대퇴골두 해면골의 미세구조 특성과 기계적 특성의 분석 (Analysis of the Micro-Structural and Mechanical Properties in Human Femoral Head Trabecular Bone with and without Osteoporosis)

  • 골다공증이 있는 대퇴골두 해면골과 골다공증이 없는 대퇴골두 해면골의 특성을 평가하고자 미세단층촬영기 및 유한요소모델을 이용하여 해면골을 미세구조 특성과 기계적 특성을 비교 및 분석하였다. 골다공증이 있는 대퇴골두는 골밀도를 측정하여 T-score 값이 -2.5 이상인 시편 15개를 사용했으며, 골다공증이 없는 대퇴골두는 사망전 골다공증으로 인한 병력이 없었던 사체 4구에서 대퇴골두 5개를 획득하여 사용하였다. 대퇴골두 해면골의 미세구조 특성을 분석하기 위해서 미세단층촬영기를 이용하여 골다공증이 있는 대퇴골두와 골다공증이 없는 대퇴골두를 촬영하여 형태학적 지수(histomorphometry index)를 구했고, 미세단층촬영기에 의해서 획득된 미세영상(pixel size=21.31㎛)을 바탕으로 유한요소모델을 재건하여 기계적 특성을 구하였다. 형태학적지수는 골다공증이 없는 대퇴골두가 골다공증이 있는 대퇴골두에 비해서 골소주 두께, 골 체적, 골 체적비, 이방성 정도 그리고 골소주 개수가 증가했으며, 골소주 간격과 구조모델지수는 감소했다. 기계적특성 지수는 골다공증이 없는 대퇴골두 해면골 시편에서 반력, 표면응력 그리고 영률이 높게 분석되었다. 골다공증이 있는 대퇴골두 해면골 시편에서 미세구조 특성이 퇴화되며, 기계적 특성이 감소된다는 사실을 알수 있었다.

열처리 조건에 따른 무전해 Ni/전해 Cr 이중도금의 계면반응 및 균열성장거동 분석 (Effects of Heat Treatment Conditions on the Interfacial Reactions and Crack Propagation Behaviors in Electroless Ni/electroplated Cr Coatings)

  • 무전해 Ni/전해Cr이중도금 구조에서 무전해 Ni의 결정화 열처리 조건이 Cr도금의 균열성장 및 Ni/Cr계면반응에 미치는 영향을 분석하였다. 비정질 무전해 Ni/전해 Cr 도금 후 $750^{\circ}C$에서 6시간 동안 1회 열처리한 시편을1단계 열처리 조건으로 정했다. 또한, 무전해 Ni도금 후 동일 열처리를 통해 결정화 시킨 후, 전해 Cr도금 후 한번 더 동일조건 열처리한 경우를 2단계 열처리 조건으로 정하여 상호 비교하였다. 두 가지 열처리 조건 모두에서 공통적으로 Ni/Cr계면에서 상호확산에 의한 Ni-Cr고용체band layer가 관찰되었다. 1단계 열처리 조건의 경우 Cr도금에 관통균열이 발생하였으며, 2단계 열처리 조건의 경우 Cr도금에 표면 미소균열만 형성되고 관통균열은 거의 발생하지 않았다. 이는 무전해 Ni도금 직후 열처리에 의해 Ni-P비정질 구조에서 Ni, $Ni_3P$상으로 결정화되면서 급격한 체적 감소가 발생하여 Cr층의 잔류응력 완화에 영향을 끼쳐서, 상부 전해 Cr도금의 관통균열 형성에 영향을 미치는 것으로 판단된다.

소변 중 4-hydroxyproline 분석에 관한 연구 (Determination of Free 4-hydroxyproline with Dansylchloride by HPLC in Human Urine)

