The effect of a titanium socket with a zirconia abutment on screw loosening after thermocycling in an internally connected implant: a preliminary study (내부연결 임플란트용 타이타늄 소켓을 이용한 지르코니아 지대주에서 열순환이 나사풀림에 미치는 영향: 예비연구)

  • Purpose: The aim of this study was to investigate the effects of a titanium component for the zirconia abutment in the internal connection implant system on screw loosening under thermocycling conditions. Materials and Methods: Internal connection titanium abutments and external connection zirconia abutments with titanium sockets were connected respectively to screw-shaped internal connection type titanium implants with 30 Ncm tightening. These implant-screw-abutment assemblies were divided into two groups of five specimens each; titanium abutments as control and zirconia abutments with titanium sockets as experimental group. The specimens were subjected to 2,000 thermocycles in water baths at $5^{\circ}C$ and $55^{\circ}C$, with 60 seconds of immersion at each temperature. The removal torque values (RTVs) of the abutment screws of the specimen were measured before and after thermocycling. RTVs pre- and post-thermocycling were investigated in statistics. Results: There was not screw loosening identified by tactile and visual inspection in any of the specimens during or after thermocycling. The mean RTV difference for the control group and the experimental group were $-1.34{\pm}2.53Ncm$ and $-1.26{\pm}2.06Ncm$, respectively. Statistical analysis using an independent t-test revealed that no significant differences were found in the mean RTV difference of the groups (P > 0.05). Conclusion: Within the limitations of this in vitro study, the titanium socket for the zirconia abutment did not show a significant effect on screw loosening under thermal stress compared to the titanium abutment in the internal connection implant.

Comparison of the outcomes of three-dimensional finite element analysis under arbitrary and realistic occlusal loading conditions in mandibular posterior region (하악 구치부에서 임의로 부여된 교합과 실제 교합의 삼차원 유한요소해석결과의 비교)

  • Purpose: The purpose of this study was to compare the biomechanical outcome in the mandibular posterior region between two different loading conditions by finite element analysis. Materials and Methods: The mandibular posterior teeth model and the implant model were generated for the study. And 2 different types of loading conditions were provided: Arbitrary occlusion and natural occlusion obtained from the digital occlusal analyzer, Accura (Accura, Dmetec Co. Ltd., Seoul, Korea). Total load of 100 N was evenly distributed over arbitrary occlusion points, and 100 N load was differentially distributed over natural occlusion points according to Accura data. The biomechanical outcome was evaluated by the finite element analysis software. Results: The result of finite element analysis showed considerable difference in both von Mises stress pattern and displacement under different loading conditions. Conclusion: In finite element analysis, it is recommended to simulate a realistic occlusal loading pattern that is based on accurate measurement.

Resistance Function of Woody Landscape Plants to Air Pollutants(II) - POD Activity - (조경수목(造景樹木)의 대기오염물질(大氣汚染物質)에 대한 방어기능(防禦機能)(II) - POD 활성(活性)을 중심으로 -)

  • This study was conducted to determine the toxic effects of air pollutants on landscaping tress, Pinus densiflora, Pinus koraiensis, Ginkgo biloba, Liriodendron tulipifera, Platanus occidentalis and their resistance to the pollutant toxicity in urban and industrial regions of Seoul and Taejon, Korea. Total sulfur contents and enzyme activities such as superoxide dismutase and peroxidase were analyzed in tree foliage of Pinus densiflora, Pinus koraiensis, Ginkgo biloba, Liriodendron tulipifera, Platanus occidentalis. In addition, POD activity was analyzed in the foliage on tree seedlings, i.e. Pinus densiflora, Pinus koraiensis, Ginkgo biloba, Lirioderdron tulipifera, with the fumigation of $SO_2$ in gas chamber 4 hours a day for six days. In Ginkgo biloba total sulfur content and POD activity had a negative correlation while other species had a positive relationship in total sulfur content and enzyme activity. Air pollutants accumulated in tree tissues were supposed to enhance the enzyme activity like POD providing the resistance mechanisms. Especially Pinus koraiensis and Platanus occidentalis had higher POD activity than other species. The increase of temporary POD activity against environmental stress appeared in sensitive trees and prolonged increase of POD activity played an important role in resistance mechanism. SOD and POD activities in all species had a positive correlation except Ginkgo biloba. Changes of SOD and POD activities were different between species and in most species SOD as well as POD seemed to participate in resistance mechanism.

