable state of the complex. The present results are consistent with the current model. The method used herein may be useful for screening residues involved in intersubunit communication.
The feasibility of the geopolymer as a cultural asset restoration material was studied by investigating compressive strength and chromaticity change. Metakaolin that was synthesized by calcination of the kaolin at $750^{\circ}C$ for 6 hours was used as a geopolymeric starting material. Kaolin lost its crystallinity and changed into non-crystalline phase during calcination. NaOH solution and water glass were used as an initiator for the geopolymeric reaction. As the concentration of NaOH solution and water glass increased the compressive strength increased. When alumina was substituted with metakaolin, the compressive strength decreased at a small amount of alumina, but increased at a large substitution. For the most composition of geopolymers, the change of chroma values remained within the limit of slight variation after exposure to sunlight for 8 and 100 days. However, even small amount of organic pigment addition increased chroma values of metakaoline. It was shown that geopolymer had excellent chroma value change over epoxy resins.
[ ${\alpha},{\beta}$ ]-Unsaturated carboxamides 12 with trifluromethylated dihydro-1,4-dithiins were synthesized for the purpose of development of new agrochemical fungicide. Chlorination of trifluoromethylated ${\beta}$-ketoester 4 followed by tile reaction with 1,2-ethanedithiol gave intermediate 1,4-dithiane 9. Without purification of 9 substitution of hydroxy by chlorine followed by dehydrochlorination in the presence of triethylamine afforded trifluoromethylated dihydro-1,4-dithiin ethyl ester 7. Activation of the hydroxy of the carboxylic acid 10 obtained from the hydrolysis of 7 and then reacted with various amines gave the corresponding trifluoromethylated dihydro-1,4-dithiin carboxamides. Antifungal screening (in vivo) against typical plant diseases, Rice Blast, Rice Sheath Blight, Cucumber Gray Mold, Tomato Late Blight, Wheat Leaf Rust, and Barley Powdery Mildew of the synthesized compounds was carried out. As a result, most of the compounds shlowed weak antifungal activities and some compounds in which isopropyl group was substituted in meta of the phenyl showed antifungal activity (99%) at 250 ppm against the disease Wheat Leaf Rust.
PURPOSES : In this study, we analyzed the compressive strength characteristics of lean base concrete in relation to changes in the outdoor temperature after analyzing the cold and hot weather temperature standards and calculated the minimum and maximum temperatures when pouring concrete. We examined the rate of strength development of lean base concrete in relation to the temperature change and derived an appropriate analysis formula for FRC base structures by assigning the accumulated strength data and existing maturity formula. METHODS : We measured the strength changes at three curing temperatures (5, 20, and $35^{\circ}C$) by curing the concrete in a temperature range that covered the lowest temperature of the cold period, $5^{\circ}C$, to the highest temperature of the hot period, $35^{\circ}C$. We assigned the general lean concrete and FRC as test variables. A strength test was planned to measure the strength after 3, 5, 7, 14, and 28 days. RESULTS : According to the results of compressive strength tests of plain concrete and FRC in relation to curing temperature, the plain concrete had a compressive strength greater than 5 MPa at all curing temperatures on day 5 and satisfied the lean concrete standard. In the case of FRC, because the initial strength was substantially reduced as a result of a 30% substitution of fly ash, it did not satisfy the strength standard of 5 MPa when it was cured at $5^{\circ}C$ on day 7. In addition, because the fly ash in the FRC caused a Pozzolanic reaction with the progress into late age, the amount of strength development increased. In the case of a curing temperature of $20^{\circ}C$, the FRC strength was about 66% on day 3 compared with the plain concrete, but it is increased to about 77% on day 28. In the case of a curing temperature of $35^{\circ}C$, the FRC strength development rate was about 63% on day 3 compared with the plain concrete, but it increased to about 88% on day 28. CONCLUSIONS : We derived a strength analysis formula using the maturity temperatures with all the strength data and presented the point in time when it reached the base concrete standard, which was 5 MPa for each air temperature. We believe that our findings could be utilized as a reference in the construction of base concrete for a site during a cold or hot weather period.