  • Objectives : The level of 4-hydroxyproline (4-Hyp) in human urine was measured using high performance liquid chromatography (HPLC) with a fluorescence detector. This method is useful for medical examinations and investigating the radicals induced by physical, chemical, mental stresses. This method is superior to many published several methods in terms of its low cost and ability to analyze many samples. Methods : The urine from workers in a tire manufacturing company (22 male pre- and post-shift workers) and 18 office-workers as controls were analyzed. Data concerning age, the cumulative drinking amount and the cumulative smoking amount was collected with a questionnaire. The optimum applied amount of dansyl-Cl, the optimum reaction temperature and time, the recoveries and the optimum pH of the eluent and buffer were determined.4-Hyp from human urine was derivatized with dansyl-Cl (dimethylamino-naphthalene-1-sulfonyl chloride) after removing the a-amino acid by a treatment with phthalic dicarboxaldehyde (OPA) and cleaned with Bond Elut C18 column. The 4-Hyp derivatives were separated on a reversed phase column by gradient elution with a phosphate buffer (5 mmol, pH 8.0) and acetonitrile, and detected by fluorescence measurements at 340 nm (excitation) and 538 nm (emission). Results : The detection limit for the urinary free 4-Hyp was $0.364{\mu}mol/l$. The recovery rate of 4-Hyp was 99.7%, and the effective pH of the phosphate buffer and borate buffer were 3.0 and 8.0, respectively. From statistical analysis, age, drinking and smoking did not affect the urinary free 4-Hyp in both the controls and workers. The range of urinary 4-Hyp in the controls, pre-shift, and post-shift workers were 0.33-16.44, N.D-49.06, and $0.32-56.27{\mu}mol/l$. From the pared-sample t-test, the urinary 4-Hyp levels in post-shift workers ($11.82{\pm}6.73\;nmmol/mg\;Cre$) were 2-fold higher than in pre-shift workers ($5.36{\pm}5.53\;nmol;/mg\;Cre$) and controls ($4.91{\pm}4.89\;nmol;/mg\;Cre$). Conclusions : This method was developed with high sensitivity, accuracy, and precision. The present method was effectively applied to analyze the urinary free 4-Hyp in both controls and workers.

꽃송이버섯 열수추출물이 HaCaT의 세포 연접 관련 유전자의 발현에 대한 영향 (Effect of a Hot Water Extract of Sparasis Crispa on the Expression of Tight Junction-Associated Genes in HaCaT Cells)

  • Purpose : Keratinocytes are the main cellular components involved in wound healing during re-epithelization and inflammation. Dysfunction of tight junction (TJ) adhesions is a major feature in the pathogenesis of various diseases. The purpose of this study was to identify the various effects of a Sparassis crispa water extract (SC) on HaCaT cells and to investigate whether these effects might be applicable to human skin. Methods : We investigated the effectiveness of SC on cell HaCaT viability using MTS. The antioxidant effect of SC was analyzed by comparing the effectiveness of ABTS to that of the well-known antioxidant resveratrol. Reverse-transcription quantitative polymerase chain reaction (qRT-PCR) is the most widely applied method Quantitative RT-PCR analysis has shown that SC in HaCaT cells affects mRNA expression of tight-junction genes associated with skin moisturization. In addition, Wound healing is one of the most complex processes in the human body. It involves the spatial and temporal synchronization of a variety of cell types with distinct roles in the phases of hemostasis, inflammation, growth, re-epithelialization, and remodeling. wound healing analysis demonstrated altered cell migration in SC-treated HaCaT cells. Results : MTS analysis in HaCaT cells was found to be more cytotoxic in SC at a concentration of 0.5 mg/㎖. Compared to 100 µM resveratrol, 4 mg/㎖ SC exhibited similar or superior antioxidant effects. SC treatment in HaCaT cells reduced levels of claudin 1, claudin 3, claudin 4, claudin 6, claudin 7, claudin 8, ZO-1, ZO-2, JAM-A, occludin, and Tricellulin mRNA expression by about 1.13 times. Wound healing analysis demonstrated altered cell migration in SC-treated HaCaT cells and HaCaT cell migration was also reduced to 73.2 % by SC treatment. Conclusion : SC, which acts as an antioxidant, reduces oxidative stress and prevents aging of the skin. Further research is needed to address the effects of SC on human skin given the observed alteration of mRNA expression of tight-junction genes and the decreased the cell migration of HaCaT cells.

MicroRNA-200a Targets Cannabinoid Receptor 1 and Serotonin Transporter to Increase Visceral Hyperalgesia in Diarrhea-predominant Irritable Bowel Syndrome Rats

  • Background/Aims MicroRNAs (miRNAs) were reported to be responsible for intestinal permeability in diarrhea-predominant irritable bowel syndrome (IBS-D) rats in our previous study. However, whether and how miRNAs regulate visceral hypersensitivity in IBS-D remains largely unknown. Methods We established the IBS-D rat model and evaluated it using the nociceptive visceral hypersensitivity test, myeloperoxidase activity assay, restraint stress-induced defecation, and electromyographic (EMG) activity. The distal colon was subjected to miRNA microarray analysis followed by isolation and culture of colonic epithelial cells (CECs). Bioinformatic analysis and further experiments, including dual luciferase assays, quantitative real-time polymerase chain reaction, western blot, and enzyme-linked immunosorbent assay, were used to detect the expression of miRNAs and how it regulates visceral hypersensitivity in IBS-D rats. Results The IBS-D rat model was successfully established. A total of 24 miRNAs were differentially expressed in the distal colon of IBS-D rats; 9 were upregulated and 15 were downregulated. Among them, the most significant upregulation was miR-200a, accompanied by downregulation of cannabinoid receptor 1 (CNR1) and serotonin transporter (SERT). MiR-200a mimic markedly inhibited the expression of CNR1/SERT. Bioinformatic analysis and luciferase assay confirmed that CNR1/SERT are direct targets of miR-200a. Rescue experiments that overexpressed CNR1/SERT significantly abolished the inhibitory effect of miR-200a on the IBS-D rats CECs. Conclusions This study suggests that miR-200a could induce visceral hyperalgesia by targeting the downregulation of CNR1 and SERT, aggravating or leading to the development and progression of IBS-D. MiR-200a may be a regulator of visceral hypersensitivity, which provides potential targets for the treatment of IBS-D.