Analysis of Nutrition and Antioxidants of Yak-Kong Chungkukjang Powder Added Black Foods (블랙푸드가 첨가된 약콩청국장분말의 영양소 및 생리활성물질 분석)

  • This study is carried out to investigate the analysis of nutrition and antioxidants of soybean Chungkukjang powder (SCP), Yak-Kong Chungkukjang powder (YCP) and Yak-Kong Chungkukjang powder added black foods (YCBP) for the management of Alzheimer's disease. The water content of YCBP was higher than that of SCP and YCP. The soluble and crude protein contents of YCP were the highest among three Chungkukjang powders. In SCP, YCP and YCBP, the ratios of unsaturated and saturated fatty acids were 5.17, 5.76, and 5.78, respectively. The mineral content of SCP was higher than that of YCP and YCBP. Antioxidants analysis showed that the content of diadzein was the highest in YCP and genistein, and anthocyanin contents were higher than the others in YCBP. In conclusion, YCP and YCBP had higher contents than SCP in soluble and crude proteins, and unsaturated fatty acids, which are needed for composition and function of the brain tissue. Also, it was found that the contents of diadzein, genistein and anthocyanin with outstanding antioxidative ability were high. Thus, this study suggests that Chungkukjang powder, which is made with Yak-Kong, black sesame, black rice and sea tangle, can be utilized for in vivo experiment for the control of oxidative stress, reported as cause and therapy for Alzheimer's Disease.

A Study on the Optimal Design of Reinforced Concrete Frames Using SUMT (SUMT 법(法)을 이용(利用)한 철근(鐵筋)콘크리트 뼈대구조물(構造物)의 최적설계(最適設計)에 관한 연구(研究))

  • This study is conserned with the optimization of reinforced concrete frames using limit state design theory. Formulations of the optimal design for reinforced concrete frame based on the limit state theory turn out to be the nonlinear programming problems which have to deal with the required steel area, the width and effective height of the beam and column section and the moment reduction factor as the design variables. The objective function is formulated as the total construction cost which considers the costs of steel, concrete and forming for the reinforced concrete frames, and the basic constraints are imposed upon both ultimate and serviciability limit state concepts. Also, the stress blocks assumpted in CP110 and Hognestad et al. theory are applied to analysis an ultimate resistant section force for the ultimate limit state and only the criteria of CP110 are used for serviciability limit state. The optimized technique which is applied to solve the nonlinear programming problems for the optimization of reinforced concrete frames is SUMT utilizing the modified Newton-Raphson method. This algorithm is used to test for the two reinforced concrete frames, and then is compared and analysized with the numerical results of reference(10) to examine its convergence, applicability and stability under the same conditions. The results of this study are discussed about the economy comparision of the optimal values for CP110 and Hognestad et al., and the applicability, stability, convergence and validity of this algorithm used herein through the numerical analyses.

Freshness Monitoring of Raw Salmon Filet Using a Colorimetric Sensor that is Sensitive to Volatile Nitrogen Compounds (휘발성 질소화합물 감응형 색변환 센서를 활용한 연어 신선도 모니터링)