회전킬른반응기를 이용한 리튬이온전지용 Si/C/CNF 음극활물질의 제조 및 전기화학적 특성 조사 (Preparation and Electrochemical Characterization of Si/C/CNF Anode Material for Lithium ion Battery Using Rotary Kiln Reactor)

  • 흑연은 리튬이온전지에 사용 되는 대표적인 음극활물질이다. 그러나 최대 이론 용량이 $372mA\;h\;g^{-1}$으로 제한되기 때문에 고용량의 리튬이온전지 개발을 위해서는 새로운 음극 소재 활물질이 필요하다. 실리콘의 최대 이론 용량은 $4200mA\;h\;g^{-1}$으로 흑연보다 높은 값을 나타내지만 부피 팽창이 400%로 크기 때문에 음극 소재 활물질로 바로 적용하기에는 적합하지 않다. 따라서 부피 팽창으로 인한 방전 용량의 감소를 최소화하기 위해 건식 방법으로 실리콘을 분쇄 하여 기계적 응력 및 반응상의 체적 변화를 감소시키고 입도 제어 된 실리콘 입자에 탄소를 코팅하여 체적의 변화를 억제하였다. 그리고 탄소 섬유를 입자 표면에 실타래처럼 성장시켜 2차적으로 부피 팽창을 제어하고 전기전도성을 개선하였다. 실험 변수에 따른 재료들의 물리화학적 특성을 XRD, SEM 및 TEM을 사용하여 측정하였고 전기화학적 특성을 평가 하였다. 본 연구에서는 실리콘의 수명 특성을 향상시켜 음극 소재 활물질로 사용 할 수 있는 합성 방법에 대하여 알아보았다.

신수(BL23) 택사약침이 Cisplatin으로 유발된 급성신부전 백서에 미치는 영향 (Effects of Alismatis Rhizoma Pharmacopuncture at BL23 on Cisplatin-Induced Acute Renal Failure in Rats)

  • Objectives : This study was designed to investigate the effects of Alismatis Rhizoma (AR) pharmacopuncture at BL23 on acute renal failure induced by cisplatin. Methods : The Sprague-Dawley rats were divided into Normal group (no injection of cisplatin and no treatment), Control group (cisplatin injection without treatment), Acu group (needling at BL23 after cisplatin injection), AR-PA1 group (treated with $0.3571mg/kg/20{\mu}l$ of AR pharmacopuncture at BL23 after cisplatin injection), and AR-PA2 group (treated with $1.7855mg/kg/20{\mu}l$ of AR pharmacopuncture at BL23 after cisplatin injection). Each treatment was given once daily for 8 days. Changes in body weight, kidney weight, tumor necrosis factor-alpha ($TNF-{\alpha}$), interleukin-6 (IL-6), Cu-Zn superoxide dismutase (Cu-Zn SOD), glutathione peroxidase (GPx), serum blood urea nitrogen (serum BUN), and serum creatinine were observed. Results : Body weight was significantly increased in AR-PA1 on $4^{th}$ and $6^{th}$ days and AR-PA2 on $2^{nd}$ day. $TNF-{\alpha}$ was significantly decreased in Acu, AR-PA1 and AR-PA2 groups. Cu-Zn SOD was significantly increased in AR-PA2 group. GPx was significantly increased in AR-PA1 and AR-PA2 groups. But kidney weight, IL-6, serum BUN and serum creatinine were not significantly changed in any groups compared to control group. Conclusions : In acute renal failure induced by cisplatin, AR pharmacopuncture has a mitigating effect on the inflammatory reaction related to the increase of $TNF-{\alpha}$ in the kidney tissue and a protective effect on the oxidative stress of the kidney tissue. However it is unlikely to restore the glomerular function or inhibit the renal swelling.