  • A colorimetric paper sensor was used to detect volatile nitrogen-containing compounds emitted from spoiled salmon filets to determine their freshness. The sensing mechanism was based on acid-base reactions between acidic pH-indicating dyes and basic volatile ammonia and amines. A sensing layer was simply fabricated by drop-casting a dye solution of bromocresol green (BCG) on a polyvinylidene fluoride substrate, and its color-change response was enhanced by optimizing the amounts of additive chemicals, such as polyethylene glycol, p-toluene sulfonic acid, and graphene oxide in the dye solution. To avoid the adverse effects of water vapor, both faces of the sensing layer were enclosed by using a polyethylene terephthalate film and a gas-permeable microporous polytetrafluoroethylene sheet, respectively. When exposed to basic gas analytes, the paper-like sensor distinctly exhibited a color change from initially yellow, then to green, and finally to blue due to the deprotonation of BCG via the Brønsted acid-base reaction. The use of ammonia analyte as a test gas confirmed that the sensing performance of the optimized sensor was reversible and excellent (detection time of < 15 min, sensitive naked-eye detection at 0.25 ppm, good selectivity to common volatile organic gases, and good stability against thermal stress). Finally, the coloration intensity of the sensor was quantified as a function of the storage time of the salmon filet at 28℃ to evaluate its usefulness in monitoring of the food freshness with the measurement of the total viable count (TVC) of microorganisms in the food. The TVC value increased from 3.2 × 105 to 3.1 × 109 cfu/g in 28 h and then became stable, whereas the sensor response abruptly changed in the first 8 h and slightly increased thereafter. This result suggests that the colorimetric response could be used as an indicator for evaluating the degree of decay of salmon induced by microorganisms.

Ataxin-2 Dysregulation Triggers a Compensatory Fragile X Mental Retardation Protein Decrease in Drosophila C4da Neurons

  • Dendrites require precise and timely delivery of protein substrates to distal areas to ensure the correct morphology and function of neurons. Many of these protein substrates are supplied in the form of ribonucleoprotein (RNP) complex consisting of RNA-binding proteins (RBPs) and mRNAs, which are subsequently translated in distal dendritic areas. It remains elusive, however, whether key RBPs supply mRNA according to local demands individually or in a coordinated manner. In this study, we investigated how Drosophila sensory neurons respond to the dysregulation of a disease-associated RBP, Ataxin-2 (ATX2), which leads to dendritic defects. We found that ATX2 plays a crucial role in spacing dendritic branches for the optimal dendritic receptive fields in Drosophila class IV dendritic arborization (C4da) neurons, where both expression level and subcellular location of ATX2 contribute significantly to this effect. We showed that translational upregulation through the expression of eukaryotic translation initiation factor 4E (eIF4E) further enhanced the ATX2-induced dendritic phenotypes. Additionally, we found that the expression level of another disease-associated RBP, fragile X mental retardation protein (FMRP), decreased in both cell bodies and dendrites when neurons were faced with aberrant upregulation of ATX2. Finally, we revealed that the PAM2 motif of ATX2, which mediates its interaction with poly(A)-binding protein (PABP), is potentially necessary for the decrease of FMRP in certain neuronal stress conditions. Collectively, our data suggest that dysregulation of RBPs triggers a compensatory regulation of other functionally-overlapping RBPs to minimize RBP dysregulation-associated aberrations that hinder neuronal homeostasis in dendrites.

Changes of Ganglioside GM3 Expression in Porcine Oocyte Maturation and Early Embryonic Development In Vitro

  • Gangliosides exist in glycosphingolipid-enriched domains on the cell membrane and regulate various functions such as adhesion, differentiation, and receptor signaling. Ganglioside GM3 by ST3GAL5 enzyme provides an essential function in the biosynthesis of more complex ganglio-series gangliosides. However, the role of gangliosides GM3 in porcine oocytes during in vitro maturation and early embryo development stage has not yet understood clear. Therefore, we examined ganglioside GM3 expression patterns under apoptosis stress during maturation and preimplantation development of porcine oocytes and embryos. First, porcine oocytes cultured in the NCSU-23 medium for 44 h after $H_2O_2$ treated groups (0.01, 0.1, 1 mM). After completion of meiotic maturation, the proportion MII (44 h) was significantly different among control and the H2O2 treated groups ($76.8{\pm}0.3$ vs $69.1{\pm}0.4$; 0.01 mM, $55.7{\pm}1.0$; 0.1 mM, $38.2{\pm}1.6%$; 1 mM, P<0.05). The expressions of ST3GAL5 in $H_2O_2$ treated groups were gradually decreased compared with control group. Next, changes of ST3GAL5 expression patterns were detected by using immunofluorescene (IF) staining during preimplantation development until blastocyst. As a result, we confirmed that the expressions of ST3GAL5 in cleaving embryos were gradually decreased (P<0.05) according to the early embryo development progress. Based on these results, we suggest that the ganglioside GM3 was used to the marker as pro-apoptotic factor in porcine oocyte of maturation and early embryo production in vitro, respectively. Furthermore, our findings will be helpful for better understanding the basic mechanism of gangliosides GM3 regulating in oocyte maturation and early embryonic development of porcine in vitro.