건식 분쇄 공정으로 Si 입도 제어를 통한 고용량 리튬이온전지 음극 소재의 개발 (Development of High Capacity Lithium Ion Battery Anode Material by Controlling Si Particle Size with Dry Milling Process)

  • 현재 리튬이온전지의 음극 소재 활물질로는 흑연이 주로 사용되고 있다. 그러나 흑연의 최대 이론 용량이 $372mA\;h\;g^{-1}$으로 제한되기 때문에 차세대 고용량 및 고에너지 밀도의 리튬이온전지 개발을 위해서는 새로운 음극 소재 활물질이 필요하다. 여러 음극 소재 활물질 중에서 Si의 최대 이론 용량은 $4200mA\;h\;g^{-1}$으로 흑연의 최대 이론 용량보다 약 10배 이상 높은 값을 나타내고 있지만 부피 팽창율이 거의 400%로 크기 때문에 사이클이 진행될수록 비가역 용량이 증가하여 충전 대비 방전 용량이 현저히 감소하는 현상을 나타내고 있다. 이러한 문제점을 해결하기 위한 방법으로 Si 음극 소재 활물질의 입자 크기를 조절하여 기계적 응력 및 반응상의 체적 변화를 감소시켜 사이클 특성을 다소 향상시킬 수 있다. 따라서 Si 입자의 부피 팽창율에 따른 충전 및 방전 용량의 감소를 최소화하기 위해 공정 시간 및 원가 절감이 우수한 건식 방법으로 Si을 분쇄하여 사이클 특성 향상에 관한 연구를 진행 하였다. 본 논문에서는 진동밀을 이용하여 Si을 나노 크기로 제어하고 실험 변수에 따른 재료들의 물리화학적 특성과 전기화학적 특성을 측정하였다.

부유식 해양구조물의 플로트오버 설치용 LMU 최적설계 (LMU Design Optimization for the Float-Over Installation of Floating Offshore Platforms)

  • LMU(Leg Mating Unit)는 해양구조물의 플로트오버 실치에서 활용되는 장비 중 하나로 충격을 흡수하는 부분과 결합부로 구성된다. 본 연구에서는 최적설계를 통해 부유식 해양구조물의 플로트오버 설치용 LMU의 성능을 개선하여 설계 요구 조건을 만족하는 설계를 개발하였다. 초기설계는 고정식 해양구조물의 플로트오버 설치용으로 개발된 것의 제원을 참조하였으며, 초탄성재료의 거동을 표현하기 위해 Mooney-Rivlin 모델을 활용하였다. 설계민감도해석 결과를 바탕으로 중요도에 따라 설계 변수들을 선별하였고, 진화알고리듬 기반 최적설계를 수행하였다. 최적설계 문제에서 목적함수는 LMU의 중량이며, 제약 조건은 LMU에 작용하는 최대 폰-미세스 응력과 LMU의 성능을 평가할 수 있는 반발력이다.

Gynostemma pentaphyllum extract and Gypenoside L enhance skeletal muscle differentiation and mitochondrial metabolism by activating the PGC-1α pathway in C2C12 myotubes

  • BACKGROUND/OBJECTIVES: Peroxisome proliferator-activated receptor-gamma co-activator-1α (PGC-1α) has a central role in regulating muscle differentiation and mitochondrial metabolism. PGC-1α stimulates muscle growth and muscle fiber remodeling, concomitantly regulating lactate and lipid metabolism and promoting oxidative metabolism. Gynostemma pentaphyllum (Thumb.) has been widely employed as a traditional herbal medicine and possesses antioxidant, anti-obesity, anti-inflammatory, hypolipemic, hypoglycemic, and anticancer properties. We investigated whether G. pentaphyllum extract (GPE) and its active compound, gypenoside L (GL), affect muscle differentiation and mitochondrial metabolism via activation of the PGC-1α pathway in murine C2C12 myoblast cells. MATERIALS/METHODS: C2C12 cells were treated with GPE and GL, and quantitative reverse transcription polymerase chain reaction and western blot were used to analyze the mRNA and protein expression levels. Myh1 was determined using immunocytochemistry. Mitochondrial reactive oxygen species generation was measured using the 2'7'-dichlorofluorescein diacetate assay. RESULTS: GPE and GL promoted the differentiation of myoblasts into myotubes and elevated mRNA and protein expression levels of Myh1 (type IIx). GPE and GL also significantly increased the mRNA expression levels of the PGC-1α gene (Ppargc1a), lactate metabolism-regulatory genes (Esrra and Mct1), adipocyte-browning gene fibronectin type III domain-containing 5 gene (Fndc5), glycogen synthase gene (Gys), and lipid metabolism gene carnitine palmitoyltransferase 1b gene (Cpt1b). Moreover, GPE and GL induced the phosphorylation of AMP-activated protein kinase, p38, sirtuin1, and deacetylated PGC-1α. We also observed that treatment with GPE and GL significantly stimulated the expression of genes associated with the anti-oxidative stress response, such as Ucp2, Ucp3, Nrf2, and Sod2. CONCLUSIONS: The results indicated that GPE and GL enhance exercise performance by promoting myotube differentiation and mitochondrial metabolism through the upregulation of PGC-1α in C2C12 skeletal muscle.