Study of Sophorae Radix on $H_2O_2$-mediated Apoptosis and Total Protein Expression Pattern in Vascular Smooth Muscle Cells (고삼이 $H_2O_2$에 의한 대동맥 평활근세포 고사 및 전체 단백질 발현에 미치는 영향)

  • Apoptosis of vascular smooth muscle cells(VSMCs) is essential in atherogenesis, being a factor that modulates its early progression rather than a terminal event in the course of the disease. Various stimuli, including oxide lipoproteins, altered hemodynamic stress and free radical, can induced VSMCs apoptosis in vitro. The protective effects of Sophorae Radix (SR) on apoptotic cell death induced by H₂O₂ were investigated in VSMCs. The viability of VSMCs was markedly decreased by H₂O₂. Sophorae Radix protected the H202-induced apoptotic death of VSMCs, which was characterized as nuclear fragmentation and increase of sub-G0/G1 fraction .. Sophorae Radix decreased the activation of caspase-3 like protease induced by H₂O₂ and recovered control level from H202-induced PARP, Bak, Bcl-XL and mitochondrial membrane potential. These results suggest that Sophorae Radix protected VSMCs apoptotic death induced by H₂O₂ via inactivation of caspase-3 and modulation of mitochondrial function. Also, the expression profile of proteins by using two-dimensional (2-D) gel electrophoresis was screened. Future investigations will need to explore the use of an anti atherosclerotic therapy of Sophorae Radix, which relies on inhibition of the proapoptotic activation of the vascular smooth muscle cells.

THE CHANGE OF EXPRESSION OF INTERLEUKIN-6 AND -8 AFTER THE APPLICATION OF THE STATIC COMPRESSIVE PRESSURE ON THE FIBROBLAST ORIGINATED FROM THE PERIODONTAL LIGAMENTS (치주인대 기원의 섬유아 세포에 압축력을 가한 경우 Interleukin-6 및 Interleukin-8의 발현 변화에 관한 연구)

  • The fibroblast in the periodontal ligaments received various stress. Among them, compression and tension are quite important and they are related to the remodeling of tooth and alveolar bone. We studied the change of expression of interleukin-6 (IL-6) and interleukin-8 (IL-8) in the fibroblasts of the periodontal ligaments by real-time RT-PCR and ELISA. In results, the relative activity of IL-6 mRNA in 2 hours after was 1.54${\pm}$0.08 and 1.00${\pm}$0.05 in control and test, respectively (P<0.05). Its 12 hours after was 1.23${\pm}$0.06 and 2.78${\pm}$0.14 in control and test, respectively (P<0.05). The relative activity of IL-8 mRNA in 2 hours after was 1.00${\pm}$0.05 and 0.24${\pm}$0.01 in control and test, respectively (P<0.05). Its 12 hours after was 1.23${\pm}$0.06 and 0.63${\pm}$0.03 in control and test, respectively (P<0.05). The concentration of IL-6 was 1.02${\pm}$0.16 ng/ml, 0.90${\pm}$0.14 ng/ml, and 1.32${\pm}$0.12 ng/ml (P<0.05) in control, 2, and 12 hours after, respectively. The concentration of IL-8 was 2.26${\pm}$0.17 ng/ml, 1.70${\pm}$0.26 ng/ml (P<0.05), and 0.84${\pm}$0.47 ng/ml (P<0.05) in control, 2, and 12 hours after, respectively. In conclusion, the expression of IL-6 was significantly increased after the application of the static compressive force, but IL-8 was significantly decreased. Considering their known function, their expression is quite important in tooth and bone resorption.
